Comment on: Changes in Aqueous Humor Cytokines and Metabolomics in Contralateral Eye After Unilateral Cataract Surgery.

UNLABELLED: We extend Li et al.'s investigation of aqueous humor (AH) metabolomics in sequential cataract surgery by referencing our prior study on interocular symmetry/asymmetry in AH metabolomic profiles from simultaneous bilateral cataract surgery in emmetropic patients, which demonstrated similar AH compositions in fellow eyes. We also illustrate variability with 2 sequential-surgery cases and highlight the most and least variable metabolites across 6 biochemical classes. Taken together with Li et al., these observations support careful attribution of second-eye changes to surgery versus b

Methodological approaches for dissecting ferroptosis in lens epithelial cells and its role in diabetic cataract progression.

Diabetic cataract has become the second-largest blind causing disease among the complications of diabetes mellitus in eyes. Ferroptosis is responsible for many complications associated with diabetes mellitus, but the role of ferroptosis in diabetic cataract has not yet been studied. Here, we first ascertained that ferroptosis was triggered during diabetic cataract, as identified by elevated levels of transferrin receptor protein 1 (CD71) and solute carrier family 7 member 11 (SLC7A11), lipid peroxidation , total iron and Fe2+ under conditions of excessive glucose. Moreover, voltage-dependent a

Red reflex examination for early detection of pediatric cataract using a novel red-reflex screening device.

BACKGROUND: Early detection of pediatric cataracts is essential to prevent irreversible visual impairment; however, effective screening remains limited in many settings. This study evaluated the diagnostic performance of the CatScreener, a self-designed handheld red-reflex screening device, when operated by non-expert examiners. METHODS: A total of 250 eyes from 250 infants and young children (≤8 years) underwent CatScreener screening and slit-lamp examination. Images were independently interpreted by two medical students and two junior ophthalmologists. Diagnostic outcomes were compared with

Age-Related Lens Membrane Composition and N101D Deamidation Modulate αA-Crystallin Membrane Interactions Relevant to Cataract.

PURPOSE: αA-crystallin (αAc), the predominant subunit comprising α-crystallin (αABc), becomes increasingly deamidated and membrane bound with cataract formation. Furthermore, the lens lipids and cholesterol (Chol) composition significantly change with age and cataract formation. Despite this, the relationship between αAc deamidation and lens membrane composition in cataractogenesis remains unclear. To address this, we analyzed the role of N101 deamidation and membrane composition in the membrane interactions of αAc. METHODS: Experimentation involved using electron paramagnetic resonance (EPR)

Hereditary Hyperferritinemia Cataract Syndrome Mimicking Iron Overload in a Pediatric Patient With Coexisting HFE H63D Homozygosity.

BACKGROUND: Hereditary hyperferritinemia cataract syndrome (HHCS) is a rare autosomal dominant disorder caused by mutations in the FTL gene, characterized by elevated serum ferritin levels without systemic iron overload and early-onset cataract. Misinterpretation of hyperferritinemia may lead to unnecessary investigations and treatment. CASE: We report a pediatric case with familial involvement in which hyperferritinemia was incidentally detected in a 9-year-old boy. Laboratory evaluation revealed markedly elevated serum ferritin levels with normal transferrin saturation and no evidence of sy
