Safety profiles of intravesical Bacillus Calmette-Guerin in bladder cancer.

Bladder cancer (BCa) exhibits significant sex disparities, and intravesical Bacillus Calmette-Guerin (BCG) is a widely used treatment for non-muscle invasive bladder cancer (NMIBC). A comprehensive evaluation of intravesical BCG, including the safety profile and adverse events (AEs), is essential. In particular, exploring the sex differences is crucial. We conducted a pharmacovigilance data analysis using real-world big data from the Food and Drug Administration Adverse Event Reporting System (FAERS) database. In our study, we analyzed 3,374 cases of intravesical BCG for signal mining. We dete

Elevated host lipid metabolism revealed by iTRAQ-based quantitative proteomic analysis of cerebrospinal fluid of tuberculous meningitis patients.

PURPOSE: Tuberculous meningitis (TBM) remains to be one of the most deadly infectious diseases. The pathogen interacts with the host immune system, the process of which is largely unknown. Various cellular processes of Mycobacterium tuberculosis (MTB) centers around lipid metabolism. To determine the lipid metabolism related proteins, a quantitative proteomic study was performed here to identify differential proteins in the cerebrospinal fluid (CSF) obtained from TBM patients (n = 12) and healthy controls (n = 12). METHODS: CSF samples were desalted, concentrated, labelled with isobaric tags

Activation of intrinsic immune responses and microglial phagocytosis in an ex vivo spinal cord slice culture model of West Nile virus infection.

UNLABELLED: West Nile virus (WNV) is a neurotropic flavivirus that causes significant neuroinvasive disease involving the brain and/or spinal cord. Experimental mouse models of WNV infection have established the importance of innate and adaptive immune responses in controlling the extent and severity of central nervous system (CNS) disease. However, differentiating between immune responses that are intrinsic to the CNS and those that are dependent on infiltrating inflammatory cells has proven difficult. We used a murine ex vivo spinal cord slice culture (SCSC) model to determine the innate imm

Neurology of inflammatory bowel disease.

Inflammatory bowel diseases (IBD) are chronic inflammatory conditions affecting the digestive system, comprising two main distinctive entities, ulcerative colitis (UC) and Crohn's disease (CD). Besides gastrointestinal manifestations, IBD causes extraintestinal manifestations in the central and peripheral nervous system. The incidence of neurological complications in IBD ranges from 0.25% to 47.5%. The pathophysiology of neurological manifestations of IBD is mostly immune mediated, but dysfunction of the brain-gut axis, arterial and venous thromboembolism, infections, nutritional deficiencies

Immunologic role of the cholinergic anti-inflammatory pathway and the nicotinic acetylcholine alpha 7 receptor.

Cytokines are small protein molecules that facilitate communication between cells of the immune system and other tissues. Their messenger function elicits responses in other cells, ranging from DNA binding and tissue remodeling to coordinating the local cellular response to inflammation. The magnitude of the cytokine response is regulated closely, because an over- or underabundance of cytokine activity can impair organ function and cause shock and tissue injury. Counterregulatory molecular and humoral mechanisms protect the host from cytokine excess, including the pituitary-adrenal-glucocortic

[Autoimmune liver diseases and their overlap syndromes].

Autoimmune diseases of the liver are chronic inflammatory diseases leading to an etiologically undefined immune-mediated attack aimed at the hepatocyte, small microscopic bile ducts, and the entire biliary system detectable by cholangiography, respectively. From the standpoint of clinical disease three entities can be distinguished: autoimmune hepatitis (AIH), primary biliary cirrhosis (PBC), and primary sclerosing cholangitis (PSC). These are not only different regarding their clinical profile but also differ in diagnostic strategy, therapeutic regimen and probability of remission, as well as

Bite of the wolf: innate immune responses propagate autoimmunity in lupus.

The etiopathogenesis of systemic lupus erythematosus (SLE), a clinically heterogeneous multisystemic syndrome that derives its name from the initial characterization of facial lesions that resemble the bite of a wolf, is considered a complex, multifactorial interplay between underlying genetic susceptibility factors and the environment. Prominent pathogenic factors include the induction of aberrant cell death pathways coupled with defective cell death clearance mechanisms that promote excessive externalization of modified cellular and nuclear debris with subsequent loss of tolerance to a wide

Beyond immune privilege: the brain as a dynamic immunological interface.

Classically viewed as an "immune‑privileged" site, the central nervous system (CNS) was believed to be shielded from peripheral immune surveillance by the blood-brain barrier (BBB) and the absence of conventional lymphatic vessels. Recent discoveries, particularly the identification of functional meningeal lymphatic vessels and the glymphatic system, recast the CNS as a dynamic immunological interface. Here, we synthesize advances that explain how immune cells access brain border tissues and parenchyma, how resident glia (microglia and astrocytes) shape inflammatory tone and repair, and how gu

Chronic Stress and Latent Virus Reactivation: Effects on Immune Aging, Chronic Disease Morbidity, and Mortality.

OBJECTIVES: Social stress has been shown to affect immune functioning. Past research has found that chronic social stress and latent viral infections accelerate immune aging, leading to chronic disease morbidity and mortality. Chronic stress may also reactivate latent viral infections, like cytomegalovirus (CMV), accelerating the aging of the immune system. METHOD: Utilizing panel survey data from 8,995 U.S. adults aged 56 or older from the Health and Retirement Study, this study investigates whether chronic stress interacts with CMV positivity to drive aging of the immune system, multimorbid

Schizophrenia and psychoneuroimmunology: an integrative view.

PURPOSE OF REVIEW: Since decades immunological aberrancies have been reported in schizophrenia patients. As schizophrenia represents a heterogenous disorder with a variety of clinical manifestations, complex interactions between the immune system in the brain might have important etiological implications. RECENT FINDINGS: Recent findings of altered expression of immune-related genes, changes of peripheral and central cytokines, antibodies and immune cells point toward dysbalanced immune response processes in schizophrenia. SUMMARY: Based on immunogenetic factors, immune dysfunctions caused b

Expression of acquired immunity to the hookworm Ancylostoma ceylanicum in hamsters.

Four experiments are described in which hamsters, initially exposed to primary infection with Ancylostoma ceylanicum, were given a homologous challenge and components of the secondary response were quantified and compared to relevant control groups. The initial establishment of the L3 larvae was not prevented in immunized hamsters but their growth was slowed and virtually all larvae were lost within a week of challenge, when the majority were still at the L4 stage of development. The loss of worms was associated with an accelerated mucosal mastocytosis and increased systemic antibody. Thus acq

Assessing the causal relationship between immune cells and prostatitis: evidence from bidirectional mendelian randomization analysis.

Prostatitis represents a common disease of the male genitourinary system, significantly impacting the physical and mental health of male patients. While numerous studies have suggested a potential link between immune cell activity and prostatitis, the exact causal role of immune cells in prostatitis remains uncertain. This study aims to explore the causal relationship between immune cell characteristics and prostatitis using a bidirectional Mendelian randomization approach. This study utilizes data from the public GWAS database and employs bidirectional Mendelian randomization analysis to inve

Single-Cell RNA Sequencing of Peripheral Blood Reveals Immune Cell Signatures in Alzheimer's Disease.

The peripheral immune system is thought to affect the pathology of the central nervous system in Alzheimer's disease (AD). However, current knowledge is inadequate for understanding the characteristics of peripheral immune cells in AD. This study aimed to explore the molecular basis of peripheral immune cells and the features of adaptive immune repertoire at a single cell level. We profiled 36,849 peripheral blood mononuclear cells from AD patients with amyloid-positive status and normal controls with amyloid-negative status by 5' single-cell transcriptome and immune repertoire sequencing usin

Immunomodulatory actions of catecholamines, cluster-specific signaling pathways, and metabolites in pheochromocytoma and paraganglioma.

Pheochromocytomas and paragangliomas (PPGLs) are rare neuroendocrine neoplasms with an exceptionally high genetic contribution, often driven by single high-penetrance mutations. Approximately one third of patients harbor germline mutations and another third carry somatic driver mutations, allowing stratification into three molecular clusters: pseudohypoxia-related (cluster 1), kinase signaling-related (cluster 2) and Wnt-signaling-related (cluster 3). These clusters differ in catecholamine secretion, malignancy, and metastatic behavior. Although PPGLs have long been viewed as immunologically '

Gut-brain-immune interactions in neonatal hypoxic-ischemic brain injury.

BACKGROUND: Neonatal hypoxia-ischemia (HI) is the leading cause of childhood mortality and neurodevelopmental disability. Despite therapeutic hypothermia as the only clinically established treatment to date, outcomes remain poor for a significant proportion of affected infants. Mechanistic understanding has been brain-oriented in the past, however the gut and immune system are increasingly recognized as active modulators of brain injury, recovery and neurodevelopment. MAIN BODY: Gut microbiota regulate microglial maturation, myelination and blood-brain barrier integrity. Neonatal HI induces g

Systemic Immunoglobulin Changes Following Adenoidectomy, Tonsillectomy, and Adenotonsillectomy in Patients With Chronic Tonsillitis and Adenoiditis: A Prospective Study.

BACKGROUND: Chronic tonsillitis and adenoiditis are common inflammatory disorders of Waldeyer's ring that may lead to persistent systemic immune stimulation due to recurrent infection of tonsillar lymphoid tissue. Because the palatine tonsils and adenoids participate in mucosal immune defense and antibody production, concerns have occasionally been raised regarding the potential immunologic consequences of surgical removal of these structures. METHODS: A prospective clinical investigation was conducted involving 100 participants, including 75 patients with chronic tonsillitis or adenoiditis a

Blood-brain barrier dysfunction and peripheral immune activation in Alzheimer's disease: an inflammation-centered review.

Alzheimer's disease (AD) is traditionally defined by amyloid-β deposition, tau pathology, synaptic failure, and progressive cognitive decline. However, growing evidence indicates that neurovascular dysfunction and systemic immune activation are not merely secondary consequences but active contributors to disease progression. The blood-brain barrier (BBB), as a dynamic immunovascular interface, regulates the communication between the central nervous system and the peripheral immune system. In AD, aging, amyloid-β toxicity, tau-related stress, vascular senescence, endothelial inflammatory signal

Inhibitors of acetylcholinesterase and butyrylcholinesterase meet immunity.

Acetylcholinesterase (AChE) inhibitors are widely used for the symptomatic treatment of Alzheimer's disease and other dementias. More recent use is for myasthenia gravis. Many of these inhibitors interact with the second known cholinesterase, butyrylcholinesterase (BChE). Further, evidence shows that acetylcholine plays a role in suppression of cytokine release through a "cholinergic anti-inflammatory pathway" which raises questions about the role of these inhibitors in the immune system. This review covers research and discussion of the role of the inhibitors in modulating the immune response

[The characteristics of the immune and neuromediator homeostasis in patients with borderline neuropsychic disorders in chronic poisoning by higher fatty-series aliphatic amines].

The authors provide the data on the etiological role played by higher aliphatic amines of the fatty series in the development of borderline neuropsychic disorders. The dopamine system was shown to play the leading part in their genesis. Activation of that system goes in parallel with disorders in immune and neuromediator homeostasis.
