Multilayered Regulatory Dynamics of p53 Mutations and Platinum Resistance in Ovarian Cancer.

TP53 mutation-driven gene expression programs define oncogenic phenotypes. While extensive studies have concentrated on the transcriptome and proteome, post-transcriptional processes, particularly translational variation, remain underexplored. This study presents a comprehensive analysis of the transcriptomics, translatiomics, and proteomics dynamics in the ovarian cancer cell line SKOV3, with a focus on the effects of p53 missense mutations (R175H, R273H, and Y220C) on gene dosage fluctuations. Despite clear transcriptional differences between wild-type and mutant p53, we find that extensive

Enhanced biomethanation in anaerobic membrane bioreactor: decarbonization efficiency, carbon mass flow, and microbial dynamics.

Methanation of carbon dioxide (CO2) is a key process for improving carbon utilization technology. In this study, we investigated a novel approach to biomethanation from hydrogen (H2) and CO2 using an anaerobic membrane bioreactor under mesophilic conditions. System performance was evaluated by varying carbon loading rates, with a focus on decarbonization efficiency, carbon mass flow, and microbial dynamics. The results demonstrated stable operation and strong resilience, achieving a decarbonization efficiency higher than 84 % and a methane (CH4) flow-out rate of 1.1 L/L/d at a carbon loading r

Hbo1 and Msl complexes preserve differential compaction and H3K27me3 marking of active and inactive X chromosomes during mitosis.

In mammals, chromosome-wide regulatory mechanisms ensure a balance of X-linked gene dosage between males (XY) and females (XX). In female cells, expression of genes from one of the two X chromosomes is curtailed, with selective accumulation of Xist-RNA, Xist-associated proteins, specific histone modifications (for example, H3K27me3) and Barr body formation observed throughout interphase. Here we show, using chromosome flow-sorting, that during mitosis, Xist-associated proteins dissociate from inactive X (Xi) chromosomes, while high levels of H3K27me3 and increased compaction of the Xi relative

Synergistic optimization of bioretention cells for antibiotic removal and denitrification by iron-copper bimetallic catalytic system with Phalaris arundinacea.

Bioretention cells have been less studied for treating stormwater runoff and removing antibiotics and denitrification. In this experiment, bioretention cells with iron-copper bimetallic catalysis and Phalaris arundinacea were constructed and synergistically optimized. The performance of antibiotic removal and denitrification under single and compound antibiotic stress was investigated, and the microecological environment of the system was analyzed. The results showed that under single and compound antibiotic stress, the removal rates of ofloxacin (OFLX) and tetracycline (TC) reached 91.69 %-99

NKX2-5 Mutation and ICD Implantation in a Pregnant AV Block Patient.

BACKGROUND: Arrhythmias and conduction disorders are relatively common during pregnancy and are often amplified by the physiological changes of gestation. Although new-onset complete heart block is uncommon, it has been linked to mutations in the NKX2-5 gene, which is associated with congenital heart disorders and progressive conduction system defects. CASE SUMMARY: We present the case of a pregnant woman with an NKX2-5 gene mutation who developed progressive atrioventricular nodal disease and chronotropic incompetence. She underwent dual-chamber implantable cardioverter-defibrillator placeme

Sitosterolemia caused by compound heterozygosis of 2 allelic variants in the ABCG5 gene-21 years of follow-up.

BACKGROUND: Sitosterolemia is a rare autosomal recessive disease characterized by elevated phytosterol levels in the bloodstream and tissues due to increased absorption and reduced biliary excretion. This condition arises from mutations in one of two genes, ABCG5 or ABCG8, located on chromosome 2p21. OBJECTIVE: We report the case of a patient, and his first-degree relatives, diagnosed with sitosterolemia at the age of 23. METHODS: The patient was monitored for 21 years, during which he was advised to adopt a low-plant sterol diet and was treated with ezetimibe. RESULTS: Over this period, he

Airway stenosis: classification, pathogenesis, and clinical management.

Airway stenosis (AS) is a fibroinflammatory disease characterized by abnormal activation of fibroblasts and excessive synthesis of extracellular matrix, which has puzzled many doctors despite its relatively low prevalence. Traditional treatment such as endoscopic surgery, open surgery, and adjuvant therapy have many disadvantages and are limited in the treatment of patients with recurrent AS. Therefore, it is urgent to reveal the pathogenesis of AS and accelerate its clinical transformation. Based on the discovered pathogenesis, including fibrosis, inflammation, epithelial-mesenchymal transiti

[Update of pathology in medullary thyroid carcinoma].

Medullary thyroid carcinoma (MTC) is the most common neuroendocrine carcinoma within the thyroid gland, characterized by strong invasiveness, high metastasis and recurrence rates. It is relatively rare among thyroid malignancies. The cytological and histological features of MTC are diverse and disperse, presenting as papillary, follicular, solid, trabecular, and spindle cell patterns. Immunohistochemical staining shows variable expression of calcitonin, carcinoembryonic antigen, and neuroendocrine markers. MTC can be classified into hereditary and sporadic types, with most cases caused by germ

Prevalence of intronic repeat expansions in the RFC1 gene in Polish patients with cerebellar syndrome.

Cerebellar ataxia with neuropathy and vestibular areflexia syndrome (CANVAS) is a recessively inherited neurodegenerative ataxic disorder, which has been associated with intronic biallelic repeat expansions in the RFC1 gene. Our objective was to assess retrospectively the prevalence of CANVAS in Polish population. We screened 2523 Polish patients in whom other repeat expansions were excluded. To determine the repeat expansions in the RFC1 gene in patients, we performed RFC1-flanking PCR and repeat primed PCR (RP-PCR) and to measure the size of the expansion we used Southern blotting and optica

Growth behavior and mRNA expression profiling during growth of IPEC-J2 cells.

OBJECTIVE: The IPEC-J2 cell line is used as an in vitro small intestine model for swine, but it is also used as a model for the human intestine, presenting a relatively unique setting. By combining electric cell-substrate impedance sensing, with next-generation-sequencing technology, we showed that mRNA gene expression profiles and related pathways can depend on the growth phase of IPEC-J2 cells. Our investigative approach welcomes scientists to reproduce or modify our protocols and endorses putting their gene expression data in the context of the respective growth phase of the cells. A combined therapy of meropenem-ZnO nanoparticles efficiently eliminates carbapenem-resistant Klebsiella pneumoniae biofilms, with reduced nephrotoxicity (in vitro).

In response to the World Health Organization's research agenda of antimicrobial resistance in human health, this study appraised the antibacterial and antibiofilm synergistic activity of meropenem and ZnO nanoparticles (ZnO-NPs) combination against carbapenem-resistant Klebsiella pneumoniae (CRKP). The minimum inhibitory concentration (MIC) of meropenem in combination was found to be ~1/12 of its MIC alone. The results of microtiter dilution assay showed that the combination was more efficient in reducing the biofilm biomass than meropenem alone or ZnO-NPs alone. The scanning-electron-microsco

HIV-1 Vif global diversity and possible APOBEC-mediated response since 1980.

HIV-1 Vif's principal function is to counter the antiretroviral activities of DNA-editing APOBEC3 cytidine deaminases. Unconstrained APOBEC3 activity introduces premature stop codons in HIV-1 genes and can lead to viral inactivation. To investigate the evolution and diversification of Vif over the HIV-1 pandemic and document evidence of APOBEC3-mediated pressure, we analyzed 4612 publicly available sequences derived from 10 dominant subtypes and circulating recombinant forms (CRFs) using the Hervé platform. We found widespread evidence of diversifying selection that was convergent across subty

Diving into the zebrafish brain: exploring neuroscience frontiers with genetic tools, imaging techniques, and behavioral insights.

The zebrafish (Danio rerio) is increasingly used in neuroscience research. Zebrafish are relatively easy to maintain, and their high fecundity makes them suitable for high-throughput experiments. Their small, transparent embryos and larvae allow for easy microscopic imaging of the developing brain. Zebrafish also share a high degree of genetic similarity with humans, and are amenable to genetic manipulation techniques, such as gene knockdown, knockout, or knock-in, which allows researchers to study the role of specific genes relevant to human brain development, function, and disease. Zebrafish

Akaluc bioluminescence offers superior sensitivity to track in vivo dynamics of SARS-CoV-2 infection.

Monitoring in vivo viral dynamics can improve our understanding of pathogenicity and tissue tropism. Because the gene size of RNA viruses is typically small, NanoLuc is the primary choice for accommodation within viral genome. However, NanoLuc/Furimazine and also the conventional firefly luciferase/D-luciferin are known to exhibit relatively low tissue permeability and thus less sensitivity for visualization of deep tissue including lungs. Here, we demonstrated in vivo sufficient visualization of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection using the pair of a codon-o

Cross-species transmission and host range genes in poxviruses.

The persistent epidemic of human mpox, caused by mpox virus (MPXV), raises concerns about the future spread of MPXV and other poxviruses. MPXV is a typical zoonotic virus which can infect human and cause smallpox-like symptoms. MPXV belongs to the Poxviridae family, which has a relatively broad host range from arthropods to vertebrates. Cross-species transmission of poxviruses among different hosts has been frequently reported and resulted in numerous epidemics. Poxviruses have a complex linear double-strand DNA genome that encodes hundreds of proteins. Genes related to the host range of poxvi

Stem Cells and Targeted Gene Therapy in Brain and Spinal Cord Tumors.

The CNS tumors, in particular those with malignant characteristics, are prominent burdens around the world with high mortality and low cure rate. Given that, researchers were curious about novel treatments with promising effectiveness which resulted in shifting the dogmatism era of neurogenesis to the current concept of postnatal neurogenesis. Considering all existing stem cells, various strategies are available for treating CNS cancers, including hematopoietic stem cells transplantation, mesenchymal stem cells transplantation, neural stem cells (NSCs) transplantation, and using stem cells as

Characteristics of intestinal microbiota in C57BL/6 mice with non-alcoholic fatty liver induced by high-fat diet.

INTRODUCTION: As a representation of the gut microbiota, fecal and cecal samples are most often used in human and animal studies, including in non-alcoholic fatty liver disease (NAFLD) research. However, due to the regional structure and function of intestinal microbiota, whether it is representative to use cecal or fecal contents to study intestinal microbiota in the study of NAFLD remains to be shown. METHODS: The NAFLD mouse model was established by high-fat diet induction, and the contents of the jejunum, ileum, cecum, and colon (formed fecal balls) were collected for 16S rRNA gene analys

Genetic Causes of Cerebral Small Vessel Diseases: A Practical Guide for Neurologists.

Cerebral small vessel disease (CSVD) includes various entities affecting the brain and, often, systemic small arteries, arterioles, venules, and capillaries. The underlying causes of CSVD are different, and some of them are genetic. Monogenic CSVDs are responsible for 1%-5% of all strokes and for several other disturbances. Despite many genes being involved, the phenotypes of monogenic CSVD partly overlap. Given that the genetic testing for different diseases can be challenging and time-consuming, the practicing neurologist should be adequately informed of the genetic background of CSVD and sh
