HLA class I depletion by citric acid, and irradiation of apheresis platelets for transfusion of refractory patients.

BACKGROUND: Patients can form antibodies to foreign human leukocyte antigen (HLA) Class I antigens after exposure to allogeneic cells. These anti-HLA class I antibodies can bind transfused platelets (PLTs) and mediate their destruction, thus leading to PLT refractoriness. Patients with PLT refractoriness need HLA-matched PLTs, which require expensive HLA typing of donors, antibody analyses of patient sera and/or crossmatching. An alternative approach is to reduce PLT HLA Class I expression using a brief incubation in citric acid on ice at low pH. METHODS AND MATERIALS: Apheresis PLT concentra

Hydrogen sulfide suppresses homocysteine-induced glial activation and inflammatory response.

Neuro-inflammation plays a critical role in hyperhomocysteinemia (HHcy)-associated neurodegenerative disorders. Hydrogen sulfide (H2S) has been suggested as an endogenous neuromodulator and potent anti-inflammatory molecule. In present study, we have investigated the effect of NaHS supplementation (a H2S source) on inflammatory response in animals subjected to HHcy. NaHS adminstration restored the decreased levels of H2S and polysulfides with a concomitant increase in the activity of cystathionase (CSE) and cystathionine β-synthase (CBS) in the brain regions of HHcy animals. NaHS supplementati

The role of exosomes and MYC in therapy resistance of acute myeloid leukemia: Challenges and opportunities.

Acute myeloid leukemia (AML) is caused by abnormal production of white blood cells, red blood cells or platelets. The leukemia cells communicate with their microenvironment through nano-vesicle exosomes that are 30-100 nm in diameter. These nano-vesicles are released from body fluids upon fusion of an endocytic compartment with the cell membrane. Exosomes function as cargo to deliver signaling molecules to distant cells. This allows cross-talk between hematopoietic cells and other distant target cell environments. Exosomes support leukemia growth by acting as messengers between tumor cells and

Inflammation, Endothelial Dysfunction and Arterial Stiffness as Therapeutic Targets in Cardiovascular Medicine.

In the last decades, many factors thought to be associated with the atherosclerotic process and cardiovascular events have been studied, and some of these have been shown to correlate with clinical outcome, such as arterial stiffness, endothelial dysfunction and immunoinflammatory markers. Arterial stiffness is an important surrogate marker that describes the capability of an artery to expand and contract in response to pressure changes. It can be assessed with different techniques, such as the evaluation of PWV and AIx. It is related to central systolic pressure and it is an independent predi

Mast cells promote proliferation and migration and inhibit differentiation of mesenchymal stem cells through PDGF.

BACKGROUND: Mast cells (MCs) dynamically participate in wound healing after myocardial infarction (MI) by releasing cytokines. Indeed, MC-deficient mice undergo rapid left ventricular dilation post-MI. Mesenchymal stem cells (MSCs) are recruited to the injured region following an MI and have potential for cardiac repair. In the current study, we evaluate the effect of MCs on MSC proliferation and myogenic differentiation. METHODS AND RESULTS: MCs were cultured from mouse bone marrow and MC granulate (MCG) was extracted from MCs via freeze-thaw cycles followed by filtration. α-SMA (smooth musc

HYAL-2-WWOX-SMAD4 Signaling in Cell Death and Anticancer Response.

Hyaluronidase HYAL-2 is a membrane-anchored protein and also localizes, in part, in the lysosome. Recent study from animal models revealed that both HYAL-1 and HYAL-2 are essential for the metabolism of hyaluronan (HA). Hyal-2 deficiency is associated with chronic thrombotic microangiopathy with hemolytic anemia in mice due to over accumulation of high molecular size HA. HYAL-2 is essential for platelet generation. Membrane HYAL-2 degrades HA bound by co-receptor CD44. Also, in a non-canonical signal pathway, HYAL-2 serves as a receptor for transforming growth factor beta (TGF-β) to signal wit

Isorhynchophylline protects against pulmonary arterial hypertension and suppresses PASMCs proliferation.

Increased pulmonary arterial smooth muscle cells (PASMCs) proliferation is a key pathophysiological component of pulmonary vascular remodeling in pulmonary arterial hypertension (PAH). Isorhynchophylline (IRN) is a tetracyclic oxindole alkaloid isolated from the Chinese herbal medicine Uncaria rhynchophylla. It has long been used clinically for treatment of cardiovascular and cerebrovascular diseases. However, very little is known about whether IRN can influence the development of PAH. Here we examined the effect of IRN on monocrotaline (MCT) induced PAH in rats. Our data demonstrated that IRN

Adenoviral dominant-negative soluble PDGFRβ improves hepatic collagen, systemic hemodynamics, and portal pressure in fibrotic rats.

BACKGROUND & AIMS: Platelet-derived growth factor (PDGF) is the most potent stimulus for proliferation and migration of stellate cells. PDGF receptor β (PDGFRβ) expression is an important phenotypic change in myofibroblastic cells that mediates proliferation and chemotaxis. Here we analyzed the relationship between PDGFRβ expression, hemodynamic deterioration, and fibrosis in CCl(4)-treated rats. Thereafter, we investigated the effects produced by an adenovirus encoding a dominant-negative soluble PDGFRβ (sPDGFRβ) on hemodynamic parameters, PDGFRβ signaling pathway, and fibrosis. METHODS: Mea

Platelet-derived growth factor BB induces nuclear export and proteasomal degradation of CREB via phosphatidylinositol 3-kinase/Akt signaling in pulmonary artery smooth muscle cells.

Cyclic AMP response element binding protein (CREB) content is diminished in smooth muscle cells (SMCs) in remodeled pulmonary arteries from animals with pulmonary hypertension and in the SMC layers of atherogenic systemic arteries and cardiomyocytes from hypertensive individuals. Loss of CREB can be induced in cultured SMCs by chronic exposure to hypoxia or platelet-derived growth factor BB (PDGF-BB). Here we investigated the signaling pathways and mechanisms by which PDGF elicits depletion of SMC CREB. Chronic PDGF treatment increased CREB ubiquitination in SMCs, while treatment of SMCs with

Drug synergies and the potential for combination therapy in onychomycosis.

Characterized by abnormal proliferation and migration of vascular smooth muscle cells (VSMCs), neointima hyperplasia is a hallmark of vascular restenosis after percutaneous vascular interventions. Vaccinia-related kinase 1 (VRK1) is a stress adaptionassociated ser/thr protein kinase that can induce the proliferation of various types of cells. However, the role of VRK1 in the proliferation and migration of VSMCs and neointima hyperplasia after vascular injury remains unknown. We observed increased expression of VRK1 in VSMCs subjected to platelet-derived growth factor (PDGF)-BB by western blott

The immunothrombosis pathway linking pulmonary disease to stroke and neurodegeneration: mechanisms and therapeutic implications.

Pulmonary diseases increasingly reshape vascular biology and coagulation beyond the lung. Across acute infections and acute respiratory distress syndrome (ARDS), chronic airway inflammation (e.g., COPD), sleep-disordered breathing, fibrotic interstitial lung disease, and particulate air pollution, convergent immune programs couple inflammation to coagulation through immunothrombosis. Physiologic immunothrombosis can confine pathogens within the microvasculature, but dysregulated thromboinflammation drives endotheliopathy, platelet-leukocyte cooperation, neutrophil extracellular trap (NET) form

Electrospun PLGA-Propolis Scaffolds Regulate Collagen Architecture in Burn Wounds.

Wound (especially burn) healing, is a complex process involving cells e.g., leukocytes, macrophages, keratinocytes, fibroblasts, endothelial cells and platelets. Additionally, glycosaminoglycans, proteoglycans and collagen participate in the remodeling of the extracellular matrix (ECM). It is well-known that collagen, especially type I and III, are products of fibroblasts. These cells proliferate in the final phase wound healing. The various stages of skin regeneration, i.e., processes such as hemostasis, inflammation, cells' growth, differentiation and migration, can be accelerated by certain

Breaking the endometriosis-stroke axis: lesion-focused hydrogel reprogramming of thromboinflammatory networks.

Endometriosis is a chronic, systemic inflammatory disorder characterized by the ectopic implantation of endometrial-like tissue and accompanied by persistent pain and infertility. Beyond the pelvis, accumulating epidemiological evidence indicates that endometriosis confers a significantly elevated risk of cardiovascular and cerebrovascular disease, including ischemic stroke. This association points to a shared pathogenic continuum in which chronic inflammation, aberrant neuroimmune signaling, dysregulated coagulation, and endothelial dysfunction jointly drive both local lesion persistence and

Interleukin-1 receptor type 1 in pancreatic cancer progression and metastasis: a review.

Pancreatic ductal adenocarcinoma (PDAC) is a global lethal malignancy, with increasing incidence and dismal survival rates. Inflammation plays a central role in shaping PDAC biology, and interleukin-1 receptor type 1 (IL-1R1) is a key mediator linking oncogenic signaling, stromal activation, and immune suppression. This review summarizes current insights into IL-1R1 signaling in PDAC, focusing on its role in tumor microenvironment remodeling, tumor progression, and metastasis. IL-1R1 activation by IL-1α or IL-1β triggers MyD88-dependent pathways, including NF-κB, MAPK, and STAT3, which induce

Improvement in health‑related quality of life in patients with heavy menstrual bleeding after treatment and its association with hereditary bleeding disorders.

BACKGROUND: The prevalence of hereditary bleeding disorders (HBDs) in heavy menstrual bleeding (HMB) and their impact on health-related quality of life (HRQoL) remain underexplored. We investigated the HRQoL of women with non-structural HMB and its association with HBDs. METHOD: A prospective longitudinal study of 100 women with HMB without structural gynecological pathology. HMB was defined as ≥ 8 days of bleeding or a PBAC score > 100 points. Four HRQoL questionnaires —SF-12®v2, EQ-5D-3 L, MBQ, and SAMANTA— were administered at baseline and 6 months. HBD diagnosis included hypermobility spec

Effect of Prostaglandin Analogues on Hyaluronan Production of Orbital Fibroblasts.

Background: In thyroid eye disease (TED) pathogenesis, a major contributor is hyaluronan (HA) overproduction by orbital fibroblasts (OF). TED patients often receive prostaglandin analogues (PGA) for elevated intraocular pressure. Their direct effects on OFs, however, remain unclear. Methods: OFs were established from connective tissues obtained from TED patients (n = 4) and non-TED controls (n = 4) during orbital surgeries. HA levels in primary OF culture media, and in the OFs' pericellular coat, were measured by ELISA. Cultures were performed with or without platelet-derived growth factor BB
