Spinal Cord CELF4 in Chronic Constriction Injury-Induced Neuropathic Pain: Association with TRPV1 and COX-2 Expression.

Despite significant advancements, the mechanisms underlying neuropathic pain remain poorly understood, making it challenging for effective pain management. This study investigated the role of CUGBP Elav-Like Family Member 4 (CELF4), an RNA-binding protein, in modulating pain pathways in neuropathic pain conditions. The results showed a reduction in both mRNA and protein levels of CELF4 in the spinal cord of a chronic constriction injury (CCI) mouse model, suggesting its potential role in response to nerve injury. To further explore the role of CELF4 in modulating pain pathways in vivo, an aden

A continuous adhesion-enhanced osteogenic pathway in artificial scaffold drives cellular infiltration and condensed mineralization for rapid bone regeneration.

Enhancing the cellular infiltration and mineralization capacity of bone scaffolds can effectively address the challenges of bone nonunion and the prolonged osteogenic repair cycle, particularly in the treatment of critical-sized bone defects. Conventional bone scaffolds, whether composed of inorganic materials or fabricated via 3D-printed titanium alloy, frequently hinder seamless cellular integration due to inherent structural discontinuities, such as granular interfaces or layer-by-layer striations. Here, we address this limitation by employing graphene, not merely as a filler, but as a cont

Natural Products in Epilepsy Treatment: From Traditional Medicine Towards Computational Drug Discovery.

Epilepsy affects approximately 50 million people worldwide, with nearly one-third of patients experiencing drug-resistant seizures despite available antiepileptic drugs (AEDs). Natural products remain an important source of bioactive scaffolds for drug discovery, offering diverse chemical structures capable of modulating key pathological pathways in epilepsy. This review examines major classes of natural compounds, including alkaloids, flavonoids, terpenoids, and phenolic compounds, and their activity against validated targets such as GABAergic and glutamatergic systems, voltage-gated ion chan

Molecular Friction Trumps Viscosity in Angstrofluidic Transport.

In the realm of ultra-confined nanofluidics, where channels barely accommodate a single molecule, classical continuum theories would inevitably falter. Fundamental properties like viscosity, wettability, and velocity profiles lose their conventional meaning, leaving a critical gap in our ability to predict and engineer flow at the molecular scale, despite these features continuing to be extrapolated from the underlying molecular behavior. Here, we tackle this conceptual anomaly by probing single-file transport of liquid argon through graphene nanopores and carbon nanotubes via molecular dynami

Carbonyl-Enriched Nonionic Covalent Organic Framework Nanochannels for High-Salinity Ionic Thermoelectrics.

High-salinity electrolytes coupled with low-grade heat offer an attractive opportunity for ionic thermoelectric conversion, yet conventional fixed-charge membranes rapidly lose ion permselectivity in concentrated media because Donnan exclusion is strongly screened. Here, we report an isoreticular series of nonionic covalent organic framework membranes with comparable one-dimensional nanochannels but systematically varied pore-wall oxygen chemistry. Hydroxyl-substitution-regulated hydrazone-to-β-ketoenamine tautomerism generates carbonyl-enriched BthTb-3OH nanochannels, which preferentially par

Comparative genomics of nicotinic acetylcholine receptors reveals early divergence of the α6 subunit in cryptic Anopheles species.

BACKGROUND: Nicotinic acetylcholine receptors (nAChRs) are pentameric ligand-gated ion channels that mediate fast cholinergic transmission in the insect central nervous system and serve as targets for several widely used classes of insecticides. nAChR-targeting formulations are being deployed in malaria vector control programs to mitigate widespread resistance to pyrethroids. However, the extent of genetic variation at insecticide target sites that may facilitate evolutionary responses to chemical exposure among vector species remains poorly understood. RESULTS: Here, we used whole-genome and

Genetic loci responsible for atrioventricular block in children.

Atrioventricular block (AVB) represents a prevalent form of bradyarrhythmia in the pediatric population, broadly classified into congenital and acquired subtypes. Historically, the underlying mechanisms of AVB were predominantly attributed to structural cardiac anomalies, maternal autoantibody-mediated inflammatory processes, and pharmacological agents; however, the contribution of genetic factors remained largely underexplored. With the advent and progressive refinement of genetic sequencing technologies, a growing repertoire of arrhythmia-associated genes has been systematically identified.

The 5'-Isobutyryl Ester Prodrug of 4'-Thiouridine Inhibits SARS-CoV‑2 Replication in Culture and in a Syrian Hamster Infection Model.

Although the SARS-CoV-2 pandemic is now largely controlled through widespread vaccination and available therapeutics, emerging variants could undermine their overall effectiveness. 4'-Thiouridine (TU) and its 5'-isobutyryl prodrug (iBu TU) were identified as potent inhibitors of SARS-CoV-2 replication across various cell models, including advanced primary human respiratory models. Both compounds exhibited no significant toxicity across a range of cell culture systems, with no signs of mitochondrial damage or elevated lactic acid production. Furthermore, TU and iBu TU tested negative for mutage

Xenon anesthesia: shedding light on brain dynamics and clinical niches.

PURPOSE OF REVIEW: Xenon is reemerging as a clinically attractive inhalational anesthetic because it combines rapid kinetics and cardiovascular stability with mechanism features that differ from GABAergic agents. Simultaneously, new membrane-biophysics and systems-neuroscience data are reshaping how models in anesthetic state transitions worked, which expands the future of how anesthetic agents are used. RECENT FINDINGS: Contemporary work strengthens the view that xenon acts primarily via noncompetitive NMDA receptor inhibition, with additional effects on excitability-related ion channels. Wh

Comparative analysis of microRNA expression in serum-derived extracellular vesicles from sudden infant death syndrome cases.

Sudden infant death syndrome (SIDS) remains one of the most common and poorly understood diagnoses of death in infants. In this study, we searched for novel biomarkers to aid in elucidating the pathogenesis of SIDS through a bioinformatics analysis of serum-derived extracellular vesicle (EV) miRNAs using next-generation sequencing. Comparative analyses between infants who died of SIDS and those who died from known causes showed that 15 and 38 miRNAs were significantly up- or down-regulated more than twofold in SIDS, respectively. Myocardial-specific miRNAs, such as miR-1, miR-208, and miR-499,

Mechanochemically Coupled Multidimensional Modulation of Calcium Overload.

Disruption of calcium ion (Ca2+) homeostasis has emerged as a promising strategy for tumor therapy. However, the intricate regulation of Ca2+ signaling and the limitations of single-dimensional modulation often hinder therapeutic efficacy. Here, we developed a Janus nanomotor platform that orchestrates mechanochemically coupled multidimensional modulation of Ca2+ overload for enhanced tumor therapy. Utilizing a liquid-nano-liquid interface-mediated anisotropic encapsulation strategy, amorphous calcium carbonate (ACC) nanoparticles were asymmetrically coated with mesoporous polydopamine (mPDA)

Controlling Enantioselectivity of Halohydrin Dehalogenase for Asymmetric Synthesis of Chiral Epichlorohydrin.

Halohydrin dehalogenases (HHDHs) are promising biocatalysts for the asymmetric synthesis of chiral epoxides. In this study, we demonstrate bidirectional enantioselectivity switch of HHDHs for the asymmetric synthesis of chiral epichlorohydrin (ECH) from 1,3-dichloro-2-propanol (1,3-DCP). HHDH from Propionicicella superfundia (PsHHDH) was identified for its high activity toward 1,3-DCP with a slight preference for (R)-ECH formation (13.6% ee). Guided by the catalytic mechanism of HHDHs and substrate binding features, we designed a rational engineering strategy targeting two distinct cavities wi

[Advances in research on the molecular mechanisms and gene therapy of hereditary hearing impairment].

Hereditary hearing impairment represents a significant etiology of language and social dysfunction in both children and adults, primarily caused by genetic factors. To date, over 150 genes have been identified in association with this disorder. The pathogenic mechanisms involve multiple molecular levels, including abnormalities in hair cell cytoskeleton and stereociliary structure, dysfunction of intercellular gap junctions (e.g., GJB2, GJB6), dysregulation of ion channels and transporters (e.g., SLC26A4, KCNQ4), alterations in extracellular matrix composition, and disruption of intracellular

Enhanced calcium activity and transcriptomic alterations in iPSC-derived neurons from BAFME patients with repeat expansions.

Benign adult familial myoclonus epilepsy (BAFME) is caused by intronic TTTCA and TTTTA repeat expansions in SAMD12 and other genes; the neuronal basis of cortical hyperexcitability, however, remains unclear. We generated induced pluripotent stem cell (iPSC)-derived glutamatergic and GABAergic neurons from three BAFME1 patients and examined functional and transcriptomic phenotypes. Patient-derived neurons retained the pathogenic repeat expansions and showed a tendency toward upstream intronic RNA accumulation. Calcium imaging revealed increased spontaneous Ca2 + transient frequency in both neur

Mechanisms contributing to the age at onset of temporal lobe epilepsy due to hippocampal sclerosis.

Temporal lobe epilepsy (TLE) secondary to hippocampal sclerosis (HS) is a common cause of drug-resistant epilepsy. HS is characterized by neuronal loss in selected hippocampal subfields and gliosis. Age at epilepsy onset (AEO) critically influences disease severity and treatment response. We compared early AEO (<10 years) and late AEO (>11 years) in hippocampi from patients undergoing surgery for HS (n = 30). HS hippocampi showed altered mitochondrial enzyme activities and membrane potential (vs controls). Early AEO showed reduced membrane potential and ATP and increased proton leak, whereas o

Pulsed focused ultrasound for seizure suppression: Mechanisms, models, and clinical translation.

Focused ultrasound (FUS) has emerged as a versatile, non-invasive therapeutic platform for drug-resistant epilepsy, offering both thermal effects for the permanent ablation of epileptogenic foci and non-thermal mechanical mechanisms for the reversible neuromodulation of neural activity, all without the need for surgical implantation. Pulsed focused ultrasound (pFUS) achieves seizure control through seizure suppression via non-thermal mechanical mechanisms. At the cellular level, pFUS modulates mechanosensitive ion channels, and may interrupt astrocyte-mediated excitatory signaling pathways. At

Motor neuron axonal excitability changes in the clinical course of amyotrophic lateral sclerosis.

Amyotrophic lateral sclerosis (ALS) is a fatal disease characterized by slowly progressive degeneration of upper motor neurons (UMNs) and lower motor neurons (LMNs). Although the pathogenesis of sporadic form of ALS has not been fully elucidated, the initial damage mostly leads to hyperexcitability of the central and peripheral motor neuron. The results of our study aimed to confirm the changes in the excitability of the peripheral motor axon and contribute to the emergence of these measurements as potential biomarkers for disease progression. A total of 56 ALS patients [24 women (43%), median

Comprehensive characterization of multi-omics landscapes between gut microbial metabolites and the druggable genome in sepsis.

BACKGROUND: Sepsis is a life-threatening condition with limited therapeutic options. Emerging evidence implicates gut microbial metabolites in modulating host immunity, but the specific interactions between these metabolites and host druggable targets remain poorly understood. METHODS: We utilized a systems biology framework integrating genetic analyses, multi-omics profiling, and structure-based virtual screening to systematically map the interaction landscape between human gut microbial metabolites and druggable G-protein-coupled receptors (GPCRs), ion channels (ICs), and kinases (termed th

Quickly Generated N, Co Codoped Porous Graphene from Waste Biomass at Low Temperature for an Effective Oxygen Reduction Reaction.

Porous carbon nanomaterials derived from waste biomass have attracted a great deal of attention as high-performance nonprecious metal electrocatalysts for the oxygen reduction reaction (ORR). However, the production of porous carbon nanocatalysts usually requires a high temperature and a long time. Here, we propose a fast low-temperature strategy to obtain nonmetal and transition metal codoped porous graphene with a tunable hole structure from waste seed husk via a triple KCl/LiCl/ZnCl2 molten salt system. The codoped porous graphene exhibited a large specific surface area, sufficient micropor

Trace Cu Doping Enabled High Rate and Long Cycle Life Sodium Iron Phosphate Cathode for Sodium-Ion Batteries.

Na4Fe3(PO4)2(P2O7) (NFPP) is currently receiving a lot of attention, as it combines the advantages of NaFePO4 and Na2FeP2O7 in terms of cost, energy density, and cycle stability. However, the issues of intrinsic poor electronic conductivity and difficult high-purity preparation may impede its practical application. Herein, the pivotal role of Cu doping in strengthening the polyanion structure and improving its electrochemical properties is comprehensively investigated. It is found that trace Cu doping not only expands the lattice volume of NFPP but also suppresses the formation of the inactive
