The Effectiveness of Myofascial Release Versus Traditional Physiotherapy Techniques for Plantar Fasciitis Among Adults: A Randomized Controlled Trial.

BACKGROUND: Plantar fasciitis is characterized by degenerative changes and irritation at the origin of the plantar fascia, primarily involving the medial calcaneal tuberosity and the surrounding perifascial soft tissues. Common causes include overuse from activities such as running, prolonged standing, or walking on hard surfaces, which can lead to small tears in the fascia. Other contributing factors include wearing unsupportive footwear, obesity, and foot conditions such as pes planus or cavus. METHODS: Fifty-eight subjects with plantar fasciitis were selected and randomly assigned to two g

Decoding cellular communication networks and signaling pathways in bone, skeletal muscle, and bone-muscle crosstalk through spatial transcriptomics in a young male mouse.

Bone and skeletal muscle are essential components of musculoskeletal system, enabling movement, load-bearing, and systemic homeostasis. These tissues communicate through dynamic bone-muscle crosstalk mediated by cytokines, growth factors, and extracellular-matrix (ECM) proteins. The spatial organization of these mediators is critical for maintaining tissue integrity, and its disruption contributes to diseases, such as osteoporosis, sarcopenia, and metabolic syndrome. Despite this importance, spatial transcriptomics (ST) studies of bone-muscle interactions remain limited. Here, we applied 10x G

Reliability and Construct Validity of the Physician's Global Assessment of Lung Disease in Systemic Juvenile Idiopathic Arthritis-Associated Lung Disease.

OBJECTIVE: The physician global assessment of lung disease (PGALD) is a recently proposed disease activity measure for patients with systemic juvenile idiopathic arthritis-associated lung disease (SJIA-LD). This study evaluates the reliability and construct validity of the PGALD. METHODS: Fifty-seven pediatric rheumatologists and pulmonologists with experience caring for children with SJIA-LD were invited to rate 20 clinical vignettes using the PGALD, a 10-point Likert scale (0 = inactive SJIA-LD; 10 = highly active SJIA-LD). Raters were also asked to rate a subset of eight repeat vignettes.

Outcomes of patients with relapsed or refractory classic Hodgkin lymphoma after frontline brentuximab vedotin.

Frontline therapy for classic Hodgkin Lymphoma (cHL) incorporating brentuximab vedotin (BV) improves outcomes compared with traditional chemotherapy, but up to 20% of patients relapse and need salvage treatment. Prior retrospective studies examining salvage therapies are mostly limited to patients who received chemotherapy-based treatment in frontline without novel agents. We evaluated outcomes in 116 patients with relapsed/refractory (R/R) cHL who received brentuximab and anthracycline-containing frontline treatment. High risk factors at relapse or progression were common, including primary r

Clinical outcomes and spatial transcriptomic profiles of CD19/20 CAR-T therapy in relapsed or refractory B-cell non-Hodgkin's lymphoma.

BACKGROUND: Relapsed or refractory (R/R) B-cell non-Hodgkin's lymphoma (B-NHL) remains a major therapeutic challenge despite CD19-directed chimeric antigen receptor (CAR) T-cell therapies, with treatment failure often driven by antigen escape and limited CAR-T persistence. Dual targeting of CD19 and CD20 may mitigate antigen loss. We conducted an expanded phase I/II study of bispecific CD19/20 CAR-T cells and incorporated spatially resolved single-cell transcriptomics to evaluate tumor-intrinsic and microenvironmental factors of clinical response. METHODS: Patients with R/R B-NHL received CD1

Thymic impairment and low IL-7 levels contribute to deficient immune recovery in relapsing visceral leishmaniasis.

Visceral leishmaniasis (VL) is a severe disease caused by Leishmania infantum in Brazil. Relapses have increased among immunocompetent patients; however, the underlying factors remain unclear. Previous studies have linked persistently low CD4+ T cell counts to VL relapses, suggesting that impaired immune reconstitution may favor recurrence. Here, we investigated whether immune activation and thymic output influence immune recovery in VL and shape the TCRVβ repertoire, predisposing patients to relapse. Patients with VL, followed for 12 months, were stratified according to relapse (R-VL,n = 5) o

Enhanced salivary correlation between Streptococcus mutans and herpes simplex virus-1 in oral squamous cell carcinoma: a case-control study.

BACKGROUND: Streptococcus mutans modulates local immune responses in the oral mucosa and has been implicated in oral squamous cell carcinoma (OSCC) pathogenesis. Emerging evidence suggests that certain oral bacteria can influence viral infections through immune modulation. This case-control study aimed to determine whether the correlation between S. mutans and herpes simplex virus type 1 (HSV-1) differs between OSCC patients and non-tumor controls, thereby elucidating potential bacterial-viral association in the tumor microenvironment. METHODS: Unstimulated whole saliva samples were collected

Integration of Metabolomic and Brain Imaging Data Highlights Pleiotropy Among Posttraumatic Stress Disorder, Glycoprotein Acetyls, and Pallidum Structure.

BACKGROUND: The development of posttraumatic stress disorder (PTSD) is attributable to the interplay between exposure to severe traumatic events, environmental factors, and biological characteristics. Blood and brain imaging markers have been associated with PTSD. However, to our knowledge, no study has systematically investigated the genetic relationship between PTSD, metabolic biomarkers, and brainwide imaging. METHODS: We integrated genome-wide data informative of PTSD, 233 metabolic biomarkers, and 3935 brain imaging-derived phenotypes (IDPs). Pleiotropy was assessed by applying global an

Preoperative Risk Stratification Using Radiographic Measures of Central Obesity.

STUDY DESIGN: Retrospective cohort study. OBJECTIVE: To evaluate whether abdominal circumference (AC) and abdominal-to-pelvic circumference (ACPC) ratio, derived from CT imaging, offer improved prediction of cardiometabolic comorbidities compared with body mass index (BMI) in orthopedic patients. BACKGROUND: BMI is a widely used measure for assessing obesity-related risk but lacks sensitivity to fat distribution and body composition. In surgery, where central adiposity contributes to surgical complexity and metabolic risk, BMI alone may be insufficient for accurate preoperative assessment.

Hearing Impairment Among Medicare Beneficiaries in the United States: Trends, Comorbidities, and Public Health Consequences.

BACKGROUND: Hearing loss is a significant public health issue in the United States, affecting an estimated 72.9 million people, or 22% of the population. Despite its prevalence and clinical impact, insurance coverage for hearing-related interventions remains inconsistent. Given these factors, particularly in the Medicare population, examining trends in hearing loss, co-occurring conditions, and utilization of audiologic services is essential to understand the burden of hearing impairment and identify gaps in care. OBJECTIVE: To examine 10-year trends in hearing loss and cochlear implant use,

Once bitten, twice shy: A modeling framework for incorporating heterogeneous mosquito biting into transmission models.

The risk and intensity of mosquito-borne disease outbreaks are tightly linked to the frequency at which mosquitoes feed on blood, also known as the biting rate. However, standard models of mosquito-borne disease transmission inherently assume that mosquitoes bite only once per reproductive cycle-an assumption commonly violated in nature. Drivers of multiple biting, such as host defensive behaviors or climate factors, also affect the mosquito gonotrophic cycle duration (GCD), a quantity customarily used to estimate the biting rate. Here, we present a novel framework for incorporating more compl
