Differential Neuronal Development in iPSC-Derived Neural Stem Cells From Monozygotic Twin Cases With Treatment-Resistant Schizophrenia and Discordant Responses to Clozapine.

Treatment-resistant schizophrenia (TRS) affects 20%-30% of individuals diagnosed with schizophrenia and is effectively managed with clozapine. However, the molecular and cellular mechanisms that underlie its efficacy remain largely unclear. We previously generated induced pluripotent stem cell (iPSC) lines from a unique pair of monozygotic twins with TRS. One twin responded to clozapine (CLZ-res), while the other did not (CLZ-non-res). Building on our previous study of these twins, we included healthy controls and focused on the early developmental stages of neuronal differentiation. To invest

Shared genetic architecture of gray matter deficits in schizophrenia and bipolar disorder: evidence from structural neuroimaging-genetic analyses.

BACKGROUND: Schizophrenia (SCZ) and bipolar disorder (BD) share substantial clinical and neuroanatomical features, yet the neurogenetic basis underlying their shared gray matter volume (GMV) deficits remains poorly understood. METHODS: We conducted meta-analyses to identify convergent GMV alterations across the two disorders. Genome-wide association studies (GWAS) were performed to uncover genetic variants associated with the shared GMV deficits region in UK Biobank participants. Polygenic risk score (PRS)-GMV associations were analyzed to examine the cumulative influence of genetic risk on G

Co-Occurrence of Achondroplasia and 16p11.2 Deletion Syndrome in a Child With Group 4 Medulloblastoma: Expanding the Spectrum of Dual Genetic Diagnoses.

Achondroplasia (OMIM #100800), the most common skeletal dysplasia, results from the recurrent FGFR3 c.1138G > A (p.Gly380Arg) gain-of-function variant, which constitutively activates fibroblast growth factor receptor 3 signaling and impairs endochondral ossification. The 16p11.2 microdeletion (OMIM #611913) is a recurrent ~600 kb copy-number variant associated with developmental delay, autism spectrum disorder, epilepsy, and obesity, encompassing dosage-sensitive genes including MAPK3, KCTD13, and PRRT2. Recent genome-wide analyses have linked 16p11.2 deletion to a 13.9-fold increased risk of

Impact of per- and polyfluoroalkyl substances (PFAS) on brain development and function.

Per- and polyfluoroalkyl substances (PFAS), persistent environmental chemicals known as "forever chemicals," demonstrate significant neurotoxic effects across the human lifespan. This review examines the evidence linking PFAS exposure to neurodevelopmental disorders and neurological dysfunction. Epidemiological studies have shown that prenatal PFAS exposure is associated with reduced intelligence quotient in children, increased attention-deficit/hyperactivity disorder and autism spectrum disorder, and impaired motor and cognitive development, with males exhibiting greater susceptibility. Intrinsic elaboration of prefrontal modularity: a dual-control model of axon bundling and synaptic docking.

The evolutionary expansion of the primate prefrontal cortex (PFC) presents a profound biological enigma: how does this region achieve a highly ordered, modular architecture in the absence of direct dense sensory templates that govern primary sensory areas? In this review, we synthesize classical neuroanatomical frameworks with recent advances in spatial transcriptomics and connectomics to delineate a model of intrinsic elaboration. We propose that PFC modularity emerges from a developmental program facilitated by expansion of the outer subventricular zone (OSVZ) and the legacy of whole-genome

Ecdysteroid signaling acts as a central switch underlying wing polyphenism in two hemipteran insects.

Wing polyphenism represents a prevalent evolutionary adaptation in insect taxa, characterized by the capacity of a single genotype to produce distinct wing morphologies in response to environmental variations. Across hemipteran lineages, the regulation of wing polyphenism involves taxon-specific endocrine signaling cascades; however, the evolutionarily conserved genetic toolkit that underlies this developmental plasticity remains elusive. Here, we find that the steroid hormone ecdysteroids serve as a central hormonal switch that orchestrates wing polyphenism through dual downstream integration

A Novel SINE Transposable Element-Associated lncRNA Regulates Zygotic Genome Activation in Porcine Parthenogenetically Activated Embryos.

Zygotic genome activation (ZGA) is a pivotal event during early mammalian embryonic development and is regulated by multiple molecular factors. In mice, previous studies have demonstrated that long non-coding RNAs (lncRNAs) and transposable elements (TEs) participate in the regulation of this process. However, the functional roles of TEs-associated lncRNAs in porcine ZGA remain largely unknown. In this study, we identified a previously unannotated lncRNA associated with a SINE transposable element sequence in porcine parthenogenetically activated embryos, which we named LincSAKP. LincSAKP exhi

Sublethal effects of matrine and azadirachtin on the wolf spider Pardosa laura: no impairment of growth but marked suppression of reproduction.

BACKGROUND: Plant-derived pesticides (PDPs) are increasingly promoted as 'eco-friendly' alternatives to agrochemicals because of their low vertebrate toxicity, yet their safety for non-target arthropod natural enemies is rarely tested, especially the enrichment effect via trophic transfer. RESULTS: We cultured fruit flies using media supplemented with two widely applied Chinese PDPs in tea plantations, matrine or azadirachtin, under commercial concentration, which were fed to newly emerged spiderlings of wolf spider Pardosa laura (Araneae: Lycosidae) throughout their entire life cycle. We exa

Rad53 orchestrates divergent pathways governing genotoxic resistance, morphogenesis, and virulence in Candida albicans: Mechanistic decoupling through overexpression analysis.

The DNA damage response (DDR), an evolutionarily conserved surveillance mechanism enabling cells to detect genomic lesions and cell cycle arrest for repair, coordinates genomic stability through key effector kinases. In Candida albicans, our prior work characterized Rad53-mediated transcriptional reprogramming based on the RAD53 deletion strain, establishing its canonical DDR functions. However, Rad53 activation dynamically orchestrates cell cycle synchronization and repair coordination during genotoxic challenge. Investigating cellular modifications resulting from increased Rad53 functionalit

FBRSL1 regulates the expression of chromatin regulators BRPF1 and KAT6A.

FBRSL1-associated syndrome is a rare congenital malformation and intellectual disability syndrome caused by heterozygous truncating variants in Fibrosin-Like 1 (FBRSL1). While FBRSL1 is known to be involved in embryonic development, its precise molecular function remains poorly understood. Therefore, the aim of this study was to elucidate the molecular function of FBRSL1, which is thought to be essential for developmental processes, and to investigate the effect of patient-derived truncating FBRSL1 variants. Using chromatin immunoprecipitation followed by sequencing (ChIP-Seq), we show that FB

Effectiveness of 6-nitrobenzo[d]thiazol-2-amine derivative (N2) in ameliorating ethanol-induced fatty liver through lipogenesis inhibition and anti-inflammatory expression in a zebrafish model.

This study examines the effectiveness of the 6-nitrobenzo[d]thiazol-2-amine derivative (N2) in mitigating the effects of ethanol-induced fatty liver disease in zebrafish by assessing its anti-inflammatory, antioxidant, and antihyperlipidemic properties. The investigation encompasses the evaluation of N2 derivatives on hemolysis, cytotoxicity, developmental toxicity, oxidative stress, and lipid metabolism within zebrafish models. Antioxidant assays highlighted N2's significant radical scavenging activity. Cytotoxic evaluations conducted on HepG2 cells and human red blood cells (RBCs) revealed m

Pyraclostrobin induces developmental toxicity and cardiotoxicity through oxidative stress and inflammation in zebrafish embryos.

Pyraclostrobin, a typical representative of strobilurin fungicides, is extensively used in agriculture to control fungi and is often detected in water bodies and food. However, the comprehensive toxicological molecular mechanism of pyraclostrobin requires further study. To assess the toxic effects and underlying mechanisms of pyraclostrobin on aquatic organisms, zebrafish embryos were exposed to pyraclostrobin (20, 40, and 60 μg/L) until 96 h post fertilization (hpf). These results indicated that exposure to pyraclostrobin induces morphological alterations, including spinal curvature, shortene

Single-Cell Profiling of Premature Neonate Airways Reveals a Continuum of Myeloid Differentiation.

Single-cell genomic technologies hold great potential to advance our understanding of lung development and disease. A major limitation lies in accessing intact cells from primary lung tissues for profiling human airway health. Sampling methods such as endotracheal aspiration that are compatible with clinical interventions could enable longitudinal studies, the enrollment of large cohorts, and the development of novel diagnostics. To explore single-cell RNA sequencing profiling of the cell types present at birth in the airway lumen of extremely premature neonates (<28 wk gestation), we isolated

Zika virus vertical transmission in interferon receptor1-antagonized Rag1-/- mice results in postnatal brain abnormalities and clinical disease.

The mechanisms by which vertically transmitted Zika virus (ZIKV) causes postnatal brain development abnormalities and congenital disease remain poorly understood. Here, we optimized the established anti-IFNAR1 treated, Rag1-/- (AIR) mouse model of ZIKV infection to examine the consequence of vertical transmission on neonate survival and postnatal brain development. We found that modulating the infectious dose and the frequency of anti-IFNAR1 treatment of pregnant mice (termed AIRlow mice) prolonged neonatal survival allowing for pathogenesis studies of brain tissues at critical postnatal time

Clinical, biochemical, and molecular characterization of mild (nonclassic) rhizomelic chondrodysplasia punctata.

Rhizomelic chondrodysplasia punctata (RCDP) is a heterogenous group of disorders due to defects in genes encoding peroxisomal proteins required for plasmalogen (PL) biosynthesis, specifically PEX7 and PEX5 receptors, or GNPAT, AGPS and FAR1 enzymes. Most patients have congenital cataract and skeletal dysplasia. In the classic form, there is profound growth restriction and psychomotor delays, with most patients not advancing past infantile developmental milestones. Disease severity correlates to erythrocyte PL levels, which are almost undetectable in severe (classic) RCDP. In milder (nonclassic

Repeated evolution of eye loss in Mexican cavefish: Evidence of similar developmental mechanisms in independently evolved populations.

Evolution in similar environments often leads to convergence of behavioral and anatomical traits. A classic example of convergent trait evolution is the reduced traits that characterize many cave animals: reduction or loss of pigmentation and eyes. While these traits have evolved many times, relatively little is known about whether these traits repeatedly evolve through the same or different molecular and developmental mechanisms. The small freshwater fish, Astyanax mexicanus, provides an opportunity to investigate the repeated evolution of cave traits. A. mexicanus exists as two forms, a sigh

Congenital lymphedema as a rare and first symptom of tuberous sclerosis complex.

Lymphedema are characterized by interstitial edema leading to swelling of extremities. They can be divided into primary and secondary lymphedema. Developmental abnormalities of the lymphatic system are responsible for the primary form of lymphedema. The secondary form of lymphedema is caused by damage of the lymphatic system due to external factors. Lymphedema can rarely be observed in patients with tuberous sclerosis complex (TSC), which is a neurocutaneous syndrome caused by pathogenic variants in the genes TSC1 or TSC2. Patients with TSC usually present with neurological manifestations and

Caudal Regression Syndrome-A Review Focusing on Genetic Associations.

Caudal regression syndrome (CRS) represents a spectrum of clinical phenotypes with varying degrees of malformation of the lower body with involvement of structures deriving from all 3 layers of the trilaminar embryo. We review areas of active investigation in the diagnosis, etiology, epidemiology, and treatment of the disease with a focus on underlying genetics. CRS pathobiology is complex and multifactorial with a significant contribution from environmental factors as evidenced in twin studies. Contemporary genomic and genetic investigations in both human primary tissue and murine in vitro an
