DHX9 Inhibition Enhances Paclitaxel Sensitivity by Inducing Mitotic Failure in Ovarian and Endometrial Cancers.

Recurrent high-grade serous ovarian carcinoma (HGSOC) and endometrial cancer remain major clinical challenges with limited effective treatment options. DExH-box helicase 9 (DHX9), a DNA/RNA helicase essential for genomic stability, has not yet been explored as a therapeutic target in gynecologic cancers. In this study, we show that a selective DHX9 inhibitor (DHX9i) suppresses proliferation in a subset of HGSOC and endometrial cancer cell lines by inducing DNA damage, chromosomal instability, and mitotic failure. This effect was independent of microsatellite instability status and prior resist

Endophytic Fungal Metabolites as Modulators of Key Signaling Pathways in Chronic Diseases and Aging.

Chronic diseases and aging-related disorders are driven by interconnected mechanisms, including oxidative stress, low-grade inflammation, metabolic dysregulation, and glycation. Targeting these overlapping pathways remains a major challenge for conventional single-target therapeutics. In this context, endophytic fungi have emerged as a promising source of bioactive metabolites with multi-target pharmacological potential. This review provides a mechanistic overview of endophyte-derived metabolites, including alkaloids, terpenoids, polyketides, and phenolic compounds, with a focus on their abili

Increased type 3 deiodinase reduces hippocampal T3 signaling and is associated with cognitive impairment in a sporadic Alzheimer's disease animal model.

Alzheimer’s disease (AD) is a progressive neurodegenerative disorder characterized by amyloid-β and tau pathology, neuroinflammation, oxidative stress, and cognitive decline. Emerging evidence indicates that thyroid hormone (TH) dysregulation may also contribute to AD pathogenesis, particularly through impaired hippocampal signaling. THs are crucial for neuronal metabolism, plasticity, and mitochondrial integrity, and their local availability is tightly controlled by deiodinases. This study investigated the role of type 3 deiodinase (D3) and its redox-dependent regulation via glutathione (GSH)

Ecological risks of microplastics in soils of major Angelica sinensis production areas and biological responses of roots to polyethylene microplastics.

Farmland microplastic pollution has become an emerging environmental issue in agricultural ecosystems; however, systematic studies on soil microplastic contamination in the main production areas of Angelica sinensis in China and its potential effects on medicinal roots remain limited. This study analyzed the composition, ecological risks, and accumulation and biological effects of microplastics in A. sinensis roots through field investigation combined with controlled experiments. The results indicate that farmland soils in the main production areas of Gansu Province contain relatively high lev

Vascular endothelial cells as signaling niches for epithelial stem cells in the skin.

In many non-skin tissues, vascular endothelial cells (VECs) are increasingly recognized as integral components of stem cell niches engaged in bidirectional molecular crosstalk, regulating stem cell behavior through angiocrine signaling. In the skin, however, the nature and physiological relevance of VEC cues influencing epithelial stem cells remain poorly defined. While many cell types, including fibroblast, immune cells, nerves, and adipocytes are known to crosstalk to the epithelial stem cells in skin contributing to their niche, recent studies began to implicate VECs as a putative niche com

RNA acetylation modification ac4C: An emerging regulatory hub of RNA metabolism disruption in Alzheimer's disease.

RNA metabolic dysregulation is a key pathological mechanism underlying the onset and progression of Alzheimer's disease (AD), involving multiple aspects such as abnormal RNA splicing, loss of function in RNA-binding proteins, dysregulation of non-coding RNAs, and impaired nuclear-cytoplasmic transport. In recent years, the emergence of epigenome research has revealed the critical role of RNA chemical modifications in regulating RNA metabolism at the post-transcriptional level. N4-acetylcytidine (ac4C) is the only known RNA acetylation modification in eukaryotes and is specifically catalyzed by

Single-cell analysis reveals IL1B+ macrophages promote lung metastasis in adenoid cystic carcinoma.

A high rate of metastasis remains a challenge for the treatment of adenoid cystic carcinoma (ACC), yet the underlying mechanism of lung metastasis remains elusive. By integrating single-cell RNA sequencing and bulk transcriptomics with multiplex immunohistochemistry, we identified a metastatic niche consisting of IL1B+ tumor-associated macrophages (TAMs), CD24+ luminal-like tumor cells, ACKR1+ endothelial cells (ECs) and HSPA1A+CD8+ T cells. Importantly, we revealed that IL1B+ TAMs play diverse roles in the processes of tumor cell intravasation and colonization, two fundamental steps supportin

lncRNA LUCAT1 regulates DNA damage response in glioma stem cells under hypoxia.

BACKGROUND: Genomic stability is essential for cell survival, particularly under stress conditions like hypoxia, which disrupt DNA repair. Glioblastoma (GBM) is markedly resistant to DNA-damaging therapies, primarily due to glioma stem cells (GSCs) and the hypoxic tumor microenvironment. Long non-coding RNAs (lncRNAs) play a significant role in maintaining genome integrity, but their involvement in the DNA damage response (DDR) under hypoxic conditions in GSCs remains unclear. We previously identified LUCAT1 as the most highly induced lncRNA in GSCs under hypoxia and a key regulator HIF1α acti

Cell surface CD11c as a neutrophil aging marker molecule.

Neutrophils exhibit substantial functional heterogeneity shaped by both developmental stage and cellular maturation, but their mechanisms remain incompletely defined. CD11c (ITGAX) is traditionally considered as a cell surface marker of dendritic cells. We recently reported that it is highly expressed intracellularly in neutrophils and contributes to their maturation; however, its role at the cell surface is unclear. Here, we showed that surface CD11c expression level was heterogeneous across neutrophils, enabling classification into CD11chi and CD11c-/lo populations. CD11chi neutrophils displ

Methamphetamine-Driven Neuroinflammation and Parkinson's Disease Pathology: Mechanistic Insight into Nrf2 and NFĸB Signaling.

Methamphetamine (METH) abuse has emerged as a significant public health concern due to its widespread use and persistent adverse effects on brain function. Accumulating evidence indicates that chronic METH exposure disrupts dopaminergic neurotransmission and induces neurotoxic processes that overlap with key pathological features of Parkinson's disease (PD). This review critically examines clinical, epidemiological, and experimental studies exploring the association between METH use and increased vulnerability to PD-related neurodegeneration. Particular emphasis is placed on findings from anim

From theory to tissue: Constitutive modeling and underlying assumptions in cartilage biomechanics.

Articular cartilage is a charged, multiphasic tissue whose mechanical response emerges from coupled solid-fluid-ion interactions. Modeling this complexity remains a major challenge in computational biomechanics. This scoping review maps cartilage constitutive models and provides a structured, mechanics-informed appraisal of their physiological representation, constitutive assumptions, and numerical implementation practices. Database searches (1995-2025) identified 84 eligible studies. Models were classified into monophasic, biphasic, triphasic, and other constitutive families. To systematicall

Cellular senescence in cancer: Friend or fraud?

Cellular senescence, a stress-induced state of stable cell cycle arrest accompanied by a senescence-associated secretory phenotype (SASP), plays a paradoxical role in cancer biology. On the one hand, senescent cells function as a barrier to tumor initiation by activating the DNA damage response (DDR) and tumor suppressor pathways such as p53/p21 and p16INK4a-retinoblastoma (RB). On the other hand, their long-term persistence promotes chronic inflammation, immune evasion, and tissue remodeling via sustained SASP, ultimately facilitating tumor progression, metastasis, and therapeutic resistance.

A versatile phenotyping platform combining three-photon microscopy, optimized tissue clearing, and deep-learning signal separation for full-depth multi-organelle phenotyping.

Imaging the internal structures of plant organs at the subcellular level is essential for understanding key processes such as photosynthesis, growth, and stress responses. However, conventional optical microscopy struggles to visualize deep tissues owing to intense light scattering and spectral crosstalk from autofluorescent components-most prominently chlorophyll. To overcome these barriers, we present an integrated platform that combines deep-penetrating three-photon microscopy (3PM), tissue clearing, and a custom deep-learning network (SE-UNet) for intelligent signal separation. First, our

PmWRKY-10 Enhances Pine Wilt Disease Resistance by Promoting Pinosylvin Monomethyl Ether Biosynthesis Through Transcriptional Activation of PmPMT2-1.

Pine wilt disease (PWD), caused by the pine wood nematode (PWN, Bursaphelenchus xylophilus), poses a severe threat to global pine forests. Plants can enhance their resistance to external stresses by eliciting secondary metabolites; however, the transcriptional regulatory mechanisms governing this defence response in pines remain largely unexplored. Here, we report a novel defence regulatory module in Pinus massoniana. PmWRKY-10 may participate in PWN resistance by regulating the biosynthesis of the nematicidal metabolite pinosylvin monomethyl ether (PME). We first demonstrated that PME accumul

Influence of peel morphology on pesticide distribution and metabolic responses in mango after postharvest immersion treatment.

Pesticide residues in agricultural products pose substantial food safety challenges, necessitating an improved understanding of pesticide behavior for optimized application. We examined the residue distribution patterns and metabolic responses of two non-systemic pesticides (thiram and tolfenpyrad) and three systemic ones (pyriproxyfen, pyraclostrobin, and tebuconazole) in two mango cultivars with distinct peel characteristics, thick-peel R2E2 and thin-peel Guifei, following postharvest immersion. The results demonstrated that peel morphology, fruit maturity, and pesticide properties collectiv

Bridging the gap: integrating hereditary cancer into precision oncology.

Advances in next-generation sequencing have expanded the identification of hereditary cancer patients, which account for approximately 10% of all cancer diagnoses. Precision medicine has profoundly reshaped the landscape of oncology, particularly in the management of hereditary cancer syndromes, through the convergence of germline genetics and targeted therapeutics. Disruption of homologous recombination repair genes such as BRCA1/2 and PALB2 confers synthetic lethal sensitivity to Poly ADP-ribose polymerase inhibitors and combination strategies targeting replication stress or cell-cycle check

The different meanings of tolerating the gut microbiome.

Multicellular life arose in a world dominated by microorganisms, a reality that has imposed a constant and pervasive selective pressure on all subsequent complex organisms. The immune system has been historically defined by its role in pathogen clearance through resistance mechanisms. However, a complementary and equally critical strategy is to enable the peaceful and inevitable coexistence with microorganisms, allowing each host species to shelter a unique associated microbiome. The term tolerance holds multiple meanings in immunology, yet all underlie a balanced and cooperative host-microorg
