Unveiling the mechanism of action and regulation of the steroidogenic acute regulatory protein.

Stimulation of steroid-producing cells of the gonads and adrenals with trophic hormone (LH, and ACTH, respectively) produces a marked increase in steroid hormone synthesis within minutes. The rate-limiting step in this acute steroidogenic response is the transport of cholesterol from the outer to the inner mitochondrial membrane, where the first committed step in steroid synthesis is performed by the side-chain cleavage enzyme system (P450scc), resulting in the production of pregnenolone. This process of cholesterol translocation is blocked by inhibitors of protein synthesis (i.e. cycloheximid

Roles of estrogen receptor-alpha gene expression in reproduction-related behaviors in female mice.

The role of gene expression of the estrogen receptor-alpha form (ER alpha) in the regulation of female reproductive behavior was investigated in estrogen receptor knockout (ERKO) mice, deficient specifically for the ER alpha, but not the ER beta, gene. Estrogen- or estrogen- plus progesterone-treated gonadectomized ERKO mice did not show any lordosis response. Detailed behavioral analysis revealed that ERKO females were also deficient in sexual behavioral interactions preceding the lordosis response. They were extremely rejective toward attempted mounts by stud male mice, which could not show

Prolactinomas resistant to standard dopamine agonists respond to chronic cabergoline treatment.

Cabergoline (CAB), a new, potent, and long-lasting PRL-lowering agent, was shown to be effective in tumoral hyperprolactinemia. The aim of this study was to investigate the effectiveness of CAB in patients with prolactinoma proven to be resistant to bromocriptine (BRC) and quinagolide (CV 205-502). Twenty-seven patients (19 macro- and 8 microprolactinomas) were treated with CAB at a weekly dose of 0.5-3 mg for 3-22 months. All patients were previously shown to be resistant to BRC, and 20 of them were resistant to CV 205-502 as well. Basal serum PRL levels before CAB treatment ranged from 108-3

Hypothalamic involvement in sexuality and hostility: comparative psychological aspects.

Evidence presented in this article shows the representation of sexual and aggressive behaviors at the level of the hypothalamus to be more prominent than in all other brain areas involved. Indeed, there are good arguments to attribute a central position to the hypothalamus within larger structural systems encompassing the limbic system, where aspects of the behaviors involved can be influenced. So far, however, the arguments are purely descriptive and factual and do not contribute much to answering questions about hypothalamic function: the grounds for and consequences of this massive represen

Annual reproductive rhythms in mammals: mechanisms of light synchronization.

Animals restrict the time of birth of offspring to the most advantageous time of year, usually spring or summer. This is achieved by controlling the preceding period of fertility and, in some cases, by delaying implantation of the zygote. Seasonal changes in daylength, the principal, though not the only cue, regulate pulsatile release of hypothalamic releasing factors that in turn activates the pituitary-gonadal axis. The role of the neuroendocrine system is therefore to translate the photoperiodic stimulus into an endocrine signal (Figure 12). The measurement of day length is a function of th

Distinguishing between hidden testes and anorchia: the role of endocrine evaluation in infancy and childhood.

OBJECTIVE: Non-palpable testes remain a diagnostic challenge, often involving exploratory laparoscopy. We evaluated the diagnostic value of a wide range of reproductive hormones in order to distinguish between bilateral cryptorchidism and bilateral anorchia. DESIGN: In this retrospective study, we identified and included 36 boys with non-palpable testes (20 with cryptorchidism, 3 with congenital hypogonadotropic hypogonadism (CHH), and 13 with anorchia) at first examination during childhood. METHODS: Information on karyotype, phenotype, surgical results from laparoscopy, and biochemistry was

Exploring the Causal Relationship Between Sex Hormones and Depression by Means of Two-Sample Mendelian Randomization Analysis.

BACKGROUND: To investigate the potential causal association between sex hormones and depression, a twosample mendelian randomization (MR) analysis was conducted. METHODS: Summary statistics from Genome-wide Association Study (GWAS) on sex hormones and depression were collected. Sex-specific instruments were used to analyze seven sex hormones, including progesterone (PROG) and bioavailable testosterone (BAT). The inverse variance weighted (IVW) method was employed as the primary analysis, and sensitivity analyses were conducted to assess the robustness of the findings. RESULTS: The IVW analys

Sex-specific mechanisms of chronic pain.

For decades, we have known that women, especially younger women, are disproportionately affected by pain disorders, including irritable bowel syndrome, endometriosis, menstrual migraines, fibromyalgia, osteoarthritis, and neuropathic pain, all of which degrade quality of life. The adoption of sex as a biological variable in 2016 has begun to transform the field, revealing distinct sex-specific pathways that initiate, amplify, and resolve pain. In this Review, we synthesize recent advances across visceral and somatic pain, highlighting the impact of sex hormones, immune-neural cross-talk, the g

Sex-based hormonal influences on asthma biomarkers.

Asthma, a chronic inflammatory airway disease affecting 25 million people in the United States, exhibits distinct sex differences in prevalence, phenotype, and treatment response. While asthma is more common in boys during childhood, adult women experience greater disease severity, likely due to hormonal and immunologic influences. Asthma biomarkers, including blood eosinophil count, fractional exhaled nitric oxide, IgE, and emerging markers, provide insight into disease phenotypes and therapeutic response. This review synthesizes current evidence on sex-specific differences in asthma biomarke

Examining the potential decoupling of reproductive-stress interactions in blackbirds with stark differences in reproductive strategy.

Stress and reproduction are often linked through interactions between the hypothalamic-pituitary-adrenal (HPA) and hypothalamic-pituitary-gonadal (HPG) axes such that stress can suppress reproduction. It is unclear, however, whether species with enhanced annual fecundity exhibit modifications in this interaction. We addressed this question using a comparative study of two closely related blackbird species that differ markedly in parental care and reproductive output: the brown-headed cowbird (Molothrus ater), an obligate brood parasite that has enhanced annual fecundity without parental care,

Advances in understanding the impact of sex on kidney health and disease.

Biological differences exist between males and females in kidney structure and function, leading to sex heterogeneity in the presentation and outcomes of chronic kidney disease (CKD) and response to novel therapeutics. However, treatment guidelines ignore sex-specific differences. This Series paper provides an integrated discussion of the complexity of sex differences in the context of kidney health and disease, from biology through clinical characteristics, treatment, evidence generation, and reporting. Throughout, we consider the effect of genetics, epigenetic regulation, and the role of sex

Reversible congenital hypogonadotropic hypogonadism: keys for clinical management.

Congenitalhypogonadotropic hypogonadism (CHH) is characterized by absent/incomplete puberty and a treatable form of male infertility resulting from deficient gonadotropin-releasing hormone (GnRH) secretion/action. Traditionally, CHH has been considered a permanent, lifelong condition. However, evidence indicates a subset men undergo reversal and recover reproductive axis function. We conducted a structured literature search (Medline, PubMed) using keywords to retrieve articles on CHH reversal (1975-2025). We synthesize the literature to provide a high-level overview of CHH and the reversal phe

Precocious puberty in boys: current insights into etiology, genetic advances, and environmental factors.

BACKGROUND: Central precocious puberty (CPP) in males results from the premature activation of the hypothalamic-pituitary-gonadal (HPG) axis, clinically diagnosed by a testicular volume >4 mL before the age of 9 years. Over the past two decades, a clear secular trend toward earlier pubertal onset has been reported. Historically, CPP in boys was strongly associated with intracranial lesions, observed in 40-50% of cases. RECENT FINDINGS: Over the last decade, converging evidence has indicated a markedly lower prevalence of brain lesions in males with CPP, approximately 6-8%. Identified risk fac

The influence of the estrous cycle in preclinical models that mimic key features of post-traumatic stress disorder: A systematic review.

Similar to other mood and anxiety conditions, post-traumatic stress disorder (PTSD) appears to be more prevalent and have a more protracted course in females. In the clinic, PTSD symptoms have been shown to worsen during phases of the menstrual cycle characterized by low estrogen and progesterone levels. Similarly, low estrogen levels have been associated with the development of fear and anxiety-like behaviour in preclinical models. Despite the commonly held belief that sex hormones influence fear and anxiety, the literature contains conflicting findings and remains controversial. To better un

Oxidative stress and apoptotic pathways mediate BDE-47-induced reproductive dysfunction and offspring skeletogenesis disruption in zebrafish.

2, 2', 4, 4'-tetrabromodiphenyl ether (BDE-47) has been recognized as a potential hazard to the reproductive systems of organisms. However, the reproductive toxicity and transgenerational effects of BDE-47 in female remain insufficiently characterized. In this study, we systematically investigated the reproductive performance, sex hormone levels, ovarian morphology, expression of genes associated with the hypothalamic-pituitary-gonadal (HPG) axis, and ovaries apoptotic responses following a 21-day exposure to BDE-47 in female zebrafish. Furthermore, we examined the intergenerational impacts of

Photoperiodic schedules during rearing and laying stages modulate photoreceptors transcription and the hypothalamic-pituitary-testicular axis in male White King pigeons.

The photoperiod during rearing and laying period is a primary environmental regulator of reproductive performance in poultry, yet its effects on male breeding pigeons have been largely unexplored. Therefore, this study evaluated the impact of four photoperiod regimes on the reproductive performance, testicular morphology, hormone profiles, and gene expression of hypothalamic-pituitary-testicular axis in male White King pigeons. From 101 d of age, the pigeons in the control group were exposed to a natural photoperiod until 160 d, and then to a photoperiod of 16 h (16 light (L): 8 dark (D)) and
