Relationships between motor-unit number estimates and isometric strength in distal muscles in ALS/MND.

Measurement of progression in amyotrophic lateral sclerosis/motor neuron disease (ALS/MND) is useful for charting the natural history and assessing efficacy in drug trials. Common measures are maximal voluntary isometric contraction (MVIC) which mainly focuses on-proximal muscles, and electrophysiologic measures of the compound muscle action potential (CMAP) and motor-unit number estimation (MUNE) which focus on distal muscles. We have undertaken a study to compare the relationships between MVIC, CMAP and MUNE recorded in intrinsic hand muscles of 10 subjects with ALS/MND. Grip and pinch (betw

Diamond-blackfan anemia: etiology, pathophysiology, and treatment.

Diamond-Blackfan anemia (DBA) is manifested by a wide variety of clinical and in vitro abnormalities. Despite this biological diversity, the hematological phenotype is remarkably similar for all patients and consists of a normochromic-macrocytic anemia in early childhood, reticulocytopenia, and a normocellular marrow with a selective deficiency of red cell precursors. Fetal hemoglobin is usually increased, distributed heterogeneously, has a fetal G gamma/A gamma pattern, and is associated with increased expression of red cell i antigen. Although most cases are sporadic, there are examples of a

Comparison between the Regenerative and Therapeutic Impacts of Bone Marrow Mesenchymal Stem Cells and Adipose Mesenchymal Stem Cells Pre-Treated with Melatonin on Liver Fibrosis.

BACKGROUND: The distinctive feature of liver fibrosis is the progressive replacement of healthy hepatic cells by the extracellular matrix protein, which is abundant in collagen I and III, with impaired matrix remodeling. The activation of myofibroblastic cells enhances the fibrogenic response of complex interactions of hepatic stellate cells, fibroblasts, and inflammatory cells to produce the excessive deposition of the extracellular protein matrix. This process is activated by multiple fibrogenic mediators and cytokines, such as TNF-α and IL-1β, accompanied with a decrease in the anti-fibroge

Falls prevention for older adults in outdoor public spaces: an interdisciplinary Delphi consensus on risks, actions, and barriers.

BACKGROUND: Falls are the leading cause of accidental injury among older adults, 30% of community-dwelling adults aged 65 and over fall each year, with nearly half occurring outdoors. These falls are complex, understudied, and insufficiently addressed in current age-friendly cities or walkability frameworks. This study aimed to build interdisciplinary consensus on risks, preventive actions, and barriers to fall prevention in outdoor public spaces through a Delphi process. METHODS: A three-phase Delphi study was conducted with 64 participants in round 1, 60 in round 2, and 49 in round 3, inclu

Two simultaneous cases of exertional heat stroke during a trail run in Guadeloupe (French West Indies): exceptional presentations or emerging trend?

BACKGROUND: Exertional heat stroke is a potentially fatal condition that can affect individuals performing intensive physical activity, particularly in hot environments. We report two simultaneous cases of EHS that occurred during the same outdoor sporting event in Guadeloupe (French West Indies) in April, in two unrelated young athletes with similar profiles but different clinical pictures. At the time of the event, the apparent temperature was 29.6 °C with a relative humidity of 85%. CASES PRESENTATION: Patient 1: A 22-year-old man with no known medical history collapsed during an endurance

Concussion history and virtual reality metrics predict core or lower extremity injury occurrence among high school athletes.

INTRODUCTION: A history of concussion is recognized as a risk factor for musculoskeletal injury, which is likely associated with physiological effects that warrant better understanding. This study aimed to assess the potential of measurements obtained from an immersive virtual reality (VR) test to identify a subtle perceptual-motor impairment that may be prospectively associated with the occurrence of a core or lower extremity sprain or strain. METHODS: A cohort of 68 high school athletes (41 female soccer players and 27 male football players) provided survey responses and completed an immers

Brain and Peripheral Atypical Inflammatory Mediators Potentiate Neuroinflammation and Neurodegeneration.

Neuroinflammatory response is primarily a protective mechanism in the brain. However, excessive and chronic inflammatory responses can lead to deleterious effects involving immune cells, brain cells and signaling molecules. Neuroinflammation induces and accelerates pathogenesis of Parkinson's disease (PD), Alzheimer's disease (AD) and Multiple sclerosis (MS). Neuroinflammatory pathways are indicated as novel therapeutic targets for these diseases. Mast cells are immune cells of hematopoietic origin that regulate inflammation and upon activation release many proinflammatory mediators in systemi

Short stem total hip arthroplasty: Potential explanations for persistent post-surgical thigh pain.

Short stem uncemented femoral implants were developed with the aim of preserving proximal bone stock for future revisions, improving biomechanical reconstruction, aiding insertion through smaller incisions and potentially decreasing or limiting the incidence of thigh pain. Despite all the advantages of short stem designs, it remains unclear whether they are able to limit post-surgical thigh pain. In patients with short stem hip arthroplasty and persistent thigh pain, it is of the utmost importance to understand the potential etiologies of this chronic pain for selecting the appropriate treatme

Centrosome amplification induces high grade features and is prognostic of worse outcomes in breast cancer.

BACKGROUND: Centrosome amplification (CA) has been reported in nearly all types of human cancer and is associated with deleterious clinical factors such as higher grade and stage. However, previous reports have not shown how CA affects cellular differentiation and clinical outcomes in breast cancer. METHODS: We analyzed centrosomes by immunofluorescence and compared to ploidy and chromosomal instability (CIN) as assessed by 6-chromosome FISH in a cohort of 362 breast cancers with median clinical follow-up of 8.4 years. Centrosomes were recognized by immunofluorescence using antibodies for per

Enthesitis: an autoinflammatory lesion linking nail and joint involvement in psoriatic disease.

The traditional model for psoriasis and psoriatic arthritis (PsA) is that autoimmunity directed against a common skin and joint autoantigen leads to chronic autoreactive T cell driven inflammation. However, recent imaging, histological and genetic studies have challenged this view, especially with respect to joint and nail disease, and provide a broader insight into the pathogenesis of PsA and associated nail involvement. Clinically unrecognized enthesitis (inflammation at tendon and ligament attachments) is commonly seen in early PsA at all sites of the disease. Specifically, enthesitis is as

Palate development.

In all vertebrates, the secondary palate arises as bilateral outgrowths from the maxillary processes. In birds and most reptiles, these palatal shelves grow initially horizontally, but do not fuse with each other resulting in physiological cleft palate. In crocodilians, shelf fusion occurs resulting in an intact secondary palate. Mammalian palatal shelves initially grow vertically down the side of the tongue, but elevate at a precise time to a horizontal position above the dorsum of the tongue and fuse with each other to form an intact palate. Palatal shelf-elevation is the result of an intrin

Distinct mobility facets and their association with intrinsic capacity domains in (pre-)frail community-dwelling older adults: an application of the unified framework for measuring mobility.

BACKGROUND: Maintaining mobility is essential for healthy aging. A recently proposed unified framework conceptualizes mobility through three facets: actual mobility, perceived mobility, and locomotor capacity. Identifying determinants of each facet is important for understanding mobility and informing tailored mobility-focused interventions. This is particularly relevant for (pre-)frail older adults, who are especially vulnerable to mobility decline. The aim of this study was to examine associations of the three mobility facets with intrinsic capacity (IC) domains and environmental factors in

Cell Cycle Phases, Their Effect on Cell Mechanical Properties, and the Impact on Candida-Host-Cell Interactions.

Cell mechanics is essential in many biological phenomena such as cell division and migration. Further, cell mechanobiological measurements can distinguish between healthy and diseased cells; thus, investigations of cell mechanics have led to the development of tools to study the elasticity and the viscosity of cells. Cell mechanics can be affected by factors such as cell morphology, cytoskeletal remodelling, and cell intrinsic factors, and these are important in understanding disease progression. Here, we use an atomic force microscopy (AFM)-microrheology with a colloidal probe for a dynamic m

Replacement of feline foamy virus bet by feline immunodeficiency virus vif yields replicative virus with novel vaccine candidate potential.

BACKGROUND: Hosts are able to restrict viral replication to contain virus spread before adaptive immunity is fully initiated. Many viruses have acquired genes directly counteracting intrinsic restriction mechanisms. This phenomenon has led to a co-evolutionary signature for both the virus and host which often provides a barrier against interspecies transmission events. Through different mechanisms of action, but with similar consequences, spumaviral feline foamy virus (FFV) Bet and lentiviral feline immunodeficiency virus (FIV) Vif counteract feline APOBEC3 (feA3) restriction factors that lead

Surface Proteins and Pneumolysin of Encapsulated and Nonencapsulated Streptococcus pneumoniae Mediate Virulence in a Chinchilla Model of Otitis Media.

Streptococcus pneumoniae infections result in a range of human diseases and are responsible for almost one million deaths annually. Pneumococcal disease is mediated in part through surface structures and an anti-phagocytic capsule. Recent studies have shown that nonencapsulated S. pneumoniae (NESp) make up a significant portion of the pneumococcal population and are able to cause disease. NESp lack some common surface proteins expressed by encapsulated pneumococci, but express surface proteins unique to NESp. A chinchilla model of otitis media (OM) was used to determine the effect various pneu

Peak bone strength is influenced by calcium intake in growing rats.

In this study we investigated the effect of supplementing the diet of the growing male rat with different levels of calcium (from low to higher than recommended intakes at constant Ca/P ratio), on multiple factors (bone mass, strength, size, geometry, material properties, turnover) influencing bone strength during the bone accrual period. Rats, age 28days were supplemented for 4weeks with high Ca (1.2%), adequate Ca (0.5%) or low Ca level (0.2%). Bone metabolism and structural parameters were measured. No changes in body weight or food intake were observed among the groups. As anticipated, com

[Angina pectoris in extracoronary diseases].

Chest pain can arise from cardiovascular or noncardiovascular causes. Among the latter are the skin, the chest wall, intrathoracic structures, or subdiaphragmatic organs. The problem to attribute the chest discomfort to either the heart or extracardiac organs arises because the heart, pleura, aorta, and esophagus are all supplied by sensory fibers from the same spinal segments. In contrast to the diseases mentioned above, angina pectoris in sensu strictu is defined as chest pain or discomfort of cardiac origin that arises because of temporary imbalance between myocardial oxygen supply and dema
