α-Synuclein triggers intercellular nanotubes formation to prevent apoptosis in astroglia by promoting stemness.

Astrocytes play a significant role in neuroprotection by internalizing neurodegenerative aggregates and facilitating their degradation. Recent studies indicate that α-Synuclein (α-SYN) protofibrils promote the transfer of pathogenic aggregates and dysfunctional mitochondria between astroglia via tunneling nanotubes (TNTs), which enhances cell survival and resistance to apoptosis. However, the underlying mechanism of TNT-driven apoptosis resistance remains unclear. We find that α-SYN protofibrils induce aberrant mitochondria with decreased membrane potential (Ψm) and promote dynamic actin remod

Next-generation hybrid nanosystems for cannabidiol: From molecular challenges to site-specific preclinical applications.

Cannabidiol (CBD) is a monoterpene phenolic compound extracted mainly from Cannabis sativa, which is produced in high amounts compared to other plants. The compound is regarded as a promising therapeutic agent with anti-inflammatory, analgesic, and neuroprotective effects for biomedical use. Furthermore, important advances have been obtained in dermatological and anticancer effects. Nowadays, the major challenges to the development of new medicine based on CBD arise from its unfavorable physicochemical properties that include reduced solubility in aqueous media (∼0.01 mg/mL), significant degra

Fluoxetine attenuates UPR-activation, neuroinflammation, and oxidative stress in a nitroglycerin-induced rat model of migraine.

OBJECTIVES: Migraine is a debilitating neurological disorder characterized by oxidative stress, neuroinflammation, and dysregulation of the unfolded protein response (UPR) in the brain. Fluoxetine, a selective serotonin reuptake inhibitor (SSRI), has demonstrated neuroprotective properties, but its effects on UPR activation, neuroinflammation, and oxidative stress in migraine are not well defined. This study evaluates the effects of fluoxetine on UPR-related gene expression, inflammatory cytokines, and oxidative stress markers in a nitroglycerin (NTG)-induced chronic migraine rat model. MATER

Edaravone-dexborneol improves neurological function after cerebral ischemia-reperfusion injury by activating NRF2/p62 and suppressing NF-κB/NLRP3 signaling.

Edaravone-dexborneol (EDB) is a prospective neuroprotective agent for ischemic stroke. However, its integrated mechanisms of action remain to be elucidated. Here, we investigated the multiple effects of EDB in a rat model of cerebral ischemia-reperfusion injury induced by transient middle cerebral artery occlusion (MCAO). Rats received intraperitoneal EDB (4 mL/kg, once daily). Acute infarct volume and biochemical parameters were assessed 24 h after reperfusion. Longitudinal behavioral and histological outcomes were evaluated after a 7-day treatment course. EDB significantly reduced neurologic

Mechanistic Integration of Beta-Alanine-Carnosine Buffering and Glutamine Metabolism in Exercise-Induced Muscle-Brain Crosstalk: Implications for Aging, Neurodegeneration, and Cognitive Decline.

Aging is marked by a gradual deterioration in muscular performance, cognitive abilities, and metabolic flexibility, phenomena that are intricately linked through the muscle-brain axis. Recent research indicates that particular amino acids, notably beta-alanine (BA) and glutamine, may not function in isolation but instead converge mechanistically to modulate this axis through synergistic influences on intracellular pH regulation, nitrogen metabolism, and redox equilibrium. BA, through its involvement in the biosynthesis of carnosine, augments intracellular buffering capacity and maintains pH st

A combined extract of Salvia miltiorrhiza and Panax notoginseng enhances collateral flow and protects the neurovascular unit after ischemic stroke.

BACKGROUND: Ischemic stroke remains a major cause of mortality and disability, with few therapeutic options outside the narrow time window for reperfusion. Collateral circulation critically sustains penumbral viability, yet no pharmacological agent is approved to enhance collateral flow. Salvia (S.) miltiorrhiza and Panax (P.) notoginseng possess complementary vascular, anti-inflammatory, and neuroprotective actions, suggesting potential synergy in acute ischemia. METHODS: A combined extract of S. miltiorrhiza and P. notoginseng (SM/PN) was tested in a permanent middle cerebral artery occlusi

Neurovascular unit uncoupling in diabetic retinopathy: molecular mechanisms and stage-adapted therapeutic strategies.

Diabetic retinopathy (DR) is a major neurovascular complication of diabetes and remains a leading cause of vision loss among working-age adults worldwide. Although DR has traditionally been classified as a microvascular complication, it is now increasingly recognized as a neurovascular degenerative disorder involving coordinated injury to neuronal, glial, vascular, and extracellular matrix components of the retinal neurovascular unit (NVU). The NVU provides the structural and functional basis for coupling neuronal activity to local blood flow and for maintaining retinal immune and barrier home

Cholecalciferol (vitamin D3): efficacy, safety, and implications in public health.

Vitamin D₃ (cholecalciferol) is a fat-soluble secosteroid with essential roles in calcium-phosphorus metabolism, bone health, and an expanding range of extraskeletal processes. Upon synthesis in the skin via ultraviolet B exposure or ingestion from dietary sources, cholecalciferol is hydroxylated in the liver and kidneys to form its active metabolite, calcitriol (1,25-dihydroxyvitamin D), which exerts pleiotropic effects through vitamin D receptor (VDR)-mediated genomic and non-genomic pathways. This narrative review synthesizes evidence on the systemic effects of high-dose cholecalciferol on

Anti-inflammatory Effects of Synaptamide in the Peripheral Immune System in a Murine Model of Traumatic Brain Injury.

Traumatic brain injury (TBI) induces not only local neuroinflammation but also systemic immune responses involving peripheral organs such as the spleen. While synaptamide (N-docosahexaenoylethanolamine), an endogenous lipid mediator derived from docosahexaenoic acid, has demonstrated neuroprotective effects, its impact on peripheral immune regulation after TBI remains unclear. In this study, we investigated the anti-inflammatory effects of synaptamide in a murine weight-drop TBI model. Male C57BL/6 mice were divided into five groups: “Control”, “Sham” (craniotomy only), “Sham + Syn” - cranioto

Neurotrophic Effects of GH and GnRH in a Full Sciatic Nerve Transection Model in Male Rats.

INTRODUCTION: Peripheral nerve injuries, such as sciatic nerve transection (SNT), are often associated with significant sensory and motor deficits. Growth hormone (GH) and gonadotropin-releasing hormone (GnRH) have been shown to exert neurotrophic effects that can promote nerve regeneration and functional reinnervation. However, the combined impact of these hormones on peripheral nerve repair remains poorly understood. METHODS: This study aimed to analyze the individual and combined effects of GH and GnRH in a rat model of SNT, using orchiectomized male rats to prevent steroid-mediated neuror
