"In vivo" murine macrophages activation by a dichloromethane extract of Tilia x viridis.

Macrophages are involved in the host defense against infectious pathogens and tumors. Tilia species have been used in folk medicine for the treatment of infectious diseases, previously it was demonstrated that a dichloromethane (DM) extract possess antiproliferative action "in vitro" on a lymphoma cell line. The aim of this work was to study the "in vivo" effect of DM extract upon mice peritoneal macrophages. DM extract-activated macrophages phagocytosis through hydrogen peroxide (H(2)O(2)) and nitric oxide (NO) production (phagocytosis (%): basal 16.93 +/- 0.18, DM extract 25.93 +/- 2.8; H(2)

Complicated urinary tract infections: practical solutions for the treatment of multiresistant Gram-negative bacteria.

Resistance in Gram-negative bacteria has been increasing, particularly over the last 6 years. This is mainly due to the spread of strains producing extended-spectrum β-lactamases (ESBLs) such as CTX-M enzymes or AmpC β-lactamases. Many of the isolates producing these enzymes are also resistant to trimethoprim, quinolones and aminoglycosides, often due to plasmid co-expression of other resistance mechanisms. CTX-M-producing Escherichia coli often occurs in the community and as E. coli is one of the commonest organisms causing urinary tract infections (UTIs) the choice of agents to treat these i

Characterization of enteroglial cells and denervation process in chagasic patients with and without megaesophagus.

Chagas disease is caused by infestation with the parasite Trypanosoma cruzi, and some patients who are serologically positive develop chronic megaesophagus, whereas others are symptom-free. Gastrointestinal form of Chagas disease involves an inflammatory invasion of the enteric plexuses and degeneration of enteric neurons and previous works related that enteroglial cells would be involved in enteric inflammatory responses. Because of this, the aims of this study were to determine the relation of enteroglial cells with the denervation process in chagasic patients with and without megaesophagus

A dicyanotriterpenoid induces cytoprotective enzymes and reduces multiplicity of skin tumors in UV-irradiated mice.

Inducible phase 2 enzymes constitute a primary line of cellular defense. The oleanane dicyanotriterpenoid 2-cyano-3,12-dioxooleana-1,9(11)-dien-28-onitrile (TP-225) is a very potent inducer of these systems. Topical application of TP-225 to SKH-1 hairless mice increases the levels of NAD(P)H-quinone acceptor oxidoreductase 1 (NQO1) and heme oxygenase 1 (HO-1) and protects against UV radiation-induced dermal thickening. Daily topical treatments of 10 nmol of TP-225 to the backs of mice that were previously subjected to low-level chronic UVB radiation (30 mJ/cm(2)/session, twice a week for 17 we

Participation of epidermal langerhans cells in human pathology and their potential as targets for drug development: a review of literature.

Langerhans cells (LC) are antigen presenting cells of the epidermis originated from bone marrow progenitors that arrive into the epidermis through the blood vessels LC are also referred to as dendritic cells. In the presence of antigens LC become activated and migrate from the skin to the lymph nodes where they induce T cells responses, therefore, LC function as sentinels of the epidermis and constitute, in part the Skin Immune System (SIS). LC have been implicated in the pathogenesis and pathophysiology of diverse diseases such as atopic dermatitis, alopecia areata, human immunodeficiency vir

Perinatal exposure to nicotine causes deficits associated with a loss of nicotinic receptor function.

We investigated the role played by beta2-containing neuronal nicotinic receptors [nicotinic acetylcholine receptors (nAChRs)] in mediating nicotine's side effects in the fetus and newborn. Pregnant WT and mutant mice lacking the beta2 nAChR subunit were implanted with osmotic minipumps that delivered either water or a controlled dose of nicotine. Subsequently, we compared the development of the sympathoadrenal system and breathing and arousal reflexes of offspring shortly after birth, a period of increased vulnerability to nicotine exposure. Newborn WT pups exposed to nicotine exhibited all of

Small-molecule inhibition of siderophore biosynthesis in Mycobacterium tuberculosis and Yersinia pestis.

Mycobacterium tuberculosis and Yersinia pestis, the causative agents of tuberculosis and plague, respectively, are pathogens with serious ongoing impact on global public health and potential use as agents of bioterrorism. Both pathogens have iron acquisition systems based on siderophores, secreted iron-chelating compounds with extremely high Fe3+ affinity. Several lines of evidence suggest that siderophores have a critical role in bacterial iron acquisition inside the human host, where the free iron concentration is well below that required for bacterial growth and virulence. Thus, siderophore

NEUROPEPTIDES AND NEUROGENIC MECHANISMS IN ORAL AND PERIODONTAL INFLAMMATION.

It is generally accepted that the nervous system contributes to the pathophysiology of peripheral inflammation, and a neurogenic component has been implicated in many inflammatory diseases, including periodontitis. Neurogenic inflammation should be regarded as a protective mechanism, which forms the first line of defense and protects tissue integrity. However, severe or prolonged noxious stimulation may result in the inflammatory response mediating injury rather than facilitating repair. This review focuses on the accumulating evidence suggesting that neuropeptides have a pivotal role in the c

Listeria monocytogenes: clinical and experimental update.

Listeria monocytogenes, a small gram-positive bacillus, causes sepsis and meningitis in immunocompromised patients and a devastating maternal/fetal infection in pregnant women. Recent outbreaks demonstrated that L. monocytogenes can cause gastroenteritis in otherwise healthy individuals and more severe invasive disease in immunocompromised patients. Centralized processing in the food industry may be the cause of these large-scale listeriosis outbreaks. The mouse model of listeriosis, which was developed in the 1960s, has been extraordinarily useful for studying T cell-mediated immunity. Contra

Interleukin-1beta and rhinovirus sensitize adenylyl cyclase in human airway smooth-muscle cells.

Rhinovirus (RV) is a major cause of wheezing in asthmatics and has been reported to cause beta2 adrenergic receptor hyporesponsiveness in human airway smooth muscle (HASM) via cellular secretion of interleukin (IL)-1beta. We studied the effects of IL-1beta and RV on cyclic adenosine monophosphate (cAMP) production in HASM cells. Chronic incubation with IL-1beta or RV caused a significant increase (approximately 3- and approximately 2-fold, respectively) in forskolin (FSK)-stimulated cAMP production, suggesting a sensitization of adenylyl cyclase (AC). The observed augmentation of FSK-stimulate

Expression and purification of Streptococcus pneumoniae hyaluronate lyase from Escherichia coli.

Pneumococcal hyaluronate lyase enzyme breaks down hyaluronan of the extracellular matrix of tissues and possibly contributes to the invasion of host tissue and to the penetration of host defenses by this bacterial pathogen. In light of the emergence of increasing numbers of antibiotic-resistant strains, the understanding of the mechanism of action of hyaluronate lyase enzyme may lead to a better understanding of interactions between a host and bacterial pathogens and may contribute to more efficient treatment of bacterial infections. The native Streptococcus pneumoniae hyaluronate lyase enzyme

Evidence that tumor necrosis factor has an important role in antibacterial resistance.

TNF is produced in the spleens of Listeria-infected mice during the first 3 days of a sublethal immunizing infection. The production of Listeria-induced TNF coincides with the time when peak numbers of bacteria are present in the liver and spleen. Evidence suggesting that TNF produced in Listeria-infected organs functions in a T cell-independent resistance mechanism comes from results showing that listeriosis is exacerbated in both T cell-intact mice and T cell-deficient (athymic nude) mice treated with a monospecific anti-murine TNF IgG. Listeriosis is exacerbated, however, only if the anti-T

Trichinella spiralis: identification and purification of superoxide dismutase.

A metalloprotein with superoxide dismutase activity was isolated and purified from muscle-stage Trichinella spiralis. The anti-genicity of the purified enzyme was demonstrated by an immunospecific reaction with T. spiralis antiserum in an enzyme-linked immunosorbent assay. In addition to its presence in somatic extracts of T. spiralis, the enzyme was also excreted into culture fluids in which the muscle-stage larvae had been incubated for periods as short as 3 hr and up to 72 hr. The enzyme was characterized as a copper- and zinc-containing, cyanide-sensitive, superoxide dismutase with a molec

Effect of oral pancreatic enzymes on the course of cholera in protein-deficient vervet monkeys.

The effect of pancreatic extract on the course of cholera infection in protein-deficient vervet monkeys (Cercopithecus aethiops) was studied. Eleven animals were made protein deficient by diet. Thier intestinal pancreatic enzyme concentration decreased as they became protein depleted. These animals were then challenged with Vibrio cholerae. Four control animals given a standard diet were similarly challenged. Immediately after challenge, 6 of the protein-deficient monkeys were given a highly purified pancreatic extract at a dose of 500 mg per day, and the other 5 protein-deficient animals rece

Prostatic antibacterial factor. Identity and significance.

Normal human prostatic fluid possesses pronounced antibacterial activity. This activity is absent or markedly diminished in the fluid of men with chronic bacterial prostatitis. Ion-probe and mass spectrographic analysis of this antibacterial factor has shown it to be a zinc salt. Prostatic fluid zinc levels in 15 men with chronic bacterial prostatitis averaged 50 mug. per milliliter (range 0 to 139 mug. per milliliter). The zinc level in the expressed prostate secretion (EPS) of 49 control men averaged 448 mug. per milliliter (range 150 to 1,000 mug. per milliliter). There was no overlap in th

ATP release and metabolism by human and guinea pig airways and P2X receptor-dependent effects on bronchopulmonary vagal afferent nerve subtypes.

BACKGROUND AND PURPOSE: ATP-dependent activation of P2X receptors has been implicated in chronic cough. The mechanisms of ATP release in cough and how P2X-receptor engagement produces cough hypersensitivity are not clear. It is also unclear whether P2X receptor blockade could compromise the essential defensive functions of cough regulated by the vagal Aδ-fibres terminating in the extrapulmonary airways. EXPERIMENTAL APPROACH: We measured ATP release from isolated human and guinea pig airways. P2X receptor-dependent regulation of vagal afferent nerve discharge in vitro and respiratory reflexes

Deciphering pUL10's mastery in duck plague virus virulence: dual coordination of host immunity and viral replication.

Interferon (IFN) critically regulates antiviral immunity, wherein its production level determines disease progression, many viruses disrupt these defenses through mechanisms that remain incompletely understood. This study investigates the effect of duck plague virus (DPV) infection on immune organs and elucidates the molecular mechanism by which its UL10 protein (pUL10) inhibits melanoma differentiation-associated protein 5 (MDA5)-mediated IFN-β production. DPV UL10 gene encodes pUL10, a virulence-associated protein critical for viral pathogenicity. The conserved cysteine residue C54 serves as

The antioxidant effect of preischemic dexmedetomidine in a rat model: increased expression of Nrf2/HO-1 via the PKC pathway.

BACKGROUND: The precise underlying mechanism of antioxidant effects of dexmedetomidine-induced neuroprotection against cerebral ischemia has not yet been fully elucidated. Activation of Nuclear factor erythroid 2-related factor (Nrf2) and Heme Oxygenase-1 (HO-1) represents a major antioxidant-defense mechanism. Therefore, we determined whether dexmedetomidine increases Nrf2/HO-1 expression after global transient cerebral ischemia and assessed the involvement of Protein Kinase C (PKC) in the dexmedetomidine-related antioxidant mechanism. METHODS: Thirty-eight rats were randomly assigned to fiv

Does yawning increase arousal through mechanical stimulation of the carotid body?

Yawning is a stereotyped event that occurs in humans and animals from fish to mammals, but neither its mechanisms nor its functions are entirely known. Its widespread nature suggests that it has important physiological functions. It is associated with stretching of muscles in a large area, but the function of this stretching is understood far from completely. It has been proposed that yawning increases arousal and that it is an arousal defense reflex, whose aim is to reverse brain hypoxia. Whilst yawning has been speculated to have an important role in reversing hypoxia, there is a structure i
