GABAA receptor pharmacology and subtype mRNA expression in human neuronal NT2-N cells.

Human NT2 teratocarcinoma cells differentiate into neuron-like NT2-N cells when treated with retinoic acid. GABA evoked concentration-dependent whole-cell currents in NT2-N cells with an EC50 of 21.8 microM and a Hill slope of 1.2. GABAA receptor (GABAR) currents reversed at ECl- and did not display voltage-dependent rectification. GABAR single channels opened in bursts to a 23 pS main conductance level and a 19 pS subconductance level, with infrequent openings to a 27 pS conductance level. Kinetic properties of the main conductance level were similar to other native and recombinant GABAR chan

Expression of class I major histocompatibility complex antigens in Epstein-Barr virus-carrying lymphoblastoid cell lines and Burkitt lymphoma cells.

Epstein-Barr virus (EBV) carrying lymphoblastoid cell lines (LCLs) and EBV-positive Burkitt lymphoma (BL) cells were compared for their expression of class I antigens of the major histocompatibility complex. Five common BL lines, LCLs, pokeweed mitogen-stimulated blasts and resting B-cells from healthy donors, and eight pairs of BL cells and LCLs, each pair originating from one patient, were tested. Quantitative analysis was performed using a radioimmunoassay; qualitative aspects were studied by one- and two-dimensional gel electrophoresis. In general, LCLs expressed significantly higher amoun

NUPR1-positive tumor-associated macrophages facilitate the progression of hepatocellular carcinoma via ferroptosis resistance.

BACKGROUND: Tumor-associated macrophages (TAMs) are major contributors to the immunosuppressive microenvironment of hepatocellular carcinoma (HCC). Nuclear protein 1 (NUPR1) is a stress-responsive regulator implicated in tumor progression; however, whether it functionally controls ferroptosis-associated stress adaptation in TAMs and thereby sustains their immunosuppressive phenotype remains unclear. PATIENTS AND METHODS: Single-cell RNA sequencing and multiplex immunohistochemistry identified and validated NUPR1-expressing TAMs in human HCC. Pharmacological and genetic approaches assessed mac

microRNA expression in macrophages regulates antibody-dependent effector function and B cell lymphoma therapy.

CD163 positive tumour associated macrophages (TAMs) are associated with poor response to immunochemotherapy in patients with diffuse large B cell lymphoma and comprise a significant proportion of the tumour microenvironment. Targeting these TAMs to reverse immunosuppression is an attractive approach for improving treatment response rates. A change in microRNA expression has the potential to modify hundreds of genes by interacting with multiple messenger RNA targets, thereby altering the wider cellular transcriptional profile. The manipulation of microRNAs that control macrophage behaviour may

BmSNX20 restricts Bombyx mori nucleopolyhedrovirus infection and is associated with autophagy-mediated antiviral immunity.

Sorting nexins (SNXs) are phosphoinositide-binding proteins that play critical roles in membrane trafficking and have been implicated in antiviral immunity. However, the function of SNX20 in insect antiviral immunity remains poorly defined. In this study, we investigated the role of Bombyx mori SNX20 (BmSNX20) during infection with Bombyx mori nucleopolyhedrovirus (BmNPV). Phylogenetic and domain analyses revealed that SNX20 is evolutionarily conserved across insect species and contains a canonical phox homology (PX) domain. Expression profiling showed that BmSNX20 is highly expressed in the m

Programmable Immunity: DNA Nanostructures in Cancer Immunotherapy.

Despite significant progress in cancer immunotherapy, challenges such as limited immune activation, tumor immune evasion, low response rates in solid tumors and off-target effects continue to hinder therapeutic efficacy. These challenges necessitate strategies to remodel the immune-suppressive tumor microenvironment (TME) into an immune activated state. Over the past decade, nanotechnology-based cancer immunotherapies have gained considerable attention for their ability to modulate the TME and stimulate immune responses. Among these, DNA nanotechnology-based systems have emerged as highly prom

Biodegradable molybdenum intramedullary nails facilitate fracture repair through neutrophil-mediated immune response.

Molybdenum (Mo) has been considered as a promising biodegradable metal for fracture fixation owing to its favorable mechanical properties and controlled degradation behavior. Although existing studies have confirmed the osteoinductive potential of molybdenum-based materials, the biological mechanism through which molybdenum facilitates bone healing remains poorly elucidated. This study systematically evaluated the fracture repair performance of biodegradable Mo intramedullary nails in a rat femoral fracture model and further characterized the early peri-implant immune microenvironment using si

Anemoside B4 upregulates DDAH2 and attenuates oxidative stress induced endothelial dysfunction.

Endothelial dysfunction, involving diminished nitric oxide (NO) bioavailability and vascular inflammation, plays a pivotal role in cardiovascular disease progression. Anemoside B4 (AB4), a major bioactive component extracted from Pulsatilla chinensis, has been reported to exert anti-inflammatory, anti-tumor, and antiviral effects. However, its potential role in regulating endothelial function remains largely unexplored. Integrating RNA-seq analysis, we revealed that AB4 may exert anti-atherosclerotic effects by upregulating DDAH2 expression and improving endothelial NO production. Using an H2O

Mediator at the Helm: Coordinating transcription and biomolecular condensates in hematopoiesis.

Hematopoiesis relies on precisely coordinated transcriptional programs that balance stem cell self-renewal, lineage commitment, and terminal differentiation. Central to this regulation is the Mediator complex, a large multi-subunit transcriptional co-regulator that integrates signals from transcription factors and chromatin regulators to control RNA polymerase Ⅱ (Pol Ⅱ) activity. The dynamic and modular composition of Mediator enables context-dependent transcriptional outputs, while individual subunits can exert specialized regulatory functions during hematopoietic lineage specification, there

ARID1A Mediates SWI/SNF-Independent Maintenance of Heterochromatin Architecture to Restrain Viral Mimicry and Immunogenicity in Colon Cancer.

UNLABELLED: AT-rich interaction domain 1A (ARID1A) is a crucial subunit of the switch/sucrose nonfermentable (SWI/SNF) chromatin-remodeling complex and the ARID1A gene is frequently mutated in human cancers. Although its tumor-suppressive activity has been ascribed exclusively to SWI/SNF-dependent chromatin remodeling, we established in this study a SWI/SNF-independent role of ARID1A in safeguarding heterochromatin architecture to silence viral mimicry and restrain immunogenicity in colorectal cancer. ARID1A deficiency triggered viral mimicry and enhanced immunogenicity in both microsatellite-

Duck lncRNA lnc455 enhances RIG-I/MAVS type I interferon signaling by modulating hnRNPAB-mediated regulation of MAVS signaling.

Long non-coding RNAs (lncRNAs) regulate antiviral immunity against influenza A viruses, yet their roles in natural reservoir hosts like ducks remain unclear. In ducks, Retinoic acid-inducible gene I (RIG-I) contributes to their resistance to infection. To identify duck lncRNAs involved in RIG-I-mediated antiviral responses, we analyzed transcriptomic data from highly pathogenic avian influenza (HPAI)-infected duck lung tissues and identified lnc455 co-expressed with IFNβ. In-silico predictions suggested lnc455 could interact with RIG-I and MAVS, two key signaling proteins in the Type I IFN pat

DHX9 Inhibition Enhances Paclitaxel Sensitivity by Inducing Mitotic Failure in Ovarian and Endometrial Cancers.

Recurrent high-grade serous ovarian carcinoma (HGSOC) and endometrial cancer remain major clinical challenges with limited effective treatment options. DExH-box helicase 9 (DHX9), a DNA/RNA helicase essential for genomic stability, has not yet been explored as a therapeutic target in gynecologic cancers. In this study, we show that a selective DHX9 inhibitor (DHX9i) suppresses proliferation in a subset of HGSOC and endometrial cancer cell lines by inducing DNA damage, chromosomal instability, and mitotic failure. This effect was independent of microsatellite instability status and prior resist

CREB5 Inhibits Neuronal Ferroptosis via Transactivating ApoL6 to Regulate Lipid Droplet Metabolism After Spinal Cord Injury.

BACKGROUND: After spinal cord injury (SCI), neuronal lipid peroxidation and excessive production of reactive oxygen species (ROS) induced by secondary injury exacerbate ferroptosis, impeding regenerative repair and functional recovery in mice. Thus, clarifying the molecular and cellular mechanisms underlying the inhibition of neuronal ferroptosis post-SCI is crucial. METHODS: Single-cell RNA sequencing (scRNA-seq) and single-cell assay for transposase-accessible chromatin sequencing (scATAC-seq) were used to analyze changes in the transcription factor CREB5 post-SCI. Combined with in vitro (p

PI3Kδ inhibition alters CD8 T cell differentiation and reprograms the tumor microenvironment following adoptive immunotherapy.

T cell exhaustion remains a significant barrier to effective adoptive cell therapy in solid tumors. Here, we demonstrate that in vitro treatment with the PI3Kδ inhibitor CAL-101 generates T cells with enhanced stemness and metabolic fitness. These cells show increased mitochondrial dependence and spare respiratory capacity while maintaining normal basal metabolism. Under chronic antigen stimulation, CAL-101-treated T cells, including human T cells, resist terminal exhaustion and maintain stem-like properties. Using single-cell RNA sequencing and spatial transcriptomics of B16 melanoma tumors,

MicroRNA-based strategies to overcome the immunotherapy barrier in pancreatic ductal adenocarcinoma.

Pancreatic ductal adenocarcinoma (PDAC) is a highly lethal malignancy that continues to pose a major clinical challenge, primarily due to the difficulty of early detection and the limited efficacy of existing therapeutic approaches. Immunotherapy, which has revolutionized the treatment of many other cancers, has shown limited success in PDAC, largely because of the complex and immunosuppressive features of its tumor microenvironment (TME). Consequently, strategies aimed at remodeling or modulating the TME have emerged as promising avenues for enhancing the therapeutical potential of immunother

Ascomycin enhances the antitumor activity of osimertinib in EGFR-TKI-resistant non-small cell lung cancer.

BACKGROUND: Resistance to epidermal growth factor receptor tyrosine kinase inhibitors (EGFR-TKIs) remains a major challenge in non-small cell lung cancer (NSCLC) treatment. Although ascomycin (FK520) has demonstrated some antitumor activity, its potential in overcoming osimertinib resistance has not been fully characterized. METHODS: Cell viability and proliferation were assessed using the Cell Counting Kit 8 (CCK8) and colony formation assays, respectively, following treatment with ascomycin, osimertinib, or interleukin (IL)-6 for 48 h. Flow cytometry was employed to characterize cell cycle

RHBDD2 Confers Insensitivity of Esophageal Squamous Cell Carcinoma to Cisplatin by Inhibiting Ferroptosis Through the Wnt3a/β-Catenin/FTH1 Axis.

As a pseudoprotease member of the rhomboid family, rhomboid domain containing 2 (RHBDD2) has been demonstrated to be associated with 5‑FU resistance. Our previous work found that RHBDD2 is also upregulated by cisplatin (DDP) treatment in esophageal squamous cell carcinoma (ESCC). However, the effect of RHBDD2 on DDP sensitivity and the underlying molecular mechanisms remain poorly understood. In this study, we demonstrate that RHBDD2 regulates the sensitivity of ESCC cells to DDP in vitro. In vivo, RHBDD2 knockdown significantly enhanced the antitumor effect of DDP in a subcutaneous xenograft
