Chimeric antigen receptor T cells for sustained remissions in leukemia.

BACKGROUND: Relapsed acute lymphoblastic leukemia (ALL) is difficult to treat despite the availability of aggressive therapies. Chimeric antigen receptor-modified T cells targeting CD19 may overcome many limitations of conventional therapies and induce remission in patients with refractory disease. METHODS: We infused autologous T cells transduced with a CD19-directed chimeric antigen receptor (CTL019) lentiviral vector in patients with relapsed or refractory ALL at doses of 0.76×10(6) to 20.6×10(6) CTL019 cells per kilogram of body weight. Patients were monitored for a response, toxic effect

A novel membrane protein, VanJ, conferring resistance to teicoplanin.

Bacterial resistance to the glycopeptide antibiotic teicoplanin shows some important differences from the closely related compound vancomycin. They are currently poorly understood but may reflect significant differences in the mode of action of each antibiotic. Streptomyces coelicolor possesses a vanRSJKHAX gene cluster that when expressed confers resistance to both vancomycin and teicoplanin. The resistance to vancomycin is mediated by the enzymes encoded by vanKHAX, but not by vanJ. vanHAX effect a reprogramming of peptidoglycan biosynthesis, which is considered to be generic, conferring res

Recirculating cardiac delivery of AAV2/1SERCA2a improves myocardial function in an experimental model of heart failure in large animals.

Abnormal excitation-contraction coupling is a key pathophysiologic component of heart failure (HF), and at a molecular level reduced expression of the sarcoplasmic reticulum (SR) Ca(2+) ATPase (SERCA2a) is a major contributor. Previous studies in small animals have suggested that restoration of SERCA function is beneficial in HF. Despite this promise, the means by which this information might be translated into potential clinical application remains uncertain. Using a recently established cardiac-directed recirculating method of gene delivery, we administered adeno-associated virus 2 (AAV2)/1S

Use of replication-competent retroviral vectors in an immunocompetent intracranial glioma model.

OBJECT: The authors had previously reported on a replication-competent retrovirus (RCR) that has been demonstrated to be stable, capable of effective transduction, and able to prolong survival in an intracranial tumor model in nude mice. The purpose of this study was further investigation of this gene therapy option. METHODS: The transduction efficiency of RCR in RG2, an immunocompetent intracranial tumor model, was tested in Fischer 344 rats. The immune response to the RCR vector was expressed by the quantification of CD4, CD8, and CD11/b in tumors. The pharmaceutical efficacy of the suicide

Inducible regulation of Runx2-stimulated osteogenesis.

Ex vivo gene therapy is a promising approach to orthopedic regenerative medicine. These strategies typically focus on the constitutive overexpression of osteogenic factors to induce osteoblastic differentiation and matrix mineralization. However, the unregulated production of osteoinductive molecules has also resulted in abnormal bone formation and tumorigenesis. To address these limitations, this work describes a retroviral system to deliver the Runx2 osteoblastic transcription factor under control of the tetracycline-inducible (tet-off) promoter in primary skeletal myoblasts. Runx2 expressio

Simple and effective osteoinductive gene therapy by local injection of a bone morphogenetic protein-2-expressing recombinant adenoviral vector and FK506 mixture in rats.

We have previously utilized a human bone morphogenetic protein-2 (BMP-2)-expressing recombinant adenoviral vector (AxCAOBMP-2) for osteoinductive gene therapy in rats. However, immunosuppression is essential for osteoinduction by AxCAOBMP-2 and this is one of the major impediments to its clinical use. Injection of AxCAOBMP-2 together with the immunosuppressant FK506 made it possible to markedly reduce the dose of the immunosuppressive agent and still induce ectopic bone reliably. We injected AxCAOBMP-2 and FK506 into the right calf muscle of rats, while the same number of plaque forming units

Transduction patterns of pseudotyped lentiviral vectors in the nervous system.

We have developed a non-primate-based lentiviral vector based on the equine infectious anemia virus (EIAV) for efficient gene transfer to the central and peripheral nervous systems. Previously we have demonstrated that pseudotyping lentiviral vectors with the rabies virus glycoprotein confers retrograde axonal transport to these vectors. In the present study we have successfully produced high-titer EIAV vectors pseudotyped with envelope glycoproteins from Rhabdovirus vesicular stomatitis virus (VSV) serotypes (Indiana and Chandipura strains); rabies virus [various Evelyn-Rokitnicki-Abelseth ER

Sustained transduction of ocular cells with a bovine immunodeficiency viral vector.

Human immunodeficiency viral (HIV) vectors mediate long-term transduction of many types of nondividing cells in vivo. Bovine immunodeficiency virus (BIV) is a lentivirus that shares many characteristics with HIV, but does not cause human disease. In this study, we investigated the potential of BIV vectors for ocular gene therapy. An enhanced green fluorescent protein (eGFP)-encoding reporter gene was packaged in recombinant BIV vector (BIV.eGFP). Adult C57BL/6 mice were given an intravitreous (5 x 10(4) or 5 x 10(5) transducing units [TU]) or subretinal (5 x 10(5) TU) injection of BIV.eGFP and

Analysis of promoters from tyrosine/dihydroxyphenylalanine decarboxylase and berberine bridge enzyme genes involved in benzylisoquinoline alkaloid biosynthesis in opium poppy.

Tyrosine/dihydroxyphenylalanine decarboxylase (TYDC) and the berberine bridge enzyme (BBE) represent the entry point and a key branch point, respectively, in the biosynthesis of benzylisoquinoline alkaloids in select species of the Papaveraceae and Fumariaceae. Genomic clones for tydc7 and bbe1 from opium poppy (Papaver somniferum L.) were isolated. Deletion analysis of tydc7 and bbe1 5'-flanking regions revealed the location of putative regulatory domains necessary for expression of the beta-glucuronidase (gus) reporter gene in a transient assay system based on the microprojectile bombardment

Transcriptional repression of PTEN in neural cells using CRISPR/dCas9 epigenetic editing.

After damage to the adult mammalian central nervous system (CNS), surviving neurons have limited capacity to regenerate and restore functional connectivity. Conditional genetic deletion of PTEN results in robust CNS axon regrowth, while PTEN repression with short hairpin RNA (shRNA) improves regeneration but to a lesser extent, likely due to suboptimal PTEN mRNA knockdown using this approach. Here we employed the CRISPR/dCas9 system to repress PTEN transcription in neural cells. We targeted the PTEN proximal promoter and 5' untranslated region with dCas9 fused to the repressor protein Krüppel-

The hologenomic stress legacy: A framework for soil as a transgenerational memory bank of plant immunity.

Plants pass transgenerational inheritance through genetic and epigenetic information; however, the soil's role as the host environment in this memory transfer remains largely unexplored. Research within this field has largely been domain-specific, with only 117 publications in 24 years integrating fields of soil persistent metabolites (soil chemistry), stress-induced microbial consortia (soil microbiology), and plant molecular science (plant epigenetics). In this perspective article, we synthesize evidence and propose a testable hypothesis, the Hologenomic Stress Legacy (HSL) hypothesis. It st

Engineered mesenchymal stem cells expressing CXCR4 and LIGHT enhance Anti-Tumor activity in a subcutaneous gastric cancer xenograft model.

Gastric cancer remains a leading cause of cancer-related mortality, with limited treatment options and a poor prognosis. Mesenchymal stem cells (MSCs) possess inherent tumor-homing ability but exhibit modest efficacy against solid tumors. Enhancing both tumor targeting and immune activation through genetic modification may improve the therapeutic potential of these approaches. To evaluate the anti-cancer efficacy of human umbilical cord–derived MSCs (HUC-MSCs) genetically modified to co-express C-X-C chemokine receptor type 4 (CXCR4) and TNFSF14 (LIGHT) against gastric cancer in vitro and in v

Comparative Analysis of Induced Pancreatic Stem Cells Generated with Different Factors.

Recently, our research group generated induced tissue-specific stem/progenitor (iTS/iTP) cells. Compared with induced pluripotent stem (iPS) cells, iTS/iTP cells offer several advantages, that is, easy generation, higher differentiation efficiency, and no teratoma formation. In this study, iTS cells were generated from mouse pancreatic tissues (iTS-P cells) using two different methods. Plasmid vectors were used for expressing Oct3/4, Sox2, Klf4, c-Myc (OSKM), or Yap1 (YAP) to evaluate the efficiency and differentiation potential of the resulting cells. No significant difference in reprogrammin

Conditionally Reprogrammed Cells Preserve Cellular Diversity and Permit Genetic Manipulation: Implications for Cancer Heterogeneity and Tissue Regeneration.

Our previous study demonstrated that a Rho kinase inhibitor (Y-27632), in combination with irradiated fibroblast feeder cells, enables both normal and tumor epithelial cells from various tissues to proliferate indefinitely in vitro, without the need for exogenous viral or cellular gene transduction. These cells are referred to as Conditionally Reprogrammed Cells (CRCs). This approach has shown great promise for applications in regenerative and personalized medicine. In the present study, we first investigated whether CRCs could be genetically manipulated and stably express exogenous genes. Usi

Engineering IL-10-Overexpressing MSCs via a Non-Viral PEG-PEI Nanoplatform for Potent Therapy of Inflammatory Bowel Disease.

Genetic engineering of therapeutic cells is a key strategy to enhance cell-based therapies, yet current gene delivery methods-viral vectors, electroporation, and commercial non-viral reagents-are limited by safety concerns, high cost, operational complexity, cytotoxicity, and poor scalability. We developed a safe, efficient, low-cost, and scalable non-viral gene delivery platform using a polyethylene glycol-polyethyleneimine (PEG-PEI) copolymer to engineer mesenchymal stromal cells (MSCs) for inflammatory bowel disease (IBD) treatment. The PEG-PEI copolymer was synthesized via covalent conjuga

Molecular Detection and Genetic Diversity of Bat-Associated Bartonella spp. in Bat Ectoparasites Collected From Ratchaburi Province, Thailand.

INTRODUCTION: Bats are known to harbour Bartonella bacteria, which can be found in several species of bats. Various ectoparasites can infest bats and harbour the bacteria, serving as important vectors for diverse Bartonella species. This study aimed to assess the prevalence and diversity of Bartonella spp. in bat ectoparasites, as well as to identify the species of ectoparasites involved. METHODS: Bat ectoparasites were collected from four species of insectivorous bats in Ratchaburi, central Thailand. The ectoparasites were identified using the Cytochrome oxidase subunit I (COI) gene (bat fli
