Prevalence and genotype distribution of cervical human papillomavirus infection in Macao.

Population-specific epidemiological data on human papillomavirus (HPV) infection are essential for formulating strategies to prevent cervical cancer. The age-specific prevalence of HPV infection was determined among 1,600 women enrolled for cervical screening in Macao. A U-shaped age-specific prevalence curve with a first peak (prevalence rate, 10%) at 20-25 years and a second peak (13%) at 51-55 years was observed. Co-infections with multiple types were detected in 32.5% of HPV-positive subjects and without significant variation among different age groups (P = 0.318). The majority (84.6%) of

The secreted and surface proteomes of the adult stage of the carcinogenic human liver fluke Opisthorchis viverrini.

Infection with the human liver fluke, Opisthorchis viverrini, is a serious public health problem in Thailand, Laos and nearby locations in Southeast Asia. Both experimental and epidemiological evidence strongly implicate liver fluke infection in the etiology of one of the liver cancer subtypes, cholangiocarcinoma (CCA). To identify parasite proteins critical for liver fluke survival and the etiology of CCA, OFFGEL electrophoresis and multiple reaction monitoring were employed to characterize 300 parasite proteins from the O. viverrini excretory/secretory products and, utilizing selective label

Application of a multiplex PCR to cervical cells collected by a paper smear for the simultaneous detection of all mucosal human papillomaviruses (HPVs) and typing of high-risk HPV types 16 and 18.

A simple paper smear (PS) method for dry collection and storage of cervical specimens was employed to develop an easy multiplex (MPX) PCR for simultaneous detection of generic human papillomaviruses (HPVs) as well as typing of the high-risk HPV-16 and -18, the two clinically most important HPV genotypes, which are responsible for more than 80 % of cervical cancers. Multiplexing was performed with a small amount of DNA eluted by boiling from a single PS punch in a single tube and using a mixture of four pairs of primers specific for the HPV L1 consensus sequence, HPV-16, HPV-18 and the β-globin

Enhancing CD8 T-cell memory by modulating fatty acid metabolism.

CD8 T cells, which have a crucial role in immunity to infection and cancer, are maintained in constant numbers, but on antigen stimulation undergo a developmental program characterized by distinct phases encompassing the expansion and then contraction of antigen-specific effector (T(E)) populations, followed by the persistence of long-lived memory (T(M)) cells. Although this predictable pattern of CD8 T-cell responses is well established, the underlying cellular mechanisms regulating the transition to T(M) cells remain undefined. Here we show that tumour necrosis factor (TNF) receptor-associat

[The role of TMPRSS2 in tumor development and viral infection].

TMPRSS2 is a member of a family of proteins localized to the cell membrane and characterized by a conserved serine protease domain. Previous studies have demonstrated its involvement in the initiation and progression of various cancers in the human body. Its serine protease activity can degrade extracellular matrix components, disrupt the integrity of the basement membrane, and promote tumor cell invasion and metastasis. In addition, its expression level is closely related to the degree of immune cell infiltration and may affect tumor immune escape by regulating local inflammatory responses. I

Emerging concepts, hot topics, and open questions in today's pancreatic cancer research.

Despite intense research, pancreatic ductal adenocarcinoma (PDAC) remains one of the most lethal malignancies to date. The low 5-year survival rate of currently 8-10% has only marginally improved over the last years, necessitating novel approaches and a thinking "outside the box". We as translational basic and medical scientists working in a clinical research unit on tumor-stroma interactions in PDAC (the DFG-funded CRU325) have sought to identify emerging concepts in current pancreatic cancer research. We have addressed recent developments and open questions, covering a wide spectrum of topic

Generation and MHC class II loading of an endogenous influenza epitope revealed by a T cell receptor-like antibody.

CD4+ T cells coordinate immune responses to infections and cancers and initiate many autoimmune diseases, yet the intracellular pathways that generate the peptide antigens they recognize remain incompletely understood. Progress has been limited in part by the scarcity of reagents that directly detect defined peptide:MHC class II (pMHC-II) complexes. Here, we established an mRNA vaccine-based pipeline to generate and characterize T cell receptor (TCR)-like antibodies. One result was 1D6, a monoclonal antibody specific for NA25:Ab, an influenza A/PR8 neuraminidase-derived epitope presented by MH

Adulthood diagnosis of Nijmegen breakage syndrome in twins with antibody deficiency clinical presentation.

Nijmegen breakage syndrome (NBS) is a rare autosomal recessive DNA repair disorder within the spectrum of inborn errors of immunity (IEI), characterized by immunodeficiency and cancer predisposition and typically diagnosed in childhood. We report a pair of 21-year-old monozygotic twins presenting with recurrent infections, hypogammaglobulinemia, ovarian dysfunction, and, in one twin, peripheral T-cell lymphoma. Both carried two novel compound heterozygous variants in the NBN gene (c.657dupA and c.1064_1124 + 9del). Immunophenotyping revealed reduced memory B cells, impaired vaccine responses,

Comprehensive genome editing and signaling pathway engineering evolve potent and safe adenoviral producer cell.

Dramatic attention has been drawn to adenovirus as a vehicle for vaccine and cancer therapy, due to the capabilities of annihilating tumor cells and stimulating host immune response. However, few producer cells are perfect for adenoviral manufacture. Herein, we orchestrate a strategy by combining homology-independent targeted integration and sgRNA library, which not only modulates the cellular gene expression profile systemically that enhance subgroup B/C adenovirus yields by 3 ~ 7 folds, but also substitutes naïve adenovirus sequence in HEK293 cells with a recombinant E1 transgene, drasticall

Overcoming cancer immunotherapy barriers via nanomaterial-mediated pyroptosis.

While cancer immunotherapy has achieved groundbreaking clinical success, its efficacy is frequently compromised by insufficient T-cell activation, the immunosuppressive tumor microenvironment (TME), and off-target toxicity. Pyroptosis, a highly immunogenic form of programmed cell death characterized by gasdermin-mediated pore formation, massive cytokine release (e.g., IL-1β and IL-18), and robust dendritic cell activation, offers a compelling strategy to overcome these limitations. This review critically examines how nanotechnology-enabled pyroptosis induction can potentiate immunotherapy by (

Targeting HPV for the prevention, diagnosis, and treatment of cervical cancer.

Despite advances in screening and prevention, cervical cancer (CC) remains an unresolved public health issue and poses a significant global challenge, particularly for women in low-income regions. Human papillomavirus (HPV) infection, especially with the high-risk strains, is a primary driver of cervical carcinogenesis. Emerging evidence indicates that integrating HPV testing with existing approaches, such as cervical cytology and visual inspection, offers enhanced sensitivity and specificity in CC screening. HPV infection-associated biomarkers, including HPV E6/E7 oncogenes, p16^INK4a, DNA me

Human alveolar echinococcosis-global, regional, and national annual incidence and prevalence rates.

BACKGROUND: Alveolar echinococcosis (AE) of humans is a disease caused by the larval (metacestode) stage of the 'fox tapeworm' Echinococcus multilocularis. AE primarily affects the liver and, like cancer, can spread to surrounding or distant organs. Its slow progression and resemblance to malignant tumours emphasise the critical importance of early detection and intervention, as there is no highly effective vaccine against AE. Although recognised as a neglected disease, there has been no recent, comprehensive review of the distribution, incidence and prevalence of AE in humans around the world

Multifunctional Liposomes Remodeling Tumor Immune Microenvironment for Tumor Chemoimmunotherapy.

In the treatment of solid tumors, the complex barriers composed of cancer-associated fibroblasts (CAFs) prevent drug delivery and T cells infiltration into tumor tissues. Although nanocarriers hold great prospects in drug delivery, fibrosis causes the biological barrier and immunosuppressive tumor microenvironment (ITM) that impairs the anti-tumor efficacy of nanocarriers. Here, a small dendritic macromolecule loaded with doxorubicin (PAMAM-ss-DOX) (DP) is synthesized and encapsulated into pH-responsive nanoliposome, together with adjuvant toll-like receptor 7/8 (TLR7/8) agonist resiquimod (R8

Case report: Radiotherapy plus pneumococcal conjugate vaccine stimulates abscopal immune response in a patient with ALK+ NSCLC.

Most patients with anaplastic lymphoma kinase-positive (ALK+) non-small-cell lung cancer (NSCLC) could benefit from the treatment with selected tyrosine kinase inhibitors (TKIs) for a period of time, but almost inevitably progress due to drug resistance. It was reported that these patients were generally unresponsive to immune-based therapies. Here, we reported that stereotactic body radiotherapy (SBRT) combined with pneumococcal conjugate vaccine (PCV) produced excellent therapeutic outcomes in a patient after multiple lines of TKI treatment. The patient's metastasis lesion experienced regres

Recent Advances in Polymer Microneedles for Drug Transdermal Delivery: Design Strategies and Applications.

In recent years, transdermal drug delivery based on microneedles (MNs) technology has received extensive attention, which offers a safer and painless alternative to hypodermic needle injections. They can pierce the stratum corneum and deliver drugs to the epidermis and dermis-structures of skin, showing prominent properties such as minimally invasiveness, bypassing first-pass metabolism, and can be self-administered. A range of materials has been used to fabricate MNs, such as silicon, metal, glass, and polymers. Among them, polymer MNs have gained increasing attention from pharmaceutical and

Metastatic melanoma in the breast and axilla: A case report.

Metastatic melanoma of the breast is rare, and demonstrates nonspecific imaging findings which may overlap with both benign and malignant pathology.1-3 Immunohistochemical stains are important to confirm the diagnosis, particularly combining S100, a sensitive marker for melanoma, with more specific tumor markers such as Melan-A and HMB-45, and lack of cytokeratin staining.4-7 We present a case of a 64-year-old female who presented for diagnostic imaging of a palpable abnormality in her right breast, with medical history notable for previously excised cutaneous melanoma, recent COVID-19 vaccina

Epidemiological projections of viral-induced hepatocellular carcinoma in the perspective of WHO global hepatitis elimination.

Hepatitis B is an eminent risk factor for hepatocellular carcinoma (HCC) in Southeast Asia and sub-Saharan Africa, whereas hepatitis C is a key risk factor for HCC in Western Europe and North America. Increased awareness of the global burden of viral hepatitis resulted, in May 2016, in the adoption of the first global health sector strategy on viral hepatitis by the World Health Assembly, which calls for the elimination of viral hepatitis as a public health threat by 2030. Although the incidence of liver cancer resulting from viral infections has increased since the 1990s, the implementation o

Structural Organization and Protein-Protein Interactions in Human Adenovirus Capsid.

Human adenoviruses (HAdVs) are large (150 MDa), complex, nonenveloped dsDNA viruses that cause self-limiting respiratory, ocular and enteric infections. They are significant health hazard in young, elderly and immuno-compromised populations. Moreover, various adenoviruses (AdVs) of mammalian origin are being used as vectors in gene, vaccine and cancer therapies. Multiple copies of at least 13 different proteins, all in all ~2800 protein molecules, come together to form an adenovirus virion packaging the ~36 Kbp geome. The details of structural organization of the adenovirus capsid and underlyi
