Neural correlates of adjunctive rivastigmine treatment to antipsychotics in schizophrenia: a randomized, placebo-controlled, double-blind fMRI study.

Facilitation of central cholinergic activity may form a potential treatment strategy for cognitive impairment in schizophrenia. In a randomized, placebo-controlled, double-blind, parallel-group design, we investigated the neural correlates of cognitive effects of rivastigmine, an acetylcholinesterase inhibitor, given as an add-on therapy to antipsychotic-treated schizophrenia patients. Thirty-six chronic schizophrenia patients with mild cognitive impairment took part. After 1 week on placebo (baseline), all patients entered a double-blind protocol; 18 were allocated to receive rivastigmine and

Visual imagery deficits, impaired strategic retrieval, or memory loss: disentangling the nature of an amnesic person's autobiographical memory deficit.

Conclusions about the duration of hippocampal contributions to our autobiographical record of personal episodes have come under intense scrutiny in recent years. Interpretation is complicated by such factors as extent and site of lesions as well as test sensitivity. We describe the case of an amnesic person, K.C., with large, bilateral hippocampal lesions who figured prominently in the development of theories of remote memory due to his severely impoverished autobiographical memory extending across his entire lifetime. However, the presence of lesions in higher-order visual cortex raises the p

Internal structure of the nucleus rotundus revealed by mapping cadherin expression in the embryonic chicken visual system.

The nucleus rotundus is the largest nucleus of the avian thalamus. It is an important center of visual information processing and conveys information from the optic tectum to the ectostriatum in the telencephalon. The nucleus rotundus is generally believed to contain internal divisions processing information on color, form, motion, and looming of visual objects. The detailed arrangement of these internal divisions is unclear. Here, we map the expression of four classic cadherins (N-cadherin, R-cadherin, cadherin-6B, and cadherin-7), which are markers for specific functional gray matter divisio

Chronic NMDA receptor blockade from birth increases the sprouting capacity of ipsilateral retinocollicular axons without disrupting their early segregation.

We have investigated the role of the NMDA glutamate receptor (NMDAR) in the genesis and regulation of structural plasticity during synaptogenesis in the visual layers of the rat superior colliculus (sSC). In this neuropil, three projections compete for synaptic space during development. By fluorescently labeling the projections of both eyes and imaging them with confocal microscopy, we can quantify the sprouting of the ipsilateral retinal projection that follows removal of a portion of the contralateral retinal and/or corticocollicular projection. Using these techniques we have studied the eff

A modular neural circuit for computing the motion of objects.

Computing the direction of a moving object requires integrating motion signals from its component edges into coherent patterns. The representation of pattern motion in visual cortex has been extensively studied, yet its underlying neural circuitry remains unknown. Using high-density recordings in macaque area MT, we show that selectivity to pattern motion emerges from a functionally and anatomically distinct cortical circuit. We demonstrate that neurons specialized for encoding component and pattern motion comprise distinct cell types arranged in a hierarchical circuit in which pattern neurons

ECoGScope: An integrated platform for real-time Electrophysiology and fluorescence imaging.

In this study, we present ECoGScope, a versatile neural interface platform designed to integrate multiple functions for advancing neural network research. ECoGScope combines an electrocorticography (ECoG) electrode array with a commercial microendoscope, enabling simultaneous recording of ECoG signals and fluorescence imaging. The electrode array, constructed from highly flexible and transparent polymers, ensures conformal contact with the brain surface, allowing unobstructed optical monitoring of neural activity alongside electrophysiological recordings. A key innovation is the compact connec

Acute Cortical Blindness Associated with Occipital Lobe Compression from Simian T Cell Lymphoma Virus Lymphoma in a Baboon (Papio anubis).

Simian T cell leukemia virus (STLV) is associated with lymphoma in many captive and wild Old World nonhuman primate (NHP) species and is readily transmitted by bodily fluids. The disease is best characterized in baboons with a predisposition for aged female animals. Asymptomatic infections are common; however, clinical signs may include generalized lymphadenopathy, lethargy, and anorexia. Herein, we report a case of disseminated lymphoma in a middle-aged female STLV-positive olive baboon (Papio anubis) that presented with generalized lymphadenopathy and a rapid onset of decreased visual acuity

Mental programming of spatial sequences in working memory in the macaque frontal cortex.

How the brain mentally sorts a series of items in a specific order within working memory (WM) remains largely unknown. We investigated mental sorting using high-throughput electrophysiological recordings in the frontal cortex of macaque monkeys, who memorized and sorted spatial sequences in forward or backward orders according to visual cues. We discovered that items at each ordinal rank in WM were encoded in separate rank-WM subspaces and then, depending on cues, were maintained or reordered between the subspaces, accompanied by two extra temporary subspaces in two operation steps. Furthermor

Intermittent Theta-Burst Stimulation Increases the Working Memory Capacity of Methamphetamine Addicts.

The present study aimed to explore the effect of intermittent theta-burst stimulation (iTBS) on visual working memory for people suffering from methamphetamine use disorder (MUD). Five sessions of iTBS were carried over the left dorsolateral prefrontal cortex (DLPFC) or the vertex as a sham control, with each session in one day. Orientation free-recall tasks were conducted before the iTBS stimulation, after the first and fifth sessions of stimulation. Results showed that when compared with the sham group, a single session of iTBS over the left DLPFC improved participants' working memory perfor

Calpain-5 gene expression in the mouse eye and brain.

OBJECTIVE: Our objective was to characterize CAPN5 gene expression in the mouse central nervous system. Mouse brain and eye sections were probed with two high-affinity RNA oligonucleotide analogs designed to bind CAPN5 RNA and one scramble, control oligonucleotide. Images were captured in brightfield. RESULTS: CAPN5 RNA probes were validated on mouse breast cancer tumor tissue. In the eye, CAPN5 was expressed in the ganglion cell, inner nuclear and outer nuclear layers of the retina. Signal could not be detected in the ciliary body or the iris because of the high density of melanin. In the br

The role of the dorsal medial frontal cortex in central processing limitation: a transcranial magnetic stimulation study.

When humans perform two tasks simultaneously, responses to the second task are increasingly delayed as the interval between the two tasks decreases (psychological refractory period). This delay of the second task is thought to reflect a central processing limitation at the response selection stage. However, the neural mechanisms underlying this central processing limitation remain unclear. Using transcranial magnetic stimulation (TMS), we examined the role of the dorsal medial frontal cortex (dMFC) in a dual-task paradigm in which participants performed an auditory task 1 and a visual task 2.

Early planning activity in frontal and parietal cortex in a simplified task.

Cortical planning activity has traditionally been probed with visual targets. However, external sensory signals might obscure early correlates of internally generated plans. We devised a nonspatial decision-making task in which the monkey is encouraged to decide randomly whether to reach or saccade in the absence of sensory stimuli. Neurons in frontal and parietal planning areas (in and around the arcuate and intraparietal sulci) showed responses predictive of the monkey's upcoming movement at early stages during the planning process. Neurons predicted the animal's future movements several sec

The effect of breakfast versus no breakfast on brain activity in adolescents when performing cognitive tasks, as assessed by fMRI.

OBJECTIVES: The study examined the feasibility of utilizing functional magnetic resonance imaging (fMRI) with a group of adolescent boys and girls to assess modifications in cognitive function, dependent upon the nutritional state of the participants. METHODS: Twenty children aged 12-14 years completed two cognitive trials, in a randomized counterbalanced order, one under fasting condition, one after consuming breakfast, during which continuous fMRI data were acquired. RESULTS: Although no statistically significant (P > 0.05) improvement in task performance was determined, significantly high

The impact of unilateral brain damage on weight perception, sensorimotor anticipation, and fingertip force adaptation.

Damage to the left parietal cortex can lead to apraxia - a selective deficit in tool use and action planning. There is conflicting evidence as to whether this disorder affects more fundamental motor parameters, such as applying the appropriate forces to lift objects based upon how heavy they look. Here we examined how individuals with left and right-lateralized brain damage lift and perceive the weight of objects of the same mass which vary in their size and material properties. No clear differences emerged between the groups in terms of how visual material properties affected their perception

Impaired critical period plasticity in primary auditory cortex of fragile X model mice.

Fragile X syndrome, the most common form of heritable mental retardation, is a developmental disorder with known effects within sensory systems. Altered developmental plasticity has been reported in the visual and somatosensory systems in Fmr1 knock-out (KO) mice. Behavioral studies have revealed maladaptive auditory responses in fragile X syndrome patients and Fmr1 KO mice, suggesting that adaptive plasticity may also be impaired in the auditory system. Here we show that, whereas tonotopic frequency representation develops normally in Fmr1 KO mice, developmental plasticity in primary auditory

Neurological examination of cortical function deficits.

Human cortical functions have been elucidated by studies of deficits in traumatic and vascular brain damage, outcomes after elective neurosurgical procedures, studies in primates and in more recent years by imaging techniques. Cortical functions are well-defined for primary cortical areas like motor, sensory and visual functions. More complex cortical functions like language and to some degree memory are also well clarified. The associative cortical areas are more difficult to study as functions are integrated to and modulate primary cortical functions. Nevertheless, the structural basis for s

An atypical autopsy case of Lewy body disease with clinically diagnosed major depression: a clinical, radiological and pathological study.

We report an 84-year-old woman who was clinically diagnosed with late-life major depression (LLMD) and having a diffuse type of dementia with Lewy bodies (DLB) neuropathologically. Clinically, this case showed depressive mood, anxiety, and irritation, but did not show cognitive dysfunction, visual hallucination, fluctuation of alertness and parkinsonism, which define the criteria for diagnosing DLB. Neuropathological examination demonstrated abundant Lewy-related pathology including Lewy bodies and neurites in the hippocampal region and the cerebral cortex, and moderate levels in brain stem nu

Influence of continual biofeedback on jaw pursuit-tracking in healthy adults and in adults with apraxia plus aphasia.

Apraxia of speech (AOS) is a disorder of motor programming resulting from damage to premotor or anterior insula cortex. The authors used a pursuit visuomotor tracking task to test whether such a disorder interferes with development of motor programs or with modification of existing programs via integration of feedback. Healthy older adults (n = 15) and adults with AOS plus aphasia and nonverbal apraxia (n = 8) performed a jaw movement task with (a) continuous visual feedback of a target movement pattern and their jaw movement and (b) no feedback. Healthy speakers were more accurate and less va

The anatomy of neglect without hemianopia: a key role for parietal-frontal disconnection?

We investigated the anatomical correlates of chronic unilateral neglect in right brain damaged patients with no visual field defects. Independently from basal ganglia or thalamus involvement, neglect patients had a subcortical area of maximal lesion overlap (maxov) in the superior longitudinal fasciculus (SLF). Neglect patients without lesion of subcortical nuclei had an additional maxov area in the rostralmost limit of the supramarginal gyrus (SmG). Patients without neglect showed a maxov area in the corticospinal tract with no involvement of the SLF or SmG. These findings call attention to t

Different brain correlates for watching real and virtual hand actions.

We investigated whether observation of actions reproduced in three-dimensional virtual reality would engage perceptual and visuomotor brain processes different from those induced by the observation of real hand actions. Participants were asked to passively observe grasping actions of geometrical objects made by a real hand or by hand reconstructions of different quality in 3D virtual reality as well as on a 2D TV screen. We found that only real actions in natural environment activated a visuospatial network including the right posterior parietal cortex. Observation of virtual-reality hand acti
