Microvascular injury is believed to be mechanistically involved in radiation fibrosis, but direct molecular links between endothelial dysfunction and radiation fibrosis have not been established in vivo. We examined radiation-induced changes in endothelial thrombomodulin (TM) and protease-activated receptor-1 (PAR-1) in irradiated intestine, and their relationship to structural, cellular, and molecular aspects of radiation injury. Rat small intestine was locally exposed to fractionated X-radiation. Structural injury was assessed 24 hours and 2, 6, and 26 weeks after the last radiation fraction

Activin, a member of the transforming growth factor-beta superfamily, has been shown to be a critical regulator in exocrine and endocrine pancreas formation. The purpose of our study was to describe the ontogeny of activin B and its inhibitor, follistatin, in developing pancreas and to elucidate potential mechanisms for exocrine and endocrine lineage selection. Mouse embryonic pancreata were dissected at various ages (day 10 [E10.5] to birth [E18.5]), sectioned, and immunostained for activin B (one of two existing isomers, A and B), follistatin, insulin, and glucagon. In addition, reverse tran

OBJECTIVE: Cuproptosis, a newly discovered cell death mechanism, has been linked to the pathogenesis of multiple diseases. However, its role in diabetic kidney disease (DKD) remains unclear.
METHODS: By analyzing datasets GPL17586 and GPL571 from the GEO database and applying machine learning, cuproptosis-related marker genes associated with DKD were identified. The expression levels of these genes were examined in Mouse Podocyte Cell Line (MPC5) podocytes cultured in vitro and treated with high glucose (30 mM) for 24, 48, and 72 h to explore their roles in the onset and progression of DKD.

BACKGROUND: Pulmonary arteriovenous malformations (PAVMs) occur in over 25% of patients with the autosomal dominant disorder hereditary haemorrhagic telangiectasia (HHT). Mutations in two genes, endoglin and ALK-1, are known to cause HHT. Each encodes a protein expressed on vascular endothelial cells and involved in signalling by members of the transforming growth factor (TGF)-beta superfamily. To date, PAVMs have not been detected in ALK-1 families. There is evidence from a single HHT family without pulmonary involvement that a third HHT gene may exist. To establish the existence of a further

We performed the first in utero correction of a unilateral right coronal craniosynostosis using 70-day gestation fetal lambs. The craniosynostosis was created in eight fetuses by excising their right coronal sutures, and then placing demineralized bone powder, transforming growth factor-beta, and bone morphogenetic protein-2 into the defect. Twenty-one days later, after suture fusion had occurred, four of the eight sheep were treated with a 4 mm x 12 mm strip craniectomy to open the entire synostosed right coronal suture. The edges of the excision were wrapped with 100-microm-thick Gore-Tex (W

In vitro incubation of Listeria-immune spleen cells (LISC) with recombinant interleukin-2 (rIL-2) for at least 3 days increased their ability to transfer antilisteria resistance to recipient mice. This effect was blocked by the in vitro addition of transforming growth factor beta 1. The level of protection afforded by the transfer of rIL-2-incubated LISC was further elevated by the in vivo administration of rIL-2 at a dose that by itself did not significantly increase antilisteria resistance. The antilisteria resistance of recipient mice remained elevated for approximately 7 days and then rapi

Immune modulatory vaccines (IMVs) are an emerging class of immunotherapies designed to expand anti-regulatory T cells (anti-Tregs) that selectively target immunosuppressive elements within the tumor microenvironment (TME). Unlike conventional cancer vaccines aimed at tumor-associated antigens on malignant cells, IMVs target tumor microenvironment antigens (TMAs), such as indoleamine 2,3-dioxygenase (IDO), PD-L1, arginase-1 (ARG1), and transforming growth factor-β (TGF-β), which are expressed by malignant, myeloid, regulatory, endothelial, and stromal populations. IMVs elicit both CD8⁺ and CD4⁺

Accumulation of adipocytes within the bone marrow is a frequently observed during aging. However, the molecular mechanisms underlying aberrant adipocyte differentiation in aged bone marrow remain largely unclear. In this study, we identified Ly-1 antibody reactive clone (Lyar) as an interacting partner of TGF-β activated kinase 1 (Tak1), a key molecule of non-canonical TGF-β signaling, through a proteomics approach, and demonstrated its involvement in the regulation of aging-related enhancement of adipogenesis. Lyar was not only implicated in the regulation of BMMSC proliferation but also may

Liver fibrosis, marked by excessive ECM deposition, can progress to cirrhosis and hepatocellular carcinoma, yet effective treatments are lacking. Since hepatic stellate cell (HSC) activation is central to fibrosis, inhibiting it is a key therapeutic strategy. Vitamin D receptor (VDR) activation can suppress HSC activation by inhibiting the TGFβ/SMAD3 pathway, making it a promising target. However, steroidal VDR agonists' clinical use is limited by hypercalcemia caused by upregulation of calcium metabolism genes. To overcome this, we designed novel steroidal VDR modulators by modifying the side

Airway remodeling is a process that occurs in chronic obstructive diseases, such as asthma and COPD. It is associated with adverse changes in the structure and function of the airways. An increasing amount of literature points to the potential protective effects of vitamin D and caffeine against inflammation and fibrosis. The aim of the study is to evaluate the effect of calcitriol and caffeine on the expression of genes and proteins associated with airway remodeling. The Calu-3 cell line was treated with TGF-β, calcitriol, and caffeine in different combinations. Subsequently, the expression o

INTRODUCTION: Sepsis is a major global health burden associated with high mortality and multiple organ dysfunction, among which liver injury is a key determinant of poor prognosis. However, effective therapeutic strategies for sepsis-associated liver injury (SALI) remain limited.
METHODS: In this study, we investigated the protective effects of MAGL-18c, a novel monoacylglycerol lipase (MAGL) inhibitor, on lipopolysaccharide (LPS)-induced SALI. Hepatic inflammation, apoptosis, mitochondrial function, and lipid metabolism were assessed using liquid chromatography-mass spectrometry (LC-MS), Wes

BACKGROUND: Platelet-rich plasma (PRP) shows promise in burn management, yet optimal platelet concentrations, timing, and standardized protocols remain unclear, leading to inconsistent clinical outcomes.
AIMS: To compare the therapeutic efficacy of PRP at different platelet concentrations in deep second-degree burns and to explore associated molecular mechanisms.
PATIENTS/METHODS: Twenty-eight patients with small-area deep second-degree burns were randomly assigned to four groups: Group A (PRP 600-1000 × 109/L), Group B (normal saline control), Group C (PRP 1000-1400 × 109/L), and Group D (P

BACKGROUND: Excessive microglial activation after subarachnoid hemorrhage (SAH) contributes to early brain injury (EBI) and poor clinical outcomes. This study explores the role of complement C3a receptor 1 (C3aR1) in microglial activation after SAH and evaluates Vorinostat as a potential therapeutic agent.
METHODS: Differentially expressed genes were identified from GEO datasets (GSE36791, GSE73378), followed by WGCNA and machine learning to pinpoint key SAH-related genes. Molecular docking and molecular dynamics simulation identified Vorinostat as a potential C3aR1-targeting compound. Functi

Obesity exacerbates rheumatoid arthritis (RA). However, the underlying mechanisms remain incompletely defined. Elucidating these mechanisms can help the identification of novel therapeutic targets. Herein, we used high-fat diet (HFD)-induced obese collagen-induced arthritis (CIA) mice to investigate these mechanisms. Immunohistochemistry revealed that obesity exacerbated joint inflammation and cartilage degradation. Next, integrated label-free quantitative proteomics and cytometry by time-of-flight (CyTOF) were used to characterize lymphocyte subsets. Proteomic profiling identified 26 differen

OBJECTIVE: Anaemia is common in rheumatoid arthritis (RA), but the role of erythroid-lineage cells is unclear. We investigated the function of CD45+ erythroid progenitor cells (CD45+ EPCs) in RA.
METHODS: We analysed CD45+ EPC frequency in patients with RA and mice with collagen-induced arthritis (CIA). Transcriptomics, functional studies and mechanistic assays (Transwell and dual-luciferase reporter assays, chromatin immunoprecipitation followed by quantitative PCR) were used. Therapeutic potential was tested in RA synovial organoids and via splenectomy/adoptive transfer in mice with CIA.

Duchenne muscular dystrophy (DMD) is an incurable muscle-wasting disorder characterized by chronic membrane damage, inflammation, and progressive fibrosis. Fibrosis in DMD is driven by sustained TGFβ signaling, which promotes extracellular matrix (ECM) accumulation. We previously showed that SETDB1 sustains the TGFβ-induced fibrotic response in DMD myotubes. Here, we further show that SETDB1 modulates the TGFβ-induced secretome, particularly by regulating ECM-related proteins. Comparison of the basal secretome from DMD patient-derived myotubes and healthy controls revealed a distinct disease-s

Emerging evidence implicates intestinal barrier dysfunction and translocation of microbial factors as key drivers of post-myocardial infarction inflammation and cardiac remodeling, yet therapeutic strategies targeting the gut-heart axis remain underdeveloped. Here we demonstrate that ALY688, an adiponectin receptor agonist peptide, confers robust cardioprotection in rat myocardial ischemia-reperfusion (IR) injury through coordinated immunoregulatory programs in cardiac and intestinal tissues. ALY688 administration during ischemia or at reperfusion and subsequently daily for 28 days significant

BACKGROUND: Traditionally, dental implants require a healing period of 4 to 9 months for osseointegration, with longer recovery times considered when bone grafting is needed. This retrospective study evaluates the clinical efficacy of demineralized dentin matrix (DDM) combined with recombinant human bone morphogenetic protein-2 (rhBMP-2) during dental implant placement to expedite the osseointegration period for early loading.
METHODS: Thirty patients (17 male, 13 female; mean age 55.0 ± 8.8 years) requiring bone grafts due to implant fixture exposure (more than four threads; ≥ 3.2 mm) were i

The Anti-Müllerian hormone (AMH) is widely recognized for promoting Müllerian duct regression in higher vertebrates and regulating key reproductive functions like steroidogenesis, folliculogenesis, and Leydig cell development. In teleost fish, which lack Müllerian ducts, Amh primarily influences male reproductive functions, including sex determination, testis differentiation, and germ cell proliferation. In adult fish, Amh supports gonad development and spermatogenesis, but its role in teleost gonadal physiology remains largely underexplored. This study reveals a novel steroidogenic function i
