Higher Serum Omega-3 Polyunsaturated Fatty Acid Content Is Associated with Improved Long-Term Cardiovascular and Renal Outcomes in Patients with Chronic Kidney Disease.

Background: Patients with chronic kidney disease (CKD) exhibit reduced serum levels of n-3 polyunsaturated fatty acids (PUFA). Alterations in fatty acid profiles may contribute to increased cardiovascular risk and potentially accelerate CKD progression. Aim: The aim of the study was to assess whether fatty acid profiles measured a decade earlier predicted CKD progression and cardiovascular events. Additionally, the impact of dietary patterns at baseline was evaluated. Methods: The study comprised 77 patients with CKD whose serum fatty acid profiles had been assessed approximately a decade earl

PLIN2-PGAM5-regulated lipid droplet-mitochondria contacts drive microglial neuroinflammation after spinal cord injury via fatty acid metabolic reprogramming.

Persistent neuroinflammation is a hallmark of central nervous system (CNS) injury, driving neuronal loss and functional deficits. While microglial lipid metabolic reprogramming, particularly lipid droplet accumulation, has been implicated in chronic inflammation, the underlying mechanisms remain poorly understood. Using single-cell transcriptomics, we identified a previously defined lipid droplet-accumulating microglia (LDAM) subpopulation in spinal cord injury (SCI), characterized by elevated PLIN2 expression. Genetic deletion of Plin2 markedly reduced lipid droplet burden, attenuated neuroin

Small intestinal compared with colonic short-chain fatty acid delivery drives distinct systemic concentrations and endocrine responses in humans: a randomized, crossover trial.

BACKGROUND: Through fermentation in the colon (Co), dietary fiber produces short-chain fatty acids (SCFAs), which modulate host physiology through endocrine actions in the gut and systemic effects on peripheral organs. OBJECTIVES: We hypothesized that delivering SCFAs to the small intestine (SI) instead of the colon would yield higher systemic SCFA concentrations (small intestinal cells do not metabolize SCFA), yet lower endocrine effects (density of enteroendocrine cells is lower in the SI). METHODS: To test our hypothesis, we conducted a randomized, single-blinded (participants), crossover

Segment-specific intestinal bacterial community structure is associated with short-chain fatty acid profiles and mucosal morphology in two high-altitude sheep breeds.

The gastrointestinal tract of grazing ruminants contains segment-specific microbial niches that contribute to fermentation metabolism and intestinal function. However, the intestinal bacterial communities and their relationships with fermentation products and mucosal morphology remain insufficiently characterized in alpine sheep breeds. In this study, Tianhua mutton sheep and Gansu Alpine fine-wool sheep maintained under the same grazing conditions were compared using the techniques of hematoxylin and eosin (H&E) staining, gas chromatography, and full-length 16S rRNA sequencing. Histological a

Establishing the Role of Liver Fatty Acid-Binding Protein in Post-Golgi Very-Low-Density Lipoprotein Trafficking Using a Novel Fluorescence-Based Assay.

The liver plays a crucial role in maintaining lipid homeostasis by converting toxic free fatty acids into VLDL, which the body uses for energy. Even minor changes in VLDL formation and secretion can result in serious health conditions such as atherosclerosis and non-alcoholic fatty liver disease. Despite the importance of VLDL, the proteins and signaling pathways involved in its regulation remain largely unknown. This study aims to develop a novel methodology to study intracellular VLDL transport events and explore the role of liver fatty acid-binding protein (LFABP) in VLDL transport and secr

Functional divergence of fabp genes in common carp and the effects of high-fat diets supplemented with linalool on growth, fatty acid composition and fabp gene expressions.

The fatty acid-binding protein (fabp) genes involved in β-oxidation were identified and characterized in the common carp (Cyprinus carpio) genome. Fish were then fed high-fat diets supplemented with various amounts of linalool, a natural peroxisome proliferator-activated receptor α ligand, for 9 weeks. We determined that C. rapio genome contains 19 fabp genes. The mRNA expression of 4R-whole genome duplication copies of fabp genes was differently regulated by the diets, except for fabp3a.1/fabp3a.2 and fabp7a.1/fabp7a.2. These findings indicate that the transcriptional regulatory mechanisms go

Bufalin Suppresses Colorectal Cancer Liver Metastasis by Inhibiting De Novo Fatty Acid Synthesis via the PI3K/AKT-Mediated SREBP1/FASN Pathway.

BACKGROUND: Colorectal cancer (CRC) is the third most common cancer worldwide, with liver metastasis being the leading cause of mortality. De novo fatty acid synthesis plays a critical role in CRC progression and metastasis. Bufalin, a cardiotonic steroid isolated from toad skin, has demonstrated anticancer activity in multiple preclinical models. However, the mechanisms underlying its suppression of CRC metastasis and modulation of fatty acid synthesis remain to be elucidated. METHODS: The effects of bufalin on CRC cell proliferation, migration, and apoptosis were assessed via colony formati

Different effects of Lorenzo's oil components against very long-chain fatty acid-induced endoplasmic reticulum stress in peroxisome-deficient CHO cells.

Adrenoleukodystrophy (ALD) is an X-linked peroxisomal disorder caused by mutations in the ABCD1 gene, leading to the accumulation of very long-chain fatty acids (VLCFAs). The accumulation of saturated VLCFAs, such as C24:0 and C26:0, is believed to impair myelination. A mixture of C18:1 (oleic acid) and C22:1 (erucic acid), known as Lorenzo's oil, has been used to reduce these saturated VLCFAs. However, despite lowering saturated VLCFA levels, Lorenzo's oil proved ineffective in preventing neurological symptoms. Previously, we found that VLCFA-induced apoptosis is prevented by C18:1 supplement

Impacts of dietary arginine supplementation on performance, nutrient digestion and expression of proteins related to milk fatty acid and casein synthesis in early lactating dairy cows.

Arginine (Arg) can potentially increase milk yields in early lactating dairy cows by stimulating the development of mammary gland and the biosynthesis of milk proteins and fatty acids (FA); however, the underlying mechanism is still unclear. The hypothesis that rumen-protected Arg addition would increase milk, milk fat, and milk protein yields was tested, aiming to dlucidate the possible mechanism. Forty-eight multiparous Chinese Holstein dairy cows (milk yield 34.9 ± 1.27 kg/d, body weight 658 ± 12.7 kg, and days in lactation [DIL] 16.4 ± 2.56) were selected on the principle of proximity and

5-Heptadecylresorcinol ameliorates high-fat diet induced non-alcoholic fatty liver disease through modulating bile acid profile and suppressing inflammation.

BACKGROUND: The risk of non-alcoholic fatty liver disease (NAFLD) has been inversely associated with whole grain consumption. 5-Heptadecylresorcinol (AR-C17) is a phenolic lipid present in the bran part of whole grain wheat, which has been proven to ameliorate obesity and atherosclerosis. However, whether AR-C17 could alleviate NAFLD and the underlying molecular mechanisms remain unclear. RESULTS: AR-C17 administration (150 mg/kg) significantly attenuated high-fat diet (HFD)-induced NAFLD in mice by reducing serum total cholesterol (TC), low-density lipoprotein cholesterol (LDL-C), and aspart

The crosstalk between fatty acid metabolism disorder and mitochondrial dysfunction is involved in hepatotoxicity co-induced by cadmium and molybdenum in ShaoXing duck (Anas platyrhyncha) livers.

Excessive molybdenum (Mo) and cadmium (Cd) exert negative influences on organism. Our prior experiments attested that Mo and Cd brought liver damage in ShaoXing ducks (Anas platyrhyncha); this injury mechanism has not yet been fully clarified. Hence, the hepatotoxicity mechanisms of Mo and Cd were roundly analyzed by network toxicology in the study, the analysis confirmed that Mo and Cd primarily induce liver injury through lipid metabolism dysregulation, oxidative stress, and mitochondrial dysfunction. Further validation demonstrated that Mo and/or Cd upregulated fatty acid synthesis, reduced

Gut microbiota composition combined with reduced intestinal fatty acid uptake prevents hepatic steatosis in obesity-resistant mice fed a high-fat diet.

The metabolic response to excessive caloric intake varies and some individuals remain resistant to obesity with different tendencies to develop hepatic steatosis. Gut microbiota may play a crucial role in these differences by influencing energy utilization. The aim of this study was to investigate gut microbiota composition that may underlie variations in the occurrence of hepatic steatosis between obese and obesity-resistant mice. After 14 weeks on a high-fat diet (60 kcal% fat), the male C57BL/6J mice with the highest body mass were categorized as obese, while the animals without weight gain

Enhancing tofacitinib's therapeutic efficacy in murine arthritis with a synbiotic formulation comprising Bacillus megaterium DSM 32963 and an Omega-3 fatty acid lysine salt.

INTRODUCTION: Omega-3 polyunsaturated fatty acids (n3-PUFA) are known for their anti-inflammatory benefits, particularly in chronic conditions like rheumatoid arthritis (RA). To resolve an acute inflammation, conversion of n3-PUFA into specialized pro-resolving mediators (SPM) is crucial. Recently, it was shown that the probiotic Bacillus megaterium DSM32963 supports this conversion. METHODS: This study evaluates a synbiotic formulation combining Bacillus megaterium DSM32963 and a unique n3-PUFA-lysine salt as adjunct nutritional supplement to tofacitinib in adjuvant-induced arthritis (AIA) i

Gut microbiota dysbiosis and short-chain fatty acid depletion in phlegm-dampness polycystic ovary syndrome: a cross-sectional 16S rRNA sequencing analysis.

BACKGROUND: Polycystic ovary syndrome (PCOS) is a prevalent endocrine and metabolic disorder characterized by hyperandrogenism, insulin resistance, and reproductive dysfunction. Accumulating evidence indicates that gut microbiota and their metabolites, particularly short-chain fatty acids (SCFAs), contribute to PCOS pathogenesis. However, subtype-specific alterations, especially in Traditional Chinese Medicine (TCM)-defined phlegm-dampness PCOS, remain insufficiently explored. OBJECTIVE: This study aimed to comprehensively compare gut microbial composition, SCFA concentrations, and their asso
