Investigation into the degradation mechanism of Cu-Fe/HA biodegradable composite materials based on experiments and simulations.

The degradation behavior and mechanisms of Cu-Fe/HA composites are investigated in this study, combining experimental analysis with cellular automata simulations to elucidate the synergistic mechanism between Cu and Hydroxyapatite (HA) during the degradation process. The results demonstrate that the corrosion of the Fe matrix exhibits periodic characteristics during corrosion, while the addition of Cu significantly influences the degradation performance of the material. In the early immersion stage (30 days), Cu and Fe form a "Fe-Cu galvanic cell", accelerating the corrosion rate of the sample

Label-Free Near-Infrared Image Cytometry for Chlorophyll Quantification and Carotenoid Prediction with Multiparametric Biophysicochemical Profiling.

Microalgae offer a promising platform for carotenoid manufacturing due to their productivity, tunable selectivity, and compatibility with large-scale bioprocessing. Yet, their cellular responses exhibit highly heterogeneous and asynchronous pigment and morphological transitions under fluctuating light stress, making precise prediction and process optimization challenging. Existing analytical methods cannot resolve these transitions in living cells because they either require destructive sampling or lack spectral specificity. In this study, we introduce near-infrared (nIR) image cytometry (NIC)

Molecular Insights into Widespread Pseudouridine RNA Modifications: Implications for Women's Health and Disease.

Pseudouridine (Ψ), the most abundant RNA modification, plays essential roles in shaping RNA structure, stability, and translational output. Beyond cancer, Ψ is dynamically regulated across numerous physiological and pathological contexts-including immune activation, metabolic disorders, stress responses, and pregnancy-related conditions such as preeclampsia-where elevated Ψ levels reflect intensified RNA turnover and modification activity. These broad functional roles highlight pseudouridylation as a central regulator of cellular homeostasis. Emerging evidence demonstrates that Ψ dysregulation

Fibronectin is required for proper extracellular matrix organization and cardiac outflow tract elongation in Xenopus laevis.

INTRODUCTION: Congenital heart defects frequently arise from alterations in cardiac outflow tract (OFT) elongation, a process that depends on the coordinated deployment of second heart field (SHF) progenitor cells and their interactions with the extracellular matrix (ECM). Among ECM components, fibronectin (Fn1) and tenascin-C (TnC) have emerged as key regulators of cardiac morphogenesis; however, the cellular organization of the SHF and the dynamics of its ECM environment remain poorly characterized in externally developing vertebrate models. METHODS: Here, we investigated the cellular and e

Design, synthesis, and biological evaluation of selective HDAC6 inhibitors featuring an N-benzyl-2-(aryl/heteroaryl)acetamide cap.

Histone deacetylase 6 (HDAC6) is predominantly localized in the cytoplasm with nucleocytoplasmic shuttling capability. Featuring two tandem catalytic domains and a C-terminal ubiquitin-binding domain, it primarily deacetylates non-histone substrates such as α-tubulin and HSP90. This distinct subcellular localization and substrate profile endow HDAC6 with the unique capacity to orchestrate pivotal roles in cytoskeletal regulation, the cellular stress response, protein degradation, and cell migration, thereby positioning it as a highly attractive target for anti-tumor drug development. Design, synthesis, and screening of novel thienopyrimidine derivatives as PI3K/HDAC dual inhibitors against cervical cancer cells.

Phosphatidylinositol 3-kinase (PI3K) and histone deacetylase (HDAC) inhibition is a potential tumor treatment strategy, including for cervical cancer. Herein, a novel series of thienopyrimidine derivatives was rationally designed, synthesized, and biologically evaluated as dual PI3K/HDAC inhibitors targeting cervical cancer. Compound B7 was evaluated for its inhibitory effects across the entire PI3K and HDAC families, and exhibited strong PI3Kα (IC50 = 25.41 ± 5.23 nM), PI3Kδ (IC50 = 41.58 ± 4.54 nM), HDAC6 (IC50 = 29.35 ± 3.41 nM), and moderate HDAC8 inhibition (IC50 = 157.27 ± 6.18 nM). The effect of surface modification of ultrasmall palladium nanoparticles (Pd NPs) with polyethylene glycol and glucose on cellular uptake and radiosensitizing potential - Comprehensive in vitro and molecular dynamics studies.

Glioblastoma multiforme (GBM) remains one of the most aggressive and radioresistant brain tumors, highlighting the urgent need for effective radiosensitizers compatible with proton radiotherapy (PRTH). Ultrasmall (< 5 nm) palladium nanoparticles (Pd NPs) were synthesized and subsequently surface-functionalized with thiolated polyethylene glycol (PEG-SH) and thiolated glucose (Glu-SH). The influence of surface chemistry on biological interactions and radiosensitizing performance was then investigated. Structural characterization confirmed the formation of highly crystalline, monodisperse Pd NPs

Chrono-mechanobiology: The complex interactions between mechanotransduction and circadian rhythms in the intervertebral disc and articular cartilage, in health and disease.

The intervertebral disc (IVD) and the articular cartilage (AC) are specialized, load-bearing tissues critical for spinal flexibility and joint mobility, respectively. Both tissues are characterized by their avascular nature and abundant extracellular matrix (ECM). They heavily rely on precisely regulated anabolic and catabolic processes to maintain structural integrity and functional performance. Disturbances can contribute to IVD degeneration and osteoarthritis that represent leading global causes of disability and pose substantial challenges to healthcare systems worldwide. Cellular and molecular pathways linking obesity to skeletal muscle dysfunction.

Obesity is increasingly recognized as a condition that directly impairs skeletal muscle structure, metabolism, and endocrine function through complex molecular and cellular mechanisms extending beyond the classical concept of sarcopenic obesity. This narrative review aimed to synthesize current evidence regarding the intracellular signaling pathways, metabolic alterations, and endocrine interactions involved in obesity-induced skeletal muscle dysfunction independent of overt sarcopenia. Relevant literature from experimental, clinical, and review studies was identified through searches of PubMe

Tetrahedral framework nucleic acids loaded with microRNA-17 for treating temporomandibular joint osteoarthritis: mechanisms of action.

Temporomandibular joint osteoarthritis (TMJOA) is a degenerative joint disease involving cartilage degeneration, synovial inflammation, and subchondral bone remodeling. Current treatments are primarily palliative, lacking precise therapies. MicroRNA-based gene therapy is promising, but free miRNAs are limited by rapid degradation and poor cellular uptake. Given its marked downregulation in osteoarthritic chondrocytes and its multi-target capacity to concurrently suppress multiple matrix-degrading enzymes and inflammatory mediators, miR17 was selected as the therapeutic payload. Tetrahedral fra

Myosin VI is involved in the structural organization and molecular composition of epididymal epithelial cells in mouse†.

Myosin VI (MYO6) is the only actin-based motor protein that moves toward the minus end of actin filaments. It participates in multiple cellular processes, including endocytosis, secretion, autophagy, and the formation of apical stereocilia and microvilli in highly specialized epithelia. These diverse functions are mediated by specific cargo-adaptor proteins that recruit MYO6 to distinct cellular compartments. We have previously demonstrated that loss of MYO6 function in Snell's waltzer mice leads to several defects during spermatogenesis, resulting in reduced male fertility. Here, we show for
