Increased activity of TNAP compensates for reduced adenosine production and promotes ectopic calcification in the genetic disease ACDC.

ACDC (arterial calcification due to deficiency of CD73) is an autosomal recessive disease resulting from loss-of-function mutations in NT5E, which encodes CD73, a 5'-ectonucleotidase that converts extracellular adenosine monophosphate to adenosine. ACDC patients display progressive calcification of lower extremity arteries, causing limb ischemia. Tissue-nonspecific alkaline phosphatase (TNAP), which converts pyrophosphate (PPi) to inorganic phosphate (Pi), and extracellular purine metabolism play important roles in other inherited forms of vascular calcification. Compared to cells from healthy

Inhibition of Eicosanoid Degradation Mitigates Fibrosis of the Heart.

BACKGROUND: Organ fibrosis due to excessive production of extracellular matrix by resident fibroblasts is estimated to contribute to >45% of deaths in the Western world, including those due to cardiovascular diseases such as heart failure. Here, we screened for small molecule inhibitors with a common ability to suppress activation of fibroblasts across organ systems. METHODS: High-content imaging of cultured cardiac, pulmonary, and renal fibroblasts was used to identify nontoxic compounds that blocked induction of markers of activation in response to the profibrotic stimulus, transforming gro

Acute vaping in a golden Syrian hamster causes inflammatory response transcriptomic changes.

E-cigarette vaping is a major aspect of nicotine consumption, especially for children and young adults. Although it is branded as a safer alternative to cigarette smoking, murine and rat models of subacute and chronic e-cigarette vaping exposure have shown many proinflammatory changes in the respiratory tract. An acute vaping exposure paradigm has not been demonstrated in the golden Syrian hamster, and the hamster is a readily available small animal model that has the unique benefit of becoming infected with and transmitting respiratory viruses, including SARS-CoV-2, without genetic alteration

[New pathogenetic insights into pseudoexfoliation syndrome/glaucoma. Therapeutically relevant?].

Pseudoexfoliation (PEX) syndrome is a genetically determined, generalized disease of the extracellular matrix leading to the progressive deposition of an abnormal fibrillar material in various intraocular and extraocular tissues including the trabecular meshwork. It thus represents the most common identifiable cause of open-angle glaucoma and a leading cause of blindness worldwide. The PEX-specific fibrotic matrix process, a stress-induced elastosis, is characterized by an excessive production and abnormal cross-linking of elastic microfibrils into fibrillar PEX aggregates. Co-modulating facto

Matrix metalloproteinase-9 and tissue inhibitor of metalloproteinase-1: expression in the lung of fetal rats with nitrofen-induced diaphragmatic hernia.

The surrounding extracellular matrix of airway wall tissues changes in response to mechanical stresses and hypoxia. The presence of matrix metalloproteinase-9 (MMP-9) and its inhibitor, tissue inhibitor of metalloproteinase-1 (TIMP-1), is correlated with collagen degradation and tissue repair in lung disorders. The aim of this study was to evaluate the expression of MMP-9 and TIMP-1 in the lung of fetal rats with nitrofen-induced congenital diaphragmatic hernia (CDH). Administering 100 mg of nitrofen dissolved in 1 ml olive oil to pregnant Wistar rats on day 9 of gestation induced left-sided C

Renal growth responses to acute and chronic injury: routes to therapeutic intervention.

Knowledge of the control of cell growth and extracellular matrix deposition has assumed center stage in the understanding of how the diseased kidney responds to injury. After acute tubular injury, there may be reversible, partial depolarization of renal cells or cell necrosis. The latter requires a regenerative response, which could be under the control of growth factors such as epidermal growth factor (EGF). Up-regulation of EGF receptors on viable cells provides the cell with an enhanced growth response despite a reduction in EGF production by the kidney. Acute glomerular injury involves a h

Myofibroblast-Derived Extracellular Vesicles Drive Profibrotic Cascade Amplification in Pulmonary Fibrosis via the Nestin-Rab7 Axis.

Idiopathic pulmonary fibrosis (IPF) is a fatal fibrotic lung disease characterized by aberrant myofibroblast activation and excessive extracellular matrix deposition, with extracellular vesicles (EVs) playing a crucial role in this pathological process. We observed that EVs levels are significantly elevated in IPF and positively correlate with nestin expression, a known marker of lung myofibroblasts. These myofibroblast-derived EVs further amplify profibrotic responses, creating a self-perpetuating cycle. To elucidate the mechanisms driving increased EVs secretion, we conducted in vitro and in

Biofilm-Forming Methicillin-Resistant Staphylococcus aureus: A Comprehensive Phenotypic and Genotypic Review.

Methicillin-resistant Staphylococcus aureus (MRSA) is a major global pathogen, capable of forming biofilms that confer enhanced antimicrobial tolerance, virulence, and persistence in both hospital and community settings. Biofilms are structured microbial communities encased in extracellular polymeric substances, composed of polysaccharides, proteins, and extracellular DNA. Complex genetic networks, including the ica operon, the agr quorum-sensing system, the sarA regulatory system, and stress-response genes such as sigB, regulate MRSA biofilm formation. Integrated 3D-QSAR and cellular profiling of sulfated and hydrophobic aminoglycoside glycans to modulate heparanase activity.

Heparanase (HPSE) plays a critical role in tumor progression by degrading heparan sulfate chains in the extracellular matrix and modulating the tumor microenvironment. As a result, it has emerged as a promising therapeutic target. Aminoglycoside-derived sulfated glycan mimetics have shown potential as HPSE inhibitors, but rational optimization is challenged by complex steric and electrostatic interactions. Here, we used three-dimensional quantitative structure-activity relationship (3D-QSAR) modeling to identify key structural features governing HPSE inhibition, providing new predictive struct

Neutrophil extracellular traps (NETs) and pulmonary fibrosis: Mechanisms and implications.

Pulmonary fibrosis is a fatal interstitial lung disease characterized by excessive fibroblast proliferation and aberrant deposition of extracellular matrix (ECM), ultimately leading to structural lung destruction and respiratory failure. Neutrophils, as key effector cells of innate immunity, have been increasingly recognized for their role in chronic inflammation and fibrotic diseases through the formation of neutrophil extracellular traps (NETs)-a distinct form of cell death. Distinguishing itself from prior reviews, this review aims to compare the cytotoxic effects of three NETosis subtypes

Innovative scaffold-based strategies for diabetic wound healing: progress and prospects.

Diabetic wounds present a major clinical concern due to chronic inflammation, impaired angiogenesis, excessive protease activity, and delayed extracellular matrix remodelling, leading to prolonged healing and high recurrence rates. Globally, over 536 million individuals were affected by diabetes in 2021, with diabetic foot ulcers impacting approximately 15-25% of these patients during their lifetime. Conventional wound dressings often fail to address the complex pathophysiology of diabetic wounds, necessitating advanced therapeutic strategies. Scaffold-based biomaterials have emerged as promis

Decoding the Tissue Characteristics of Perihematomal Edema in Intracerebral Hemorrhage: Poroelastic and Viscoelastic Perspectives.

Intracerebral hemorrhage (ICH) causes severe brain damage, with perihematomal edema (PHE) contributing to poor outcomes. However, the biomechanical properties of PHE tissue remain incompletely characterized. In this study, we examined poroviscoelastic behavior in edematous brain tissue using a rat ICH model, together with associated structural and molecular alterations. Load-relaxation tests were used to characterize viscoelastic and poroelastic responses, while MRI (magnetic resonance imaging) and histological analyses were performed to assess edema progression and tissue integrity. Senescence-associated extracellular vesicles in skin aging and regeneration: from pathophysiology to therapeutic opportunities.

Cellular senescence is an important factor which significantly impairs the homeostasis and regeneration capability of skin through causing chronic inflammation, degradation of the extracellular matrix, depletion of stem cells, and angiogenesis impairment. In addition to the already known senescence associated secretory phenotype (SASP), senescent cells secrete senescence associated extracellular vesicles (SA-EVs) serving as powerful signalling molecules. SA-EVs are distinguished by the increased secretion rates, altered biogenesis and selective cargo loading which include proinflammatory micro

Bioprinting of bespoke islet-specific niches to promote maturation of stem cell-derived islets.

Pancreatic islets are densely packed cellular aggregates containing various hormonal cell types essential for blood glucose regulation. Interactions among these cells markedly affect the glucoregulatory functions of islets along with the surrounding niche and pancreatic tissue-specific geometrical organization. However, stem cell (SC)-derived islets generated in vitro often lack the three-dimensional extracellular microenvironment and peri-vasculature, which leads to the immaturity of SC-derived islets, reducing their ability to detect glucose fluctuations and insulin release. Here, we bioengi

Extracellular Vesicles Derived From Endometrial Stem Cells Preconditioned With PGE2-Reverse Myofibroblast Phenotype in Mare Endometrial Cells: A Novel Anti-Fibrotic Approach.

Endometrial fibrosis in mares compromises fertility through aberrant extracellular matrix deposition and sustained myofibroblast activation. Conventional interventions fail to reverse these pathological alterations, necessitating innovative, mechanism-focused therapies. In this study, we pioneered the use of prostaglandin E2 (PGE2) preconditioning of equine endometrial-derived mesenchymal stem cells (ET-eMSCs) and their extracellular vesicles (EVs) to target fibrotic processes directly. ET-eMSCs were isolated from mare endometrial biopsies pretreated with PGE2 to enhance their anti-fibrotic se

Comprehensive Analysis of Potential Common Pathogenic Mechanisms for COVID-19 Infection and Gastric Cancer.

A growing body of data suggests that the prevalence of COVID-19 pneumonia in patients with stomach cancer is much higher than in the general population. However, these mechanisms are still not fully understood. After a thorough examination of shared differentially expressed genes (DEGs) for gastric cancer (GC) and COVID-19 pneumonia, we performed functional annotation, protein-protein interaction (PPI) networks, module design, and pivot gene identification. qPCR was used to verify the expression of hub genes in GC. Finally, a pivotal gene transcription factor-gene regulatory network was create

Succinic Acid-Induced Macrophage Endocytosis Promotes Extracellular Vesicle-Based Integrin Beta1 Transfer Accelerating Fibroblast Activation and Sepsis-Associated Pulmonary Fibrosis.

Sepsis-associated pulmonary fibrosis (SAPF) is a life-threatening condition driven by persistent fibroblast activation and excessive extracellular matrix (ECM) deposition. While metabolic reprogramming, profibrotic extracellular vesicles (EVs), and integrin activation are implicated in pulmonary fibrosis, their interplay remains unclear. This study reveals that succinic acid, a product of glycometabolic reprogramming, promotes macrophage-mediated endocytosis, driving the release of profibrotic EVs. These EVs transfer integrin beta1 (ITGβ1) from macrophages to fibroblasts, activating fibroblast

PAI-1 uncouples integrin-β1 from restrain by membrane-bound β-catenin to promote collagen fibril remodeling in obesity-related neoplasms.

The paracrine actions of adipokine plasminogen activator inhibitor-1 (PAI-1) are implicated in obesity-associated tumorigenesis. Here, we show that PAI-1 mediates extracellular matrix (ECM) signaling via epigenetic repression of DKK1 in endometrial epithelial cells (EECs). While the loss of DKK1 is known to increase β-catenin accumulation for WNT signaling activation, this epigenetic repression causes β-catenin release from transmembrane integrins. Furthermore, PAI-1 elicits the disengagement of TIMP2 and SPARC from integrin-β1 on the cell surface, lifting an integrin-β1-ECM signaling constrai

HAPLN1 potentiates peritoneal metastasis in pancreatic cancer.

Pancreatic ductal adenocarcinoma (PDAC) frequently metastasizes into the peritoneum, which contributes to poor prognosis. Metastatic spreading is promoted by cancer cell plasticity, yet its regulation by the microenvironment is incompletely understood. Here, we show that the presence of hyaluronan and proteoglycan link protein-1 (HAPLN1) in the extracellular matrix enhances tumor cell plasticity and PDAC metastasis. Bioinformatic analysis showed that HAPLN1 expression is enriched in the basal PDAC subtype and associated with worse overall patient survival. In a mouse model for peritoneal carci
