Dose-dependent, opioid-induced sleep disruption is partially mediated by motor activity but not by body temperature in C57BL/6J mice.

States of sleep and wakefulness, motor activity, and body temperature are temporally linked, yet these variables are often studied independently. Here, we report novel results from male C57BL/6J mice (n = 24) that quantify the causal and correlational relationships between these temporally linked measures. The initial experiments used 12 mice to identify the lowest antinociceptive doses of fentanyl (0.1 mg/kg) and morphine (1 mg/kg). Twelve additional mice were implanted with telemeters to simultaneously record electroencephalogram, electromyogram, motor activity, and subcutaneous body tempera

Arousal burden is highest in supine sleeping position and during light sleep.

STUDY OBJECTIVES: Arousal burden (AB) is defined as the cumulative duration of arousals during sleep divided by the total sleep time. However, in-depth analysis of AB related to sleep characteristics is lacking. Based on previous studies addressing the arousal index, we hypothesized that the AB would peak in the supine sleeping position and during non-rapid eye movement stage 1 and show high variability between scorers. METHODS: Nine expert scorers analyzed polysomnography recordings of 50 participants, the majority with an increased risk for obstructive sleep apnea. AB was calculated in diff

The use of the "group traumatic episode protocol (G-TEP) " in an outpatient randomized and hospital setting: Implementation and effects in heterogeneous patient samples.

OBJECTIVE: The resource-based eye movement desensitization and reprocessing group intervention group traumatic episode protocol G-TEP showed promising results in reducing posttraumatic stress disorder, depression, and anxiety. Using a high structured G-TEP worksheet, the patients conduct self-bilateral stimulation while (re)processing their targets. We examined the effectiveness on multiple symptoms and practicability of G-TEP in an outpatient setting and its feasibility in hospital treatment. METHOD: Study A: Forty-four patients suffering from different symptoms and diagnoses received four a

Deep-learning detection of mild cognitive impairment from sleep electroencephalography for patients with Parkinson's disease.

Parkinson's disease which is the second most prevalent neurodegenerative disorder in the United States is a serious and complex disease that may progress to mild cognitive impairment and dementia. The early detection of the mild cognitive impairment and the identification of its biomarkers is crucial to support neurologists in monitoring the progression of the disease and allow an early initiation of effective therapeutic treatments that will improve the quality of life for the patients. In this paper, we propose the first deep-learning based approaches to detect mild cognitive impairment in t

Bace1 Deletion in the Adult Reverses Epileptiform Activity and Sleep-wake Disturbances in AD Mice.

Alzheimer's disease (AD) increases the risk for seizures and sleep disorders. We show here that germline deletion of β-site amyloid precursor protein (APP) cleaving enzyme-1 (BACE1) in neurons, but not in astrocytes, increased epileptiform activity. However, Bace1 deletion at adult ages did not alter the normal EEG waveform, indicating less concern for BACE1 inhibition in patients. Moreover, we showed that deletion of Bace1 in the adult was able to reverse epileptiform activity in 5xFAD mice. Intriguingly, treating 5xFAD and APPNL-G-F/NL-G-F (APP KI) mice of either sex with one BACE1 inhibitor

Association between sleep apnea severity and symptoms of depression and anxiety among individuals with obstructive sleep apnea.

OBJECTIVE: We determined whether the severity of sleep apnea is associated with symptoms of depression and anxiety in patients with obstructive sleep apnea (OSA) and whether symptoms of OSA, other than respiratory events, are associated with depression and anxiety symptoms. METHODS: Depressive and anxiety symptoms were defined as a Patient Health Questionnaire-9 score of ≥10 and a Generalized Anxiety Disorder-7 score of ≥8, respectively. Sleep apnea severity and rapid eye movement-related OSA were evaluated using the apnea-hypopnea index (AHI). Subjective symptoms of OSA were assessed using t

[A case suspected of dystonia with marked cerebellar atrophy with torsion dystonia of the neck and cerebellar ataxia that developed during pharmacologic schizophrenia treatment].

A 46 year-old man with schizophrenia had taken several anti-psychotic drugs since 25 years of age. From ~35 years of age, he noticed occasional neck torsion to the left, and later an ataxic gait; both symptoms gradually worsened. On admission, the patient was taking olanzapine (5 mg/day) and biperiden hydrochloride (1 mg/day) because his schizophrenia was well controlled. His parents were not consanguineous, and there was no family history of neuropsychiatric diseases. On neurological examination, he showed mild cognitive impairment, saccadic eye pursuit with horizontal gaze nystagmus, mild dy

Modeling Neurodegenerative Spinocerebellar Ataxia Type 13 in Zebrafish Using a Purkinje Neuron Specific Tunable Coexpression System.

Purkinje cells (PCs) are primarily affected in neurodegenerative spinocerebellar ataxias (SCAs). For generating animal models for SCAs, genetic regulatory elements specifically targeting PCs are required, thereby linking pathological molecular effects with impaired function and organismic behavior. Because cerebellar anatomy and function are evolutionary conserved, zebrafish represent an excellent model to study SCAs in vivo We have isolated a 258 bp cross-species PC-specific enhancer element that can be used in a bidirectional manner for bioimaging of transgene-expressing PCs in zebrafish (bo

Time-of-day influences on respiratory sequelae following maximal electroshock-induced seizures in mice.

Sudden unexpected death in epilepsy (SUDEP) is the leading cause of death in refractory epilepsy patients. Although specific mechanisms underlying SUDEP are not well understood, evidence suggests most SUDEP occurs due to seizure-induced respiratory arrest. SUDEP also tends to happen at night. Although this may be due to circumstances in which humans find themselves at night, such as being alone without supervision or sleeping prone, or to independent influences of sleep state, there are a number of reasons why the night (i.e., circadian influences) could be an independent risk factor for SUDEP

Saccadic Palsy following Cardiac Surgery: Possible Role of Perineuronal Nets.

OBJECTIVE: Perineuronal nets (PN) form a specialized extracellular matrix around certain highly active neurons within the central nervous system and may help to stabilize synaptic contacts, promote local ion homeostasis, or play a protective role. Within the ocular motor system, excitatory burst neurons and omnipause neurons are highly active cells that generate rapid eye movements - saccades; both groups of neurons contain the calcium-binding protein parvalbumin and are ensheathed by PN. Experimental lesions of excitatory burst neurons and omnipause neurons cause slowing or complete loss of s

Complex sleep apnea.

Complex sleep apnea currently refers to the emergence and persistence of central apneas and hypopneas following the application of positive airway pressure therapy in patients with obstructive sleep apnea. However, this narrow definition is an "outcome" and does not capture the spectrum of pathological activation of the respiratory chemoreflex in sleep apnea. The International Classification of Sleep Disorders - 3rd edition recognizes the phenomenon of Treatment-Related Central Sleep Apnea, but the phenotype is usually evident prior to onset of therapy. The key polysomnographic characteristics

REM and NREM sleep-state distribution of respiratory events in habitually snoring school-aged community children.

BACKGROUND: Studies ascribe different functions to rapid eye movement (REM) and non-rapid eye movement (NREM) sleep, such that their disruption could result in discrepant clinical outcomes. Although sleep architecture is globally preserved in children with obstructive sleep apnoea (OSA), it is considered to be an REM sleep REMS disorder. Furthermore, body position during sleep affects the occurrence of respiratory events, while the presence of obesity has been claimed to affect sleep-state distribution of respiratory disturbance. METHODS: To explore the distribution of respiratory events duri

Prostaglandin D synthase (beta-trace) in healthy human sleep.

STUDY OBJECTIVES: The prostaglandin D system plays an important role in animal sleep. In humans, alterations in the prostaglandin D system have been found in diseases exhibiting sleep disturbances as a prominent symptom, such as trypanosoma infection, systemic mastocytosis, bacterial meningitis, major depression, or obstructive sleep apnea. Assessment of this system's activity in relation to human physiologic sleep was the target of the present study. DESIGN: Serum concentrations of lipocalin-type prostaglandin D synthase (L-PGDS, former beta-trace), and plasma levels of the pineal hormone me
