Antibacterial action of anacardic acids against methicillin resistant Staphylococcus aureus (MRSA).

The structural and antibacterial activity relationship of 6-alk(en)ylsalicylic acids, also known as anacardic acids, was investigated against Gram-positive bacteria, emphasizing the methicillin resistant Staphylococcus aureus ATCC 33591 (MRSA) strain. The unsaturation in the alkyl side chain is not essential in eliciting activity but is associated with increasing the activity. The antibacterial activity of methicillin against MRSA strains was significantly enhanced in combination with C(12:0)-anacardic acid, and the fractional inhibitory concentration index for this combination was calculated

Antibiotic-resistant organism infection.

Bacteria possess a remarkable number of ways to become resistant to antibiotics. Antibiotic resistance has become a major problem in the treatment of Gram-positive infections. Resistance to methicillin and vancomycin in staphylococci and enterococci has resulted in organisms that are resistant to all known antibiotics. Although it is important to continue to search for newer and more effective antibiotics, it is imperative that we develop a surgical mindset of appropriate antibiotic stewardship. The use of single-dose prophylactic regimens, using narrow-spectrum agents when possible for therap

Diagnosis and treatment challenges in a rare Clostridium infection: A case report.

The Clostridium genus includes >180 species of Gram-positive, anaerobic, sporulating bacteria. Under certain conditions, these can cause a wide range of invasive infections in humans. Clostridium paraputrificum occurs in the commensal intestinal flora and related bacteremia typically occurs secondary to an injury to the intestinal mucosa and in the presence of predisposing conditions, such as gastrointestinal disorders, malignancies, diabetes, HIV infection or neutropenia. The current study presents the case of a 70-year-old male patient, a rural resident living in poverty, with a history of a

Phylogeny of Catabacter hongkongensis Strains Responsible for Bacteremia Is Not Associated with Clinical Outcomes or Therapeutic Efficacy.

Catabacter hongkongensis is a Gram-positive rod, isolated in 2007 in blood culture. Fewer than 15 infections have been reported. Herein, we present a lethal case of bacteremia due to C. hongkongensis identified through phylogenetic analyses. A woman was found unconscious in a context of chronic diarrhea. An abdominal abscess with a hydroaeric level was discovered, associated with sigmoid adenocarcinoma and peritoneal carcinomatosis. Despite hospitalization in an ICU and the adaptation of antibiotic therapy, the patient died. Blood cultures were positive in the final stage of the disease (>60 h

Plasmid profile of ESBL producing Gram-negative bacteria and correlation with susceptibility to beta-lactam drugs.

BACKGROUND: Members of family Enterobacteriaceae can acquire resistance to extended spectrum beta lactams by a number of mechanisms; most important being the plasmid encoded extended spectrum beta lactamase (ESBL) and AmpC beta lactamase. This study has been designed to look for the presence of plasmids and their correlation with drug resistance. METHODS: ESBL production was studied in different gram-negative bacteria and susceptibility testing of ESBL positive isolates was done for various beta lactams, cephalosporins and other commonly used drugs against them. Plasmid DNA isolation of all t

Presumptive diagnosis of anaerobic bacteremia by gas-liquid chromatography of blood cultures.

By quantitative gas-liquid chromatography of glucose-containing blood cultures at the moment of first signs of growth, a presumptive diagnosis of anaerobic bacteremia could be made in 24 out of 26 cultures yielding obligate anaerobes upon subsequent culture. With Bacteroides sp. (20 strains isolated), elevated levels of isovaleric acid (greater than or equal to 0.1 mumol/ml) and/or succinic acid (greater than or equal to 5 mumol/ml) were detected in the medium used. An exception to this were two strains of Bacteroides thetaiotaomicron that did not produce sufficient quantities of these acids.

Fungal L-Asparaginase from environmental isolates with antimicrobial and antibiofilm activities.

Fourteen fungal species were isolated from sewage water and soil, of which ten strains showed positive extracellular L-asparaginase production by plate assay. Among them, Aspergillus nidulans AUMC17371, A. flavus AUMC17373, A. flavus AUMC17374, and A. terreus AUMC17372 exhibited the highest enzyme activities (1.35, 1.25, 0.97, and 0.79 U/mL, respectively). Optimization of culture conditions revealed that maximum production was achieved using fructose as carbon source, asparagine as nitrogen source, pH 6, and 30 °C. SDS-PAGE analysis confirmed enzyme production, with protein bands ranging from

A Daptomycin Resistance-Conferring cls1 I269T Mutation in Enterococcus faecium and its Global Prevalence.

Daptomycin is widely considered as a ‘last-resort’ antibiotic used for the treatment of antibiotic-resistant Gram-positive bacteria, including vancomycin-resistant Enterococcus. Resistance to this antibiotic can lead to severe consequences, including inability to find a cure for Gram positive infections. Our previous investigations showed that a mutated cls1 gene mediated low-level daptomycin resistance in Enterococcus faecium strain EF332, distinguishing it from previous reports that only linked cls2 mutations to daptomycin resistance. In this study, through additional genetic and microbiolog

Crosstalk Between BmToll9-2, the Toll Pathway, and Antimicrobial Peptides in the Silkworm (Bombyx mori) Larval Fat Body.

Insects lack adaptive immunity and rely exclusively on innate immune system for pathogen defense. However, the specific role of BmToll9-2, a key Toll receptor in the silkworm (Bombyx mori), in mediating immune responses, particularly against bacterial challenges in the larval fat body, remains incompletely understood. To address this gap, this study employed a combination of molecular approaches, including RNA interference (RNAi) targeting BmToll9-2, bacterial challenges with heat-inactivated Escherichia coli (Gram-negative) and Staphylococcus aureus (Gram-positive), and quantitative real-time

Development of pyrazole and pyrazolopyrimidine derivatives as promising anti-MRSA agents targeting penicillin-binding protein (PBP2a) and mecA gene.

Global health is threatened by methicillin-resistant S. aureus (MRSA). The rising prevalence of MRSA complicates anti-infective treatment strategies, highlighting the urgent need for novel therapeutics targeting MRSA. In this study, novel pyrazole and pyrazolopyrimidine derivatives were synthesized and comprehensively characterized using IR, 1H/13C NMR, and elemental analysis. Significant anti-microbial activity of these derivatives was demonstrated (using MIC test) against both Gram-positive (S. aureus ATCC 25923 and MRSA ATCC 43300) and Gram-negative bacteria (E. coli ATCC 25922 and K. pneum

Stabilization of a Cu-binding site by a highly conserved tryptophan residue.

Copper serves as an essential cofactor for nearly all living organisms. There are still many gaps remaining in our knowledge of how Gram-positive bacteria import copper and maintain homeostasis. To obtain a better understanding of how these processes work, here we focus on the ycnKJI operon responsible for regulating copper levels in the Gram-positive bacterium Bacillus subtilis. This operon encodes three Cu-related proteins: a copper-dependent transcriptional repressor (YcnK), a putative copper importer (YcnJ), and a copper-binding protein of unknown function (YcnI). We previously found that

Vancomycin dosing, monitoring and toxicity: Critical review of the clinical practice.

Vancomycin is an antibiotic used in the treatment of infections caused by multidrug-resistant Gram-positive bacteria, especially methicillin-resistant Staphylococcus aureus. In the last decades, vancomycin has been widely used in hospital environments due to the increasing incidence of sepsis and septic shock. Sepsis may lead to multiple organ failure; it is considered a risk factor for the development of acute kidney injury (AKI), with an overall mortality rate of around 45%, which can reach the surprising rate of 70% with the combination of AKI and sepsis. Considering the high mortality rate

Signals triggered by a bacterial pore-forming toxin contribute to toll-like receptor redundancy in gram-positive bacterial recognition.

BACKGROUND: Toll-like receptor (TLR) 2 is the principal recognition receptor for gram-positive microbes. However, in some gram-positive bacterial infections, TLR2 is dispensable. One of the outstanding questions regarding host-bacteria interactions is why TLR2 is essential in some infections but dispensable in others. METHODS: We used a combination of bacterial plating, flow cytometry, enzyme-linked immunosorbent assay, and reverse-transcriptase polymerase chain reaction to analyze the inflammatory responses induced by Listeria monocytogenes and its toxin listeriolysin O (LLO) in vitro and in

Epidemiology of vascular catheter-related bloodstream infections in a large university hospital in Israel.

Intravascular catheter-related infection and associated bacteraemia constitute a serious and increasing problem among nosocomial infections. As a part of an ongoing survey of positive blood cultures, all catheter-related bloodstream infections (CR-BSI) were reviewed in the authors' Medical Center in 1996, in order to evaluate the magnitude and seriousness of this problem. The largest group (28%) of hospital-acquired bacteraemia by 1 source of infection during 1996 was CR-BSI, identified in 110 patients with 126 episodes. The vascular line was central in 83 (66%), peripheral in 24 (19%), tunnel
