Plasma concentrations of leptin in a bulimic patient.

Recently, the obese gene in the ob/ob mouse was cloned, along with its human homologue. The gene product leptin is important in the regulation of body weight. Excessive food intake during a binge might affect leptin synthesis. Alternatively, fluctuations in leptin synthesis might induce binge eating. Therefore, plasma leptin levels of a patient with bulimia nervosa were determined over a period of 48 hr in a natural setting. Amount, type, time of food intake, and binging and purging episodes were concomitantly assessed. Although binging and purging episodes were quite frequent, leptin levels r

Mouse myocilin (Myoc) gene expression in ocular tissues.

Human myocilin is identical to TIGR (trabecular meshwork inducible glucocorticoid response) which is responsible for the pathogenesis of juvenile-onset primary open angle glaucoma (GLCIA). We have isolated cDNA for mouse myocilin (Myoc) and investigated mouse myocilin gene expression in ocular tissues with in situ RNA hybridization. Hybridization signals were observed in the iris, ciliary body, trabecular meshwork, sclera, and retina in the mouse eye. The marked signals were seen in trabecular meshwork cells and the anterior portion of sclera. These findings suggest that myocilin mutation coul

Anatomy of the orbit.

The normal orbit arguably contains the most complex anatomy of the human body. The orbit's small size, complex anatomy, and elaborate function make it a diagnostic challenge to evaluate. Magnetic resonance imaging (MRI) and computed tomography (CT) are essential studies for visualization of most normal and abnormal orbital structures. These studies also can help to understand some of the functional anatomical relationships within the orbit. This article attempts to review and illustrate the normal anatomical structures that can be identified on CT and MR images. These structures include the bo

Hyaluronan in the exfoliation syndrome.

PURPOSE: To histochemically analyze if the glycoseaminoglycan component of pseudoexfoliation material consists of hyaluronan. METHODS: The hyaluronan binding region of cartilage proteoglycans was utilized in a staining specific for hyaluronan. The technique has been described and employed previously. Human donor eyes with and without the presence of visible pseudoexfoliations were analyzed. RESULTS: Hyaluronan was found to coat the fibrillar exfoliation material on the lens, the zonules, the iris epithelium and the ciliary body. It was also found on a capsular bag following cataract surgery.

Acoustic trauma caused by lightning.

Lesions produced by exposure to noise are frequent in everyday life. Injuries may be found in all systems of the human body, from the digestive to the endocrine, from the cardiovascular to the nervous system. Many organs may be damaged, the ear being one of them. It is known that noise produced by factories, airports, musical instruments and even toys can cause auditory loss. Noises in nature can also cause acoustic trauma. This report is the case history of acoustic trauma caused by lightning. The patient was studied with CAT scan, electroencephalogram, and brain mapping, impedance audiometry

Tissue distribution of human acetylcholinesterase and butyrylcholinesterase messenger RNA.

Cholinesterase inhibitors occur naturally in the calabar bean (eserine), green potatoes (solanine), insect-resistant crab apples, the coca plant (cocaine) and snake venom (fasciculin). There are also synthetic cholinesterase inhibitors, for example man-made insecticides. These inhibitors inactivate acetylcholinesterase and butyrylcholinesterase as well as other targets. From a study of the tissue distribution of acetylcholinesterase and butyrylcholinesterase mRNA by Northern blot analysis, we have found the highest levels of butyrylcholinesterase mRNA in the liver and lungs, tissues known as t

Molecular basis of mouse microphthalmia (mi) mutations helps explain their developmental and phenotypic consequences.

Mutations in the mouse microphthalmia (mi) gene affect the development of a number of cell types including melanocytes, osteoclasts and mast cells. Recently, mutations in the human mi gene (MITF) were found in patients with Waardenburg Syndrome type 2 (WS2), a dominantly inherited syndrome associated with hearing loss and pigmentary disturbances. We have characterized the molecular defects associated with eight murine mi mutations, which vary in both their mode of inheritance and in the cell types they affect. These molecular data, combined with the extensive body of genetic data accumulated f

Abnormal accumulation of phospholipase C-delta in filamentous inclusions of human neurodegenerative diseases.

We have previously demonstrated that an antibody to phosphoinositide-specific phospholipase C (PLC) isozyme, PLC-delta, intensely stained neurofibrillary tangles (NFT) in the brain tissue of Alzheimer's disease (AD). This study was performed to determine if abnormal PLC-delta accumulation might be present in the filamentous inclusions of other neurodegenerative diseases. We found that the anti-PLC-delta antibody stained neuronal inclusions of Pick's disease, progressive supranuclear palsy and diffuse Lewy body disease while the inclusions of idiopathic Parkinson's disease lacked PLC-delta accu

Human UDP-glucuronosyl transferases: chemical defence, jaundice and gene therapy.

Human UDP-glucuronosyltransferases (UDPGTs) are a family of enzymes which detoxify many hundreds of compounds by their conjugation to glucuronic acid, rendering them both harmless and more water soluble, hence, excretable. The level of expression of each UDPGT isoform in the body is the result of interplay between temporal, tissue-specific and environmental regulators. This complexity contributes to the difficulty in predicting the metabolic fate of compounds. Genetic defects and polymorphisms affecting individual isoform activities have deleterious and potentially lethal effects, as exemplifi

Penetration of industrial chemicals across the skin: a predictive model.

The recently reported dermal absorption and toxicity potential of industrial chemicals is reconsidered using an alternative physicochemically based model of skin penetration. In this model, the outermost, and least permeable, component of the skin [namely, the stratum corneum (SC)] is considered to provide only a lipoidal transport pathway into the body for chemicals that come into contact with the skin. The predictive algorithm of the model is biophysically compatible with known SC properties, and is based on experimental determinations of permeability coefficients through human skin in vitro

The pathogenesis of Plasmodium falciparum-associated tissue lesions--what role for C3b receptors?

Human tissue sections from cerebral and cerebellar cortices, the liver, the stomach pylorus, the kidney and the placenta, were examined for the presence of C3b receptors. This was carried out in order to determine whether these receptors play any role in the pathological effects of malarial infections on these tissues. The sheep erythrocyte/rabbit antibody/complement system was used. C3b receptors were only detected in the kidney glomeruli, but not in the other tissues. It is suggested that during malarial infections complement may be indirectly involved in the pathogenesis of tissue damage in

Hyperbaric diuresis and nocturia--a review.

Human divers are now able to engage in a multi-day dive to a considerable depth using the mixed gas saturation diving technique, however their daily activities are somewhat limited in the depth. One of the major problems that has not been generally appreciated is the potential for body fluid disturbances at high pressure due to the development of a sustained diuresis. In addition, a marked nocturia has been observed in recent saturation dives at 26-31 atmospheres absolute (ATA), which disturbs divers' sleep during the night. In this review, we will briefly discuss the basic characteristics and

Serologic differences in strains of Sporothrix schenckii.

To obtain evidence that Sporothrix scheneckii enters the body by contact with contaminated materials, the antigenic property of strains from different sources was investigated. The reciprocal absorption test of the antisera against a soil isolate and a human isolate (KO 4606) showed that the absorbed antisera against KO 4606 possessed unique antigen(s) in addition to the common antigen of both strains. Twenty-three clinical isolates were tested with absorbed antisera. Not all of them possessed the unique antigen(s), but there were serologic varieties among S. schenckii strains, regardless of t

The problem of chondromalacia patellae.

Daily activity subjects the human patella to forces often several times the individual's body weight. Healthy cartilage can adjust to these forces if they are not too excessive, concentrated, or repetitive. Such abnormal stresses most frequently occur with the disturbance of normal patellar mechanical function. Chondromalacia patellae is the result common to a wide variety of unusual traumata. Treatment must be directed primarily not toward the damaged patellar cartilage but toward a correction of the mechanical abnormality causing it. Until proven otherwise, a young female complaining of knee

Evidence of in-vivo myofascial force transfer in humans- a systematic scoping review.

BACKGROUND: The fascial system not only enables the body to operate in an integrated manner but modifies its tension in response to the stress on it. Recent animal, cadaveric and in-vitro trials have shown that "myofascial force transmission" (MFT) can play a major role in homeostasis, musculoskeletal function and pain. Human evidence for the in-vivo existence of MFT is scarce. OBJECTIVE: This scoping review attempts to gather and interpret the available evidence of the in-vivo existence of MFT in humans, its role in homeostasis, and musculoskeletal function. METHOD: A search of major databa

Detecting volatile organic compounds to locate human remains in a simulated collapsed building.

The occurrence of mass disasters has increased worldwide due to changing environments from global warming and a heightened threat of terrorism acts. When these disasters strike, it is imperative to rapidly locate and recover human victims, both the living and deceased. While search and rescue dogs are used to locate the living, cadaver detection dogs are typically tasked with locating the dead. This can prove challenging because commingling of victims is likely to occur during disasters in populated areas which will impact the decomposition process and the resulting odour produced. To date, th

'The Internet Both Reassures and Terrifies': exploring the more-than-human worlds of health information using the story completion method.

Lay people are now encouraged to be active in seeking health and medical information and acting on it to engage in self-care and preventive health practices. Over the past three decades, digital media offering ready access to health information resources have rapidly expanded. In this article, I discuss findings from my study that sought to investigate health information practices by bringing together the social research method of story completion with more-than-human theory and postqualitative inquiry. Narratives of health, illness and embodiment are powerful ways to portray people's experien

Metabolism of 26,26,26,27,27,27-F6-1 alpha,23S,25-trihydroxyvitamin D3 by human UDP-glucuronosyltransferase 1A3.

26,26,26,27,27,27-Hexafluoro-1alpha,25-dihydroxyvitamin D(3) [F(6)-1alpha, 25(OH)(2)D(3)], which is now clinically used as a drug for secondary hyperparathyroidism, is a hexafluorinated analog of the active form of vitamin D(3). Our previous studies demonstrated that CYP24A1 is responsible for the metabolism of F(6)-1alpha,25(OH)(2)D(3) in the target tissues and that F(6)-1alpha,25(OH)(2)D(3) was successively converted to F(6)-1alpha,23S,25(OH)(3)D(3) and F(6)-23-oxo-1alpha,25(OH)(2)D(3). In this study, we examined the metabolism of F(6)-1alpha,25(OH)(2)D(3),F(6)-1alpha,23S,25(OH)(3)D(3), and
