Induction of a secondary human anti-HLA alloimmune response in severe combined immunodeficient mice engrafted with human lymphocytes.

BACKGROUND: Experimental manipulation of transfusion-induced alloimmunization is limited in humans by ethical considerations. Conversely, studies of alloimmunization in animal models may not reflect the human immune system closely enough to be of optimal benefit. The development of an in vivo model of human alloimmunization that is amenable to experimental manipulation is thus desirable. STUDY DESIGN AND METHODS: An in vivo model of human alloimmunization was evaluated by using mice with severe combined immunodeficiency (SCID). SCID mice underwent gamma-radiation (200 cGy) and received an int

The influence of petrochemicals and stress on the immune system of seabirds.

There is increasing attention directed to the role of environmental pollutants in altering immune function. Only with the identification of the responsible environmental toxicants, and an understanding of their mechanisms of action, can we hope to treat immunotoxic injuries. This situation is exemplified by the exposure of wild birds to oil spills, the subsequent potential for direct toxicity from the oil, and the secondary toxicity of stress-induced immune modulation. Immunosuppressive mechanisms related to oil ingestion and handling stress are implicated in the morbidity and mortality of sea

Biotin-avidin immobilization of platelet glycoproteins (BAIPG): a new capture assay for the detection of anti-platelet antibodies.

Several 'capture' assays are currently employed to identify specific platelet antibodies, but all require the use of murine monoclonal antibodies (MoAbs) against the antigen of interest. We have developed a new antigen capture assay for the detection of platelet reactive antibodies, based on platelet surface sialoglycoprotein labelling with biotin hydrazide, and a following immobilization of the biotinylated platelet proteins to microtiter wells that had been coated with streptavidin. The resulting solid phase can then be used in a simple ELISA to detect serum and platelet associated antibodie

Oral desensitization in the treatment of human immune diseases.

Oral desensitization or oral tolerance is induced by giving antigenic peptides by the mucosal route. In man only the oral route has been used up to now. Experiments in animal models of human autoimmune diseases, have shown that it is not necessary to use the primary antigen responsible for disease induction. Antigens implicated in secondary immune phenomenon can act similarly by means of the so-called "bystander suppression". Thus for diseases such as multiple sclerosis (MS) and rheumatoid arthritis (RA) candidate antigens for desensitization are available. Many patients with MS have immunity

Rabbit model of disseminated syphilis: immunoblot and immunohistologic evidence for a role of specific immune complexes in lesion pathogenesis.

Circulating immune complexes (CIC) containing Treponema pallidum proteins have been preliminarily implicated as inducers of a neutrophilic vascular reaction in early human cutaneous lesions of secondary syphilis. To clarify the role of specific CIC in producing cutaneous and renal lesions, 12 rabbits were studied at the following intervals after induction of disseminated syphilis: 20 days (4 rabbits: biopsies of normal and lesional skin for direct immunofluorescence (IMF) for (IgG, IgM, IgA, Clq, C3, C4), fibrin, and T. pallidum proteins; routine histology; and immunoblots of serum for CIC con

Glymphatic system dysfunction in stroke: the relationship between clearance failure and neuroinflammation.

Stroke triggers a complex secondary injury cascade, in which neuroinflammation plays a central role. The glymphatic system is a perivascular clearance network that facilitates cerebrospinal fluid (CSF) and interstitial fluid (ISF) exchange and removes metabolic waste from the brain. Recent studies suggest that glymphatic dysfunction is not simply a consequence of stroke, but also contributes to disease progression. Impaired glymphatic clearance promotes the accumulation of damage-associated molecular patterns, inflammatory mediators, and toxic metabolites within the brain parenchyma, sustainin

Gut Microbiota-Targeted Nutrition for Healthy Aging: Mechanistic Roles of Polyphenols and Dietary Fiber in Geroscience.

Age-related alterations in the gut microbiota contribute to chronic low-grade inflammation, immune dysregulation, metabolic dysfunction, frailty, sarcopenia, and cognitive decline. Dietary polyphenols and fermentable fiber modulate microbial composition and metabolism, promoting the production of bioactive metabolites, including short-chain fatty acids, secondary bile acids, indole derivatives, and urolithins, which regulate intestinal barrier integrity, immune homeostasis, mitochondrial function, and gut-organ communication. This narrative review critically synthesizes evidence from experimen

Cellular senescence and inflammageing: from mechanisms to senotherapeutic interventions.

Cellular senescence is a context-dependent cellular state characterised by persistent cell-cycle arrest, epigenetic remodelling, metabolic reprogramming and acquisition of a senescence-associated secretory phenotype. Transient senescence contributes to embryogenesis, tissue repair and tumour suppression, whereas persistent senescent cell populations accumulate with advancing age across multiple tissues, in part owing to declining immune-mediated clearance and intrinsic resistance to apoptosis, thereby promoting chronic systemic inflammation, tissue fibrosis, stem-cell dysfunction and propagati

Multi-omics atlas of the bovine coronavirus-infected calf jejunum: reduction of Phocaeicola coprophilus and deoxycholic acid linked to Th17/Treg imbalance.

Optimal calf rearing is fundamental for ensuring efficient milk production. Bovine coronavirus (BCoV) poses a significant threat to calf health and leads to substantial economic losses in the dairy industry. However, the mechanisms by which the intestinal mucosal microbiome regulates the host immune response during infection remain unclear. In this study, we constructed a high-resolution map of the jejunal mucosal microenvironment in BCoV-infected calves. Our findings revealed that BCoV infection led to severe microbial dysbiosis, characterized by a marked reduction in Phocaeicola coprophilus

Case Report: PD-1/CTLA-4 dual checkpoint blockade(QL1706) in advanced pulmonary squamous cell carcinoma complicated by multidrug-resistant tuberculosis after multiple lines of immunotherapy.

Immune checkpoint inhibitors (ICIs) are standard care for advanced non-small cell lung cancer (NSCLC), yet patients with active tuberculosis (TB) are consistently excluded from clinical trials. Furthermore, data guiding the use of dual PD-1/CTLA-4 blockade in the presence of multidrug-resistant TB (MDR-TB) is lacking. We report a 56-year-old man with stage IVA pulmonary squamous cell carcinoma (SqNSCLC) who progressed on multiple lines of therapy, including chemotherapy plus pembrolizumab, docetaxel, and the PD-1/VEGF bispecific antibody ivonescimab. During treatment, he developed secondary MD

Acetylcholinesterase Inhibition Reverses Age-Related Pulmonary Decline and Increases Bronchus-Associated Lymphoid Tissue Formation in Aged Mice.

Age-related chronic, low-grade inflammation, known as inflammaging, contributes to tissue damage and disease. In the lungs, inflammaging leads to abnormal tissue remodeling, reduced function, and decreased immunity. A key factor in inflammaging is declining acetylcholine signaling, which normally suppresses inflammation and promotes tissue repair. We tested whether increasing acetylcholine responsiveness could reverse age-related lung damage. Aged mice were treated with donepezil to increase acetylcholine availability. After six months, blood oxygen saturation and voluntary activity were signi

Failed resolution of inflammation in intracerebral haemorrhage.

Intracerebral haemorrhage (ICH) is a subtype of stroke, with resulting long-term disability determined by secondary brain injury. The prevailing biphasic model of neuroinflammation, which conceptualizes a temporal transition from acute to chronic stages, fails to reconcile persistent clinical trial failures or to explain the mechanisms underpinning the progression to chronic neurological deficits. We propose that the core pathology is the failure of acute inflammation to resolve in a timely manner owing to a functional collapse of physiological pro-resolution programmes, governed by dysregulat

The complement system in the pathophysiology and treatment of depression: From synaptic pruning to pharmacological targeting.

Major depressive disorder remains unsatisfactorily addressed by monoaminergic antidepressants, and the search for mechanistically novel targets has converged on the immune system. Among these, the complement system offers a defined molecular route through which locally synthesized complement proteins may tag synapses for microglial engulfment. Preclinical studies implicate the C1q-C3-CR3 pruning axis and the anaphylatoxin receptors C3aR and C5aR in linking immune activation to region-dependent synaptic and neuroinflammatory changes, but causal evidence in patients remains absent. This review s

Designing a novel multiepitope vaccine candidate against Treponema pallidum via adhesins using reverse vaccinology.

Syphilis, caused by Treponema pallidum (T. pallidum), represents a significant worldwide public health threat. This spiral-shaped, Gram-negative pathogen is a strict human-specific obligate parasite primarily transmitted through sexual contact. This pathogen induces a multistage and multisystem progressive disease, against which no effective prophylactic vaccine currently exists. This study focuses on syphilis prevention and control by employing a reverse vaccinology approach to investigate the immunogenic properties of T. pallidum adhesin proteins. Fifteen T-cell epitopes and seven B-cell epi

EBV and Concomitant Acute Motor and Sensory Axonal Neuropathy in a Healthy 15-Year-Old Female.

Acute motor and sensory axonal neuropathy (AMSAN) is a rare and severe form of acute axonal injury caused by immune damage to the axonal membrane. AMSAN is an axonal variant of GBS. GBS occurs from immune injury to the myelin sheath, axonal variants of GBS (AMSAN and AMAN) differ in that insult is to the axonal membrane. AMSAN is seldom seen, especially in pediatric and adolescent patients. Unlike acute motor axonal neuropathy (AMAN), which has been well-described in literature to be secondary to Campylobacter jejuni infection, there is no known etiology of AMSAN. Here, we present a case of an

Genomic and Transcriptomic Profiling of High-risk Bladder Cancer Reveals Diverse Molecular and Microenvironment Ecosystems.

BACKGROUND AND OBJECTIVE: High-risk bladder cancer recurs in 30% of cases and causes fatal outcomes in 10% within 2 yr despite surgical resection, endoscopic surveillance, and bacillus Calmette-Guérin (BCG) immunotherapy. The global shortage of BCG highlights the urgent need for alternative or complementary strategies. This study aimed to identify molecular subtypes and develop a precision framework to predict recurrence risk and guide treatment. METHODS: Transcriptomic profiling and targeted genomic sequencing were performed with validation by single-cell RNA sequencing and spatial transcrip

Expression profiles of NOD1 and NOD2 and pathological changes in gills during Flavobacterium columnare infection in yellow catfish, Tachysurus fulvidraco.

NOD-like receptors are significant contributors to the immune response of fish against different types of pathogen invasion. NOD1 and NOD2 genes of yellow catfish (Tachysurus fulvidraco) were identified and characterized in this study. Yellow catfish NOD1 and NOD2 have open reading frames (ORFs) of 2841 and 2949 bp, encoding 946 and 982 amino acids, respectively. Both NOD1 and NOD2 are intracellular proteins lacking transmembrane regions and signal peptides. Sequence homology analysis revealed that the protein sequences of NOD1 and NOD2 of yellow catfish are highly similar to those of channel

Relapsing Chronic Immune Thrombocytopenia and Thrombosis: A Case Report.

Immune thrombocytopenia (ITP) involves an acquired, immune-mediated disorder involving autoantibodies, which target platelet antigens. It may be primary or secondary to an underlying condition. While some may be asymptomatic, others may experience easy bruising, petechiae, purpura, epistaxis, bleeding from the gums, menorrhagia, hematuria, melena, or hematochezia. This is a case of a 21-year-old female who presented to the Emergency Room after experiencing several days of intermittent bruising and menorrhagia. She was found to have a platelet count of 16,000/μL and hemoglobin of 9.7 g/dL with

Systemic sclerosis: pathogenic mechanisms and their implications for treatment.

Systemic sclerosis (SSc) is a rare systemic autoimmune disease characterized by a triad of pathogenic mechanisms, including: a) microvascular hyperreactivity secondary to endothelial dysfunction, b) dysregulated immune activation of both innate and adaptive immunity, with the production of autoantibodies targeting nuclear antigens (e.g., anticentromere antibodies, anti-RNA polymerase III antibodies, and anti-topoisomerase I antibodies), and c) fibrosis of the skin and internal organs, such as the lungs, due to excessive extracellular matrix deposits produced by dysregulated myofibroblasts. Expansion of myeloid suppressor cells and suppression of Lassa virus-specific T cells during fatal Lassa fever.

Lassa fever is a highly lethal hemorrhagic fever endemic to West Africa. In the absence of efficient prophylactic or therapeutic countermeasures, it poses a substantial threat to public health in this region. The pathophysiological mechanisms underlying the severity of the disease are poorly known because Lassa virus (LASV), its causative agent, has to be handled in BSL-4 laboratories and access to clinical samples is difficult. The control of Lassa fever is associated with a rapid and well-balanced immune response and viral clearance. However, severe disease is characterized by uncontrolled i
