Urinary free cortisone and the assessment of 11 beta-hydroxysteroid dehydrogenase activity in man.

OBJECTIVE: Two isoforms of 11 beta-hydroxysteroid dehydrogenase (11 beta-HSD) catalyse the interconversion of cortisol to hormonally inactive cortisone; defects in the 11 beta-HSD2 isoform result in hypertension. The kidney, expressing high levels of 11 beta-HSD2, is the principal source of cortisone in man. We have validated the measurement of urinary free cortisone (UFE) excretion in normals and in patients with disorders of the pitultary-adrenal axis in an attempt to more accurately measure the activity of 11 beta-HSD2 in vivo. SUBJECTS: Forty-one normal adults, 12 normal children < 12 yea

Investigating Bidirectional Causal Relationships Between Imaging-Derived Brain Phenotypes and Sedative-Hypnotic Use Disorder: A Mendelian Randomization Study.

Despite sedative-hypnotic use disorder (SHUD) being a major public health priority due to its high abuse liability, the underlying causal neurobiological mechanisms of individual susceptibility remain largely unknown. We conducted a large-scale, bidirectional two-sample Mendelian randomization (MR) study utilizing genome-wide association study (GWAS) summary statistics for 3935 brain imaging-derived phenotypes (IDPs) from the UK Biobank (N = 33 000 participants) and SHUD data from the FinnGen R10 release (N = 2258 cases). Causal effects were primarily estimated using the inverse-variance weigh

Prepartum organic selenium supplementation influences postpartum inflammatory response by regulating plasma oxylipin profiles in periparturient dairy cows.

Excessive lipolysis and enhanced inflammation in periparturient dairy cows may lead to metabolic disorders and diseases. Membrane phospholipids and lipid mobilisation-released polyunsaturated fatty acids (PUFAs) can be oxidised into oxylipins that have pro- and/or anti-inflammatory roles. Selenium has immunomodulatory and anti-inflammatory effects, but the mechanisms are not yet fully understood. The objective of this study was to examine the effects of prepartum selenium-yeast (SY) supplementation on the inflammatory response in dairy cows during the periparturient period by regulating plasma

Early ACL Reconstruction in Children With Congenital ACL Deficiency: A Prospective Case Series.

BACKGROUND: Congenital absence or hypoplasia of the anterior cruciate ligament (ACL) is an uncommon condition. Children with this anomaly frequently present with knee instability, altered gait patterns, and may develop progressive deformities or early degenerative changes if untreated. Management remains debated, as some patients compensate well functionally, while some ACL reconstructions in young, skeletally immature children pose a potential risk to open physes. Conversely, early surgical stabilization may help protect the developing knee and support future reconstructive procedures, includ

The role of obstetric events and care-related factors in perceived threat and childbirth-related post-traumatic stress symptoms.

BACKGROUND: Childbirth is a complex event where the quality of care may profoundly be linked to women's experiences and subsequent psychological wellbeing. Adverse childbirth experiences can be associated with childbirth-related post-traumatic stress disorder. Conversely, positive care experiences, specifically supportive interactions and respectful treatment, may act as a protective factor associated with maternal resilience. AIM: This study aimed to investigate how medical events during childbirth, including birth complications and interventions, align with the quality of childbirth care. I

Glyceraldehyde-3-phosphate dehydrogenase/1,3-bisphosphoglycerate-NADH as key determinants in controlling human retinal endothelial cellular functions: Insights from glycolytic screening.

Maintaining barrier integrity, along with cell adhesion to the extracellular matrix and the subsequent process of cell spreading, are essential functions of endothelial cells, including human retinal endothelial cells (HRECs). Disruptions in these processes can lead to vision-threatening conditions like diabetic retinopathy. However, the bioenergetic mechanisms that regulate HREC barrier function and cell spreading remain incompletely understood. This study investigates the role of lower glycolytic components in modulating these critical functions of HRECs. In vitro, Electric Cell-Substrate Im

Neuropsychiatric symptoms and apolipoprotein E genotypes in neurocognitive disorders.

Complex genetic relationships between neurodegenerative disorders and neuropsychiatric symptoms have been shown, suggesting shared pathogenic mechanisms and emphasizing the potential for developing common therapeutic targets. Apolipoprotein E ( APOE ) genotypes and their corresponding protein (ApoE) isoforms may influence the biophysical properties of the cell membrane lipid bilayer. However, the role of APOE in central nervous system pathophysiology extended beyond its lipid transport function. In the present review article, we analyzed the links existing between APOE genotypes and the neurob

Mechanistic Insights into Systemic Targets of Magnolia Mediating Beneficial Effects in Metabolic Syndrome through Biochemical and In Situ Analyses.

Metabolic syndrome (MetS) is characterized by a cluster of biochemical and physiological abnormalities. Healthy eating, including the consumption of natural products, holds promise for promoting health and preventing disease. Among these, Magnolia officinalis bark extract (MAGBE) is widely used as a dietary supplement due to its potent antioxidant, anti-inflammatory, and neuroprotective properties. In this study, we aimed to investigate the therapeutic potential of MAGBE in a rat model of MetS and to identify its target proteins. One group of 2-month-old male Wistar rats was supplemented with
