Pharmacotherapy of amphetamine-type stimulant dependence: an update.

ISSUES: Methamphetamine- or amphetamine-type stimulants are the second most frequently used illicit drug worldwide, second only to cannabis. Behavioural treatments are efficacious, but their impact is limited underscoring the need for other treatment options, notably, pharmacotherapy. APPROACH: A review of randomised controlled trials of pharmacotherapies for methamphetamine- or amphetamine-type stimulants was performed using PubMed and Google Scholar databases. Evidence for efficacy of medications is reported. KEY FINDINGS: Clinical trials have yielded no broadly effective pharmacotherapy.

Decreased frontal white-matter volume in chronic substance abuse.

There is quite a body of work assessing functional brain changes in chronic substance abuse, much less is known about structural brain abnormalities in this patient population. In this study we used magnetic resonance imaging (MRI) to determine if structural brain differences exist in patients abusing illicit drugs compared to healthy controls. Sixteen substance abusers who abused heroin, cocaine and cannabis but not alcohol and 16 age-, sex- and race-matched controls were imaged on a MRI scanner. Contiguous, 5-mm-thick axial slices were acquired with simultaneous T2 and proton density sequenc

Epidemiology of Depressive Disorders in Patients With Liver Cirrhosis: A Population-Based Study in the United States.

Objective: Major depressive disorder (MDD) is a chronic, debilitating mood disorder associated with poor medical outcomes. MDD has a multifactorial etiology with numerous biopsychosocial factors implicated as risk factors. Functional and psychiatric impairments have been evaluated in patients with liver cirrhosis; however, less is known about the prevalence and risk factors for the development of MDD in those patients. The objective of this study was to evaluate the risk of developing depression among adult patients with liver cirrhosis in the United States. Methods: Data were collected using

Trends in treatment attendance for substance use disorders among adolescents and emerging adults in Australia, 2003-2020.

BACKGROUND: Substance use disorders frequently begin in adolescence and emerging adulthood. Knowledge of trends in alcohol and other drug (AOD) service utilisation during these periods can inform policy development to align service delivery with treatment need. AIMS: Characterise trends in AOD treatment episodes among adolescents (aged 10-17) and emerging adults (aged 18-24) across Australia, and compare to adult trends (aged ≥25). METHODS: National data from the Alcohol and Other Drug Treatment Services - National Minimum Data Set were analysed (N = 2904,012). Joinpoint regression analysis

Comparative analysis of five correction methods for thyroid volume by ultrasound and their recommended reference values in Chinese children aged 8-10 years.

Considering the influence of body's growth and development on thyroid volume (TVOL), whether five existed corrected methods could be applied to correct TVOL remains unclear, in terms of Chinese children's increased growth and development trends. This study aimed to compare the applicability of five correction methods: Body Surface Area corrected Volume (BSAV), Body Mass Indicator corrected Volume (BMIV), Weight and Height corrected Volume Indicator (WHVI), Height corrected Volume Indicator 1 (HVI1) and Height corrected Volume Indicator 2 (HVI2) and to establish the reference values for correct

Brain-derived neurotrophic factor as a biomarker for obsessive-compulsive disorder: A meta-analysis.

BACKGROUND: Brain-derived neurotrophic factor (BDNF) is a growth factor that plays many critical functions in the central nervous system (CNS) and may be involved in the development of a range of psychopathologies, including depression, dementia, and neurodegenerative disorders. METHODS: In the present study, we performed the first systematic review with a meta-analysis to quantitatively compare the peripheral blood BDNF levels between patients with obsessive-compulsive disorder (OCD) and healthy controls (HCs). A systematic search was conducted using PubMed and Web of Science databases to id

White matter microstructure and macrostructure brain charts across the human lifespan.

Normative reference charts are widely used in healthcare, especially for assessing the development of individuals by benchmarking anatomic and physiological features against population trajectories across the lifespan. Recent work has extended this concept to gray matter morphology in the brain, but no such reference framework currently exists for white matter (WM) even though WM constitutes the essential substrate for neuronal communication and large-scale network integration. Here, we present the first comprehensive WM brain charts, which describe how microstructural and macrostructural feat

Maternal Paracetamol Intake During Pregnancy-Impacts on Offspring Reproductive Development.

Paracetamol (acetaminophen) is the preferred antipyretic/analgesic for pregnant women as it is believed there are no adverse fetal effects at the recommended dose. However, emerging evidence suggests that intrauterine paracetamol exposure may be associated with certain urogenital/reproductive disorders in the offspring. In this mini-review, we describe human fetal sex development and possible pharmacological mechanisms by which paracetamol may disrupt this process, including reduced testicular production of testosterone and/or insulin-like peptide 3. We then review the available epidemiologica

Association of childhood trauma with cognitive function in healthy adults: a pilot study.

BACKGROUND: Animal and human studies suggest that stress experienced early in life has detrimental consequences on brain development, including brain regions involved in cognitive function. Cognitive changes are cardinal features of depression and posttraumatic stress disorder. Early-life trauma is a major risk factor for these disorders. Only few studies have measured the long-term consequences of childhood trauma on cognitive function in healthy adults. METHODS: In this pilot study, we investigated the relationship between childhood trauma exposure and cognitive function in 47 healthy adult

Mesocorticolimbic connectivity and motivational sensitivity: sex-specific effects of puberty in early adolescence.

Puberty demarcates the start of adolescence, a critical period of marked changes in motivated behavior (e.g., approach, avoidance) and socio-affective processing supported by development of the mesocorticolimbic circuitry-prefrontal cortex (PFC), amygdala, and nucleus accumbens (NAcc). Puberty-related increases in psychiatric risk have been linked to alterations in mesocorticolimbic circuitry function and sensitivity to rewards and punishments. Yet, how puberty influences the development of mesocorticolimbic circuitry supporting motivational traits remains unclear. We examined resting-state fu

Identification of a Rare Variant in the SRD5A2 Gene in Siblings With 46,XY Disorders of Sexual Development.

The SRD5A2 gene encodes the steroid 5α-reductase-2 isozyme, which converts testosterone to dihydrotestosterone and plays a key role in sexual development and androgen physiology. Deficiency of this enzyme leads to an autosomal recessive sex-linked disorder associated with ambiguous genitalia and hypovirilization/complete feminization of external genitalia in individuals with a 46,XY karyotype. The present study emphasizes the indispensable role of molecular genetic testing in siblings (Probands 1 and 2) presented with disorders of sexual development (DSD). Long-term outcomes after adolescent in-patient treatment due to alcohol intoxication: A control group study.

BACKGROUND: The long-term psychosocial development of adolescents admitted to in-patient treatment with alcohol intoxication (AIA) is largely unknown. METHODS: We invited all 1603 AIAs and 641 age- and sex-matched controls, who had been hospitalized in one of five pediatric departments between 2000 and 2007, to participate in a telephone interview. 277 cases of AIA and 116 controls (mean age 24.2 years (SD 2.2); 46% female) could be studied 5-13 years (mean 8.3, SD 2.3) after the event. The control group consisted of subjects who were admitted due to conditions other than alcohol intoxication

Normative scores for the three factor eating questionnaire (TFEQ-R18) in a UK population-based cohort.

Eating behaviour traits (EBTs), individuals' reactions to food, food-related cues, and food intake, play an important role in the development and maintenance of overweight and obesity. We provide population-based norms for cognitive restraint, uncontrolled eating, and emotional eating as measured by the Three Factor Eating questionnaire (short form, TFEQ-18) in a UK cohort of middle-aged adults (the Fenland Study). Participants included 7830 individuals recruited across Cambridgeshire General Practices; 51.8 % identified as female and participants reported a mean BMI of 26.8. Unadjusted linear

Tracing the development and lifespan change of population-level structural asymmetry in the cerebral cortex.

Cortical asymmetry is a ubiquitous feature of brain organization that is subtly altered in some neurodevelopmental disorders, yet we lack knowledge of how its development proceeds across life in health. Achieving consensus on the precise cortical asymmetries in humans is necessary to uncover the developmental timing of asymmetry and the extent to which it arises through genetic and later influences in childhood. Here, we delineate population-level asymmetry in cortical thickness and surface area vertex-wise in seven datasets and chart asymmetry trajectories longitudinally across life (4-89 yea

Effects of prenatal alcohol exposure on the development of white matter volume and change in executive function.

Prenatal alcohol exposure can cause a wide range of deficits in executive function that persist throughout life, but little is known about how changes in brain structure relate to cognition in affected individuals. In the current study, we predicted that the rate of white matter volumetric development would be atypical in children with fetal alcohol spectrum disorders (FASD) when compared to typically developing children, and that the rate of change in cognitive function would relate to differential white matter development between groups. Data were available for 103 subjects [49 with FASD, 54

A developmental examination of amygdala response to facial expressions.

Several lines of evidence implicate the amygdala in face-emotion processing, particularly for fearful facial expressions. Related findings suggest that face-emotion processing engages the amygdala within an interconnected circuitry that can be studied using a functional-connectivity approach. Past work also underscores important functional changes in the amygdala during development. Taken together, prior research on amygdala function and development reveals a need for more work examining developmental changes in the amygdala's response to fearful faces and in amygdala functional connectivity d

Baseline characteristics of the North American prodromal Synucleinopathy cohort.

OBJECTIVE: Rapid eye movement (REM) sleep behavior disorder (RBD) is widely considered a prodromal synucleinopathy, as most with RBD develop overt synucleinopathy within ~10 years. Accordingly, RBD offers an opportunity to test potential treatments at the earliest stages of synucleinopathy. The North American Prodromal Synucleinopathy (NAPS) Consortium has created a multisite RBD participant, primarily clinic-based cohort to better understand characteristics at diagnosis, and in future work, identify predictors of phenoconversion, develop synucleinopathy biomarkers, and enable early stage clin

Spinal deformity surgery in patients with movement disorders: Trade-off between increased complications and improved long-term quality of life.

INTRODUCTION: Movement disorders are associated with poor outcomes after adult spinal deformity (ASD) surgery. In a cohort of patients undergoing ASD surgery, we sought to: 1) describe the operations performed in patients with movement disorders, 2) compare outcomes between patients with movement disorders vs. without movement disorders in the areas of mechanical complications, reoperations, and patient-reported outcome measures (PROMs). METHODS: A single-institution, retrospective cohort study was performed for patients with a movement disorder undergoing ASD surgery from 2009 to 2021. Inclu
