Single-cell profiling uncovers kaempferol-mediated immunoregulation in the protection against experimental autoimmune hepatitis.

BACKGROUND: Autoimmune hepatitis (AIH) is triggered by an aberrant immune response directed against hepatocyte autoantigens, ultimately leading to liver inflammation and hepatocellular damage. Kaempferol (KAE), a natural flavonoid, exhibits potent anti-inflammatory and hepatoprotective properties. However, the precise immunoregulatory mechanisms of KAE in alleviating AIH are still undefined. OBJECTIVES: This study utilized comprehensive single-cell analysis to investigate the therapeutic efficacy and underlying immunoregulatory mechanisms of KAE in alleviating ConA-induced experimental AIH.

Intersection Between Systemic Autoimmune Diseases, Primary Immunodeficiency and Cancer: a Field in its Infancy.

PURPOSE: Systemic autoimmune diseases and primary immunodeficiencies/inborn errors of immunity (PIDs/IEIs) have traditionally been viewed as opposing immunological entities. However, growing genetic, immunological, and clinical evidence challenges this dichotomy, revealing a shared spectrum of immune dysregulation, genetic vulnerability, and convergent phenotypes. Advances in next-generation sequencing have identified monogenic IEIs presenting with autoimmune manifestations indistinguishable from polygenic systemic autoimmune diseases, blurring diagnostic boundaries and carrying therapeutic im

Immunological profiling and treatment patterns of primary Sjögren's disease patients with concomitant rheumatoid arthritis: a retrospective cohort study.

BACKGROUND: The coexistence of primary Sjögren's disease (pSS) and rheumatoid arthritis (RA) is not uncommon, but its immunological characteristics and treatment patterns remain poorly defined. This study aimed to compare the immune profiles and therapeutic strategies between pSS patients with and without RA overlap. METHODS: A total of 148 patients with pSS were retrospectively enrolled, including 89 without RA and 59 with concomitant RA. Demographic characteristics, inflammatory markers, cytokines, lymphocyte subsets, autoantibodies, and treatment regimens were analysed. Logistic regression

PIDgeon: An Explainable AI Model for Improved Flow Cytometry-Based Screening of Lymphoid Primary Immunodeficiencies.

BACKGROUND: Primary immunodeficiencies (PIDs) are rare disorders caused by immune system defects that are commonly screened using multi-parameter flow cytometry (FCM). To counter the subjective and time-consuming manual data analysis of FCM data, we present PIDgeon, a fully automated computational pipeline based on artificial intelligence (AI) techniques. PIDgeon is designed to characterize PID immune profiles, suggest PID subtypes based on altered immune profiles, age, and immunoglobulin levels, and generate interpretable reports. METHODS: The PIDgeon pipeline, including FlowSOM and extreme

Intratumoral spatiotemporal heterogeneity of HPV DNA status in HPV-associated oropharyngeal squamous cell carcinoma with clinical record.

BACKGROUND: Recently, the heterogeneity of human papillomavirus (HPV) status within HPV-associated oropharyngeal squamous cell carcinoma (HPV-OPSCC) has garnered substantial attention. However, the origin and relevance of a subset of cancer cells with HPV-loss remain largely unknown. METHODS: We encountered seven HPV-OPSCCs with spatial heterogeneity identified by HPV-DNA in situ hybridization among 83 OPSCCs, including 48 HPV-OPSCCs. We analyzed the characteristics of HPV-loss and HPV-positive cancer cells using histomorphology and immunohistochemistry. Spatial transcriptomic profiling was pe

Immune and HPV landscape in oral multiple primary cancers compared to single primary cancer.

BACKGROUND: Oral squamous cell carcinoma (OSCC) is the most common type of head and neck tumor. Notably, OSCC patients develop multiple primary cancers (MPCs) with distinct clinicopathological and molecular profiles compared to oral single primary cancer (SPC). However, the immune microenvironment of oral MPCs remains poorly characterized, particularly in contrast to SPC. METHODS: We conducted a paired case-control study using 60 samples from 30 MPCs patients and 60 samples from 60 SPC patients, matched by gender, lifestyle factors (smoking/alcohol), and tumor stage/grade. Bone Marrow CD8 + Abundance Inversely Correlates with Progressive Marrow Fibrosis and Myelodysplastic Evolution in GATA2 Deficiency: Case Report.

PURPOSE: GATA2 deficiency, a rare inborn error of immunity, presents with highly variable phenotypes. Bone marrow (BM) changes such as hypocellularity and myelodysplastic syndrome (MDS) are common, with hematopoietic stem cell transplantation being the only curative option due to the risk of progression to acute myeloid leukemia. Although traditional markers like cytogenetic abnormalities and somatic mutations (e.g., ASXL1) identify the risk of leukemic transformation, efforts to identify novel predictors of disease evolution are needed. CD8+ T cells are known to play a key role in MDS immune

From feather pecking to immunity: Immune differences between lines selected for high and low feather pecking.

Feather pecking (FP) is a serious behavioral disorder in laying hens, leading to feather damage, skin lesions, and often resulting in cannibalism. The mechanisms underlying FP are not clear yet, but recently the role of the immune system as a cause has been discussed. In humans, the interrelation between personality traits and the immune system is well-documented, with impulsivity and hyperactivity linked to distinct alterations in blood immune cell numbers and to elevated levels of pro-inflammatory cytokines. Similarly, FP in hens is associated with impulsivity and hyperactivity, suggesting a

Detection of disease-specific signatures in B cell repertoires of lymphomas using machine learning.

The classification of B cell lymphomas-mainly based on light microscopy evaluation by a pathologist-requires many years of training. Since the B cell receptor (BCR) of the lymphoma clonotype and the microenvironmental immune architecture are important features discriminating different lymphoma subsets, we asked whether BCR repertoire next-generation sequencing (NGS) of lymphoma-infiltrated tissues in conjunction with machine learning algorithms could have diagnostic utility in the subclassification of these cancers. We trained a random forest and a linear classifier via logistic regression bas

Destabilisation of T cell-dependent humoral immunity in sepsis.

Sepsis is a heterogeneous condition defined as life-threatening organ dysfunction caused by a dysregulated host response to infection. For some, sepsis presents as a predominantly suppressive disorder, whilst others experience a pro-inflammatory condition which can culminate in a 'cytokine storm'. Frequently, patients experience signs of concurrent hyper-inflammation and immunosuppression, underpinning the difficulty in directing effective treatment. Although intensive care unit mortality rates have improved in recent years, one-third of discharged patients die within the following year. Half

Effects of PM2.5 exposure on hematopoiesis and coupled immune disorder in adult male mice.

Fine particulate matter, namely PM2.5, increases the risk of morbidity and mortality in various systems, including the hematopoietic and immune systems. However, there is limited experimental evidence supporting the association between PM2.5 exposure and hematopoietic outcomes as most studies focus on epidemiological data. In this study, adult male mice were exposed to PM2.5 for a long time to investigate hematopoietic toxicity. There were significant increases in red blood cells (RBC), hemoglobin (HGB), and white blood cells (WBC) in peripheral blood after 5-month real-environmental PM2.5 exp

Activated CD4+ T Cell Proportion in the Peripheral Blood Correlates with the Duration of Cytokine Release Syndrome and Predicts Clinical Outcome after Chimeric Antigen Receptor T Cell Therapy.

Objective Chimeric antigen receptor (CAR) T cell therapy is an emerging and effective therapy for relapsed or refractory diffuse large B cell lymphoma (R/R DLBCL). The characteristic toxicities of CAR T cell therapy include cytokine release syndrome (CRS) and prolonged cytopenia. We investigated the factors associated with these complications after CAR T cell therapy by analyzing lymphocyte subsets following CAR T cell infusion. Methods We retrospectively analyzed peripheral blood samples on days 7, 14, and 28 after tisagenlecleucel (tisa-cel) infusion by flow cytometry at our institution betw
