Zellweger syndrome, neonatal adrenoleukodystrophy, and infantile Refsum's disease are genetic disorders characterized by the virtual absence of catalase-positive peroxisomes and a general impairment of peroxisomal functions. Recent studies in these three disorders have provided morphologic evidence of peroxisomal "ghosts" of density 1.10 g/cm3 that contain membrane proteins but lack a majority of the matrix enzyme activities. We report here the biochemical studies in a female infant with clinical features of infantile Refsum's disease whose liver and fibroblasts contained cytosolic catalase bu

Previous reports have indicated the association of periodontal diseases with elevated levels of serum immunoglobulin G (IgG) antibodies to periodontally relevant bacteria. Recent results from this laboratory suggest that enzymes proteolytic for immunoglobulins are important virulence factors of several periodontal bacteria. Specifically, enzymes from Porphyromonas (Bacteroides) gingivalis culture supernatant fluid (SF) cleaved human IgG (4 subclasses), IgA1 and IgA2, IgM, IgD and IgE. Proteolytic enzymes from Actinobacillus actinomycetemcomitans culture SF cleaved IgG, IgA and IgM. An enriched

The limiting membranes of pituitary growth hormone and prolactin secretory granules contain a Mg2+-ATPase sensitive to anions. This enzyme is in many ways similar to mitochondrial ATPase. The enzyme was potently inhibited by oligomycin (Ki 6.5 X 10(-9) M), and was much more sensitive to the inhibitor than pituitary mitochondrial ATPase (Ki 2.7 X 10(-7) M). In contrast, the enzyme activity of intact secretory granules was only sparingly inhibited by oligomycin (maximal inhibition close to 30% at 5 X 10(-4) M). However, oligomycin (5 microM) did diminish to basal levels the enhanced granule ATPa

A sensitive and quantitative assay for the detection of cellobiose:quinone oxidoreductase (CBQase) is described. The assay is based on the ability of CBQase to reduce the cation radicals formed by the laccase-mediated oxidation of chlorpromazine (CPZ). Formation of the CPZ radical cation is readily followed at 530 nm, and the net rate of its formation is decreased in proportion to the amount of CBQase activity present. Advantages of this assay are its increased sensitivity due to the high extinction coefficient of the CPZ radical, the high solubility of the substrate in water, and the assay's

The authors give a short report about the first-trimester prenatal detection of Hunter's disease (MPS II) inherited as X-linked disorder. There is written about a family having one affected child with Hunter's syndrome. Chorionic villus sample was taken at 10th weeks of gestation in the new pregnancy of the mother. The sex of the fetus was a male determined by DNA analysis. The activity of sulphoiduronate sulphatase was very low. The enzyme activity was also extremely low in the cultured cells from amniotic fluid taken at 16th weeks of gestation. On the basis of these results the pregnancy was

Maintenance of rats for 2 weeks on a diet consisting of 50% inert cellulose and 50% laboratory chow resulted in hypertrophy of the parotid gland and a 4-fold increase in total membrane-associated 4 beta-galactosyltransferase enzyme activity (EC 2.4.1.38). Localization of the increased specific activity to the cell surface of the enlarged gland was shown by subcellular fractionation of Golgi and plasma membranes. This observation was confirmed by enzyme assays of intact cells; quantification of immunofluorescence was made by using a fluorescence activated cell sorter. Parotid gland hypertrophy

Ferrochelatase activity was examined both in growing MEL cells and in in vitro assays of the purified enzyme to determine what effect a variety of divalent cations would have. Data obtained with the purified enzyme demonstrated that Mn2+ strongly inhibits the activity in a competitive fashion with respect to Fe2+ with a calculated Ki of 15 microM. Cadmium ion is also inhibitory (Ki: 50 microM), as is Hg2+ and arsenite, but Pb2+ is a poor inhibitor. All other metals tested had no effect. When these same metal ions were tested on differentiating MEL cells it was found that their ability to inhib

Microfilariae of bovine filarial parasite Setaria cervi are equipped with the enzymes of glycolysis, pentose phosphate and PEP-succinate pathways and thus resemble the adult form in its metabolic pattern. Malate dehydrogenase was the most active enzyme in microfilariae followed by lactic dehydrogenase and fumarase, while phosphoglucoisomerase, PEP-carboxykinase and FDP-aldolase were comparatively less active. The very low ratio of PK/PEPCK in S. cervi microfilariae indicates active fixation of CO2 into PEP to produce oxalacetate. Centperazine and diethylcarbamazine significantly inhibited PEP-

NADH-diaphorase and cytochrome b5 reductase activities of platelets and leucocytes, as well as erythrocytes, were found to be deficient in a patient with hereditary methaemoglobinaemia associated with moderate mental retardation and non-progressive neurological disturbance, in which hyperactive reflexes and involuntary movements were notable. In another methaemoglobinaemic patient with no mental or neurological abnormalities, these enzyme activities were defective in erythrocytes but normal in platelets and leucocytes. The first case was a generalised cytochrome b5 reductase deficiency with no

Admittedly, the disastrous emergence of drug resistance in prokaryotic and eukaryotic human pathogens has created an urgent need to develop novel chemotherapeutic agents. Onosma chitralicum is a source of traditional medicine with cooling, laxative, and anthelmintic effects. The objective of the current research was to analyze the biological potential of Onosma chitralicum, and to isolate and characterize the chemical constituents of the plant. The crude extracts of the plant prepared with different solvents, such as aqueous, hexane, chloroform, ethyl acetate, and butanol, were subjected to an

Sialidase (SD, neuraminidase, EC 3.2.1 18) is an enzyme which releases terminal sialic acid residues from glycoproteins, glycolipids nad oligosaccharides. In this study, we report some characteristics of this enzyme in rat salivary glands, brain, liver and kidney. SD activity was measured fluorometrically by using the synthetic substrate 4-methylumbelliferyl-alpha-D-N-acetylneuraminic acid (4MU-AcNeu). 1. SD activity in tissue homogenates was maximal at pH 4.0 in the submandibular gland (SMG), and at pH 4.5 in both the sublingual gland (SLG) and parotid gland (PG), the values being very simila

ATP-citrate lyase (ACLY) is a key enzyme in cellular metabolism that catalyses the cleavage of citrate to produce acetyl coenzyme A (acetyl-CoA), thereby bridging energy metabolism and epigenetic regulation. Acetyl-CoA serves as a common substrate for de novo lipogenesis (DNL) and histone acetylation. Aberrant ACLY activity is closely associated with various pathological conditions, including metabolic diseases, Immune-mediated inflammatory diseases (IMIDs), cancer, and neurodegenerative diseases, positioning ACLY as an important therapeutic target for these diseases. However, existing inhibit

Pharmacokinetic (PK) predictions in humans are important for both the parent compounds and their metabolites, as these predictions help decide whether to progress to human clinical trials. More human PK prediction studies have been conducted on unchanged compounds than on their metabolites. Human PK predictions are more complex for metabolites than for unchanged compounds due to the large number of metabolic enzymes involved and the need to consider species-related differences in these enzymes. Mice with humanized livers are expected to serve as simple and useful tools for assessing a wide ran

Keteleeria is a small genus of conifer in the family Pinaceae (the extant pine family). Fortunefuroic acids (FFAs) have recently been documented as characteristic specialized metabolites of this genus. FFAs constitute a unique class of triterpenoids characterized by the presence of a furoic acid moiety in the side chain. Using 1H NMR and LC-PDA-MS guided isolation and dereplication approaches, four previously undescribed (1-4, fortunefuroic acids Q-T, resp.) and one known (fortunefuroic acid H, 5) FFA-type analogues were isolated from the renewable leaves and twigs of Keteleeria pubescens W. The insect chitin metabolism system is an ideal target for species-selective pesticide development, in which OfChi-h and OfChtI jointly mediate chitin degradation. However, existing Chitinase inhibitors face limitations in efficacy and specificity, necessitating innovative computer-aided driven lead discovery strategies. Herein, a dual-target virtual screening was performed to discover triazinobenzoxazepine derivatives (TBZs) as novel Chitinase inhibitors. Compound TBZ-15 emerged as a potent dual inhibitor with Ki values of 0.64 μM and 0.43 μM against OfChi-h and OfChtI, respectively. TBZ-4 an

Streptococcus mutans diadenylate cyclase (SmDAC) catalyzes the cyclization of two ATP molecules into cyclic di-AMP, a second messenger that regulates many cellular processes including biofilm formation. Aided by structure-based drug design and subsequent structure-activity relationship studies, we identified a coumarin chemotype as low-micromolar inhibitors of SmDAC. Optimized lead compounds inhibited both biofilm formation and planktonic growth of S. mutans. Biofilm inhibition marginally exceeded growth inhibition at tested doses, indicating relative selectivity toward biofilm inhibition. Add

A novel series of sulfur-containing analogs derived from the 3-hydroxy-6-methyl-2-((4-phenylpiperazin-1-yl)methyl)-4H-pyran scaffold was synthesized. Spectroscopic techniques and elemental analysis confirmed their structures. The biological potential of these compounds, along with their previously reported bioisosteres, was assessed as tyrosinase inhibitors, as well as antitubercular and antidermatophytic agents. In all derivatives where the carbonyl group is converted to a thiocarbonyl group, tyrosinase inhibition increases significantly, with the most effective compounds being identified as

Pyruvate kinase (PK) activators enhance glycolytic flux, increasing ATP production and lowering red blood cell (RBC) 2,3-diphosphoglycerate (2,3-DPG), thereby improving oxygen affinity and potentially reducing hemoglobin S polymerization in sickle cell disease (SCD). Through these biochemical effects, PK activation may decrease hemolysis and improve anemia. This systematic review and meta-analysis evaluated the impact of PK activators on hematologic and hemolytic parameters in adults with SCD. We searched PubMed, Embase, and the Cochrane Central Register of Controlled Trials from inception thr

BACKGROUND/AIMS: Homeopathic medicines, including Arsenicum iodatum, have attracted increasing interest as potential adjunctive approaches in cancer research. However, the cellular mechanisms underlying their reported biological effects remain poorly understood. This study investigated the effects of homeopathic Arsenicum iodatum on apoptosis, cell cycle progression, and caspase activation in A549 human lung adenocarcinoma cells.
METHODS: A549 cells were treated with Arsenicum iodatum potencies (6C, 12C, 30C, and 200C). Apoptosis and cell cycle distribution were assessed by flow cytometry, wh
