PARP-1 couples β-catenin/TCF4 signaling to epithelial-mesenchymal transition in endometriosis.

Endometriosis exhibits epithelial–mesenchymal transition (EMT)-aligned traits that promote invasion and lesion persistence. We investigated whether poly(ADP-ribose) polymerase-1 (PARP-1) coordinates β-catenin/TCF4 activity with EMT programs and whether these features are amenable to pharmacologic inhibition. PARP-1 and EMT markers were profiled in normal endometrium and ovarian endometriotic lesions. In endometriotic epithelial cells, PARP-1 levels were modulated by overexpression or siRNA and effects on EMT markers and motility were quantified. Association of PARP-1 with β-catenin/TCF4 comple

[Jianwei Shoutai Pill reduces spontaneous abortion in mice by inhibiting Exo-miR-410-3p uptake and promoting trophoblast epithelial-mesenchymal transition, migration, and invasion].

OBJECTIVES: To investigate the therapeutic effect of Jianwei Shoutai Pill (JSP) on spontaneous abortion (SA) in mice and explore its therapeutic mechanism. METHODS: The miR-410-3p mimic was electroporated into milk exosomes to prepare exosomal miR-410-3p (Exo-miR-410-3p), which was characterized using transmission electron microscopy (TEM), nanoparticle tracking analysis (NTA), and Western blotting. CCK-8, scratch wound healing, and Transwell assays were used to test the effects of control and JSP-medicated rat serum on proliferation, migration, and invasion of trophoblast HTR-8/Svneo cells.

Regulation of FN1 levels in endometrial cancer cells: Probiotics activate the p62-dependent autophagy-lysosome pathway to reduce tumor metastasis.

BACKGROUND: Endometrial cancer (EC) is a common female reproductive system malignancy, with metastasis being a major cause of poor prognosis. Intestinal flora imbalance is linked to gynecological tumor progression, and probiotics exhibit anti-tumor potential, but their regulatory mechanisms in EC migration/invasion/metastasis remain unclear. METHODS: Bioinformatics analysis: TCGA database screened 1063 metastasis-related differentially expressed genes (DEGs), and 3915 DEGs after probiotic intervention, with 301 overlapping DEGs enriched in "ECM assembly", "autophagy regulation", and "epitheli

Modulation of cell migration and invasion of thyroid cancer cells using miRNA-enriched extracellular vesicles.

BACKGROUND: Extracellular vesicles (EVs) have been investigated as promising vehicles for delivering drugs and microRNAs (miRNAs) in distinct types of cancer. However, their use as a delivery system in papillary thyroid carcinoma (PTC) remains underexplored. PTC is the most prevalent subtype of thyroid cancer, and despite its good prognosis, a fraction of the cases evolve into aggressive and fatal forms. Here, we investigated whether non-tumor thyroid cells can be engineered to produce EVs-enriched with tumor suppressor miRNAs that could be delivered to PTC cells. METHODS: EVs from NThy-ORI c

Identification of lactylation-related biomarkers for diagnosis, prognosis, and treatment responsiveness in triple-negative breast cancer.

BACKGROUND: Triple-negative breast cancer (TNBC), a highly aggressive subtype of invasive breast cancer, lacks effective therapeutic targets and robust prognostic biomarkers. Lactylation, an emerging post-translational modification, is increasingly recognized for its potential role in tumorigenesis and progression. However, the functional significance of lactylation-related genes (LRGs) in TNBC clinicopathology and prognosis remains poorly defined. METHODS: We employed a combination of differential expression analysis and weighted gene co-expression network analysis (WGCNA) to identify hub LRG

Ferroptosis-dependent small extracellular vesicles ULK1 enhances mitophagy and suppresses breast cancer migration.

BACKGROUND: Breast cancer progression is closely influenced by the tumor microenvironment, including macrophage polarization. Small extracellular vesicles (sEVs) derived from tumor cells serve as key mediators of intercellular communication that regulate immune responses. Ferroptosis, characterized by iron-dependent lipid peroxidation, plays a role in tumor progression. However, whether ferroptosis alters the cargo of breast cancer cell-derived sEVs and affects macrophage function remains largely unknown. RESULTS: In this study, we identify that sEVs derived from ferroptotic breast cancer cell

Bacteriophage Cocktail in Hydrogel Dressing Modulates Macrophage Responses and Induces Skin Cell Migration.

BACKGROUND: Bacteriophage mixtures have been explored as biomaterials to promote tissue repair. In this study, we tested the hypothesis that incorporating a phage mixture into a wound dressing could modulate immune cell responses and skin cell migration within the wound environment. METHODS: Phage strains specific to three common bacterial pathogens-Escherichia coli, Salmonella enterica, and Pseudomonas aeruginosa-were embedded in an alginate hydrogel (E-Alg) or grafted onto its surface (S-Alg). The viability of the phages released from the samples were tested. 3T3 fibroblasts in a transwell

Polycystin-1 Orchestrates Tumor Context-Dependent Mechanotransduction Programs Driving Epithelial-to-Mesenchymal Transition and Invasion in Solid Cancers.

Mechanotransduction critically shapes tumor progression by regulating cytoskeletal remodeling, epithelial-to-mesenchymal transition (EMT), invasion, and immune evasion. Polycystin-1 (PC1) and polycystin-2 (PC2), established mechanosensitive proteins in epithelial systems, have recently been implicated in tumor biology; however, their roles across diverse solid malignancies remain insufficiently defined. We assessed PC1 and PC2 expression patterns and their associations with clinicopathological features in human lung, breast, prostate, and brain tumors. PC1 functional modulation was performed i

Inhibition of SRC prevents bone metastasis of breast cancer by blocking metastatic cell motility and bone directionality.

RATIONALE: Breast cancer bone metastasis remains a major cause of mortality with limited effective therapies. Although SRC is one of the earliest identified oncogenic kinases and has been extensively studied as a regulator of cancer progression and migration, it has not yet been successfully translated into an effective therapeutic target in solid tumors, highlighting the need to redefine SRC-targeted strategies in this context. METHODS: Genetic deletion and pharmacological approaches were employed to interrogate SRC function, including comparative evaluation of conventional kinase inhibitors

Auranofin Suppresses Cancer Cell Invasion by Inhibiting Heparanase-1 Expression via the aPKC-NF-κB Pathway.

Heparanase 1 (HPSE1) is the only mammalian endoglycosidase that cleaves heparan sulfate (HS), a glycosaminoglycan (GAG), and is frequently upregulated in cancers, thereby promoting tumor progression. Despite extensive efforts to develop inhibitors of its HS-degrading activity, its non-enzymatic functions limit therapeutic efficacy and pose a major challenge for therapeutic development. Thus, inhibiting HPSE1 expression is critical for controlling its enzymatic and non-enzymatic functions; however, no FDA-approved inhibitors are currently available. Here, we identify auranofin (AUF), an oral go

Retinal pigment epithelium drives macrophage migration during Toxoplasma gondii infection in vitro.

BACKGROUND: Ocular toxoplasmosis is a leading cause of infectious posterior uveitis worldwide. The retinal pigment epithelium (RPE), a key barrier and immunomodulatory layer in the eye, is directly targeted by Toxoplasma gondii during infection. However, its role in orchestrating the local immune response remains unclear. OBJECTIVES: To investigate whether RPE cells actively drive macrophage migration during T. gondii infection in vitro, and to identify associated cytokine profiles. METHODS: Adult retinal pigment epithelial cells (ARPE)-19 and primary RPE cells were exposed to tachyzoites, s

MAP4 kinase-regulated reduced CLSTN1 expression in medulloblastoma is associated with increased invasiveness.

De-regulated protein expression contributes to tumor growth and progression in medulloblastoma (MB), the most common malignant brain tumor in children. MB is associated with impaired differentiation of specific neural progenitors, suggesting that the deregulation of proteins involved in neural physiology could contribute to the transformed phenotype in MB. Calsynthenin 1 (CLSTN1) is a neuronal protein involved in cell-cell interaction, vesicle trafficking, and synaptic signaling. We previously identified CLSTN1 as a putative target of the pro-invasive kinase MAP4K4, which we found to reduce CL

Silencing of O-GlcNAc Transferase Attenuated O-GlcNAcylation and Metastatic Potentials of Melanoma Cells Through Suppression of Akt-NFκB Signaling Pathway.

O-GlcNAcylation is an important biological process in regulating the function of many nucleocytoplasmic proteins in cells. Enhancement of O-GlcNAcylation was associated with cancer development and progression. Here, we demonstrated the involvement of O-GlcNAcylation in melanoma metastasis. Using the data from GEO database, we found that O-GlcNAcylation and its related enzymes, including glutamine fructose-6-phosphate amidotransferase (GFAT), O-GlcNAc transferase (OGT), and O-GlcNAcase (OGA); were elevated in metastatic melanoma compared with primary tumors and normal tissues. Inclusion of ΑVβ3 integrin into extracellular vesicles in a caveolin-1 tyrosine-14- phosphorylation dependent manner and subsequent transfer to recipient melanoma cells promotes migration, invasion and metastasis.

Caveolin-1 (CAV1) is a membrane protein that promotes migration, invasion and metastasis of cancer cells when phosphorylated on tyrosine-14 (Y14) by a cell intrinsic mechanism involving the activation of a novel Rab5-Rac1 signaling axis. Moreover, CAV1 expressed in aggressive cancer cells is included into extracellular vesicles (EVs) and such EVs increase the metastatic potential of recipient lower grade cancer cells. However, the relevance of CAV1 Y14 phosphorylation in these extrinsic EV-stimulated events remained to be determined. Here we used B16F10 mouse melanoma cells over-expressing wil

P-Rex1 controls phagocytosis and the killing of bacteria by murine neutrophils independently of its catalytic activity.

INTRODUCTION: P-Rex1 is a guanine-nucleotide factor for the small GTPase Rac (Rac-GEF) that is known to mediate neutrophil migration and ROS production in response to the activation of GPCRs. These roles of P-Rex1 are assumed to require its activation of Rac. METHODS: Here, we used mice which lack P-Rex1 (Prex1 -/-) and mice with catalytically inactive P-Rex1 (Prex1GD ) to investigate the importance of the Rac-GEF activity in migration and ROS production. We also studied the requirement for P-Rex1 in degranulation, phagocytosis, NET formation, and the bactericidal activity of neutrophils. RE

Augmenting AMPA receptor signaling after spinal cord injury increases ependymal-derived neural stem/progenitor cell migration and promotes functional recovery.

Ependymal cells in the adult spinal cord become activated after spinal cord injury (SCI), gaining stem/progenitor cell properties. Although growing evidence has implicated these cells as potential players in the endogenous repair process after injury, their activation to a stem-cell-like state is transient and insufficient for adequate regeneration. Moreover, the drivers of their activation state remain largely unknown. Previous work suggested that AMPA receptors (AMPARs) regulate cultured ependymal-derived neural stem/progenitor cells (epNSPCs). In this study, we identified an AMPAR-dependent

Slc26a9 promotes the perineural invasion of pancreatic cancer in mice.

Perineural invasion (PNI) is a pivotal prognostic factor in pancreatic ductal adenocarcinoma (PDAC), associated with aggressive tumor behavior and poor patient outcomes. This study investigated the role of Slc26a9 in regulating peripheral nerve invasion in pancreatic cancer. Bioinformatic analysis of GEO datasets showed markedly higher Slc26a9 expression in PNI-positive patients than in those without PNI. A PDAC model was established by implanting Panc02-luc cells into C57BL/6 mice. Abdominal mechanical hyperalgesia, hunching behavior, and anxiety-like behaviors were assessed using von Frey, h

PDK4 expression and tumor aggressiveness in prostate cancer.

PURPOSE: Prostate cancer ranks as the second most common cancer in men globally, representing a significant cause of cancer-related mortality. Metastasis, the spread of cancer cells from the primary site to distant organs, remains a major challenge in managing prostate cancer. Pyruvate dehydrogenase kinase 4 (PDK4) is implicated in the regulation of aerobic glycolysis, emerging as a potential player in various cancers. However, its role in prostate cancer remains unclear. This study aims to analyze PDK4 expression in prostate cancer cells and human samples, and to explore the gene's clinical s

Investigating the Therapeutic Potential of Agarwood Nanoemulsion in Modulating TGF-β-Induced Airway Remodelling in BEAS-2B Cells.

Chronic respiratory diseases such as asthma, chronic obstructive pulmonary disease (COPD), and pulmonary fibrosis are significant global health concerns, characterised by inflammation, oxidative stress, and airway remodelling. These processes are driven by multiple cytokines, with transforming growth factor-beta (TGF-β) playing a central role in the remodelling process. TGF-β triggers pathways that promote epithelial-mesenchymal transition (EMT), excessive extracellular matrix deposition, and increased oxidative stress, all of which contribute to airway remodelling. Despite availability of the
