Freeze-fracture of the normal and pathological megakaryocyte lineage in chronic megakaryocytic-granulocytic myelosis.

Freeze-fracture and thin sections were performed on human bone marrow of chronic megakaryocytic-granulocytic myelosis (CMGM) to study the three-dimensional fine structure and maturation of normal and atypical megakaryocytes and thrombocytes. In the many normally maturing megakaryocytes the development of the demarcation membrane system (DMS) was best investigated by comparison of thin sections with freeze-fracture replicas. The DMS shows no connections with the Golgi apparatus or rough-surfaced endoplasmic reticulum, but originates from tubular infoldings of the plasma membrane. These infoldin

Left atrial ultrastructure in mitral valvular disease.

Light microscopic and ultrastructural observations were made on left atrial tissues obtained from 14 patients at the time of operation for correction of mitral valvular disease. Cardiac muscle cells varied in size but most frequently were hypertrophied. In fibrotic areas, present in all left atria, the muscle cells tended to be isolated from adjacent cells and exhibited degenerative changes of varying severity. These changes consisted or proliferation of Z-band material and cytoskeletal filaments, myofibrillar loss, proliferation of elements of free and extended junctional sarcoplasmic reticul

Ultrastructural changes in tension zones of rat molar periodontium incident to orthodontic tooth movement.

The purpose of this study was, by introducing transmission electron microscopy, to clarify fine details of tissue reactions in the periodontal ligament on the tension side. A preliminary investigation was undertaken on nine rats in which experimental tooth movement lasted 2 days, 14 days, and 28 days. It was hoped that these new observations might help in the planning and directing of further studies. A marked increase of interstitial spaces between the formed ligament structures of the periodontal ligament was characteristic of the initial phases of tension. The cells and the endoplasmic reti

Brain ultrastructure in Reye's syndrome.

Cerebral biopsies were obtained for electron microscopy 48 and 72 hours after the onset of encephalopathy from a child with severe Reye's syndrome. Gravely ill at the time of craniectomy to relieve cerebral hypertension, the child survived and recovered good brain function; therefore, the biopsy findings appear to reflect the organelle pathology of the brain at a severe yet reversible stage in the disease process. The cardinal ultrastructural changes in the brain in Reye's syndrome are astrocyte swelling and partial deglycogenation, myelin bleb formation and universal injury of neuron mitochon

Myonecrosis induced by rattlesnake venom. An electron microscopic study.

The myonecrotic effect of rattlesnake (Crotalus viridis viridis) venom on mouse skeletal muscle was studied. The biceps femoris muscle was examined with the electron microscope after one-fourth the LD(50) of the crude venom was injected into the gracilis and semimembranosus muscles. Focal areas of myonecrosis were abundant. Injured fibers contained dilated sarcoplasmic reticulum, disoriented, coagulated myofilamentous components and condensed, rounded and enlarged mitochondria. The external lamina and sarcolemma remained intact in many fibers. Hemorrhage was apparent in the endomysial connecti

Oxysophocarpine attenuates acute liver injury by targeting FBP1 to suppress ferroptosis and enhance cellular resilience via the PPARα-GPX4 axis.

Acute liver injury (ALI) is a life-threatening clinical condition with limited therapeutic options. Through network pharmacology screening, we identified oxysophocarpine (OSC), a natural compound derived from traditional Chinese medicine, as a potent reactive oxygen species-scavenging agent. In LPS/D-GalN-induced ALI models, OSC administration dose-dependently improved survival, attenuated histopathological damage, reduced serum transaminase levels and inflammatory cytokines, and enhanced antioxidant capacity. Notably, we report for the first time that OSC directly bound to fructose-1,6-bispho

Mitochondrial calcium depletion in Darier disease is rescued by dantrolene sodium partly via MCU.

Darier disease (DD), caused by ATP2A2 mutations, disrupts epidermal adhesion via endoplasmic reticulum (ER) calcium defects. While ER calcium depletion is documented, its impact on mitochondrial calcium homeostasis remains unknown. This study investigates mitochondrial calcium downregulation in DD and evaluates repurposing the ryanodine receptor (RyR) antagonist dantrolene to restore mitochondrial function. Using primary, immortalised DD and ATP2A2 knockout keratinocytes as well as patient fibroblasts, we measured mitochondrial morphology and calcium level. DD models showed fragmented mitochon

Vaccinia-related kinase 2 inhibition elicits vulnerability of glutathione metabolism in pancreatic cancer.

Metabolic reprogramming has garnered significant attention in recent years due to its therapeutic potential in cancer treatment. However, identifying responsive tumor subpopulations remains a major obstacle in developing metabolism-targeted therapies, as metabolic vulnerabilities vary among cancers with different oncogene expression profiles. Therefore, elucidating the association between oncogene expression and metabolic characteristics could enable more precise metabolic interventions in clinical settings. Using pharmacological approaches, we demonstrate that VRK2-deficient pancreatic cancer

Targeting DGAT1 reprograms lipid landscape and restores CD8⁺ T cell immunity in pancreatic cancer.

Lipid accumulation is a hallmark of the pancreatic ductal adenocarcinoma (PDAC) tumor microenvironment, yet effective strategies to reprogram this lipid-rich niche and restore anti-tumor immunity remain limited. Here, we show that diacylglycerol O-acyltransferase 1 (DGAT1) as a tumor-intrinsic metabolic checkpoint that promotes immune evasion. DGAT1 inhibition rewires tumor lipid metabolism by promoting increased fatty acid uptake and redistribution, thereby depleting extracellular free fatty acids that impair CD8⁺ T cell function. Mechanistically, decreased palmitate availability alleviates e

Structure and function of TM6SF1 reveals role in mTORC1 signaling.

The transmembrane 6 superfamily (TM6SF) comprises two members: TM6SF1, a ubiquitously expressed lysosomal membrane protein of unknown function, and TM6SF2, an endoplasmic reticulum protein required for bulk lipidation of Apolipoprotein B-containing lipoproteins. Here, we used cryo-electron microscopy (cryo-EM) to determine the structure of human TM6SF1 at 2.9-Å resolution. TM6SF1 forms a polytopic homodimer, with each protomer comprising 10 transmembrane helices (TMs). TMs 1-6 form a pocket that accommodates a cholesterol molecule. Cell-based assays revealed that loss of TM6SF1 perturbs mTORC1

Cellular retinoic acid-binding protein 1, CRABP1, in thyroid gland aging.

Aging differentially affects disease risks of various organs1. The endocrine organs undergo significant changes along aging. Clinical reports showed increased prevalence of thyroid disorders with age, especially hypothyroidism2. Primary hypothyroidism includes congenital, autoimmune, and iatrogenic causes, which can occur throughout the lifespan. But those with unknown aetiology increase dramatically (~5 fold) in the elderly (≥75 years old)3,4. In thyrocytes, accumulated thyroglobulin requires efficient protein folding in the endoplasmic reticulum (ER)5,6. When ER experiences protein folding d

Heterogeneity of Cardiomyocyte Lipid Droplets and Their Pathophysiological Implications in Cardiovascular Diseases.

The adult heart relies on fatty acid β-oxidation for energy, and cardiomyocyte lipid droplets (CLDs) are central to myocardial lipid homeostasis. CLDs are now recognized to protect cardiomyocytes against lipotoxicity, endoplasmic reticulum (ER) stress, oxidative stress, and nutrient deprivation. Moreover, accumulating evidence suggests that CLDs within the same cardiomyocyte are not uniform but may exhibit heterogeneity in size, subcellular localization, protein composition, and metabolism. Here, we propose a conceptual framework that classifies CLD heterogeneity along three dimensions: morpho

Therapeutic Potential of Gene Modified MSC-Exos in Insulin Resistance: Mitochondrial Crosstalk and miRNA Regulation.

Insulin resistance (IR) is the key driver of type 2 diabetes mellitus, metabolic dysfunction-associated steatotic liver disease, and cardiometabolic disorders. Mesenchymal stem cell-derived exosomes (MSC-Exos) as a potent cell-free alternative can deliver miRNAs, proteins, and mitochondrial regulators to reactivate the IRS1/PI3K/Akt/GLUT4 axis. Beyond restoring insulin signalling, these vesicles can protect β-cells, alleviate endoplasmic reticulum stress, rescue mitochondrial mitophagy and respiration. The genetic engineering strategies including improving the expression of miR-21, miR-3075, o

Pathogenic Modulation of Organelle Crosstalk in Helicobacter pylori-Associated Neurodegeneration.

Neurological disorders are increasingly linked to dysfunction of key cellular organelles, including mitochondria, endoplasmic reticulum (ER), lysosomes, endosomes, and peroxisomes. These organelles coordinate essential neuronal processes via tightly regulated crosstalk. Disruption in one organelle can propagate dysfunction across others, amplifying neurodegenerative cascades. Emerging evidence suggests that neurological diseases can result not only from disturbances in brain homeostasis but also from imbalances in gut homeostasis, highlighting the significant role of the gut-brain axis in main

Butyrate ameliorates ulcerative colitis through targeting STING-dependent ER stress signaling and limiting CD4+ TRM T cells accumulation.

This study aimed to investigate the role of butyrate in regulating STING-induced endoplasmic reticulum stress (ERS) and CD4+ tissue-resident memory (TRM) T cells responses during the progression of ulcerative colitis (UC). Our results demonstrated that butyrate significantly alleviated dextran sulfate sodium (DSS)-induced colitis, as evidenced by restored intestinal epithelial architecture, reduced inflammatory cytokine, and decreased CD4+ TRM T cells. These protective effects were likely mediated through modulation of the STING-ERS pathway. Using a CT26 cell model, we further confirmed that S

MAO-B inhibition by selegiline blunts cardiac functions improved by high-fat diet: Role of inflammation, apoptosis, and calcium-handling.

BACKGROUND: Obesity is a major risk factor for the development of cardiovascular disease. However, recent research shows that moderate obesity reduces the risk of developing cardiovascular disease. We evidenced before that MAO-B inhibitor selegiline reduced visceral adiposity. AIM: Therefore, our aim was to investigate cardiac effects of selegiline in moderate obesity in rats treated with a high-fat diet (HFD). KEY FINDINGS: We demonstrated that HFD improved cardiac contractility parameters, which were reversed by selegiline. Enhanced contractility might be attributed to an increased sarcopl

Dihydropyrazine activates the CAMKK2-AMPK-ULK1 signaling pathway in human hepatoma HepG2 cells.

Dihydropyrazines (DHPs) are glycation intermediates produced by nonenzymatic glycation reactions in vivo and in foods. We previously reported that 3-hydro-2,2,5,6-tetramethylpyrazine (DHP-3) activates endoplasmic reticulum (ER) stress and inhibits autophagy, although its effect on autophagy initiation remained unclear. In this study, we examined the effect of DHP-3 on the autophagy initiation pathway in HepG2 cells. DHP-3 exposure resulted in activation of UNC-51-like kinase 1 (ULK1), the catalytic subunit of the ULK complex essential for autophagy initiation. Notably, phosphorylation of AMP-a

Post-translational modifications in the pathophysiological process of metabolic dysfunction‑associated steatotic liver disease.

In recent years, the prevalence of metabolic dysfunction‑associated steatotic liver disease (MASLD), which was called non-alcoholic fatty liver disease (NAFLD), has been progressively increasing in populations. The progression of MASLD encompasses a spectrum from simple steatosis to metabolic dysfunction-associated steatohepatitis (MASH), and ultimately to cirrhosis or even hepatocellular carcinoma. During the early stages of the disease, lipid accumulation and endoplasmic reticulum stress may lead to abnormalities in hepatic DNA expression, protein synthesis, and post-translational modificati

Molecular aspects of cytoprotection by Optineurin during stress and disease.

Optineurin/OPTN is an adapter protein that plays a crucial role in mediating many cellular functions, including autophagy, vesicle trafficking, and various signalling pathways. Mutations of OPTN are linked with neurodegenerative disorders, glaucoma, and amyotrophic lateral sclerosis (ALS). Recent work has shown that OPTN provides cytoprotection from many types of stress, including oxidative stress, endoplasmic reticulum stress, protein homeostasis stress, tumour necrosis factor α, and microbial infection. Here, we discuss the mechanisms involved in cytoprotective functions of OPTN, which possi
