Benzodiazepines are among the most widely prescribed psychotropic agents, exhibiting anxiolytic, hypnotic, anticonvulsant, and myorelaxant properties primarily mediated through positive allosteric modulation of type A γ-aminobutyric acid receptors (GABAARs)-the principal inhibitory receptors in the mammalian brain. However, prolonged benzodiazepine administration frequently results in the development of tolerance, a phenomenon characterized by reduced pharmacological efficacy and the need for escalating doses to sustain therapeutic benefit. Accumulating evidence indicates that benzodiazepine t

Ventral tegmental area (VTA) dopamine (DA) neurons are highly implicated in reward learning, motivated behaviors, and substance use disorders. DA neurons in the VTA are traditionally characterized by expression of genes involved in DA synthesis, release, or reuptake, such as tyrosine hydroxylase (encoded by the Th gene), which is the rate-limiting step in DA synthesis. However, recent transcriptomic studies have revealed substantial cellular heterogeneity within the VTA, including multiple subtypes of VTA DA neurons. Using single nucleus RNA sequencing, we previously identified two transcripti

This narrative review provides evidence regarding the impact of trace amine-associated receptor 1 (TAAR1) agonists on rodent models of substance use disorders (SUDs). We delineated the fundamental aspects of the pharmacodynamics and pharmacokinetics of TAAR1 agonists and collected in vivo studies that investigated TAAR1 activation in behavioral sensitization, conditioned behaviors, or self-administration models. TAAR1 agonists have demonstrated efficacy in suppressing addiction-relevant behaviors associated with cocaine. Their effectiveness is largely consistent across intrinsic activity level

BACKGROUND: Neuropathic pain is a major source of disability and distress with few pharmacological options for treatment. Opioid drugs can be effective, but high doses are needed, leading to unwanted effects. BMS-986122 is a positive allosteric modulator of the mu opioid receptor that potentiates acute opioid antinociception without increasing opioid-induced constipation, reward, or respiratory depression. Therefore, we asked if BMS-986122 could increase the effects of low-dose opioid analgesics in chronic neuropathic pain.
METHODS: We employed the spared nerve injury and tibial neuroma model

Chronic opioid use facilitates changes in brain reward and affective systems. Cessation of opioid use induces negative affective states characterized by increased anxiety- and despair-like behaviors. In this study, the potential role of transient receptor potential ankyrin 1 channel (TRPA1) was assessed in morphine-induced dependence and negative emotional behaviors. Male and female C57BL/6J mice were exposed to escalating doses of morphine to induce dependence. Naloxone was administered to precipitate withdrawal symptoms. TRPA1 antagonism with A967079 treatment reduced withdrawal syndrome in

For decades, dietary guidance has been a signal lost in the noise of our modern food environment, resulting in stagnant indices of diet quality despite clear dietary recommendations. Emerging obesity medications (EOMs), including glucagon-like peptide-1-based therapies, appear to quiet-intrusive, cue-driven food noise and recalibrate reward responsivity to energy-dense foods. We propose that this pharmacological dampening of automatic motivation creates a transient "pharmacological window" in which the enduring elements of dietary guidance-emphasis on fruits, vegetables, legumes, whole grains,

BACKGROUND: Brain entropy reflects the brain's functional complexity and encompasses both cognitive and affective processing. However, brain entropy remains to be systematically examined in patients with treatment-resistant depression (TRD).
METHODS: The study cohort comprised 48 adults with TRD and 38 healthy controls. All participants underwent resting-state functional magnetic resonance imaging. Sample entropy, an index of brain entropy, was calculated from blood oxygenation level-dependent time series for each of 166 regions of interest.
RESULTS: At an embedding dimension (m) of 1 and a

Second-generation (atypical) antipsychotics are central to the treatment of schizophrenia and related disorders but are frequently associated with significant metabolic adverse effects, particularly weight gain and obesity. These complications can impair treatment adherence and substantially increase the risk of cardiovascular and metabolic comorbidities. Accumulating evidence indicates that antipsychotic-induced metabolic dysregulation arises from complex interactions between central neurotransmitter systems and neuroendocrine regulators of appetite and energy balance. Among these, the orexig

Brain insulin resistance (BIR) has emerged as a critical link between metabolic dysfunction and cognitive and behavioral impairment. As the twin epidemics of metabolic and brain disorders accelerate globally, understanding the precise mechanisms of BIR has become a clinical and scientific priority. However, growing evidence indicates that BIR does not manifest uniformly across the central nervous system. Instead, insulin signaling dysfunction exhibit marked regional and circuit-specific heterogeneity across the hippocampus, prefrontal cortex, hypothalamus, amygdala, and mesolimbic reward pathw

The pedunculopontine nucleus (PPN) is a heterogeneous cluster within the dorsolateral pontomesencephalic tegmentum, functioning as a critical neuromodulatory hub. A key organizing principle is its rostrocaudal functional gradient rostral glutamatergic Chx10 + neurons mediate global motor arrest, while caudal Vglut2 + neurons drive locomotor initiation and maintenance. This model, emphasizing topographically organized microcircuits, provides a coherent framework for reconciling heterogeneous outcomes of PPN stimulation across experimental and clinical settings. BACKGROUND AND OBJECTIVE: Obsessive-compulsive disorder (OCD) and hoarding disorder often co-occur but differ in onset, course, symptoms, and treatment response. This study compared diagnosis-based OCD genetic liability with hoarding-symptom liability, rather than direct expression differences between diagnosed patient groups.
METHODS: Large-scale OCD and hoarding genome-wide association study (GWAS) summary statistics were integrated with brain expression quantitative trait loci (eQTL) reference panels using a transcriptome-wide association approach. Gene-set enrichment was assessed across s

Beyond the well-established roles of serotonin and calcitonin gene-related peptide (CGRP), the contribution of dopamine (DA) to migraine pathophysiology remains a subject of intense investigation. DA acts as a neuromodulator, functioning both as a homeostatic pain filter and a driver of premonitory and autonomic symptoms. This review provides a comprehensive synthesis of DA's involvement in migraine, proposing an integrated model that spans from molecular signaling to a systemic phenotype. A comprehensive search of the PubMed database (1976-2026) was performed. The analysis evaluates data acro

BACKGROUND AND OBJECTIVES: Rates of tobacco use disorder (TUD) are high among individuals with opioid use disorder (OUD), who consistently show lower response rates to evidence-based treatment approaches for TUD relative to the general population. This systematic review aims to provide a comprehensive and updated examination of the current evidence regarding TUD treatments for individuals with OUD. We first summarize potential mechanisms driving TUD and treatment nonresponse among those with OUD, given the likely roles of overlapping reward and withdrawal processes in co-occurrence and treatme

The mu opioid receptor system has long been recognized for its role in regulating ingestive behaviors and food palatability. However, beyond their robust expression in the brain, opioid receptors are also found throughout the body, including the stomach, intestines, and pancreas. Previous studies suggest that opioids may directly regulate glycemia through actions in the endocrine pancreas, but whether these metabolic effects are linked to their influence on ingestive behavior remains unclear. Here, we used metabolic, ingestive, and genetic approaches to determine how the mu opioid receptor (MO

Large language models have become central infrastructures of contemporary digital economies while raising persistent ethical concerns regarding linguistic inequality, opacity, data governance, and the concentration of technological power. Much of the current debate on AI ethics focuses on normative principles such as fairness, transparency, and accountability. While these principles remain essential, they often do not sufficiently explain why ethically problematic outcomes persist under competitive market conditions. This paper addresses that gap by applying Karl Homann's institutional economi

Fo Mentality is a culturally embedded psychological orientation rooted in the Chinese sociocultural context and prevalent among contemporary young adults. Although labeled with the term "Fo" (Buddha), the construct represents a secular psychological orientation rather than Buddhist beliefs or religious practices. Existing measures of Fo Mentality have been developed primarily in workplace contexts, and whether this conceptualization applies to college students, whose experiences differ in academic demands, evaluation systems, and developmental tasks, remains unclear. The present study aimed to

This article summarizes the key literature describing the effects of the pancreatic beta-cell hormone amylin on eating. One of the first described and best investigated effects of amylin on eating is its physiological effect to control meal size by inducing satiation. This effect is very rapid, short-lasting and probably directly reflects the meal-induced increase in circulating amylin levels. Evidence provided by many groups suggests that the effect of amylin on eating is directly mediated by humoral action in the central nervous system rather than by peripheral receptors. It is also clear th

Drug addiction is a chronic and potentially deadly disease that is considered a global health problem and describes the alteration of brain function by psychostimulant drugs through changes in the reward system. However, there is still no ideal strategy for the management of drug addiction. Previous studies have suggested that microglia are involved in events associated with neuroplasticity and memory, which are also related to drug addiction. Many studies have shown that psychoactive substances may act directly on immune cells, altering their function and inducing the production of various in

The neurobiology of chronic pain is complex and multifaceted, intertwining with the mesocorticolimbic system to regulate the behavioral and perceptional response to adverse stimuli. Specifically, the ventral tegmental area (VTA), the dopaminergic hub of the reward pathways located deep within the midbrain, is crucial for regulating the release of dopamine (DA) throughout the central nervous system (CNS). To better understand the nuances among chronic pain, VTA response, and therapeutics, implementing progressive approaches for mapping and visualizing the deep brain in real time during nocicept

Substance use disorders (SUDs) remain a major public health challenge, exacerbated by limited access to care, low treatment engagement, and the disruption of services during the COVID-19 pandemic. Mobile phone applications (MPAs) offer a scalable mechanism for delivering behavioral health interventions; however, existing apps suffer from low engagement, limited methodological rigor, and inflexibility for research and clinical adaptation. This article presents Senyo, a proof-of-concept virtual treatment and research platform designed to support individuals with SUD while enabling systematic eva
