Type 2 diabetes mellitus is associated with the fibrosis severity in patients with nonalcoholic fatty liver disease in a large retrospective cohort of Japanese patients.

BACKGROUND: The prevalence of nonalcoholic fatty liver disease (NAFLD) and metabolic syndrome have been increasing worldwide. The associations between metabolic factors and the histologic severity of NAFLD have not yet been clarified. Therefore, we studied the relationships between relevant metabolic factors and the histological severity of NAFLD. METHODS: In a cross-sectional multicenter study conducted in Japan, we examined 1,365 biopsy-proven NAFLD patients. The frequencies of underlying lifestyle-related diseases and their relationships to the NAFLD histology were investigated. RESULTS:

LDL receptor knock-out mice are a physiological model particularly vulnerable to study the onset of inflammation in non-alcoholic fatty liver disease.

BACKGROUND & AIMS: Non-alcoholic steatohepatitis (NASH) involves steatosis combined with inflammation, which can progress into fibrosis and cirrhosis. Exploring the molecular mechanisms of NASH is highly dependent on the availability of animal models. Currently, the most commonly used animal models for NASH imitate particularly late stages of human disease. Thus, there is a need for an animal model that can be used for investigating the factors that potentiate the inflammatory response within NASH. We have previously shown that 7-day high-fat-high-cholesterol (HFC) feeding induces steatosis an

High fat diet-induced non alcoholic fatty liver disease in rats is associated with hyperhomocysteinemia caused by down regulation of the transsulphuration pathway.

BACKGROUND: Hyperhomocysteinemia (HHcy) causes increased oxidative stress and is an independent risk factor for cardiovascular disease. Oxidative stress is now believed to be a major contributory factor in the development of non alcoholic fatty liver disease, the most common liver disorder worldwide. In this study, the changes which occur in homocysteine (Hcy) metabolism in high fat-diet induced non alcoholic fatty liver disease (NAFLD) in rats were investigated. METHODS AND RESULTS: After feeding rats a standard low fat diet (control) or a high fat diet (57% metabolisable energy as fat) for

Diet, commensals and the intestine as sources of pathogen-associated molecular patterns in atherosclerosis, type 2 diabetes and non-alcoholic fatty liver disease.

Stimulation of the innate immune receptors Toll-like receptor (TLR)-2 and TLR4 has been shown to promote the development of a variety of diseases involving dysregulated metabolism, including atherosclerosis, type 2 diabetes and non-alcoholic fatty liver disease. However, the origin and nature of the agents responsible for stimulating TLR2 or TLR4 signalling in these conditions remain to be clearly identified. This review summarises the evidence supporting the proposal that 'pathogen-associated molecular patterns' (PAMPs) derived from dietary and commensal sources may contribute to the chronic

Effects of conjugated linoleic acid supplementation on fatty acid composition in the plasma, liver, and epididymal fat pads of male-Sprague Dawley rats.

The purpose of this study was to investigate the effects of conjugated linoleic acid (CLA) on the fatty acid composition of plasma, liver, and epididymal fat pads in rats. Seventy-two male Sprague-Dawley rats were divided into four groups and received beef tallow (BT), fish oil (FO), beef tallow with CLA (BTC), and fish oil with CLA (FOC). All the rats were fed an experimental diet containing 12% (wt/wt) total fat, including CLA at 1%, for 30 weeks. The fatty acid analyses of the plasma, liver, and epididymal fat pads were performed by the one-step methylation method, followed by gas chromatog

Postgastrectomy development or accentuation of focal fatty change in segment IV of the liver: correlation with the presence of aberrant venous branches of the parabiliary venous plexus.

BACKGROUND: Focal fatty change in segment IV (S4) of the liver is occasionally a diagnostic dilemma in the postoperative follow-up of gastric cancer patients. GOAL: To determine whether development of focal fatty change in S4 is related to the presence of aberrant venous branches of the parabiliary venous plexus (PBP). STUDY: Two hundred seventy-six patients with gastrectomy who had been imaged by thin-slice preoperative computed tomography (CT) and at least one postoperative CT, and had no apparent liver masses were retrospectively analyzed. Preoperative CT were evaluated for the presence o

The total body mass of fatty acid ethyl esters in skeletal muscles following ethanol exposure greatly exceeds that found in the liver and the heart.

AIMS: Skeletal muscle appears to be susceptible to chronic and acute excess alcohol intake, giving rise to alcoholic myopathy, a common disease among alcoholics. Fatty acid ethyl esters (FAEE), non-oxidative metabolites of ethanol, have been shown to be toxic to cells in vitro and in vivo. We hypothesized that accumulation of FAEE in skeletal muscle could contribute to the development of alcoholic myopathy. METHODS: Male wistar rats were treated either with 75 mmol ethanol/kg body weight or saline, in the fed state or starved for 1 or 2 days before administration. Rats were thus divided into

Decreased concentration of serum apolipoprotein C-III in cows with fatty liver, ketosis, left displacement of the abomasum, milk fever and retained placenta.

Apolipoprotein (apo) C-III is a low molecular mass protein mainly distributed in the high-density lipoprotein (HDL) fraction. In cows with postparturient diseases such as ketosis, concentrations of cholesterol, phospholipids and apoA-I and the activity of lecithin:cholesterol acyltransferase, which are mainly distributed in or functionally associated with HDL, are reduced. The purpose of the present study was to examine whether the serum concentration of apoC-III was similarly decreased in the postparturient diseases. Compared with healthy controls, the apoC-III concentration was significantly

Aberrant mitochondrial respiration in the livers of rats infected with Fasciola hepatica: the role of elevated non-esterified fatty acids and altered phospholipid composition.

The non-esterified fatty acid (NEFA) content and phospholipid composition of mitochondria isolated from the livers of Wistar rats infected with Fasciola hepatica were examined in relation to the aberrant mitochondrial respiration previously reported [Rule, Behm, and Bygrave (1989) Biochem. J. 260, 517-523]. At 2 weeks post-infection, elevated NEFA levels were associated with uncoupling of mitochondrial respiration that was reversible in vitro by the addition of BSA. State IV respiration rates showed a strong correlation with NEFA content. At 3 weeks post-infection, NEFA content had increased f

Interrelationships between biotin, choline and other B-vitamins and the occurrence of fatty liver and kidney syndrome and sudden death syndrome in broiler chickens.

1. Addition of supplemental choline to a biotin-deficient diet decreased the biotin status of chicks and increased mortality from fatty liver and kidney syndrome (FLKS). 2. Mortality was also increased by dietary supplementation with a mixture of other B-vitamins, excluding biotin, and was highest when the choline and B-vitamin supplements were combined. 3. The occurrence of sudden death syndrome (SDS) was unaffected by dietary biotin concentration. 4. A previously unreported condition was observed in which birds died showing post-mortem signs characteristic of both FLKS and SDS and whose occu

Ethyl Gallate Ameliorates High-Fat Diet-Induced Non-Alcoholic Fatty Liver Disease by Suppressing Inflammation and Modulating the Gut Microbiota-Intestinal Barrier Axis.

Non-alcoholic fatty liver disease (NAFLD) is a metabolic disorder with rising global prevalence, yet effective therapeutic options remain limited. Chronic inflammation and gut microbiota dysbiosis are key contributors to NAFLD progression. Ethyl gallate (EG), a natural polyphenolic compound, exhibits anti-inflammatory and antioxidant properties. However, its therapeutic potential and underlying mechanisms in NAFLD remain unclear. This study aimed to evaluate the effects of EG in a high-fat diet (HFD)-induced NAFLD mouse model. Mice were fed an HFD for 12 weeks, with EG administered (10 or 20 m

Multi-target synergistic mechanisms of flavonoid compounds from traditional Chinese medicine in non-alcoholic fatty liver disease: insights for human and veterinary medicine.

INTRODUCTION: Non-alcoholic fatty liver disease (NAFLD) is a prevalent chronic liver disorder characterized by dysregulated hepatic lipid metabolism, with a continuously rising global incidence and limited safe and effective therapeutic options. Importantly, NAFLD-like conditions, namely hepatic lipidosis or fatty liver syndrome, also prevail in veterinary clinical practice, affecting companion animals (obese cats and dogs) and livestock (periparturient dairy cows, fattening pigs, and broiler chickens). These metabolic liver disorders are primarily induced by inappropriate feeding management a
