The effects of benfluramate on carbohydrate metabolism in obesity.

Glucose turnover rate, glucose oxidation and lactate production from glucose were investigated in 5 obese subjects using the primed infusion technique, before and four weeks after a course on benfluramate therapy. All subjects lost weight on the drug. Mean glucose turnover rate, but not CO2 and lactate production increased after treatment. The changes in metabolism appear to reflect the combined effects of weight loss and drug action.

GLP-1 medications: use and interest in a representative survey of Finns.

BACKGROUND AND OBJECTIVES: GLP-1-based medications have rapidly reshaped the landscape of obesity treatment. Semaglutide was approved for obesity treatment in 2021 in the US and 2022 in Europe, sparking global interest, and the GLP-1/GIP dual-agonist tirzepatide has demonstrated even greater efficacy. However, survey-based data on who uses or considers these medications-particularly across socioeconomic groups, BMI categories, and weight management experiences-remain limited. SUBJECTS/METHODS: A nationally representative online survey of Finnish adults (n = 1729, of which 1693 were included)

A habenula-enriched GPCR, GPR151, regulates behavioral sensitivity to inflammation.

BACKGROUND: Inflammation-associated depression is a subtype of major depressive disorder that is often resistant to conventional pharmacotherapies, which act in a regionally non-specific manner and therefore also produce unwanted side effects. Here we test GPR151, an orphan GPCR associated with inflammation and highly expressed in the habenula-a region linked to negative valence and depression-as a therapeutic target for inflammation-associated depression. METHODS: We integrated mouse and human habenular expression analyses with genetic loss-of-function and adult habenular re-expression appro

Dynamic prediction of Radiotherapy toxicities in Head and neck cancer using clinical and imaging data.

BACKGROUND AND PURPOSE: Head and neck cancer (HNC) radiotherapy (RT) is effective but causes significant toxicity. We aimed to develop a dynamic deep learning model to predict three major HNC RT toxicities-nasogastric (NG) tube placement, hospitalization, and radionecrosis-by integrating clinical data and daily cone-beam computed tomography (CBCTs), assessing whether serial imaging or dosimetry features improve early prediction. MATERIALS AND METHODS: We retrospectively analyzed 1,012 HNC patients treated with RT between 2017 and 2022. A multibranch 3D ResNet50 and multilayer perceptron model

Effects of a High-Intensity Exercise Program on Weight Regain and Cardio-metabolic Profile after 3 Years of Bariatric Surgery: A Randomized Trial.

Weight regain is one of the most common problems in the long-term after bariatric surgery. It is unknown if high-intensity exercise programs applied in late phases of post-surgical follow-up could counteract this trend. After a 3-year follow-up, 21 patients underwent sleeve gastrectomy were randomized into an exercise group (EG, n = 11), that performed a 5-month supervised exercise program, and a control group (CG, n = 10), that followed the usual care. Body composition, cardiorespiratory fitness, glycaemia and blood cholesterol were evaluated before and after the intervention. Finally, the EG

Inter-Eye Association of Visual Field Defects in Glaucoma and Its Clinical Utility.

PURPOSE: To investigate intereye associations of visual field (VF) defects. METHODS: We selected 24-2 VF pairs of both eyes from 63,604 patients tested on the same date with mean deviation (MD) ≥ -12 dB. VFs were decomposed into one normal and 15 defect patterns previously identified using archetypal analysis. VF pattern weighting coefficients were correlated between the worse and better eyes, as defined by MD. VF defect patterns (weighting coefficients > 10%) in the better eye were predicted from weighting coefficients of the worse eye by logistic regression models, which were evaluated by a

Single Anastomosis Sleeve Ileal Bypass Could Be the Future of Redo Surgery? A Single-Center Cohort Study.

BACKGROUND: Obesity is a leading global health challenge associated with increased morbidity and mortality from cardiovascular disease, type 2 diabetes, hypertension (HTN), and other metabolic comorbidities. Bariatric surgery is currently the most effective long-term treatment for severe obesity. However, 20-30% of patients may experience weight regain or unsatisfactory metabolic outcomes after primary surgery, requiring revisional (redo) procedures. The Single Anastomosis Sleeve Ileal Bypass (SASI) is an innovative bariatric technique, recently proposed as a redo option due to its favorable b

Cancer Cachexia Prevalence Is Underestimated in Medical Records of Patients in a Regional Tertiary Hospital.

BACKGROUND: Widespread lack of awareness and limited real-world prevalence evidence have impeded cachexia care and research. We hypothesized that healthcare professionals may identify the term cachexia, leading to International Classification of Diseases (ICD) coding for this term, with or without records of body weight loss for diagnosing cancer cachexia, and that frequently, ICD coding does not accurately reflect weight data. METHODS: We assessed cachexia prevalence in patients diagnosed with cancer, using the Clinical Data Warehouse of a French hospital containing two types of real-world d
