The Interrelationship of the Genetic Risks for Different Forms of Substance Use Disorder in a Swedish National Sample: A Top-Down Genetic Analysis.

OBJECTIVE: The purpose of this study was to determine, using a top-down genetic analysis, the degree of specificity of the genetic risk factors for individual forms of substance use disorders (SUD). METHOD: We examined and followed to December 31, 2018, all individuals born in Sweden between 1960 and 1990 (N = 2,772,752) diagnosed with any of the following six SUDs: alcohol use disorder (AUD), drug use disorder (DUD), or one of four specific forms of DUD involving cannabis (CUD), cocaine and other stimulants (CSUD), opioids (OUD), or sedatives (SeUD). We examined population subsamples at high

Genetic and phenotypic correlations between surrogate measures of insulin release obtained from OGTT data.

AIMS/HYPOTHESIS: We examined the extent to which surrogate measures of insulin release have shared genetic causes. METHODS: Genetic and phenotypic correlations were calculated in a family cohort (n = 315) in which beta cell indices were estimated based on fasting and oral glucose-stimulated plasma glucose, serum C-peptide and serum insulin levels. Furthermore, we genotyped a large population-based cohort (n = 6,269) for common genetic variants known to associate with type 2 diabetes, fasting plasma glucose levels or fasting serum insulin levels to examine their association with various indice

Population-specific optimization of polygenic risk scores for breast cancer in East Asians.

PURPOSE: Polygenic risk scores (PRS) have shown promise in predicting disease susceptibility, but their accuracy varies across populations due to the limited representation of non-European ancestries in genomic studies. While European-derived PRS demonstrate some transferability to Asian populations, optimizing these scores for specific populations remains crucial for improving risk prediction. METHODS: Using data from the Taiwan Precision Medicine Initiative (n = 46,422) and an external validation cohort (n = 891), we developed an approach to optimize European-derived PRS for breast cancer p

Phylodynamic reconstruction of global evolution and geographic structure of maize chlorotic mottle virus.

Maize chlorotic mottle virus (MCMV) represents a major quarantine pathogen that poses a serious threat to global maize production, yet its spatiotemporal evolutionary dynamics remain incompletely characterized. In this study, we reconstructed the global molecular epidemiology of MCMV by analysing 117 complete genome and 214 coat protein (CP) gene sequences. We employed whole-genome data for Bayesian phylodynamic inference while utilizing CP sequences for phylogenetic reconstruction and population genetic analyses. Our phylodynamic analyses estimated a mean evolutionary rate of 2.07×10-4 substi

Detection of relatedness in chemical alarm cues by a selfing vertebrate depends on population and the life stage producing the alarm cue.

Aquatic prey have impressive abilities to extract information from a variety of chemical cues. For example, they can use the alarm cues released by wounded individuals during a predator attack to learn about predation risk, and they can also distinguish kin from non-kin individuals during interactions. However, it remains unclear whether animals can combine this information on predation risk with kin recognition of the particular individuals under threat. To examine how the relatedness of the individuals in alarm cue affects behaviour we used the self-fertilizing hermaphroditic mangrove rivulu

Application of a new method for GWAS in a related case/control sample with known pedigree structure: identification of new loci for nephrolithiasis.

In contrast to large GWA studies based on thousands of individuals and large meta-analyses combining GWAS results, we analyzed a small case/control sample for uric acid nephrolithiasis. Our cohort of closely related individuals is derived from a small, genetically isolated village in Sardinia, with well-characterized genealogical data linking the extant population up to the 16(th) century. It is expected that the number of risk alleles involved in complex disorders is smaller in isolated founder populations than in more diverse populations, and the power to detect association with complex trai

Experimental evidence that source genetic variation drives pathogen emergence.

A pathogen can readily mutate to infect new host types, but this does not guarantee successful establishment in the new habitat. What factors, then, dictate emergence success? One possibility is that the pathogen population cannot sustain itself on the new host type (i.e. host is a sink), but migration from a source population allows adaptive sustainability and eventual emergence by delivering beneficial mutations sampled from the source's standing genetic variation. This idea is relevant regardless of whether the sink host is truly novel (host shift) or whether the sink is an existing or rela

Do anthropogenic transports facilitate stored-product pest moth dispersal? A molecular approach.

Stored-product moths cause large economic damage in food processing industries and storage facilities. Control of indoor pests is currently dealt with locally, and control strategies seldom include different mills or cooperative industries in joint efforts to reduce infestations. In colder climates where conditions hinder flight dispersal of stored-product moths, we hypothesize that human transport between mills will facilitate dispersal. Albeit considered intuitive, this hypothesis has so far never been tested. Male moths from three mills (populations) in southern Sweden and Denmark were coll

Widespread movement of invasive cattle fever ticks (Rhipicephalus microplus) in southern Texas leads to shared local infestations on cattle and deer.

BACKGROUND: Rhipicephalus (Boophilus) microplus is a highly-invasive tick that transmits the cattle parasites (Babesia bovis and B. bigemina) that cause cattle fever. R. microplus and Babesia are endemic in Mexico and ticks persist in the United States inside a narrow tick eradication quarantine area (TEQA) along the Rio Grande. This containment area is threatened by unregulated movements of illegal cattle and wildlife like white-tailed deer (WTD; Odocoileus virginianus). METHODS: Using 11 microsatellite loci we genotyped 1,247 R. microplus from 63 Texas collections, including outbreak infest

Regional replication of association with refractive error on 15q14 and 15q25 in the Age-Related Eye Disease Study cohort.

PURPOSE: Refractive error is a complex trait with multiple genetic and environmental risk factors, and is the most common cause of preventable blindness worldwide. The common nature of the trait suggests the presence of many genetic factors that individually may have modest effects. To achieve an adequate sample size to detect these common variants, large, international collaborations have formed. These consortia typically use meta-analysis to combine multiple studies from many different populations. This approach is robust to differences between populations; however, it does not compensate fo

Eating disorders genetics in Asia.

A complete understanding of the genetic architecture of eating disorders requires adequately large sample sizes from individuals of all ancestries. Failure to include representative samples truncates understanding and may even exacerbate health disparities. Several countries in Asia have made rich contributions in psychiatric genetics; however, the eating disorders field requires concerted global efforts to increase representation from Asian ancestry populations to ensure that our global efforts accurately reflect the true distribution of eating disorders around the world and across ancestries

The Role of Human Leukocyte Antigen in Celiac Disease Diagnostics.

Celiac disease is an autoimmune disease affecting the small intestine, triggered by gluten sensitization in genetically susceptible individuals worldwide. Celiac disease development is strongly linked to the presence of HLA-DQ2 and/or DQ8, which present the immunogenic gluten peptides and trigger the immune response leading to pathogenesis. Because of the variability of clinical symptoms, the disease is often underdiagnosed. Intestinal biopsy and the presence of antibodies to deamidated gliadin and tissue transglutaminase are recommended diagnostic tools. Genetic testing for HLA DQ2 and DQ8 ca

Genetic Aspects of Myoclonus-Dystonia Syndrome (MDS).

Myoclonus-dystonia (M-D) is an autosomal-dominant movement disorder with onset in the first two decades of life. Mutations in the epsilon-sarcoglycan gene (SGCE, DYT11) on chromosome 7q21-q31 represent the major genetic cause of M-D in some populations. The syndrome was related with mutations in two other genes (DRD2 and DYT1). A second locus has been reported in one large M-D family (DYT15, 18p11), but no gene has been identified yet. In this review, we discuss genetic aspects of myoclonus-dystonia.

Linkage of a commoner form of recessive amyotrophic lateral sclerosis to chromosome 15q15-q22 markers.

Autosomal recessive familial amyotrophic lateral sclerosis (RFALS) is a rare form of ALS that usually presents at an early age with slow progression of symptoms. RFALS is clinically and genetically heterogeneous and the locus of RFALS type 3 was mapped to 2q33 (ALS2) in a single family. We now report linkage of a more-common form of RFALS to chromosome 15q15-q22 markers (ALS5) and show further genetic locus heterogeneity in RFALS. ALS5 is the locus for most families with RFALS and appears to be present in both North African and European populations.

[Biochemical bases for the antiseizure role of the pineal gland].

Metabolism of serotonin was studied in pineal gland of Wistar rats with low and high rate of audio excitability as well as in the Krushinsky-Molodkina rat population genetically predisposed to audio-dependent convulsions. Intensive turnover of serotonin via N-acetyl serotonin into melatonin was found in pineal gland of rats insensitive to sensory stimulus, whereas the rate of this response was decreased in the gland tissue of animals sensitive to audio effects as well as inhibition of serotonin turnover into melatonin was noticed in pineal gland of rats predisposed genetically to audio-depende

The genetics of thyroiditis as a prototype of human autoimmune disease.

The predisposition to autoimmune disease has been termed an autoimmune diathesis. The genetic basis for this predisposition has been analyzed, using three different models: juvenile thyroiditis in humans, spontaneous thyroiditis in chickens, and experimental thyroiditis in mice. At least three different categories of genetic factors contribute to the development of autoimmunity. They include an increased immune response due to immunoregulatory genes associated with the major histocompatibility complex, the balance of thymus-derived lymphocyte population with helper/inducer vs suppressor proper

Meta-analysis of 990 EBV genomes identifies EBER2 SNP7048 and its interaction with HLA-A*11:01 driving nasopharyngeal carcinoma risk.

Nasopharyngeal carcinoma (NPC) is a major health burden in East and Southeast Asia (SEA). While EBV infection is a primary driver, the specific viral strains responsible for NPC development and their interaction with host genetics across geographically diverse endemic regions remain poorly understood. We performed a whole-genome sequencing of 388 EBV isolates and matched host whole-exome sequencing in a Singaporean cohort. To define the global significance of these findings, we conducted a meta-GWAS and phylogenetic analysis across three independent cohorts (Singapore, Hong Kong, and Guangdong

Anatomical Characteristics of Tongue-Innervating Mechanoreceptors.

Mechanosensory innervation of the oral cavity enables us to detect the texture and location of the foods we consume. Our goal was to find a genetic identifier for a mechanoreceptive subtype. Toward this goal, we examined neurons expressing the neurotrophin receptor TrkC (TrkC-tdTomato), parvalbumin (Pvalb), or glutamate transporter, Vglut3. We found that the majority of Pvalb-lineage, Vglut3-lineage, and TrkC-tdTomato neurons innervating the tongue are separate trigeminal neuron populations. Because different papilla types contribute to food detection in different ways, we examined the innerva

Transcriptomics and resequencing reveal dual evolutionary strategies for salt tolerance in foxtail millet (Setaria italica (L.)).

INTRODUCTION: Soil salinity poses a major constraint to cereal production, yet the genetic basis underlying salt tolerance in foxtail millet (Setaria italica (L.)) remains largely elusive. METHODS: To dissect these tolerance mechanisms, we integrated phenotyping, physiological assays, transcriptomics, and population genomics across 18 extreme salt-tolerant panel (STP) and 18 salt-sensitive panel (SSP) accessions selected from over 500 germplasm resources, with evaluations conducted under NaCl treatment. RESULTS: Under salt stress, STP accessions exhibited better growth and higher chlorophyll

Vitamin D Receptor (VDR) Polymorphisms and Cardiometabolic Profiles in Orthopedic Patients: A Cluster-Based Analysis.

Genetic polymorphisms contribute to inter-individual variability in cardiometabolic risk and quality-of-life outcomes, yet their clinical relevance often remains unclear due to population heterogeneity and reliance on single-variant analyses. Integrative approaches combining genetic and phenotypic data may improve the characterization of complex disease profiles, particularly in orthopedic populations burdened by cardiometabolic comorbidities. This study included 289 patients scheduled for orthopedic surgery. Polymorphisms in the vitamin D receptor (VDR; ApaI, FokI, BsmI), catechol-O-methyltra
