Alteration of transporter activities in the epididymides of infertile initial segment-specific Pten knockout mice.

A fully functional initial segment, the most proximal region of the epididymis, is important for male fertility. Our previous study generated a mouse model to investigate the importance of initial segment function in male fertility. In that model, phosphatase and tensin homolog (Pten) was conditionally removed from the initial segment epithelium, which resulted in epithelial de-differentiation. When spermatozoa progressed through the de-differentiated epithelial duct, they developed angled flagella, suggesting compromised sperm maturation, which eventually resulted in male infertility. To unde

NF-YB Regulates Spermatogonial Stem Cell Self-Renewal and Proliferation in the Planarian Schmidtea mediterranea.

Gametes are the source and carrier of genetic information, essential for the propagation of all sexually reproducing organisms. Male gametes are derived from a progenitor stem cell population called spermatogonial stem cells (SSCs). SSCs give rise to male gametes through the coordination of two essential processes: self-renewal to produce more SSCs, and differentiation to produce mature sperm. Disruption of this equilibrium can lead to excessive proliferation of SSCs, causing tumorigenesis, or can result in aberrant differentiation, leading to infertility. Little is known about how SSCs achiev

Improving litter size by boar spermatozoa: application of combined H33258/CTC staining in field trial with artificial insemination.

Conventional semen analysis offers basic information on infertility; however, its clinical value in predicting fertility status is unclear. To establish an accurate diagnosis of male fertility, semen analysis under capacitation condition is necessary because only capacitated spermatozoa are capable of fertilizing oocytes. The objective of this study was to verify male fertility based on conventional semen analysis before and after capacitation, including the assessment of motility (%), motion kinematics, and capacitation status of spermatozoa. A computer-assisted sperm analysis system and chlo

Basal testosterone concentrations after the application of a slow-release GnRH agonist implant are associated with a loss of response to buserelin, a short-term GnRH agonist, in the tom cat.

Slow-release GnRH agonist implants are considered an effective, reversible alternative to surgical castration in male tom cats. Individual differences exist regarding the onset of efficacy and might be delayed in some animals. Single measurements of testosterone (T) might result in basal concentrations also in intact male cats. Consequently, GnRH stimulation tests are performed to measure T increase in intact animals and to differentiate castrated from intact male cats. In this study, five tom cats were treated with a 4.7-mg deslorelin implant and GnRH stimulation tests using buserelin were pe

Successful thrombolysis of normotensive pulmonary embolism with life-threatening hypoxia in a young man with Klinefelter syndrome.

Klinefelter syndrome (KS) affects males born with an additional X chromosome giving the genotype 47XXY classically. This syndrome has primary features of infertility and hypogonadism along with other features including a genetically hypercoagulable state. When associated with other risk factors, KS further increases the risk of venous thromboembolism and could result in life-threatening pulmonary embolism (PE). There should be a lower threshold in suspecting PE as a cause of acute respiratory failure in this patient group and thrombolysis should be considered early in normotensive PE with seve

Generation of an iPSC cohort of isogenic iPSC lines (46-XY and 47-XXY) from a non-mosaic Klinefelter Syndrome patient (47-XXY) (KAUSTi008-A, KAUSTi008-B, KAUSTi008-C, KAUSTi008-D, KAUSTi008-E, KAUSTi008-F, KAUSTi008-G).

Klinefelter Syndrome (KS) is the most common X chromosome aneuploidy in males characterized by highly heterogeneous clinical manifestations including a subtle cognitive impairment and multisystemic disorders such as infertility, metabolic syndrome, gynecomastia and cardiovascular diseases. To date dosage-dependent correlation studies of X-linked genes and low- and high-grade KS clinical phenotypes have not been performed. Here we generated multiple isogenic 47-XXY and 46-XY iPSC lines from one 47-XXY patient. Leveraging on a fully matched genetic background, our cohort represents a highly info

[A family with blepharophimosis, ptosis, epicanthus inversus and telecanthus. Occurrence of the hereditary marker in five generations].

The condition with blepharophimosis, ptosis, epicanthus inversus and telecanthus is reported in one family over five generations. The syndrome is transmitted as an autosomal-dominant characteristic with a very high penetrance and expressivity, preferentially affecting and being transmitted by males. Affected females were infertile. There were no other accompanying systemic disorders. A three-year-old boy, who develops deprivation amblyopia, was first treated by levator attachment according to Friedenwald's method. Medical canthal surgery followed two years later to correct the epicanthus and t

Analysis of sexual dysfunction development among male and female living kidney donors.

INTRODUCTION: Living kidney donations (LKDs) face a persistent demand for patients with end-stage renal disease, emphasizing the importance of LKDs' growth and success. Although living kidney donors generally exhibit excellent survival rates, little research has explored the development of long-term sexual dysfunction following LKD. OBJECTIVES: This study aimed to analyze differences in 5-year sexual dysfunction outcomes between male and female living kidney donors, utilizing the TriNetX database, a federated network of electronic medical records from multiple U.S. healthcare organizations.

Large-scale discovery of male reproductive tract-specific genes through analysis of RNA-seq datasets.

BACKGROUND: The development of a safe, effective, reversible, non-hormonal contraceptive method for men has been an ongoing effort for the past few decades. However, despite significant progress on elucidating the function of key proteins involved in reproduction, understanding male reproductive physiology is limited by incomplete information on the genes expressed in reproductive tissues, and no contraceptive targets have so far reached clinical trials. To advance product development, further identification of novel reproductive tract-specific genes leading to potentially druggable protein ta

Zinner Syndrome: The Diagnosis and Management of a Rare Urogenital Malformation.

This case highlights an atypical but important consideration in young males presenting with persistent gastrointestinal and/or genitourinary symptoms. Zinner syndrome (ZS) develops from embryologic maldevelopment of the distal mesonephric duct, resulting in ejaculatory duct atresia with consequent obstruction of the seminal vesicle and concomitant ureteral bud malformation, leading to renal agenesis/dysplasia. The lack of distinct clinical symptoms makes ZS a difficult diagnosis to reach: Abdominal pain and dysuria are often mistaken for prostatitis or cystitis. However, the use of modern imag

Characterization of Motor Activity of Spermatozoa in Adult Progeny of Female Rats with Experimental Type 1 Diabetes Mellitus.

We studied the influence of maternal experimental streptozotocin-induced type 1 diabetes mellitus on motility of spermatozoa in adult progeny. A decrease in the summary count of spermatozoa and changes in their subpopulation composition were revealed. Specifically, the count of nonfertile spermatozoa (immotile and jerking cells) increased, while fertile fraction (progressively motile and low motile cells) decreased. These shifts were responsible for lower index of spermatozoon motility (proportion of fertile to infertile sums of spermatozoa) in the progeny of females with experimental type 1 d

[What are the consequences of smoking on pregnancy and delivery].

According to a WHO report of 1997, more than one-third of the world population of more than 15 years old smokes. Since the 90s, smoking progressed amongst women and teenagers. The noxious roles of smoking addiction to smoking on male and female fertility, pregnancy and delivery are often little known or underestimated by the public and most healthcare professionals. In France, smoking addiction provokes 60,000 deaths every year and is one of the first avoidable perinatal causes of mortality and morbidity. The alarming extension of the epidemic of the feminine addiction to smoking has increased

Insulin-like 3 signalling in testicular descent.

Undescended testis is one of the most common congenital defects in the newborn boys and the common cause of cryptorchidism. If left untreated, this condition is strongly associated with infertility and drastically increased risk of testicular cancer in adulthood. Testis position in developing males is defined by sexual dimorphic differentiation of two gonadal ligaments, gubernaculum and cranial suspensory ligament. Recent transgenic mouse studies identified testicular hormone insulin-like 3 (INSL3), and its receptor, GREAT/LGR8, as the critical regulators of the gubernacular differentiation. M

[Luliberin and lutropin--their significance].

Human lutropin (hLH) is a heterodimeric, glycoprotein hormone released from pituitary by decapeptide luliberin (LH-RH). The both hormones plays an important role in axis hypothalamus-pituitary-gonads responsible for procreation. Many isoforms of hLH have been found and the occurrence of several genetic variants of the hormone in infertile female patients were described. Also genetic mutants of human lutropin choriogonadotropin receptor (LH/CG-R) in male young patients with precocious puberty were proved. Until now several thousands of LH-RH analogs were synthesized and some of them were applie

The effect of excess body fat on female and male reproduction.

The dramatic increase in the prevalence of obesity coincides with a decline in reproductive health indices in both sexes. Energy excess mediates changes to the regulatory mechanisms of the reproductive system. Obese individuals exhibit increased estrogen concentrations, due to the overexpression of aromatase in the adipose tissue; via a negative feedback loop, men present with symptoms of hypogonadotropic hypogonadism. These hormonal changes, along with increased oxidative stress, lipotoxicity and disturbances in the concentrations of adipokines, directly affect the gonads, peripheral reproduc

Cell-mediated immunity in infertility.

A role of cell-mediated immunity in immunologic infertility is indicated by a number of animal immunization studies and by reported clinical associations between cellular immune responses and infertility. Furthermore, recent technical advances in the field of cellular immunology have enabled advanced studies on the cellular and soluble mediators of cell-mediated immune responses and their interactions with reproductive processes. In this article we review some of the recent technical and conceptual advances in cellular immunology, the classic literature on cellular immune responses in infertil

[Sterility of immunologic origin].

Antispermatozoal antibodies in the woman, the man, or both, may be the cause of nonorganic infertility in a small number of couples. The antibodies are most often found in the serum or in the genital fluids. Diagnosis is based on the postcoital test and on various laboratory tests to assess sperm-cervical mucous compatibility and to detect antispermatozoal antibodies. In women the strongest relation between results of laboratory tests and infertility is between the presence of serum antispermatozoal antibodies and infertility of more than 3 years' duration. In men there is a relation between h

KIF2C is essential for meiosis and manchette dynamics in male mice.

In gametogenesis, microtubules undergo dramatic changes known as microtubule dynamics, and which is important for fertility both male and female. In spermatogenesis, spindle microtubule dynamics occur during meiosis and manchette microtubule dynamics occur in elongated spermatids. In oogenesis, spindle microtubule dynamics occur during meiosis. The microtubule depolymerization protein kinesin-13 family (KIF2A, KIF2B, and KIF2C) plays an important role in microtubule dynamics, and KIF2C is a well-known microtubule depolymerization factor in mitosis. Although the function of KIF2C in mitosis has
