Macrophage-mediated antibiotic evasion and competitive dominance of mcr-3-carrying Escherichia coli.

Mobile colistin resistance (mcr) genes compromise the efficacy of last-resort polymyxin antibiotics. Notably, the global prevalence of mcr-3 has continued to increase despite reductions in colistin use, suggesting that selective forces beyond direct antibiotic pressure contribute to its persistence. Here, mcr-3-positive Escherichia coli is shown to confer a survival advantage by reprogramming macrophage immunity. MCR-3-mediated lipid A modification blunted TLR4-NF-κB signaling, suppressed macrophage reactive oxygen species generation, and delayed phagosome-lysosome fusion, allowing mcr-3-posit

Microglial Bmal1 Contributes to Diurnal Physiology and Retinal Homeostasis.

Circadian rhythms govern various physiological processes, including innate and adaptive immune responses. Microglia, the sentinels of the central nervous system (CNS), mediate synaptic remodeling and local immune responses that contribute to tissue homeostasis. Recent studies have uncovered that microglial surveillance behavior and cytokine production exhibit rhythmicity. Furthermore, disruption of clock gene expression in microglia impairs phagocytic capacity, metabolism, and inflammatory responses, suggesting that their dynamic functions are regulated in part by circadian rhythms. Given the

Prospects of the therapeutic approaches of ketogenic diet on the clinical outcomes of cancer: A scoping review.

The scientific community has become more enthusiastic about ketogenic diet (KD) to improve the outcomes in medical conditions, including cancer by exploiting the reprogramed metabolism of cancer cells, making the diet a promising candidate as an adjuvant cancer therapy. From this perspective, the aim of this study was to provide a broad overview covering the therapeutic effects, evaluating the clinical evidence of clinical studies underlying the implementation of the KD in the context of cancer treatment and prognosis. A scoping literature search between the years 1990 and 2023 was carried out

[Transcriptomic Profile of the Trastuzumab-Resistant Breast Cancer Cell Line BT-474].

The development of resistance to trastuzumab in HER2-positive breast cancer is a serious clinical problem that limits the effectiveness of targeted therapy. In a significant proportion of patients, the mechanisms in the development of resistance remain poorly understood. The BT-474 cell line was selected as an optimal model for study because it represents a HER2-positive luminal B subtype breast cancer cell line. To identify the molecular mechanisms of resistance, a comprehensive transcriptomic analysis based on RNA-seq data comparison of three independent datasets including both sensitive and

Injectable cationic dynamic hydrogel with supramolecular drug loading reprograms neutrophil fate to alleviate diabetic periodontitis.

The pervasive infiltration of tissue-destructive yet microbicidal-impaired neutrophil extracellular traps (NETs) constitutes a primary mechanistic driver of diabetic periodontitis refractoriness. Apoptosis facilitates nonphlogistic turnover of neutrophils, but excessive activation leads to the release of an excessive amount of dysfunctional NETs, causing tissue damage in diabetic periodontitis. To co-target pathological NETosis and dysbiotic infection, we engineered an injectable cationic dynamic hydrogel (ICDH) with supramolecular drug loading via grafting phenylboronic acid onto cationic pol

Canagliflozin Reprograms the Aging Hippocampus in Genetically Diverse UM-HET3 Mice and Attenuates Alzheimer's-Like Pathology.

Aging is the strongest risk factor for cognitive decline and Alzheimer's disease (AD), yet the mechanisms underlying brain aging and their modulation by pharmacological interventions remain poorly defined. The hippocampus, essential for learning and memory, is particularly vulnerable to metabolic stress and inflammation. Canagliflozin (Cana), an FDA-approved sodium-glucose co-transporter 2 inhibitor (SGLT2i) for type 2 diabetes, extends lifespan in male but not female mice, but its impact on brain aging is unknown. Here, we used a multi-omics strategy integrating transcriptomics, proteomics, a

Coronavirus NSP14 Drives Internal m 7 G Modification to Rewire Host Splicing and Promote Viral Replication.

SARS-CoV-2, the causative agent of COVID-19, manipulates host gene expression through multiple mechanisms, including disruption of RNA processing. Here, we identify a novel function of the viral nonstructural protein 14 (NSP14) in inducing N 7-methylguanosine (m 7 G) modification in the internal sequences of host mRNA. We demonstrate that NSP14 catalyzes the conversion of guanosine triphosphate (GTP) to m 7 GTP, which is subsequently incorporated into mRNA by RNA polymerase II, resulting in widespread internal m 7 G modification. This activity is dependent on NSP14's N 7-methyltransferase ( N

Viral outbreak dynamics and evolution in wildlife at the interface with humans.

BACKGROUND: Neonates are susceptible to a wide range of microbial infection and at a high risk to develop severe sepsis and septic shock. Emerged evidence has shown that induction of trained immunity triggers a much stronger inflammatory response in adult monocytes/macrophages, thereby conferring protection against microbial infection. METHODS: This study was carried out to examine whether trained immunity is inducible and exerts its protection against microbial sepsis in neonates. RESULTS: Induction of trained immunity by Bacillus Calmette-Guerin (BCG) plus bacterial lipoprotein (BLP) prote

Unravelling ecophysiological and molecular adjustments in the photosynthesis-respiration balance during Fusarium graminearum infection in wheat spikes.

Wheat responses to F. graminearum result in a deep and sharp reprogramming of a wide range of biological processes, including energy-associated functions and related metabolisms. Although these impacts have been thoroughly described at the molecular scale through proteomics and transcriptomics studies, phenotypic studies are still needed to fill the gap between the observed molecular events and the actual impacts of the disease on the ecophysiological processes. Taking advantage of the gas exchange method, the effects of two F. graminearum strains of contrasting aggressiveness on spike's photo

Innate immune training in the neonatal response to sepsis.

Neonates, especially those born prematurely, are highly vulnerable to infection-induced mortality. Numerous observational and immunological studies in newborns have shown that live attenuated vaccines have beneficial, non-specific effects (NSEs) against secondary infections to unrelated pathogens. These beneficial effects have been attributed to trained immunity, and emergency granulopoiesis plays an essential role. However, trained immunity has been shown to affect multiple myeloid subsets and how trained immunity influences the host protective response is still undefined. Here we show that B

Glutamine modulates neutrophil recruitment and effector functions during sterile inflammation.

During sterile inflammation, tissue damage induces excessive activation and infiltration of neutrophils into tissues, where they critically contribute to organ dysfunction. Tight regulation of neutrophil migration and their effector functions is crucial to prevent overshooting immune responses. Neutrophils utilize more glutamine, the most abundant free α-amino acid in the human blood, than other leukocytes. However, under inflammatory conditions, the body's requirements exceed its ability to produce sufficient amounts of glutamine. This study investigates the impact of glutamine on neutrophil

An Immunomodulating Regenerating Hydrogel That Rescues the Oxidative Microenvironment and Reverses Cell Senescence for Osteoporotic Bone Defects.

Osteoporosis, which is characterized by reduced bone mass and structurally compromised bone tissue, along with aberrant levels of reactive oxygen species (ROS) and inflammation, has been a pressing clinical challenge. A large accumulation of ROS and pro-inflammatory factors can result in mitochondrial dysfunction and progressive cellular senescence, impeding efficacious regeneration of bone defects. Herein, in this work, a ROS-responsive hydrogel system containing HA-PBA coated Ce-ZOL nanocomposites (GHCZ) for excessive ROS scavenging and reversal of cellular senescence to accelerate bone rege

rBCG-LTAK63 enhances protection against tuberculosis by inducing autophagy and circadian gene regulation.

Tuberculosis (TB) remains a global public health challenge, with the current BCG vaccine providing limited efficacy in adults, and available treatments being lengthy and debilitating. To overcome these challenges, we have previously developed a recombinant BCG strain expressing the detoxified E. coli Heat-Labile Toxin (LTAK63), providing increased protection in mouse models and reduced lung pathology. Here, using systems biology and RNA sequencing of lung tissues in a murine model, we uncover the molecular mechanisms underlying rBCG-LTAK63's increased protection. Immunization triggered early a

Therapeutic approaches to microglial dysfunction in Alzheimer's disease: Enhancing phagocytosis and metabolic regulation.

Microglia are essential in neurogenesis, synaptic pruning, and homeostasis. Nevertheless, aging, and cellular senescence may modify their role, causing them to shift from being shields to being players of neurodegeneration. In the aging brain, the population of microglia increases, followed by enhanced activity of genes related to neuroinflammation. This change increases their ability to cause inflammation, resulting in a long-lasting state of inflammation in the brain that harms the condition of neurons. In Alzheimer's Disease (AD), microglia are located inside amyloid plaques and exhibit an

Iron Deficiency in Tomatoes Reversed by Pseudomonas Strains: A Synergistic Role of Siderophores and Plant Gene Activation.

An alkaline pH in soils reduces Fe availability, limiting Fe uptake, compromising plant growth, and showing chlorosis due to a decrease in chlorophyll content. To achieve proper Fe homeostasis, dicotyledonous plants activate a battery of strategies involving not only Fe absorption mechanisms, but also releasing phyto-siderophores and recruiting siderophore-producing bacterial strains. A screening for siderophore-producing bacterial isolates from the rhizosphere of Pinus pinea was carried out, resulting in two Pseudomonas strains, Z8.8 and Z10.4, with an outstanding in vitro potential to solubi

Sperm miR-142-3p Reprogramming Mediates Paternal Pre-Pregnancy Caffeine Exposure-Induced Non-Alcoholic Steatohepatitis in Male Offspring Rats.

Numerous studies have suggested a strong association between paternal adverse environmental exposure and increased disease susceptibility in offspring. However, the impact of paternal pre-pregnant caffeine exposure (PPCE) on offspring health remains unexplored. This study elucidates the sperm reprogramming mechanism and potential intervention targets for PPCE-induced non-alcoholic steatohepatitis (NASH) in offspring. Here, male rats are administrated caffeine (15-60 mg kg-1/d) by gavage for 8 weeks and then mated with females to produce offspring. This study finds that NASH with transgeneratio

The mechanism of PFK-1 in the occurrence and development of bladder cancer by regulating ZEB1 lactylation.

BACKGROUND: Bladder cancer (BC) is one of the most common malignancies of the genitourinary system. Phosphofructokinase 1 (PFK-1) is one of member of PFK, which plays an important role in reprogramming cancer metabolism, such as lactylation modification. Zinc finger E-box-binding homeobox 1 (ZEB1) has been demonstrated to be a oncogene in many cancers. Therefore, this study was performed to explore the effects of PFK-1 on the lactylation of ZEB1 in BC development. METHODS: Cell viability was measured using the CCK-8 kit. The glucose assay kit and lactate assay kit were used to detect glucose

Proteomic characterization of Mycobacterium tuberculosis subjected to carbon starvation.

Mycobacterium tuberculosis ( Mtb ) is the causative agent of tuberculosis (TB), the leading cause of infectious-disease related deaths worldwide. TB infections present as a spectrum from active to latent disease. In the human host, Mtb faces hostile environments, such as nutrient deprivation, hypoxia, and low pH. Under these conditions, Mtb can enter a dormant, but viable, state characterized by a lack of cell replication and increased resistance to antibiotics. These dormant Mtb pose a major challenge to curing infections and eradicating TB globally. In the current study, we subjected Mtb to
