Capsid assembly modulators (CAMs) are a novel class of antiviral agents in clinical development for the treatment of chronic hepatitis B. CAMs inhibit hepatitis B virus (HBV) replication by binding to a hydrophobic pocket, i.e., HAP pocket, between HBV capsid protein (Cp) dimer-dimer interfaces to misdirect its assembly into empty capsids or aberrant structures and designated as CAM-E and CAM-A, respectively. Because the emergence of CAM-resistant variants results in the failure of antiviral therapy, it is important to rationally design CAMs with a high barrier of resistance for development. T

Liver injury is a globally serious problem that may be observed post-chemotherapeutic administration in chronic crises such as tuberculosis (TB). Rifamycin (RIF), particularly, is an indispensable treatment regimen for TB with a significant negative hepatic impact. Therefore, this research aims to assess the restorative role of melatonin (MEL) against rifampicin (RIF)-associated hepatic damage in rats. Moreover, to investigate the ultimate mechanism of the antioxidant potential of MEL with multimodal assessment and in silico molecular conformation. Adult male Wistar albino rats were weighed an

ADCK3 is a member of the UbiB family of atypical protein kinases in humans, with homologues in archaea, bacteria, and eukaryotes. In lieu of protein kinase activity, ADCK3 plays a role in the biosynthesis of coenzyme Q10 (CoQ10), and inactivating mutations can cause a CoQ10 deficiency and ataxia. However, the exact functions of ADCK3 are still unclear, and small-molecule inhibitors could be useful as chemical probes to elucidate its molecular mechanisms. In this study, we applied structure-based virtual screening (VS) to discover a novel chemical series of ADCK3 inhibitors. Through extensive s

Plant-based diets (PBDs) are renowned for managing and developing bioactive chemical inhibitors to combat obesity, a well-known global public health concern. There are currently no published research studies examining the effects of food plant mucilage dietary supplements on animal models of obesity induced by high-fat diets (HFD). The present research investigated the anti-obesity properties of the culinary plant Pedalium murex L. mucilage (PMM) in obese albino male rats models fed HFD. PMM's HR-LCMS phytochemical profiling and in silico evaluation of anti-obesity and drug-likeness using Schr

Monkeypox, a viral zoonotic disease resembling smallpox, has emerged as a significant national epidemic primarily in Africa. Nevertheless, the recent global dissemination of this pathogen has engendered apprehension regarding its capacity to metamorphose into a sweeping pandemic. To effectively combat this menace, a multi-epitope vaccine has been meticulously engineered with the specific aim of targeting the cell envelope protein of Monkeypox virus (MPXV), thereby stimulating a potent immunological response while mitigating untoward effects. This new vaccine uses T-cell and B-cell epitopes fro

Nicotinamide-nucleotide adenylyl transferase (Rv2421c) was selected as a potential drug target, because it has been shown, in vitro, to be essential for Mycobacterium tuberculosis growth. It is conserved between mycobacterium species, is up-regulated during dormancy, has a known 3D crystal structure and has no known human homologs. A model of Rv2421c in complex with nicotinic acid adenine dinucleotide and magnesium ion was constructed and subject tovirtual ligand screening against the Prestwick Chemical Library and the ZINC database, which yielded 155 potential hit molecules. Of the 155 compou

A new series of N,N-dimethylbarbituric-pyridinium derivatives 7a-n was synthesized and evaluated as Helicobacter pylori urease inhibitors. All the synthesized compounds (IC50 = 10.37 ± 1.0-77.52 ± 2.7 μM) were more potent than standard inhibitor hydroxyurea against urease (IC50 = 100.00 ± 0.2 μM). Furthermore, comparison of IC50 values of the synthesized compounds with the second standard inhibitor thiourea (IC50 = 22.0 ± 0.03 µM) revealed that compounds 7a-b and 7f-h were more potent than thiourea. Molecular modeling study of the most potent compounds 7a, 7b, 7f, and 7g was also conducted. Ad

Human glutaminyl cyclase (hQC) appeared as a promising new target with its inhibitors attracted much attention for the treatment of Alzheimer's disease (AD) in recent years. But so far, only a few compounds have been reported as hQC inhibitors. To find novel and potent hQC inhibitors, a high-specificity ZBG (zinc-binding groups)-based pharmacophore model comprising customized ZBG feature was first generated using HipHop algorithm in Discovery Studio software for screening out hQC inhibitors from the SPECS database. After purification by docking studies and drug-like ADMET properties filters, f

Huntington's disorder (HD), caused by mutations of the IT-15 gene, is an autosomal genetic disease that causes the breakdown of the nerve cells in the brain. The IT-15 gene encodes the huntingtin (Htt) protein. Htt, along with its interacting partners, are involved in maintaining proper communication among neurons. Our work is based on the interaction behavior between Htt (in three polyglutamine (polyQ) states that is Htt 0Q, 17Q and 36Q) and SH3GL3 interacting protein by using computational methods. We used the HADDOCK docking platform to find out the extent of interaction between Htt polyQ m

The ultimate goal of the present work is to aid in the development of tools to assist in the treatment of cardiovascular disease. Gaining an understanding of hemodynamic parameters for medical implants allow clinicians to have some patient-specific proposals for intervention planning. In the present work an experimental and digital computational fluid dynamics (CFD) arterial model consisting of a number of major arteries (aorta, carotid bifurcation, cranial, femoral, jejunal, and subclavian arteries) were fabricated to study: (1) the effects of local hemodynamics (flow parameters) on global he

Eye-tracking is a key enabling technology for smart eyewear, supporting hands-free interaction, accessibility, and context-aware human-machine interfaces under strict constraints on size, power consumption, and computational complexity. While camera-based solutions provide high accuracy, their integration into lightweight and low-power wearable platforms remains challenging. This paper is a feasibility study for the design, simulation, and experimental evaluation of a photosensor oculography (PSOG) eye-tracking system that is fully integrated into an eyewear frame, based on near-infrared (NIR)

High-resolution hyperspectral computational imaging is critical for applications such as environmental monitoring, urban planning, and precision agriculture. In practical hyperspectral imaging systems, physical hardware constraints inevitably lead to coupled degradations across spatial and spectral dimensions, making it difficult to simultaneously achieve high spatial resolution and high spectral fidelity. As a representative and widely studied hyperspectral computational imaging task, hyperspectral pansharpening aims to reconstruct high-resolution hyperspectral images by integrating low-resol

Epilepsy is a debilitating neurological disorder that impacts approximately 50 million people worldwide. The treatment of epilepsy with antiepileptic drugs has not achieved effective seizure management and thus requires new therapeutic options. This study investigated the catechins' affect on epilepsy-related molecular targets using a computational method that combined network pharmacology, molecular docking, and molecular dynamics (MDs) simulation. We fetched 84 catechins-related and 5356 disease-associated targets from various databases, yielding 31 common targets. The protein-protein intera

The Skp2-Cks1 protein-protein interaction is essential for SCFSkp2/Cks1-mediated recognition and ubiquitination of cell-cycle regulators, making this interface an attractive target for anticancer drug discovery. In this study, we identified new small-molecule Skp2-Cks1 disruptors from a focused [1,2,4]triazolo[1,5-a]pyrimidine library using an integrated computational and experimental workflow. A scaffold-based set of 40 compounds was filtered by diversity, purchasability, and drug-likeness, followed by docking into the Skp2-Cks1 interfacial pocket. Five hits (C1-C5) were prioritized and furth

OBJECTIVE: Arteriovenous fistula (AVF) is the gold standard for hemodialysis access, but traditional dual-needle cannulation is limited by a restricted cannulation area and progressive stenosis; single-puncture systems may reduce these risks if hemodynamics can be optimized to prevent recirculation. This study aimed to design a novel single-puncture dual-lumen coaxial catheter and to evaluate its hemodynamic performance, flow efficiency, and recirculation rate using high-resolution computational fluid dynamics (CFD).
METHODS: In this CFD and design-optimization study, a dual-lumen coaxial cat

BACKGROUND: Postinfarction ventricular tachycardia (VT) arises from reentrant circuits within partially conductive myocardial tissue. Despite antiarrhythmic medication, many patients require ablation therapy, yet recurrence rates remain as high as 50% at 5 years. Virtual-heart arrhythmia ablation targeting (VAAT) methods aim to improve outcomes by simulating VT activity preoperatively, but they are computationally intensive and often lack validation against clinical VT and electrocardiographic (ECG) data.
OBJECTIVE: This study aimed to optimize a VAAT pipeline for speed and clinical relevance

CONTEXT: Down syndrome is a genetic condition caused by trisomy of chromosome 21, leading to intellectual and physical disabilities. Overexpression of the dual-specificity tyrosine-phosphorylation-regulated kinase 1A (DYRK1A) gene, located on chromosome 21, plays a critical role in neurodevelopmental abnormalities and synaptic dysfunction associated with the disorder. Identifying regulatory mechanisms capable of suppressing DYRK1A expression represents a promising therapeutic strategy.
METHOD: In this study, a consensus-based computational pipeline was employed to identify microRNAs (miRNAs)

Human immunodeficiency virus (HIV) is one of the viruses that has co-evolved within human populations for a considerable time. With the evolution of new drug-resistant HIV strains, the necessity to discover novel drugs has become an issue of great concern, especially those that have increased binding affinities and inhibitory activity towards RT enzymes. This study used an in silico approach to identify potential HIV-RT inhibitory candidates from Origanum vulgare (oregano). An initial in silico screening of approximately 820 compounds was conducted, and based on molecular docking-derived bindi
