American journal of orthodontics and dentofacial orthopedics : official publication of the American Association of Orthodontists, its constituent societies, and the American Board of Orthodontics1990eng
Recently, the mechanism of action of prostaglandins in promoting bone resorption has received wide attention. Prostaglandins are used in orthodontics to shorten the period of tooth movement. PGE1 was administered locally (5 micrograms/kg every 12 hours) and systemically (7.5 ng/kg/min) to rats to study the difference in the efficacy of the two methods of administration in accelerating bone resorption. The Waldo method was used on 72 Wistar rats (8 weeks old) for 5 days to induce mesial movement of the first molar. The body weight of the rats was measured every 12 hours from the start of the ex

Stimulators of bone resorption, such as PTH, 1,25-dihydroxycholecalciferol [1,25-(OH)2D3], or prostaglandin E2 (PGE2), do not cause calcium release from cultured calvaria of the genetically determined osteopetrotic microphthalmic (mi/ mi) mouse, due to a defect in the function of osteoclasts. To investigate the capacity of cells of mi/mi bone to degrade collagen, calvaria of 1- to 3-day-old normal and mi/mi littermates were labeled in vivo with [3H]proline 16 h before removal, followed by culture in resorption medium. PTH, 1,25-(OH)2D3, and PGE2 stimulated the release of 3H-labeled material in

Hand bone calcium content [Ca] has been measured by local neutron analysis in hemodialyzed patients free of signs of osteomalacia before and 6-12 months after treatment with vitamin D metabolites, and it has been compared to some iliac bone histomorphometric parameters. With 1,25-(OH)2D3 alone, [Ca] increases significantly at the 6th month of treatment (p less than 0.001) but not from the 6th to the 12th month. With 1 alpha-(OH)D3 + 25-(OH)D3 the same phenomenon is observed at the 6th and 12th months of treatment. Before treatment, [Ca] was correlated negatively with the osteoid surfaces (OS)

The developmental potential of isolated rabbit bone marrow cell populations was examined following autogeneous implantation into diffusion chambers. After 4 weeks, the implants were harvested and processed for light and electron microscopy. More total tissue was formed in abdominally implanted chambers than in corresponding femoral chambers. Two of the separated marrow cell populations produced significantly greater amounts of fibrous connective tissue, cartilage, and bone than did whole-marrow controls. These two populations, which were defined by density gradients of 1.050-1.055 gm/cm3 and 1

Stem cell-associated microRNAs (miRNAs) have emerged as pivotal regulators of osteogenesis, influencing lineage commitment, differentiation, and skeletal tissue homeostasis. Their dysregulation has been increasingly linked to a spectrum of osteogenic disorders, including osteoporosis, osteoarthritis, and impaired fracture healing. This narrative review synthesizes current evidence on the diagnostic potential of stem cell-associated miRNAs in bone-related pathologies. We highlight their roles in modulating osteoblast and osteoclast activity, maintaining mesenchymal stem cell fate, and orchestra

Osteomodulin (OMD) is a class II small leucine-rich proteoglycan enriched in mineralized extracellular matrices. This review evaluates OMD-specific evidence linking structural features to matrix interactions and tissue responses. Biochemical and mutational studies support roles in type I collagen recognition and fibril assembly, whereas hydroxyapatite affinity does not establish direct control of mineral nucleation. Cell and animal studies implicate OMD in osteogenic differentiation, osteoclast regulation, and inflammatory responses, but support for individual mechanisms ranges from direct lig

Osteoarthritis (OA) is a common degenerative joint disease, characterized by oxidative stress, excessive inflammatory responses, cartilage matrix degradation, and subchondral bone loss. However, a therapeutic strategy that simultaneously addresses these interconnected pathologies remains elusive in current nanomedicine. In this study, we prepare a DNA nanostructure with antioxidant enzyme-mimetic activities through the coordination-driven co-assembly of oligonucleotides and inorganic cerium pyrophosphate crystals. After intra-articular injection, the valence transition property of cerium ions

Diabetic craniofacial bone defects exhibit unsatisfactory repair attributed to synergistic oxidative stress, local acidification and disordered bone immunity, whereas conventional mono-functional biomaterials are incapable of synchronously resolving these interlocked pathological cascades. Herein, a hierarchical nanoplatform (CM/LDH) is fabricated by immobilizing Ce-MOF nanozyme onto CaAl-LDH substrate for synergistic bone remediation. As an alkaline and bioactive support, CaAl-LDH improves peri-defect microenvironment acidification to preserve the redox catalytic activity of loaded Ce-MOF, ac

Natural bone exhibits piezoelectric properties due to its collagen structure. Piezoelectric biomaterials, which can replicate the electromechanical behavior of bone, have thus become a key focus in the development of artificial biomaterials. Their piezoelectricity is expressed as the direct effect under mechanical stimulation and the inverse effect under electrical stimulation. Studies have shown that appropriate mechanical or electrical cues can enhance bone defect repair by activating the piezoelectric properties of these materials, influencing osteogenesis, angiogenesis, osteoclastogenesis,

Osteoinductive calcium phosphate ceramics (CaPs) hold great promise for bone repair, yet the metabolic principles governing their efficacy are poorly defined. Here, we identify mitochondrial oxidative phosphorylation (OXPHOS) as an early and indispensable pathway activated specifically by an osteoinductive tricalcium phosphate (TCPS) in material-induced bone formation. Our findings demonstrate that OXPHOS critically governs the M2 macrophage-osteoclast axis for bone formation by regulating both cellular energy supply and reactive oxygen species (ROS) homeostasis. Disrupting either function-by

Type 2 diabetes is a major risk factor for fragility fractures, yet the contributors to skeletal fragility remain unclear. Through integrated clinical metabolomics, in vivo, and in vitro analyses, we identify maltol-a widely used food additive-as a previously unrecognized risk factor for hyperglycemia-associated bone fragility. Metabolomic profiling of femoral neck tissue from individuals with fragility fractures showed diabetes-associated maltol accumulation, and elevated circulating maltol levels correlated with increased fracture incidence. Mechanistically, maltol inhibits osteoblast differ

Prostate cancer (PCa) is a common malignancy in men, and bone metastasis is a leading cause of mortality in advanced-stage PCa. This study aims to identify critical genes involved in PCa bone metastasis, exploring biomarkers for prognosis and precision treatment. Forkhead Box Q1 (FOXQ1) was identified as a potential key gene through screening of public databases, and was found to be markedly upregulated in bone metastatic PCa compared to primary PCa. FOXQ1 promotes PCa cell proliferation and metastasis while inhibiting apoptosis. Additionally, FOXQ1 recruits macrophages, promotes M2 polarizati

Musculoskeletal diseases are a major health burden. Development of bone-active therapies has been hindered by limited understanding of the cells and genes that regulate the skeleton. We exploited the value of cross-species analysis and developed single-cell methodologies in skeletal tissues to define the critical endosteal compartment that regulates bone turnover. Thirty-four distinct cell types were identified, and disease-relevant cells prioritized using enrichment for rare skeletal disorder genes and bone-mineral-density-associated genes in an extended UK Biobank genome-wide association stu

Ankylosing spondylitis (AS) is a chronic autoimmune disease characterised by inflammation, bone destruction and pathological new bone formation in the spine and sacroiliac joints. Macrophages, as central innate immune cells, play a pivotal role in AS pathogenesis through dynamic polarisation into pro-inflammatory M1 and anti-inflammatory M2 phenotypes. M1 macrophages dominate early disease stages, producing cytokines such as TNF-α, IL-6 and IL-1β, activating Th1/Th17 responses, and driving tissue inflammation and osteoclast-mediated bone loss.M2 macrophages prevail in later stages, secreting I

Osimertinib, an epidermal growth factor receptor tyrosine kinase inhibitor, improves the progression-free and overall survival of patients with inoperable or recurrent non-small cell lung cancer. However, it is reportedly associated with osteonecrosis of the jaw and oral mucositis due to its ability to reduce osteoclast differentiation and inhibit angiogenesis. A 73-year-old woman had been receiving osimertinib for recurrence after lung adenocarcinoma surgery; 3 years later she presented with bone exposure and buccal mucosal necrosis. After the clinical diagnosis of medication-related osteonec

The gut microbiota plays a multifaceted role in calcium homeostasis and bone metabolism -acting through metabolic, immunological, and hormonal pathways that collectively constitute the gut-bone axis. The microbiota influences calcium bioavailability through several overlapping mechanisms that act in the intestine. Moreover, microbial fermentation products may directly impact the osteoblast-osteoclast interplay and, by modulating immune and endocrine functions, are crucial for bone metabolism. A healthy microbiota supports bone formation; however, intestinal dysbiosis may impair bone structure

Osteopetrosis is an uncommon inherited skeletal condition arising from impaired osteoclast activity, with its autosomal recessive forms being linked to mutations in the TNFRSF11A gene. This study reports on a three-year-old Chinese girl presenting with recurrent fractures, short stature, and macrocephaly. Radiological examinations revealed diffusely elevated bone mineral density and constricted medullary spaces. Meanwhile, whole-exome sequencing detected compound heterozygous alterations in the TNFRSF11A gene: c.1567?+?2T>A inherited from the father and c.630C>G from the mother. The child unde

Denosumab is a human monoclonal antibody that inhibits RANKL, thereby suppressing osteoclast-mediated bone resorption and increasing bone mass in patients with osteoporosis. Although generally well tolerated, denosumab is associated with hypocalcemia, particularly in individuals with impaired calcium and vitamin D homeostasis. We report the case of a 40-year-old woman with a history of sleeve gastrectomy who developed severe, symptomatic hypocalcemia following a single dose of denosumab despite receiving vitamin D supplementation. She presented with neuromuscular symptoms, QTc prolongation, pr

Keratin positive giant cell rich tumor is a rare mesenchymal tumor first described in 2020. It can occur in both soft tissue and bone and predominantly affects young women. The tumor's biological behavior remains uncertain despite its low-grade classification. Characterized by keratin expression and a HMGA2::NCOR2 gene fusion, keratin positive giant cell rich tumors resemble conventional giant cell tumors but lacks H3-3A gene mutations. This article presents a 31-year-old woman with a keratin positive giant cell rich tumor in the pelvic bone, detailing her clinical presentation, diagnostic pro

This study identified six novel azaphilones, isochromophilones G-L (1-6), and three novel biosynthetically related congeners (7-9) from Diaporthe sp. SCSIO 41011. The structures and absolute configurations were elucidated through comprehensive spectroscopic analyses combined with experimental and calculated electronic circular dichroism (ECD) spectra. Significantly, three highly oxygenated azaphilones contain an acetyl group at the terminal chain (4) or linear conjugated polyenoid moieties (5 and 6), which occur infrequently in the azaphilone family. Additionally, several compounds demonstrate
