CRMP-2 is involved in kinesin-1-dependent transport of the Sra-1/WAVE1 complex and axon formation.

A neuron has two types of highly polarized cell processes, the single axon and multiple dendrites. One of the fundamental questions of neurobiology is how neurons acquire such specific and polarized morphologies. During neuronal development, various actin-binding proteins regulate dynamics of actin cytoskeleton in the growth cones of developing axons. The regulation of actin cytoskeleton in the growth cones is thought to be involved in axon outgrowth and axon-dendrite specification. However, it is largely unknown which actin-binding proteins are involved in axon-dendrite specification and how

[Effect of incubation medium dielectric permeability on enzymatic activity of functionally different ATPases of smooth muscles].

Some organic solvents (2-10%) have been comparatively studied for their effect on purified transporting Ca2+, Mg(2+)-ATPase, solubilized from the plasma membrane of smooth muscle cells and on actomyosine ATPase of the smooth muscle. The inhibiting effect of solvents on the initial maximum specific activity of Ca2+, Mg(2+)-ATPase corresponds to the sequence dioxane > acetone > ethanol > dimethyl sulfoxide (DMSO). Like the case with Ca2+, Mg(2+)-ATPase, dioxane inhibits actomyosine ATPase; acetone, ethanol and DMSO stimulate ATP-hydrolase reaction which is catalyzed by the complex of contractile

Mutagenesis of conserved residues at the yellow fever virus 3/4A and 4B/5 dibasic cleavage sites: effects on cleavage efficiency and polyprotein processing.

Flavivirus proteins are produced by co- and post-translational proteolytic processing of a large polyprotein using both host- and virus-encoded enzymes. The flavivirus serine proteinase, which consists of NS2B and NS3, is responsible for cleavages of at least four dibasic sites in the nonstructural region. In this study, a number of substitutions for the conserved amino acids flanking the 3/4A and 4B/5 dibasic cleavage sites [Arg(P2)-Arg(P1) decreases Gly(P1')] were examined for their effects on yellow fever virus (YF) polyprotein processing. The substrate for these studies was a truncated YF

Stigmasterol suppresses macrophage-mediated inflammatory amplification in acute pancreatitis through PPAR-γ-dependent macrophage reprogramming.

ETHNOPHARMACOLOGICAL RELEVANCE: Glycine max (L.) Merr. (G. max; soybean), a widely cultivated medicinal and edible plant, has been traditionally used in Chinese medicine and dietary practices for maintaining digestive health. Stigmasterol (STG), a naturally occurring dietary phytosterol widely distributed in G. max and other edible and medicinal plants, is investigated here as a bioactive dietary constituent rather than as an isolate from a documented traditional remedy specifically used for acute pancreatitis (AP). STG has attracted increasing attention due to its anti-inflammatory and immuno

Human umbilical cord-derived mesenchymal stem cells alleviate insulin resistance in diet-induced obese mice via an interaction with splenocytes.

BACKGROUND: Previous research has demonstrated that the spleen plays an important role in mesenchymal stem cell (MSC)-mediated alleviation of acute inflammation, as MSC infusion increases the spleen-derived anti-inflammatory cytokine interleukin 10 (IL-10) levels. However, studies on splenic involvement in MSC-induced protection against chronic inflammatory diseases are limited. Obesity is characterized by chronic low-grade inflammation, a key driver of insulin resistance. This study aims to evaluate the effects of MSCs on obesity-related insulin resistance and explore the underlying mechanism

Meisoindigo Protects Against Focal Cerebral Ischemia-Reperfusion Injury by Inhibiting NLRP3 Inflammasome Activation and Regulating Microglia/Macrophage Polarization via TLR4/NF-κB Signaling Pathway.

Ischemic stroke is a devastating disease with long-term disability. However, the pathogenesis is unclear and treatments are limited. Meisoindigo, a second-generation derivative of indirubin, has general water solubility and is well-tolerated. Previous studies have shown that meisoindigo reduces inflammation by inhibiting leukocyte chemotaxis and migration. In the present study, we investigated the hypothesis that meisoindigo was also protective against ischemic stroke, then evaluated its underlying mechanisms. In vivo, adult male C57BL/6J wild-type mice were used to produce a middle cerebral a

Characterization of the Plasmacytoid Dendritic Cell Response to Transmitted/Founder and Nontransmitted Variants of HIV-1.

Human immunodeficiency virus type 1 (HIV-1) infection often arises from a single transmitted/founder (TF) viral variant among a large pool of viruses in the quasispecies in the transmitting partner. TF variants are typically nondominant in blood and genital secretions, indicating that they have unique traits. The plasmacytoid dendritic cell (pDC) is the primary alpha interferon (IFN-α)-producing cell in response to viral infections and is rapidly recruited to the female genital tract upon exposure to HIV-1. The impact of pDCs on transmission is unknown. We investigated whether evasion of pDC r

The role of pro- and anti-inflammatory cytokines in the pathogenesis of spontaneous canine CNS diseases.

Dogs are comparatively frequently affected by various spontaneously occurring inflammatory and degenerative central nervous system (CNS) conditions, and immunopathological processes are a hallmark of the associated neuropathology. Due to the low regenerative capacity of the CNS a sophisticated understanding of the underlying molecular basis for disease initiation, progression and remission in canine CNS diseases represents a prerequisite for the development of novel therapeutical approaches. In addition, as many spontaneous canine CNS diseases share striking similarities with their human count

Cytokine gene expression in innately susceptible BALB/c mice and relatively resistant C57BL/6 mice during infection with virulent Burkholderia pseudomallei.

Production of cytokines including gamma interferon (IFN-gamma) and tumor necrosis factor alpha (TNF-alpha) is an important early-stage host response following infection with intracellular pathogens. Development of immunity to these pathogens is determined to a large extent by the timing and relative level of expression of the cytokines. Numerous studies have shown that early cytokine responses involving interleukin-12 (IL-12) and IFN-gamma are important for resistance to intracellular pathogens, whereas responses involving IL-4 and IL-10 increase host susceptibility. These often-indistinct ear

The tumor microenvironment shapes gastric cancer progression by coordinating immune suppression and metabolic reprogramming.

Gastric cancer (GC) remains a leading cause of cancer mortality, largely owing to metastasis driven by a highly dynamic tumor microenvironment (TME). Immunosuppressive regulatory T cells (Tregs) and tumor-associated macrophages (TAMs) orchestrate immune evasion through checkpoint signaling and polarization programs, while cancer-associated fibroblasts (CAFs) reshape stromal architecture and promote hypoxia. Concurrently, ECM remodeling-mediated by integrins, growth factors, and matrix metalloproteinases-activates oncogenic pathways such as PI3K/AKT/mTOR, MAPK/ERK, and TGF-β to drive disseminat

Chronic stress promotes colorectal cancer progression by inducing M2 macrophage polarization and activating the CCL2-CCR2 axis.

Chronic stress is associated with poor prognosis in colorectal cancer (CRC), but the underlying immune mechanisms remain poorly defined. This study investigates how β-adrenergic signaling, a key component of the stress response, modulates the tumor microenvironment. We demonstrate that stressed CRC patients have elevated serum CCL2, and high tumoral expression of CCL2 and its receptor CCR2 predicts poor survival. Single-cell analysis reveals that within the tumor microenvironment of colorectal cancer, macrophages constitute the primary subset of immune cells responsible for CCL2 secretion. Mec

Frustrated supermolecules: The high-pressure phases of crystalline methane.

Methane is the simplest hydrocarbon, yet it exhibits an extraordinarily complicated series of crystal phases. Notably, the non-plastic phases have large unit cells with nearly, but not quite, cubic symmetry. Furthermore, although non-polar molecules interact very weakly, their reorganization across phase transitions is very sluggish. Here, we demonstrate that these complex structures can be understood as a simple packing of near-spherical supermolecular clusters of methane molecules: the departure from cubic symmetry arising from the non-spherical nature of the molecules. We use molecular dyna

Novel Dual-Pharmacophore TrxR Inhibitors Integrating Ferroptosis and Antitumor Immunity: A Closed-Loop Strategy for TIME Remodeling.

Thioredoxin reductase (TrxR) overexpression in tumors is a marker of poor prognosis for liver cancer. Current TrxR inhibitors primarily feature a single pharmacophore. However, dual-pharmacophore TrxR inhibitors have been rarely reported, lacking a clear report on the relationship between their antitumor and potential regulation of tumor immune microenvironment (TIME). Here, we developed a series of dual-pharmacophore TrxR inhibitors via traditional Chinese medicine-ligand synergistic strategy (TLSS). The superior Fa-Au potently inhibited TrxR, suppressed tumor growth, and remodeled TIME by pr

Targeting Iron Regulatory Protein 2 in Tumor-associated Macrophages Activates Antitumor Immunity in NSCLC via Modulating SphK1-PD-L1 Axis.

Non-small cell lung cancer (NSCLC) is the leading cause of cancer-related mortality worldwide. As a predominant immune cell population in the tumor microenvironment (TME), tumor-associated macrophages (TAMs) play a critical role in tumor progression and resistance to immune checkpoint inhibitors (ICIs). TAMs can promote either antitumor immunity or immunosuppression depending on their polarization status. Iron regulatory protein 2 (IRP2) is a key regulator of intracellular iron homeostasis and has been reported to be upregulated in several tumor types. However, the role of IRP2 in the TME rema

Macrophage-Centric Phenotypic Screening Identifies Tetrazolone-Based HDAC6 Inhibitors That Reprogram the Tumor Immune Microenvironment and Improve Immune Checkpoint Blockade.

Tumor-associated macrophages (TAMs) play a pivotal role in shaping the tumor microenvironment (TME) and influencing the outcomes of immunotherapy. However, most drug screening strategies emphasize tumor cell cytotoxicity and neglect immune effector modulation. Here, we describe a macrophage-centric phenotypic screening platform to identify selective HDAC6 inhibitors that reprogram TAMs toward an antitumor phenotype. Building on the HDAC6 inhibitor SS-208, we synthesized a novel class of tetrazolone-based compounds with potent selectivity and minimal cytotoxicity. Among these, SM-06-09 emerged

In situ-formed multifunctional nanozyme hydrogel for synergistic radio-immunotherapy of osteosarcoma.

Osteosarcoma faces the challenges of limited drug delivery and an immunosuppressive tumor microenvironment. M1 macrophages, a type of tumor-associated macrophage, play a pivotal role in shaping tumor immune responses. A dual enzyme-mimicking nanozyme coordinated to risedronate sodium and encapsulated in a sodium alginate gel (RZRSA) was developed to enhance the radiosensitivity of osteosarcoma. After intratumoral injection, RZRSA formed a calcium alginate hydrogel in osteosarcoma owing to the presence of Ca2+. Because of its peroxidase-like activity, RZRSA generated abundant toxic hydroxyl rad

Invasion dynamics of multifocal breast tumor spheroids in three-dimensional extracellular matrix.

Breast cancer in human patients often occurs as multifocal lesions, and the mechanisms through which primary tumors in proximity interact with each other is not well understood. Here, we study an in vitro model of bifocal breast tumors by embedding tumor spheroids of MDA-MB-231 cells in 3D extracellular matrices (ECMs) made of type I collagen at different concentrations. Combining quantitative experiment, mathematical modeling, and numerical simulation, we show that collective traction force from the spheroids creates dipolar deformation in the ECM. As a result, in the inter-spheroid region, w

Darapladib Ameliorates Radiation-Induced Skin Injury and Fibrosis by Lipoprotein-Associated Phospholipase A2 Inhibition.

Current studies have elucidated the mechanisms of radiation-induced skin injury (RISI) and identified several medical countermeasures to reduce its severity. However, no treatment has yet proven effective in preventing or reversing radiation-induced skin fibrosis. Here, we show that radiation upregulates lipoprotein-associated phospholipase A2 (Lp-PLA2) expression and induces endothelial cell dysfunction, characterized by an increased DNA damage response and reduced tube-forming capacity and mitochondrial function. To define the role of Lp-PLA2 in the progression of RISI, we treated human derm

Targeting peripheral 5-HT2AR enhances antitumor immunity in colorectal cancer.

Cancer remains a leading cause of morbidity and mortality worldwide. While classical psychedelics have been used clinically to treat cancer-associated psychiatric disorders, their impact on tumor progression is unclear. Here, we show that by targeting the serotonin receptor 5-HT2AR, lysergic acid diethylamide (LSD) enhances CD8+ T cell-mediated antitumor immunity and suppresses colorectal cancer (CRC) growth. To harness this activity while avoiding psychedelic effects, we developed IHCH-8110, a non-brain-penetrant 5-HT2AR agonist that selectively targets peripheral 5-HT2AR. We show that IHCH-8

MicroRNAs as multifaceted regulators and therapeutic targets in ulcerative colitis.

Ulcerative colitis (UC) is a chronic inflammatory bowel disease characterized by mucosal barrier disruption, immune dysregulation, and gut microbiota imbalance. MicroRNAs (miRNAs), small non-coding regulators of gene expression, have emerged as pivotal modulators of UC pathogenesis. By orchestrating epithelial cell apoptosis, tight junction integrity, and mucin secretion, miRNAs such as miR-223, miR-151-5p, and miR-429 contribute to barrier dysfunction. Additionally, miRNAs shape the innate and adaptive immune responses by influencing macrophage polarization, dendritic cell maturation, and T c
