Causal effect of gut microbiota of Defluviitaleaceae on the clinical pathway of "Influenza-Subacute Thyroiditis-Hypothyroidism".

INTRODUCTION: Hypothyroidism has been found to be influenced by gut microbiota. However, it remains unclear which a taxon of gut microbiota plays a key role in this function. Identifying the key bacteria affects hypothyroidism and through what mechanism will be helpful for the prevention of hypothyroidism through specific clinical pathways. MATERIALS AND METHODS: In Study A, 35 families and 130 genera of gut microbiota are used as exposures, with hypothyroidism as the outcome. The causal effect of the gut microbiota on hypothyroidism is estimated through two-sample Mendelian randomization. Co

Women's reproductive traits and cerebral small-vessel disease: A two-sample Mendelian randomization study.

BACKGROUND: Observational studies have suggested that women's reproductive factors (age at menarche (AAM), age at first birth (AFB), age at first sexual intercourse (AFS), age at natural menopause (ANM), and pregnancy loss) may influence the risk of cerebral small-vessel disease (CSVD) although the causality remains unclear. METHODS: We conducted two-sample univariable Mendelian randomization (UVMR) and multivariable MR (MVMR) to simultaneously investigate the causal relationships between five women's reproductive traits and CSVD clinical [intracerebral hemorrhage (ICH) by location or small-v

The causal relationship between air pollution, obesity, and COVID-19 risk: a large-scale genetic correlation study.

OBJECTIVE: Observational evidence reported that air pollution is a significant risk element for numerous health problems, such as obesity and coronavirus disease 2019 (COVID-19), but their causal relationship is currently unknown. Our objective was to probe the causal relationship between air pollution, obesity, and COVID-19 and to explore whether obesity mediates this association. METHODS: We obtained instrumental variables strongly correlated to air pollutants [PM2.5, nitrogen dioxide (NO2) and nitrogen oxides (NOx)], 9 obesity-related traits (abdominal subcutaneous adipose tissue volume, w

Interrogating Causal Effects of Body Composition and Puberty-Related Risk Factors on Adolescent Idiopathic Scoliosis: A Two-Sample Mendelian Randomization Study.

Adolescent idiopathic scoliosis (AIS) is the most common form of pediatric musculoskeletal disorder. Observational studies have pointed to several risk factors for AIS, but almost no evidence exists to support their causal association with AIS. Here, we applied Mendelian randomization (MR), known to limit bias from confounding and reverse causation, to investigate causal associations between body composition and puberty-related exposures and AIS risk in Europeans and Asians. For our two-sample MR studies, we used single nucleotide polymorphisms (SNPs) associated with body mass index (BMI), wai

The Metabolome-Wide Signature of Major Depressive Disorder.

Major Depressive Disorder (MDD) is an often-chronic condition with substantial molecular alterations and pathway dysregulations involved. Single metabolite, pathway and targeted metabolomics platforms have indeed revealed several metabolic alterations in depression including energy metabolism, neurotransmission and lipid metabolism. More comprehensive coverage of the metabolome is needed to further specify metabolic dysregulation in depression and reveal previously untargeted mechanisms. Here we measured 820 metabolites using the metabolome-wide Metabolon platform in 2770 subjects from a large

Disentangling sex differences in the shared genetic architecture of posttraumatic stress disorder, traumatic experiences, and social support with body size and composition.

There is a well-known association of traumatic experiences and posttraumatic stress disorder (PTSD) with body size and composition, including consistent differences between sexes. However, the biology underlying these associations is unclear. To understand the genetic underpinnings of this complex relationship, we investigated genome-wide datasets informative of African and European ancestries from the Psychiatric Genomic Consortium, the UK Biobank, the GIANT Consortium, and the Million Veteran Program. We used genome-wide association statistics to estimate sex-specific genetic correlations (r

Assessing the Relationship Between Leukocyte Telomere Length and Cancer Risk/Mortality in UK Biobank and TCGA Datasets With the Genetic Risk Score and Mendelian Randomization Approaches.

BACKGROUND: Telomere length is an important indicator of tumor progression and survival for cancer patients. Previous work investigated the associations between genetically predicted telomere length and cancers; however, the types of cancers investigated in those studies were relatively limited or the telomere length-associated genetic variants employed often came from genome-wide association studies (GWASs) with small sample sizes. METHODS: We constructed the genetic risk score (GRS) for leukocyte telomere length based on 17 associated genetic variants available from the largest telomere len

CRP in cardiovascular disease.

In primary prevention, traditional risk factors are a useful first step in determining who is at cardiovascular risk, however, it has been noted that a considerable number of those at risk cannot be identified on the basis of traditional risk factors alone. Among blood biomarkers, C-reactive protein (CRP), measured by high-sensitivity assays (hsCRP), has received widespread interest and a large database has been accumulated on its potential role as a predictor of cardiovascular risk, although observed associations between circulating CRP and coronary heart disease (CHD) are unlikely to be caus

Causal Effects Between Antibody-Mediated Immune Responses and Parkinson's Disease: Insights from Genetic Studies.

BACKGROUND: Parkinson's disease (PD) is a multifactorial neurodegenerative disorder with a complex etiology. Increasing evidence suggests that antibody-mediated immune responses elicited by prior infectious exposures may contribute to its pathogenesis; however, whether these associations reflect true causal relationships remains unresolved, largely due to confounding and other inherent limitations of observational study designs. METHODOLOGY: We conducted a two-sample Mendelian randomization (MR) analysis to interrogate the causal associations between 46 antibody-mediated immune responses and

Role of Serum Metabolites in Mediating the Effect of Circulating Inflammatory Proteins on NK/T Cell Lymphoma.

INTRODUCTION: NK/T Cell Lymphomas (NKTCL) are highly aggressive types of Non-Hodgkin's Lymphoma (NHL) that have a predilection for Asian and South American populations. Identifying new drug targets or prognostic markers is essential for developing future strategies for NKTCL treatment. METHODS: This study employed a two-step and bidirectional Mendelian Randomization (MR) study with an Inverse Variance Weighted (IVW) approach to investigate the potential association of Circulating Inflammatory Proteins (CIPs) with NKTCL and the mediation effect of serum metabolites in the relationship between

A palmitoylation-related signature predicts prognosis and immune microenvironment features in diffuse large B-cell lymphoma.

BACKGROUND: Diffuse large B-cell lymphoma (DLBCL) is the most common aggressive lymphoma and remains highly heterogeneous in prognosis. Palmitoylation, an important post-translational modification, has been implicated in tumor biology, but its role in DLBCL, particularly in relation to the immune microenvironment, remains unclear. METHODS: Transcriptomic and clinical data from 706 patients with DLBCL were obtained from the Gene Expression Omnibus database. Prognosis-associated palmitoylation-related genes were identified by differential expression and univariate Cox regression analyses. An op

Alcohol consumption and molecular subtypes of colorectal cancer: pooled observational and Mendelian randomization analyses.

BACKGROUND: Alcohol consumption is associated with colorectal cancer (CRC) risk, yet its association with distinct molecular subtypes remains unclear. Clarifying this could reveal insights into alcohol's carcinogenic mechanisms. OBJECTIVES: We examined the association between alcohol consumption and the risk of CRC subtypes defined by individual tumor markers (and marker combinations), namely microsatellite instability (MSI) status, CpG island methylator phenotype (CIMP) status, BRAF, and KRAS mutations. METHODS: Pooled observational (ncases = 11,826, ncontrols = 10,888; nstudies = 10) and g

Gut microbiota pathways linking primary sclerosing cholangitis to colorectal cancer: the Lachnospiraceae family and PCBP1.

BACKGROUND: Colorectal cancer (CRC) remains a leading cause of cancer-related death worldwide. Primary sclerosing cholangitis (PSC) increases the risk of CRC, but the biological link is not well understood. Changes in the gut-liver axis, including gut microbiota and microbial metabolites, may contribute to CRC development in PSC. OBJECTIVE: To explore pathways connecting PSC with CRC through gut microbiota signals, and to identify candidate host genes for follow-up analyses. METHODS: Mendelian randomization (MR) was used to estimate the causal effect of genetically predicted PSC on CRC risk

Histone chaperone-based stratification combined with two-sample Mendelian randomization identifies ADORA2B and SAPCD2 as prognostic biomarkers in esophageal cancer.

BACKGROUND: Esophageal cancer (EC) lacks robust biomarkers to guide prognosis and therapy. Histone chaperone-related genes (HCRGs) shape chromatin states, but their roles in EC remain unclear. We integrated histone chaperone-based transcriptomic stratification with two-sample Mendelian randomization (MR) to identify genes with genetic evidence consistent with EC susceptibility and clinical relevance. METHODS: RNA-seq and clinical data from TCGA-ESCA (184 tumors with available RNA-seq data and 13 normal samples) and an external cohort (GSE53624; 119 tumors, 119 normals) were analyzed. Prognosi

An evaluation of age-varying genetic effects underlying body-mass index and blood pressure in the UK Biobank.

Genome-wide association studies (GWAS) are conventionally conducted in cohorts spanning a wide age-range. These studies typically assume that genetic associations are constant across different ages. Some traits, however, may have age-varying genetic associations. This has implications for the interpretation of genetic effects derived in downstream applications, such as Mendelian randomization (MR) analyses. In this study we conducted a series of age-stratified GWAS on individuals aged 40-69 years in the UK Biobank, for body-mass index (BMI) and three blood pressure traits (systolic, diastolic

High-fat diet reshapes microbial interactions to promote breast cancer stemness via Streptococcus mutans-induced leucine accumulation.

BACKGROUND: Obesity and high-fat diet (HFD) are established risk factors for breast cancer, but the mechanisms by which dietary-induced gut microbiota alterations influence cancer progression are not fully understood. The interplay between microbial composition, metabolites, and cancer progression warrants further exploration. METHODS: We employed Mendelian randomization (MR) and Bayesian colocalization analyses based on genome-wide association studies (GWAS) to identify gut microbial genera causally linked to breast cancer risk. Estimating the Causal Effect of Alcohol Consumption on Military-Relevant Mental Health Conditions: A Mendelian Randomization Study Using the All of Us Research Program.

INTRODUCTION: Depression, anxiety, and post-traumatic stress disorder (PTSD) are common, militarily relevant mental health disorders that severely impact operational readiness and can disqualify service members from flying class and special duty positions. While observational studies frequently correlate heavy alcohol consumption with these conditions, distinguishing direct causation from reverse causality (e.g., "chemical coping") remains difficult. This study uses Mendelian Randomization (MR) to estimate the causal effect of alcohol consumption on depression, anxiety, PTSD, and alcohol use d
