Cancer vaccines: comprehensive review.

BACKGROUND: There has been an increasing focus on the potential of cancer vaccines as a therapeutic oncological option, aiming to harness the immune system to recognize and eliminate malignant cells. Unlike traditional therapies, vaccines exploit tumour-associated antigens and neoantigens to generate durable, tumour-specific immune responses. METHODS: This review provides a comprehensive overview of current vaccine strategies, their clinical applications, and implications for surgical oncology. RESULTS: Clinical trials highlight promising results in high-mutational burden cancers such as mel

Vaccine-Associated Anterior Uveitis.

Vaccine-associated anterior uveitis (VAAU) is an uncommon ocular inflammatory phenotype reported after several vaccine platforms. It describes anterior segment inflammation occurring after vaccination when infectious, autoimmune, idiopathic, medication-related, and other possible causes have been reasonably excluded. The current literature indicates that post-vaccination uveitis is rare relative to the scale of immunization, although anterior uveitis is frequently reported among post-vaccination ocular inflammatory events. Evidence remains limited by reliance on case reports, case series, retr

Advances in Adjuvanted Rabies Vaccines.

Rabies is an acute and fatal zoonotic disease caused by the rabies virus, responsible for approximately 59,000 deaths worldwide each year. Once clinical symptoms manifest, the case fatality rate approaches 100%. Vaccination remains the only effective strategy for prevention and control. Currently, human rabies vaccines approved by regulatory authorities such as the U.S. Food and Drug Administration (FDA), and the China National Medical Products Administration (NMPA) are all inactivated, adjuvant-free formulations. These vaccines are associated with several limitations, including weak immunogen

The Lyme Disease Vaccine Paradox.

Lyme disease causes a significant financial and health burden. Recent advances in developing new Lyme disease vaccines have renewed optimism that vaccination may serve as an additional strategy to reduce this burden. Vaccination alone will not address the broader relevant public health challenges posed by ticks. Our experience with prior Lyme disease vaccines suggests that even an effective vaccine would not fully reduce the incidence of human disease. Importantly, vaccination against Borrelia genospecies would not affect tick abundance or exposure risk. It would also not mitigate the transmis

The recent advances in vaccine adjuvants.

Vaccine adjuvants, as key components in enhancing vaccine immunogenicity, play a vital role in modern vaccinology. This review systematically examines the historical evolution and mechanisms of vaccine adjuvants, with particular emphasis on innovative advancements in aluminum-based adjuvants, emulsion-based adjuvants, and nucleic acid adjuvants (e.g., CpG oligonucleotides). Specifically, aluminum adjuvants enhance immune responses through particle formation/antigen adsorption, inflammatory cascade activation, and T-cell stimulation. Emulsion adjuvants amplify immunogenicity via antigen depot e

Microorganism delivery of cancer vaccines.

Cancer immunotherapeutic vaccines offer a promising approach which can activate the immune system to target and kill cancer cells. Despite great progresses, challenges still exist in clinical practice, including immunosuppressive tumor microenvironment, tumor heterogeneity as well as low delivery efficiency. Microbe-based cancer vaccines, leveraging the abilities of bacteria and viruses to target hypoxic tumor regions/infect cancer cells as well as enhance the immunogenicity of tumor microenvironment, have attached prominent attention for the potential to revolutionize vaccine designing. Vaccinating patients with autoimmune diseases.

PURPOSE: Vaccine preventable diseases, especially respiratory infections, occur at higher frequency in patients with rheumatic diseases who are immunosuppressed. The focus of this review is to highlight vaccinations that patients with rheumatic diseases should optimally receive, with a focus on efficacy and safety of vaccines. MAIN FINDINGS: In general, vaccines are effective in reducing the burden of infection. However, due to underlying immunosuppression, their efficacy is most likely reduced compared to the general population. This is even true of the novel vaccines, mRNA and subunit vacci

Initiation of a coronavirus vaccine library.

Coronaviruses (CoVs) continue to pose a global health threat as they spill over from animal hosts to humans. The Coalition for Epidemic Preparedness Innovations (CEPI) is a global partnership focused on vaccine research and development. CEPI's CoV Vaccine Library initiative aims to generate knowledge and tools to accelerate vaccine development against emergent CoVs, prioritizing those with the highest assessed risk of spillover, vaccine feasibility, and phylogenetic coverage. Based on expert consultations and published data, we developed a prioritized list of 26 CoVs to serve as a foundation f

Nasal vaccines for respiratory infections.

Beginning with Edward Jenner's discovery of the smallpox vaccine, the ever-expanding repertoire of vaccines against pathogens has saved many lives. During the COVID-19 pandemic, a revolutionary mRNA injectable vaccine emerged that effectively controlled the severity of disease caused by SARS-CoV-2. This vaccine induced potent antigen-specific neutralizing serum IgG antibodies, but was limited in its ability to prevent viral invasion at the respiratory surfaces. Nasal vaccines have attracted attention as a potential strategy to combat respiratory infections and prepare for future pandemics. Inp

The History of Measles and Vaccine Development.

Measles is one of the oldest human diseases, but the fact that it was separate from smallpox, scarlatina and rubella was only disclosed a century ago. It is extremely contagious, as the infection may spread for up to 4 days before the first symptoms appear. One patient may infect 12-18 secondary cases, who then spread the infection further. Before the vaccine era, measles caused 2-3 million deaths a year, with remote populations facing chaotic consequences from epidemics. Safe and inexpensive live-virus vaccines, often combined with mumps and rubella components, have been available for 50 year

Visualising Uncertainty of Vaccine Data.

As healthcare data grows more complex, effectively conveying the inherent uncertainties in medical research and clinical decision-making is crucial to avoid misinterpretation. This study explores visualising the effectiveness and safety of the herpes zoster vaccine, focusing on conveying associated uncertainties to the general public. Two visualisation formats (static and interactive) were developed. A questionnaire-based evaluation assessed their effectiveness and impact on vaccination intentions. Both formats effectively conveyed data, with most participants accurately interpreting the infor

Corneal complications of common vaccinations.

PURPOSE OF REVIEW: To summarize and evaluate the current literature concerning corneal complications associated with common vaccinations, particularly keratitis and corneal graft rejection following vaccination against SARS-CoV-2, Varicella-Zoster, and Influenza. RECENT FINDINGS: Small case series and reports have documented keratitis/herpesvirus reactivation and corneal graft rejection following vaccination, prompting concern from ophthalmologists regarding vaccination timing and management of patients. Recent population-based studies have reported higher risks of herpesvirus keratitis and h

Tailoring biomaterials for vaccine delivery.

Biomaterials are substances that can be injected, implanted, or applied to the surface of tissues in biomedical applications and have the ability to interact with biological systems to initiate therapeutic responses. Biomaterial-based vaccine delivery systems possess robust packaging capabilities, enabling sustained and localized drug release at the target site. Throughout the vaccine delivery process, they can contribute to protecting, stabilizing, and guiding the immunogen while also serving as adjuvants to enhance vaccine efficacy. In this article, we provide a comprehensive review of the c

Vaccine value profile for schistosomiasis.

Schistosomiasis is caused by parasitic flatworms (Schistosoma). The disease in humans can be caused by seven different species of Schistosoma: S. mansoni, S. japonicum, S. haematobium, S. malayensis, S. mekongi, S. guineensis and S. intercalatum, as well as by hybrids between species, including livestock schistosome species. People are infected when exposed to infested water and the parasite larvae penetrate the skin. Poor and rural communities are typically the most affected, and the general population who lives in affected areas and is exposed to contaminated water is at risk. Areas with poo

Vaccine Strategies for Cryptococcus neoformans.

Cryptococcus neoformans infections are a major worldwide concern as current treatment strategies are becoming less effective in alleviating the infection. The most extreme and fatal cases are those of immunocompromised individuals. Clinical treatments for cryptococcosis are limited to a few classes of approved drugs, and due to a rise in drug resistance, these drugs are becoming less effective. Therefore, it is essential to develop innovative ways to control this infection. Vaccinations have emerged as a safe, viable, and cost-effective solution to treat a number of diseases over the years. Discovering vaccines: the trial tale.

This paper reviews the importance of vaccine trials to public health. Vaccines are developed to build immunity to disease, which have helped to completely eliminate smallpox, and reduced incidence of polio and measles. A variety of breakthroughs made many years ago, such as the smallpox vaccine by Edward Jenner and the discoveries by Louis Pasteur, also set the stage for modern international immunization programs. A few events, such as the licensing of the polio vaccine and the passage of the Vaccination Assistance Act helped to improve the study of vaccines. In particular, vaccine trials may
