Microdissection combined with the polymerase chain reaction to identify potentiating viral co-infection in patients with HIV/AIDS with ocular infection.

BACKGROUND: In the presence of several coexisting infections, superimposed tissue necrosis or tissue metaplasia, it may be difficult to recognize standard histologic morphology on hematoxylin-eosin slides. Tissue microdissection combined with the polymerase chain reaction (PCR-MD) offers the advantages of high specificity and relative speed. The objective of this study was to describe the use of PCR-MD in identifying potentiating viral co-infection in patients with HIV/AIDS with retinitis and choroiditis. METHODS: Eyes from two patients with HIV/AIDS with several ocular infections were studie

[Factors for the onset of and the exacerbation of tuberculosis. 5. The infection and prognosis of tuberculosis among patients with immunodeficiency, especially HIV-infected patients].

The number of people infected with human immunodeficiency virus (HIV) is gradually increasing in Japan, and the morbidity rate from tuberculosis in the Japanese people is high. Accordingly, the number of cases with both infections is considered to increase in the future. Our hospital has already encountered 22 cases of HIV associated tuberculosis. HIV infects mainly CD4-positive cells. The extreme decrease in the cell count results in serious cellular immunological disorder. CD4-positive cell disorder induces disorders of B lymphocytes, cytotoxic T cells, natural killer cells, and macrophage f

Liquid Chromatography-Mass Spectrometry Metabolomic Analysis of Terminalia ferdinandiana Exell. Fruit Extracts That Inhibit HIV-1 Cell Infection, HIV-1 Reverse Transcriptase and HIV-1 Protease.

The emergence of HIV strains resistant to the current anti-retroviral drugs has necessitated the search for new anti-retroviral medications. Methanolic and aqueous T. ferdinandiana fruit extracts have potent inhibitory activity against several phases of the HIV-1 replicative cycle. Cell infectivity studies using a non-resistant HIV-1 pseudovirus demonstrated that the methanolic (IC50 16 µg/mL) and aqueous extracts (IC50 19 µg/mL) were potent inhibitors of viral infection in a non-replicating HIV-1 assay. Both extracts also inhibited HIV-1 reverse transcriptase (IC50 values of 35 and 33 µg/mL f

The Effects of Human Immunodeficiency Virus Type 1 (HIV-1) Antigen-Expanded Specific T-Cell Therapy and Vorinostat on Persistent HIV-1 Infection in People With HIV on Antiretroviral Therapy.

BACKGROUND: The histone deacetylase inhibitor vorinostat (VOR) can reverse human immunodeficiency virus type 1 (HIV-1) latency in vivo and allow T cells to clear infected cells in vitro. HIV-specific T cells (HXTCs) can be expanded ex vivo and have been safely administered to people with HIV (PWH) on antiretroviral therapy. METHODS: Six PWH received infusions of 2 × 107 HXTCs/m² with VOR 400 mg, and 3 PWH received infusions of 10 × 107 HXTCs/m² with VOR. The frequency of persistent HIV by multiple assays including quantitative viral outgrowth assay (QVOA) of resting CD4+ T cells was measured

Alarming incidence of hepatitis C virus re-infection after treatment of sexually acquired acute hepatitis C virus infection in HIV-infected MSM.

BACKGROUND: Recent data indicate that seroprevalence of sexually transmitted hepatitis C virus (HCV) infection among MSM is stabilizing in Amsterdam. However, little is known about the incidence of HCV re-infection in MSM who have cleared their HCV infection. We, therefore, studied the incidence of re-infection in HIV-infected MSM who were HCV RNA-negative following HCV treatment of acute primary infection. METHODS: Our study population comprised HIV-infected MSM at two large HIV outpatient clinics in Amsterdam, who were previously diagnosed with a sexually transmitted acute HCV infection and

The heart rate variability changes in measles, HIV, and SARS-CoV-2 infections; a subtle autonomic involvement - a pilot study.

INTRODUCTION: Inflammation may induce dysautonomia, which is revealed by the decrease in heart rate variability (HRV) parameters. Our pilot study compares changes in HRV and the association between HRV and inflammatory markers in three RNA viral infections: acute (SARS-CoV-2, measles) and chronic (HIV). METHODS: We evaluated 25 patients with viral infections and 8 control patients without viral disease, with similar mean age and comorbidities. Patients with measles and COVID-19 were hospitalized for mild to moderate respiratory symptoms, while those with HIV were assessed during follow-up vis

Undiagnosed burden of latent tuberculosis, active tuberculosis and tuberculosis-HIV co-infections in Africa-status quo, needs, priorities, and opportunities.

Tuberculosis (TB) remains a leading cause of morbidity and mortality in Africa, with latent TB infection (LTBI) and TB co-infections posing significant public health challenges. This viewpoint examines the undiagnosed burden of LTBI and TB co-infections across the continent, emphasizing findings from autopsy studies. Data from the World Health Organization's Global Tuberculosis Report 2024 and recent autopsy research highlight the prevalence of undetected TB cases, particularly among individuals with HIV. Autopsy studies reveal a substantial proportion of TB cases that were undiagnosed ante mo

Sexually transmitted infections and bacterial vaginosis and preterm birth in pregnant people living with HIV: A population-based cohort study.

BackgroundWhile individual sexually transmitted infections are linked with preterm births, their synergistic impact among pregnant people living with HIV (PLWH) remain unclear. We aimed to identify the impact of antenatal sexually transmitted infections and bacterial vaginosis on preterm birth in PLWH.MethodsWe completed a population-based cohort study using the British Columbia Perinatal HIV Surveillance Database, capturing all births in PLWH from January 1997 to December 2022. Univariate risk factors for preterm birth were identified using chi-squared tests, Fisher's exact tests and t-tests,

Intervention to Prevent Recurrent Intestinal Parasitic Infections in People Living with HIV in Selected Parts of Eastern Cape, South Africa.

Interactions between parasites and hosts are not fully understood, though the dynamic pattern of infection and reinfection in humans varies with different demographic variables and behavioral changes. A community-based non-equivalent control group post-test-only design, an aspect of quasi-experimental design (QED), was carried out between March 2019 and February 2020. For the extraction of data from respondents, structural questionnaires were filled. Their CD4 count and viral load from the database of the National Health Laboratory Services, Mthatha were recorded. The method applied for the id

Integrated management of cryptococcal meningitis and concurrent opportunistic infections to improve outcomes in advanced HIV disease: a randomised strategy trial.

BACKGROUND: Mortality associated with HIV-associated cryptococcal meningitis remains high even in the context of clinical trials (24-45% at 10 weeks); mortality at 12-months is up to 78% in resource limited settings. Co-prevalent tuberculosis (TB) is common and preventable, and likely contributes to poor patient outcomes. Innovative strategies to increase TB preventative therapy (TPT) provision and uptake within this high-risk group are needed. PROTOCOL: The IMPROVE trial (Integrated management of cryptococcal meningitis and concurrent opportunistic infections to improve outcomes in advanced

Persistence and risk factors of occult hepatitis B virus infections among antiretroviral therapy-naïve people living with HIV in Botswana.

AIM: This study aimed to determine the kinetics of occult hepatitis B virus infections (OBI) among people with HIV (PWH). METHODS: The study used archived plasma samples from longitudinal HIV natural history studies. We identified new OBI cases and assessed risk factors for OBI using Cox proportional hazards regression analysis. RESULTS: At baseline, 8 of 382 [(2.1%) (95% CI: 1.06-4.1)] samples tested positive for hepatitis B surface antigen (HBsAg+). Of the 374 HBsAg-negative samples, 76 had sufficient sample volume for HBV DNA screening. OBI positivity (OBI+) at baseline was reported in 11

Management of Antiretroviral Therapy and Opportunistic Infections in People Living with HIV Undergoing Hematopoietic Stem Cell Transplant in British Columbia.

BACKGROUND: A growing number of people living with HIV (PLWH) are developing an indication for hematopoietic stem cell transplantation (HSCT). While overlapping immunosuppression and medication interactions make this a complicated situation, the risk is mitigable, and PLWH should have similar access to HSCT as the general population. There are currently no guidelines available for the management of HSCT in PLWH, and through this document we hope to provide initial guidance. METHODS: We performed a non-systematic review of published English-language literature regarding medication and opportun

Modelling and analysis of the HIV/AIDS epidemic with fast and slow asymptomatic infections in China from 2008 to 2021.

The aim of this paper is to investigate the spread of the HIV/AIDS epidemic in China during 2008-2021. A new mathematical model is proposed to study the dynamics of HIV transmission with acute infection, fast asymptomatic infections, and slow asymptomatic infections. The basic reproduction number is obtained by the next-generation matrix method. A quantitative analysis of the model, including the local behavior, global behavior, and permanence, is performed. Numerical simulations are presented to enhance the results of these analyses. The behavior or the model's parameters are estimated from r
