The goal of targeted immunotherapy in cancer is to damage both malignant and tumor-supporting cells of the microenvironment but spare unaffected tissue. The malignant cells in classical Hodgkin lymphoma (cHL) selectively express CD30. They release this receptor on extracellular vesicles (EVs) for the tumor-supporting communication with CD30 ligand (CD30L)-positive bystander cells. Here, we investigated how CD30-positive EVs influence the efficacy of the CD30 antibody drug conjugate (ADC) Brentuximab Vedotin (SGN-35). The malignant cells and the EVs expressed the active sheddase ADAM10. ADAM10

Clinical definition of eczema is based on the presence of multiform erythematous, papular and vesicular lesions which are followed by marked desquamation. When the underlying mechanism is allergic, skin lesions are mediated by inflammatory Th1 lymphocytes recognizing hapten determinants, i.e. IVth allergic mechanism in allergic contact dermatitis. This mechanism is responsible also for allergy to bacterial proteins in nummular eczema, or to fungal and bacterial proteins, atopens and nickel in dyshidrotic eczema. Eczematous lesions can develop after damage to the skin barrier by toxic chemical

CD40/CD40 ligand interactions are required for the development of T cell-dependent Ab responses in vivo. The role of these cell surface molecules in contributing to T cell cytokine production and the development of effector populations other than B cells and T cells is, however, less well defined. We have examined the in vivo effects of blocking CD40/CD40 ligand interactions on the type 2 mucosal immune response that follows oral inoculation of mice with the nematode parasite, Heligmosomoides polygyrus. Administration of anti-gp39 (CD40L) mAb (MR1) blocked H. polygyrus-induced elevations in se

Secretory immunity is central in primary defense of the airway mucosa. B cells involved in this local immune system are initially stimulated in mucosa-associated lymphoid tissue, including tonsils and adenoids, and then migrate to secretory effector sites where they become immunoglobulin (Ig)-producing plasma cells. Locally produced Ig consists mainly of J-chain-containing dimers and larger polymers of IgA (pIgA) that are selectively transported through glandular cells by an epithelial receptor called secretory component or pIgR. Secretory antibodies perform surface protection by immune exclus

AIMS/HYPOTHESIS: There are substantial differences in the clinical presentation of type 1 diabetes depending on the first-developing autoantibody, although the underlying mechanisms are poorly understood. The DR3-DQ2 (DR3) and DR4-DQ8 (DR4) haplotypes at the MHC locus associate with GADA and IAA as the first-developing autoantibodies, respectively, and can therefore be used as proxies for first autoantibody development in large cohort studies of type 1 diabetes cases and control individuals.
METHODS: We performed the first genome-wide association study of type 1 diabetes stratified by DR3 and

The adherence of tumour cells to microvascular endothelium is believed to be a necessary step in their migration to sites of metastasis. It has been proposed that this process occurs when cell surface molecules on tumour cells bind to complementary sites on endothelial cells. The expression of these endothelial-derived cell adhesion molecules appears to be modulated by cytokines, a broad class of protein mediators which play important roles in immune and inflammatory reactions. It has been found by ourselves and others that exposure of endothelium to some cytokines augments the adhesion of inf

Trichinella spiralis infection causes hyperexcitability in enteric after-hyperpolarising (AH) sensory neurons that is mimicked by neural, immune or inflammatory mediators known to stimulate adenylyl cyclase (AC)/cyclic 3',5'-adenosine monophosphate (cAMP) signaling. The hypothesis was tested that ongoing modulation and sustained amplification in the AC/cAMP/phosphorylated cAMP related element binding protrein (pCREB) signaling pathway contributes to hyperexcitability and neuronal plasticity in gut sensory neurons after nematode infection. Currently, the exact role of telocytes within fascial structures is unknown. The morphology, distribution and behaviour of fascial telocytes as well as the mutual relationship between telocytes and other cellular fascia constituents should be definitely a subject of further studies. It will contribute to better understanding of the role of the fascial system in health and diseases, may shed light on the regeneration potential of these tissues and may help to find targets for future treatments for locomotor disorders, including fascial diseases. Last but not least, confirmation of the presence

When simple chemical are applied to the skin, a series of events ensues that under some conditions ultimately results in contact sensitivity (CS), a proven cell-mediated immune response (CMI). Since the discovery of CMI using picryl-Cl (PCl) in 1942, we have learned a great deal on the cellular mechanisms involved in CS, including the potential to treat warts, skin cancer, autoimmune disorders, and allergies. In this review we summarize some of the basic mechanisms of both the innate and acquired immune systems involved in CS.

P815 murine mastocytoma cells were separated to plastic-adherent and -nonadherent cell populations by repetitive in vitro selections. Their abilities of experimental and spontaneous metastases were investigated in the syngeneic DBA/2 mice. While the plastic-adherent populations were found to be liver-metastatic, the plastic-nonadherent populations were liver-nonmetastatic. The inability of plastic-nonadherent P815 cells to metastasize to the liver did not mean that these cells were not tumorigeneic because they could metastasize to tissues and/or organs other than the liver. Hence it could be

Chlamydia trachomatis, is a kind of obligate intracellular pathogen. The removal of C. trachomatis relies primarily on specific cellular immunity. It is currently considered that CD4+ Th1 cytokine responses are the major protective immunity against C. trachomatis infection and reinfection rather than CD8+ T cells. The non-specific immunity (innate immunity) also plays an important role in the infection process. To survive inside the cells, the first process that C. trachomatis faces is the innate immune response. As the "sentry" of the body, mast cells attempt to engulf and remove C. trachomat

BACKGROUND & AIMS: Dysbiosis of the intestinal microbiota has been associated with development of allergies in infants. However, it is not clear what microbes might contribute to this process. We investigated what microbe(s) might be involved in analyses of infant twins and mice.
METHODS: We studied fecal specimens prospectively in a twin cohort (n = 30) and age-matched singletons (n = 14) born at National Taiwan University Children's Hospital, Taipei, Taiwan, from April 2011 to March 2013. Clinical parameters (gestational age, birth body weight, mode of delivery and feeding, immunizations, a

Recent studies have highlighted an emerging role for neuro-immune interactions in mediating allergic diseases. Allergies are caused by an overactive immune response to a foreign antigen. The peripheral sensory and autonomic nervous system densely innervates mucosal barrier tissues including the skin, respiratory tract and gastrointestinal (GI) tract that are exposed to allergens. It is increasingly clear that neurons actively communicate with and regulate the function of mast cells, dendritic cells, eosinophils, Th2 cells and type 2 innate lymphoid cells in allergic inflammation. Several mecha

Trichomonas vaginalis causes the most prevalent sexually transmitted infection worldwide. Trichomonads have been detected in prostatic tissues from prostatitis, benign prostatic hyperplasia (BPH), and prostate cancer. Chronic prostatic inflammation is known as a risk factor for prostate enlargement, benign prostatic hyperplasia symptoms, and acute urinary retention. Our aim was to investigate whether T. vaginalis could induce inflammatory responses in cells of a benign prostatic hyperplasia epithelial cell line (BPH-1). When BPH-1 cells were infected with T. vaginalis, the protein and mRNA of

Allergic contact dermatitis (ACD) is an inflammatory skin disease of great and steadily increasing importance as an occupational health problem. The disease is induced by chemicals and metal ions which penetrate the skin and form complexes with host proteins. This process is accompanied by a strong, allergen-induced inflammatory reaction and leads to the migration of allergen-carrying dendritic cells (DC) from the skin to regional lymph nodes, where they promote generation of allergen-specific T cells. The latter are the ultimate effector cells of the disease. Re-exposure to the causative agen

Normal and malignant plasma cells (PC), follicular dendritic cells (FDC), myofibroblasts (MFB) and perineurial cells (PNC) were investigated for the expression of MUC-1 glycoprotein (MUC-1gp) by immunohistochemical and immunoelectron microscopic techniques using monoclonal antibodies E29, 115D8, DF3 and a combination of the three. MUC-1 glycoprotein-positive PC detected by the combined antibodies were frequently seen in a variety of pathological lesions tested, including chronic cervicitis, chronic synovitis, Hodgkin's disease, allergic rhinitis and sinusitis, tuberculous lymphadenitis, foreig

PURPOSE OF REVIEW: Until recently, the management of anaphylaxis has been dependent on the avoidance of known triggers combined with the emergent treatment of immediate and, possibly, fatal hypersensitivity reactions. The approval of omalizumab for the prevention of food-induced anaphylaxis heralds a new era for the use of biologics to curtail anaphylactic reactions in susceptible individuals.
RECENT FINDINGS: As more knowledge is gleaned about the immunologic mechanisms of anaphylaxis, the application of biologics for prophylaxis of anaphylaxis has accelerated. While omalizumab is currently

Hereditary alpha tryptasemia was first described in 2016 and is the most common (up to 72%) cause of elevated serum basal tryptase (TPS). The clinical presentation of this condition, which is caused by copy number gains in the TPSAB1 gene encoding serum α TPS, is variable for each patient. Some patients are asymptomatic, whereas in others, especially those with increased mast cell activation, it has been associated with a higher risk of anaphylaxis. Better characterization of this entity is important to identify at-risk patients and to develop new treatment strategies. This review provided an
