Developmental toxicity of dinocap in the mouse is not due to two isomers of the major active ingredients.

Technical-grade dinocap, a complex-mixture fungicide, is teratogenic in the CD-1 mouse, causing cleft palate and otolith defects. In this study we compared the developmental toxicities of 2,4-dinitro-6-(1-methylheptyl)phenyl crotonate and 2,6-dinitro-4-(1-methylheptyl)phenyl crotonate, model isomers of the major active ingredients of technical dinocap, to the known teratogenicity of the technical compound. Individual isomers, both isomers combined, or technical dinocap were administered to pregnant mice on days 7-16 of gestation. Some dams were killed at term and litters were removed, dead fet

Tolerance to and cross-tolerance among ethanol, pentobarbital and chlordiazepoxide.

The acute administration of ethanol, pentobarbital and chlordiazepoxide impaired, in a dose-dependent manner, the performance of rats on the moving-belt and two-way shuttle-box avoidance tests. Administration of these drugs for three weeks resulted in tolerance to their motor-impairing effects. Tolerance to ethanol or pentobarbital was characterized by a parallel shift of the dose-response curve to the right. Tolerance to chlordiazepoxide, however, was of greater extent and was accompanied by an apparent flattening of the dose-response curve. Symmetrical cross-tolerance developed between ethan

Behavior of rats during one-year administration of fluphenazine and subsequent withdrawal period.

During one-year administration of fluphenazine [FLU], 1 or 5 mg/kg/d ip. two phases in the behavior of rats were observed. During the first four months of treatment the locomotor and exploratory activities were depressed, while during the following months the reversal of this phenomenon was observed, and the activity of FLU-treated rats increased, particularly in the open-field test. The irritability of rats showed a triphasic change during one year treatment with 5 mg/kg/d FLU. Catalepsy declined with the increase in length of treatment and dose of FLU during the first three months of experim

Diurnal patterns in homecage behavior of rats after acute exposure to triethyltin.

Diurnal patterns of eating, drinking, locomotor activity, and rearing in male Fischer-344 rats were examined for 11 days after a single oral dose of triethyltin bromide (TET) at 0, 1.5, 3, or 5 mg/kg. The 5 mg/kg group exhibited a time-related drop in food consumption and body weight until 3 of 10 rats were sacrificed moribund 11 days after dosing. Doses of 1.5 and 3 mg/kg TET did not reduce body weight or consumption of food and water. In contrast, food consumption was significantly increased 7 and 11 days after 3 mg/kg TET, and diurnal patterns of eating and drinking were disrupted 7 days af

Eyelid movement disorders and motor impersistence in acute hemisphere disease.

The aim of this study was to evaluate, in an acutely brain-damaged population, the occurrence of motor impersistence and its relation to the side of lesion. All the patients hospitalized in a 10-month period for a stroke involving one hemisphere and able to cooperate were given, in the early days of illness, a motor impersistence battery requiring the maintenance of a position of eyelids, gaze, mouth, and limbs for 10 seconds. A control group of 50 patients without brain damage provided the basis for evaluating pathologic performance. Failure to keep eyes closed was the most common manifestati

Sensitivity of inbred mice to methylxanthines is not determined by plasma xanthine concentration.

Male CBA/J and SWR/J mice were tested with doses of caffeine, theophylline and 8-p-sulfophenyltheophylline (xanthine). Caffeine produced dose-related decreases in locomotor activity and colonic temperatures in SWR/J mice. However, caffeine produced increases in locomotor activity and failed to lower the body temperature of CBA/J mice. Theophylline produced a decrease in body temperature of SWR/J mice. Comparison of brain caffeine levels demonstrated no difference in brain pharmacokinetics. The peripherally active xanthine failed to alter body temperature at the same molar dose as that of theop

Social behaviour in pairs of C57BL/6 mice of both sexes in the open field: effects of saline drinking and of naloxone.

We have previously found that saline drinking increases fighting in male pairs and decided to test this treatment (0.9% NaCl for 24 hours before test day 1; SAL) on social behaviour of both males and females. Paired C57BL/6 mice (same-sex pairs) were observed in the open field in daily sessions for three days. One member of each pair (test mouse) was given either SAL treatment, a control injection of saline (SI), an injection of naloxone (1 mg/kg IP; NLX) or a combination of both treatments (NLX + SAL). NLX alone had previously been found to increase aggression in resident/intruder tests at th

Behavioral aspects of serotonin-dopamine interaction in the monkey.

The effects of serotonin (5-hydroxytryptamine; 5-HT) antagonists and 5-HT uptake inhibitors on the behavioral response to amphetamine and haloperidol in monkeys (cercopithecus aethiops) were investigated. Amphetamine increased locomotor activity and reactivity and induced repetitive movements of head, limbs and trunk, but no oral hyperkinesia. Haloperidol induced dystonia and parkinsonism. Pretreatment with the 5-HT antagonists cyproheptadine and mianserin increased amphetamine-induced locomotor activity, reactivity and repetitive movements and decreased haloperidol-induced dystonia and parkin

Differing effects of short-chain alcohols on body temperature and coordinated muscular activity in mice.

Administration of single doses of ethanol, 1-propanol, 1-butanol or 1-pentanol to mice caused hypothermia and impairment of rotarod performance. Repetitive doses, at 24-72 hr intervals led to development of tolerance to the hypothermic effects of ethanol but not of the other alcohols. No tolerance was seen in the impairment of rotarod performance with repeated doses of any of the alcohols. Ethanol did show an intersession tolerance on rotarod performance; at 20 and 80 min after injection, blood levels were similar, while performance was impaired at 20 but not at 80 min.

Can social and agonistic interactions be used to detect anxiolytic activity of drugs?

Majority of adult male albino random-bred mice housed singly or in small groups show agonistic behavior on interaction with a strange male mouse: some of them are predominantly aggressive ('aggressive' mice) while others show defenses or escapes even though their partners are not aggressive ('timid' mice). The remaining males not exhibiting agonistic behavior ('sociable' mice) show more social investigation then aggressive or timid mice and more locomotion then timid mice. Active defensive-escape behavior ('timidity') and inhibition of social investigation and of locomotion is much stronger in

Anatomical and behavioral recovery from the effects of spinal cord transection: dependence on metamorphosis in anuran larvae.

This study of spinal cord injury in bullfrog (Rana catesbeiana) tadpoles using the neuroanatomical tracer horseradish peroxidase (HRP) was undertaken to determine (1) whether the same anatomical regions that normally give rise to ascending or descending spinal tracts do so following complete spinal cord transection and (2) whether the course of behavioral recovery could be related to the anatomical results. The results of this study show that (1) spinal cord continuity is readily restored in tadpoles subjected to spinal cord transection, but nerve fibers crossing the site of injury end within

[Psychopharmacological study of enkephalins, with special reference to the relation between behavioral profiles and brain monoamines].

The effects of intracerebral injection of methionine-enkephalin (met-enk) and leucine-enkephalin (leu-enk) on the behavioral patterns were examined by using multi-dimensional behavioral analyser (Animex II) in relation to the mouse brain monoamine metabolism. The multi-dimensional behavioral analyser which is able to record 9 different measures of behavior exhibited by mice has been used. The results have indicated that met-enk (100 micrograms) and leu-enk (200 micrograms) caused a remarkable but transient increase in the linear locomotion recorded on the counter 1/1. In the biochemical study,

The effects of postpartum exposure to triethyl tin on the neurobehavioral functioning of rats.

Male and female Fischer rat pups received sc injections of 1.5 or 3 mg/kg of triethyl tin bromide (TET), ethanol vehicle or distilled water on day 5 postpartum. At 21, 28, 60, and 90 days of age, the rats were weighed and assessed in a battery of tests which included fore- and hindlimb grip strength, negative geotaxis, startle responsiveness and spontaneous motor activity. Animals at 60 days of age were also tested in a two-way shuttle box avoidance procedure. Exposure to TET did not affect body weight at any time during testing, nor did it affect negative geotaxis of grip strength, Startle re

[Pharmacological activity of the optical isomers of sydnocarb].

Of the two optical isomers of sydnocarb, only 1- (or 1R)-isomer exerts a stimulant effect on the central nervous system. By its stimulant action it differs from both optical isomers of amphetamine. The activity of 1-(IR)-sydnocarb as measured by the locomotor excitation test is similar to that of d(1S)-amphetamine but is significantly less powerful as indicated by stereotypy, hyperthermia, anorexigenic action and antagonism to the depressant effects of reserpine.

Evaluation of teratogenicity and behavioral toxicity with inhalation exposure of maternal rats to trichloroethylene.

Female rats were exposed by inhalation to trichloroethylene (TCE) vapors at a concentration of 1800 +/- 200 ppm to determine whether exposure before mating and during pregnancy is more detrimental to reproductive outcome than exposure either before mating alone or during pregnancy alone. Four treatment groups were utilized in a two by two factorial design: exposure to TCE for 2 weeks before mating and during the first 20 days of pregnancy; TCE before mating and filtered air during pregnancy; filtered air before mating and TCE during pregnancy; and filtered air before and during pregnancy. Predation, aggression, and activity levels in food-deprived sunfish (Lepomis macrochirus and L. gibbosus): motivational interactions.

Three experiments investigated the effects of food deprivation on several behavioral categories in two species of sunfish. In Experiment 1, predatory behavior and general activity were observed under five levels of deprivation. For both species, predation measures increased in a similar negatively accelerating manner with increasing deprivation, while activity changed in a more complex fashion. Experiment 2 examined the effects of deprivation on activity in a novel environment and showed that the deprivation effects of Experiment 1 were masked by the response to the new setting. In Experiment

Pharmacological activity of three fractions obtained by smoking cannabis through a water pipe.

In a collaborative work, six chemical fractions obtained from smoke of cannabis, prepared by the Department of Biological Chemistry, University of Athens, were assayed pharmacologically at the Department of Psychobiology, Escola Paulista de Medicina, Brazil. The chemical fractions, obtained through burning cannabis mixed with a special tobacco through a water pipe similar to that used by Greek smokers, were: fraction IS composed of water-soluble compounds, fraction IIS made up of water-insoluble substances that condensed at the top of the water phase due to their low solubility; and fraction I
