Protected endogenous retroviral sequences copurify with infectivity in experimental Creutzfeldt-Jakob disease.

Scrapie and Creutzfeldt-Jakob disease (CJD) are caused by infectious agents that are defined phenomenologically. No agent-specific molecules or particles have been identified. Biological properties, such as exponential agent replication and strain variation, as well as physical characteristics of infectivity indicate a protected viral structure. A host membrane glycoprotein of 34 kDa ("prion" protein) that aggregates at end stages of disease is clearly important in pathology and susceptibility to infection, but has no demonstrable infectivity in any purified or recombinant form. Thus a charact

LPS and IL-4 Stimulation Favor Murine Norovirus Infection of B Cells.

Noroviruses are single-stranded positive-polarity RNA viruses classified in the family Caliciviridae. Human noroviruses (HuNoVs) are a leading cause of acute viral gastroenteritis worldwide. Despite their ability to infect various epithelial and nonepithelial cell types, establishing robust in vitro culture systems for HuNoVs remains challenging. As a result, murine norovirus 1 (MNV-1) has become a widely used model for investigating norovirus biology and pathogenesis, due to its ability to replicate efficiently in primary dendritic cells and macrophages. Although different B-cell lines are su

Tissue-specific transcriptomic responses and skeletal-muscle ultrastructural observations in Semah mahseer (Tor douronensis) under acute thermal and hypoxic conditions.

Semah mahseer (Tor douronensis) is a high-value freshwater cyprinid native to Southeast Asia. This study characterized acute transcriptomic responses in liver and white skeletal muscle following 6 h exposure to hyperthermia (35 °C), hypoxia (dissolved oxygen 2 mg/L), or their combination. Thirty-two RNA-seq libraries yielded approximately 48,504 annotated unigenes. Relative to the control, hyperthermia produced 7776 differentially expressed genes (DEGs) in liver and 1121 in muscle, hypoxia produced 2451 and 318 DEGs, and combined exposure produced 4952 and 717 DEGs, respectively. The response

PrfA, the master virulence regulator of Listeria monocytogenes: structural-functional insights, regulatory architecture, and antivirulence strategies.

Listeria monocytogenes causes listeriosis, a foodborne illness with case fatality rates of 20-30%. This bacterium tolerates a broad range of environments and can invade and replicate within human cells, properties that depend critically on PrfA (Positive Regulatory Factor A), a CRP/FNR-family transcriptional regulator. PrfA has two functional domains: an N-terminal β-barrel that binds ligands and a C-terminal helix-turn-helix motif that contacts PrfA-box sequences in target promoters. Activation is triggered by host-derived glutathione, which induces conformational changes permitting transcrip

Role of GPCR Signaling in Anthracycline-Induced Cardiotoxicity.

Anthracyclines are a class of chemotherapeutics commonly used to treat a range of cancers. Despite success in improving cancer survival rates, anthracyclines have dose-limiting cardiotoxicity that prevents more widespread clinical utility. Currently, the therapeutic options for these patients are limited to the iron-chelating agent dexrazoxane, the only FDA-approved drug for anthracycline cardiotoxicity. However, the clinical use of dexrazoxane has failed to replicate expectations from preclinical studies. A limited list of GPCRs have been identified as pathogenic in anthracycline-induced card

Mutations in the small nuclear RNA gene RNU2-2 cause a severe neurodevelopmental disorder with prominent epilepsy.

The major spliceosome includes five small nuclear RNA (snRNAs), U1, U2, U4, U5 and U6, each of which is encoded by multiple genes. We recently showed that mutations in RNU4-2, the gene that encodes the U4-2 snRNA, cause one of the most prevalent monogenic neurodevelopmental disorders. Here, we report that recurrent germline mutations in RNU2-2 (previously known as pseudogene RNU2-2P), a 191-bp gene that encodes the U2-2 snRNA, are responsible for a related disorder. By genetic association, we identified recurrent de novo single-nucleotide mutations at nucleotide positions 4 and 35 of RNU2-2 in

Multi-zonal liver organoids from human pluripotent stem cells.

Distinct hepatocyte subpopulations are spatially segregated along the portal-central axis and are critical to understanding metabolic homeostasis and injury in the liver1. Although several bioactive molecules, including ascorbate and bilirubin, have been described as having a role in directing zonal fates, zonal liver architecture has not yet been replicated in vitro2,3. Here, to evaluate hepatic zonal polarity, we developed a self-assembling zone-specific liver organoid by co-culturing ascorbate- and bilirubin-enriched hepatic progenitors derived from human induced pluripotent stem cells. Single-cell multi-omics characterize colorectal tumors, adjacent healthy tissue and matched (tumor) organoids identifying CRC-unique features.

Colorectal cancer (CRC) arises in the colorectal tissue driven by genetic disorder or the accumulation of somatic mutations, leading to abnormal epithelial cell growth. In this study, we employed single-nucleus multi-omics analysis, including single-nucleus RNA-seq and single-nucleus ATAC-seq, on over 100,000 high-quality nuclei to investigate the molecular landscape of both primary tissue and patient-derived organoids (PDOs). Our analysis showed that normal PDOs (N-PDOs) derived from tissue adjacent to tumors replicate the cellular composition and differentiation trajectory of colorectal cryp

An Organ-on-Chip Platform for Strain-Controlled, Tissue-Specific Compression of Cartilage and Mineralized Osteochondral Interface to Study Mechanical Overloading in Osteoarthritis.

Altered joint loading due to articular malalignment, instability, or trauma is an important risk factor for osteoarthritis (OA), the most prevalent musculoskeletal disease worldwide. However, the molecular links between aberrant mechanics and OA initiation/progression remain unclear due to the lack of human models capturing the interplay of joint tissues in a mechanically active environment. Replicating the strain gradient across the osteochondral interface remains an unmet challenge. Here, an OsteoChondral Unit (OCU)-on-Chip platform is engineered and functionally validated where composite hy

Cervical extracellular matrix hydrogel optimizes tumor heterogeneity of cervical squamous cell carcinoma organoids.

Cervical cancer, primarily squamous cell carcinoma, is the most prevalent gynecologic malignancy. Organoids can mimic tumor development in vitro, but current Matrigel inaccurately replicates the tissue-specific microenvironment. This limitation compromises the accurate representation of tumor heterogeneity. We collected para-cancerous cervical tissues from patients diagnosed with cervical squamous cell carcinoma (CSCC) and prepared uterine cervix extracellular matrix (UCEM) hydrogels. Proteomic analysis of UCEM identified several tissue-specific signaling pathways including human papillomaviru

Loss of Mfn1 but not Mfn2 enhances adipogenesis.

OBJECTIVE: A biallelic missense mutation in mitofusin 2 (MFN2) causes multiple symmetric lipomatosis and partial lipodystrophy, implicating disruption of mitochondrial fusion or interaction with other organelles in adipocyte differentiation, growth and/or survival. In this study, we aimed to document the impact of loss of mitofusin 1 (Mfn1) or 2 (Mfn2) on adipogenesis in cultured cells. METHODS: We characterised adipocyte differentiation of wildtype (WT), Mfn1-/- and Mfn2-/- mouse embryonic fibroblasts (MEFs) and 3T3-L1 preadipocytes in which Mfn1 or 2 levels were reduced using siRNA. Hunting Drosophila viruses from wild populations: A novel isolation approach and characterisation of viruses.

Metagenomic studies have demonstrated that viruses are extremely diverse and abundant in insects, but the difficulty of isolating them means little is known about the biology of these newly discovered viruses. To overcome this challenge in Drosophila, we created a cell line that was more permissive to infection and detected novel viruses by the presence of double-stranded RNA. We demonstrate the utility of these tools by isolating La Jolla virus (LJV) and Newfield virus (NFV) from several wild Drosophila populations. These viruses have different potential host ranges, with distinct abilities t

Evaluation of Broad Anti-Coronavirus Activity of Autophagy-Related Compounds Using Human Airway Organoids.

To deal with the broad spectrum of coronaviruses, including severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), that threaten human health, it is essential to not only drugs develop that target viral proteins but also consider drugs that target host proteins/cellular processes to protect them from being hijacked for viral infection and replication. To this end, it has been reported that autophagy is deeply involved in coronavirus infection. In this study, we used airway organoids to screen a chemical library of autophagic modulators to identify compounds that could potentially be use

Key role of exportin 6 in exosome-mediated viral transmission from insect vectors to plants.

Exosomes play a key role in virus exocytosis and transmission. The exportin family is usually responsible for cargo nucleocytoplasmic trafficking, and they are frequently found in exosomes. However, the function of exportins sorted in exosomes remains unknown. Here, we successfully isolated "cup holder"-like exosomes from the saliva of ∼30,000 small brown planthoppers, which are vectors of rice stripe virus (RSV). RSV virions were packed in comparatively large exosomes. Four viral genomic RNAs at a certain ratio were identified in the saliva exosomes. The virions contained in the saliva exosom

Influenza D Virus: A Review and Update of Its Role in Bovine Respiratory Syndrome.

Bovine respiratory disease (BRD) is one of the most prevalent, deadly, and costly diseases in young cattle. BRD has been recognized as a multifactorial disease caused mainly by viruses (bovine herpesvirus, BVDV, parainfluenza-3 virus, respiratory syncytial virus, and bovine coronavirus) and bacteria (Mycoplasma bovis, Pasteurella multocida, Mannheimia haemolytica and Histophilus somni). However, other microorganisms have been recognized to cause BRD. Influenza D virus (IDV) is a novel RNA pathogen belonging to the family Orthomyxoviridae, first discovered in 2011. It is distributed worldwide i

Analysis and pharmacological modulation of senescence in human epithelial stem cells.

Human epithelial stem cells (ESCs) are characterized by long-term regenerative properties, much dependent on the tissue of origin and varying during their lifespan. We analysed such variables in cultures of ESCs isolated from the skin, conjunctiva, limbus and oral mucosa of healthy donors and patients affected by ectrodactyly-ectodermal dysplasia-clefting syndrome, a rare genetic disorder caused by mutations in the p63 gene. We cultured cells until exhaustion in the presence or in the absence of DAPT (γ-secretase inhibitor; N-[N-(3, 5-difluorophenacetyl)-L-alanyl]-S-phenylglycine T-butyl ester

A sex-specific evolutionary interaction between ADCY9 and CETP.

Pharmacogenomic studies have revealed associations between rs1967309 in the adenylyl cyclase type 9 (ADCY9) gene and clinical responses to the cholesteryl ester transfer protein (CETP) modulator dalcetrapib, however, the mechanism behind this interaction is still unknown. Here, we characterized selective signals at the locus associated with the pharmacogenomic response in human populations and we show that rs1967309 region exhibits signatures of positive selection in several human populations. Furthermore, we identified a variant in CETP, rs158477, which is in long-range linkage disequilibrium

Pathogenicity of novel goose-origin astrovirus causing gout in goslings.

BACKGROUND: A novel goose-origin astrovirus (GoAstV) has broken out across China in recent years, causing gout in goslings with a mortality rate of around 50%. However, our understanding of the dynamic distribution, tissue tropism and pathogenesis of GoAstV is incomplete. In order to assess its pathogenicity, one-day-old goslings were inoculated separately with GoAstV via oral and subcutaneous injection routes. RESULTS: Clinical symptoms, gross and microscopic lesions, blood biochemical parameters and viral loads were detected and recorded for 20 days after infection. Typical gout was observe

Effects of atmospheric ammonia on the production performance, serum biochemical indices, and liver RNA-seq data of laying ducks.

1. Ammonia is a toxic gas and an air pollutant of great concern. The effects of ammonia-induced damage in ducks are rarely reported. 2. A total of 176 laying ducks were randomly allocated to one of two groups with four replicates and were exposed to ammonia at 5 ± 5 ppm (low ammonia, LA) or 75 ± 5 ppm (high ammonia, HA), respectively, for 30 days. 3. The results showed that atmospheric ammonia exposure significantly decreased body weight and laying performance (P < 0.05). It significantly changed serum biochemical indices by increasing alanine aminotransferase and aspartate aminotransferase (P
