Therapeutic Potential of Phytoconstituents in the Management of Hypertension.

Hypertension poses a significant global health burden due to its prevalence and serves as a major risk factor for cardiovascular diseases (CVDs) such as stroke, heart failure, myocardial infarction and renal dysfunction. Current clinical guidelines emphasize pharmacological management and lifestyle modifications. Despite the availability of effective pharmacological therapies, including diuretics, beta-blockers, calcium channel blockers, and ACE (angiotensin-converting enzyme) inhibitors, hypertension control remains suboptimal due to side effects, limited patient adherence, and the inability

A Method to Evaluate the Frequency Dependence of Retinal Vascular and Neural Metrics to Stimulus Modulation.

PURPOSE: Flickering light stimulation induces functional hyperemia, characterized by vasodilation, blood flow augmentation, and venous oxygen elevation. We present a new method to investigate the frequency dependence of metrics associated with functional hyperemia. METHODS: A novel optical imaging system was developed to quantify retinal blood vessel diameter (D), oxygen saturation (SO2), and the inner retinal oxygen extraction fraction (OEF) before and after light flicker at different frequencies. Measurements were performed in 10 visually normal subjects (20-62 years) at flicker frequencies

Functional coupling between peripheral nociceptor terminals in vivo is enhanced during acute inflammation.

Primary nociceptors are essentially characterized as afferent neurons carrying noxious sensory information from the periphery to the central nervous system. However, the information flow on primary nociceptors is bidirectional. Nociceptor peripheral terminals release a variety of mediators to the target organ near the injured area. These mediators promote sensitization of adjacent sensory neurons, vasodilation, and edema, affecting innate and adaptive immunity, leading to hyperalgesia and inflammation that often expands beyond the injured areas. While many studies associate these phenomena wit

Association between vitamin D deficiency and subarachnoid hemorrhage: a systematic review.

INTRODUCTION: Vitamin D functions also include regulating blood pressure and supporting the health of endothelial and brain cells. As such, vitamin D levels may be a determining factor for the occurrence of subarachnoid hemorrhage (SAH). OBJECTIVE: This study aims to investigate the relationship between low vitamin D levels and the occurrence of SAH, particularly non-traumatic cases. METHODS: Following PRISMA guidelines, we conducted a systematic review of studies published from 2008 to April 2023 in various databases, including SciELO, PubMed, EBSC HOST, and Google Scholar. We analyzed the

Green tea's secret weapon: a review on the protective effects of epigallocatechin-3-gallate against ischemia/reperfusion damage.

Epigallocatechin gallate (EGCG), a major polyphenol in green tea, has gained attention for its therapeutic potential in ischemia/reperfusion (I/R) injury—a condition in which restored blood flow to ischemic tissue paradoxically causes oxidative stress, inflammation, and cell death. EGCG exhibits potent antioxidant, anti-inflammatory, and anti-apoptotic properties that help mitigate these effects through modulation of various molecular signaling pathways. A key mechanism is the activation of the phosphoinositide 3-kinase/protein kinase B (PI3K/Akt) signaling pathway, which enhances Akt phosphor

The H2S-donor Erucin modulates SIRT1 and rescues obesity-induced vascular dysfunction in human vessels.

Evidence highlights the potential role of hydrogen sulfide (H₂S)-donors in preventing vascular dysfunction by modulating pathways associated with oxidative stress. We investigate the H₂S-donor Erucin for its potential to reduce oxidative stress and endothelial dysfunction in obesity. Patients undergoing laparoscopic surgery were categorized by body mass index in controls and patients with obesity. Microvascular endothelial function was measured by micromyography before and after incubation with Erucin 1 µM. Mitochondrial reactive oxygen species (mtROS) were assessed by fluorescence microscopy.

Orai1 Expression and Vascular Function in Kidney Donors Determine Graft Outcomes at Short/Mid-Term.

We aimed to determine the influence of donors' vascular function on renal function in recipients and to evaluate the role of Orai1 calcium channels as a potential marker. A prospective collaborative multicenter study was designed. Blood, aorta (HA), mesenteric arteries (HMAs) and corpus cavernosum (HCC) specimens were obtained from organ donors at the kidney procurement procedure (n = 60). Evolution (up to 2 years) of renal function measured as serum creatinine (SCr) and glomerular filtration rate (GFR) was evaluated in respective recipients (n = 64). Vascular responses were determined in HA,

Indole-3-Propionic Acid Improves Vascular Function in High-Fat Diet-Induced Obese Mice via eNOS.

BACKGROUND: The gut microbiota-derived metabolite indole-3-propionic acid (IPA) has been implicated in vascular homeostasis; however, its role in mesenteric vascular function and blood pressure regulation under obese conditions remains unclear. This study aimed to investigate the effects of IPA on mesenteric vascular endothelial function and blood pressure in high-fat diet-induced obese (DIO) mice. METHODS: C57BL/6 mice were fed a high-fat diet for 3 months to induce the DIO model. DIO mice were gavaged daily with IPA (20 mg/kg) for 6 weeks. At the end of the treatment, inflammatory markers a

[Anaphylactic shock].

Anaphylactic shock is a severe, potentially life-threatening systemic allergic reaction. It can involve multiple organ systems and is triggered by medication, food, or insect stings. Epidemiological data vary due to differing definitions, but studies estimate that 0,26% of hospital admissions are due to anaphylaxis, with 1-3 deaths per million annually. The incidence of allergic reactions has increased, though anaphylaxis-related mortality remains stable. Triggers vary by age, with food allergies being common in children, and insect stings and medication predominant in adults. An initial allerg

Peritoneal delivery of capsinoids, nonpungent TRPV1 agonists, induces mild hypothermia in conscious mice through TRPV1 activation of visceral vagal afferents.

Therapeutic hypothermia (TH) has demonstrated neuroprotection in instances of cardiac arrest and neonatal hypoxia/ischemia but faces different challenges in application to stroke due to the activation of cold defense mechanisms in conscious patients. This study examined the efficacy and specificity of capsinoids (a purified mixture of capsiate and dihydrocapsiate) to induce a sustained fall in core body temperature in conscious mice. Capsinoids function as TRPV1 agonists. However, unlike capsaicin, capsinoids are vulnerable to esterase-mediated breakdown, thus significantly restricting their a

Multidimensional study on mitochondrial dysfunction in pulmonary hypertension.

Pulmonary hypertension (PH), as a complex clinical syndrome, can be caused by multiple pathophysiological factors. Its characteristics are similar to hemodynamic abnormalities, significant increase of pulmonary artery pressure, contraction and remodeling of blood vessels, which eventually lead to serious complications such as increased pulmonary vascular resistance, hypertrophy of the right ventricle, and heart failure. The etiology of PH is multifaceted and highly variable, with a common pathological basis primarily characterized by mitochondrial dysfunction. Endothelial cell dysfunction, whi

Ticks' tricks: immunomodulatory effects of ixodid tick saliva at the cutaneous tick-host interface.

Due to changes in global climate, the geographic distribution of ticks and tick-borne infections is increasing and represents a growing global health concern for humans. Ticks of the genus Ixodidae are globally abundant and transmit a wide variety of pathogens that cause human infections, including tick-borne encephalitis and Lyme borreliosis. The transmission of pathogens into human skin while blood feeding causes changes in the local immune cell network and has various effects on structural skin cells, including sensory neurons. Recent studies have focused on the effect of tick saliva on cel

Clinical Outcomes of Carotid Artery Stenting for Carotid Artery Stenosis in Maintenance Hemodialysis Patients.

Most previous large studies of carotid artery stenting (CAS) in maintenance hemodialysis patients are old-era, do not describe the types of stents and method of protection, and their effectiveness is unknown. CAS has progressed remarkably, and tailor-made CAS is now possible in hemodialysis patients according to the lesion. We aimed to analyze the outcomes of CAS in maintenance hemodialysis patients treated in our institution. We retrospectively investigated the data of patients who underwent elective CAS in our institution between January 2012 and April 2023. Firstly, we verified the outcomes

Integrated Feedforward and Feedback Mechanisms in Neurovascular Coupling.

Neurovascular coupling (NVC) is the mechanism that drives the neurovascular response to neural activation, and NVC dysfunction has been implicated in various neurologic diseases. NVC is driven by (1) nonmetabolic feedforward mechanisms that are mediated by various signaling pathways and (2) metabolic feedback mechanisms that involve metabolic factors. However, the interplay between these feedback and feedforward mechanisms remains unresolved. We propose that feedforward mechanisms normally drive a swift, neural activation-induced regional cerebral blood flow (rCBF) overshoot, which floods the

Green-Light-Triggered and Self-Calibrated Cascade Release of Nitric Oxide and Carbon Monoxide for Synergistic Glaucoma Therapy.

Glaucoma is an optic degenerative neuropathy that is driven by a vicious cycle of oxidative stress and mechanical stress. Current clinical treatments aim exclusively at alleviating mechanical stress by reducing the intraocular pressure (IOP). With the unattended oxidative stress, recurrence and deterioration of mechanical stress are inevitable. Nitric oxide (NO) and carbon monoxide (CO) are endogenous gaseous signaling molecules for vasodilation and anti-inflammation, respectively. Mounting evidence suggests an intricate interplay between NO and CO to mediate their biological roles, like how i

Nitric oxide: Potential therapeutic target in Heat Stress-induced Multiple Organ Dysfunction.

As climate change intensifies, urgent action is needed to address global warming and its associated health risks, particularly in vulnerable regions. Rising global temperature and increasing frequency of heatwaves present a hidden health risk, disrupting the body's temperature regulation and leading to severe consequences such as heat stress-induced multiple organ dysfunction (HS-MOD). Multiple organ injury triggered by heat stress involves complex molecular pathways such as nitric oxide dysregulation, inflammation, oxidative stress, mitochondrial dysfunction, calcium homeostasis disruption, a

The Effects of Dobutamine in Septic Shock: An Updated Narrative Review of Clinical and Experimental Studies.

The key objective in the hemodynamic treatment of septic shock is the optimization of tissue perfusion and oxygenation. This is usually achieved by the utilization of fluids, vasopressors, and inotropes. Dobutamine is the inotrope most commonly recommended and used for this purpose. Despite the fact that dobutamine was introduced almost half a century ago in the treatment of septic shock, and there is widespread use of the drug, several aspects of its pharmacodynamics remain poorly understood. In normal subjects, dobutamine increases contractility and lacks a direct effect on vascular tone. Th
