Cardiovascular Calcification in Chronic Kidney Disease-Therapeutic Opportunities.

Patients with chronic kidney disease (CKD) are highly susceptible to cardiovascular (CV) complications, thus suffering from clinical manifestations such as heart failure and stroke. CV calcification greatly contributes to the increased CV risk in CKD patients. However, no clinically viable therapies towards treatment and prevention of CV calcification or early biomarkers have been approved to date, which is largely attributed to the asymptomatic progression of calcification and the dearth of high-resolution imaging techniques to detect early calcification prior to the 'point of no return'. Non-coding RNAs in aortic dissection: From biomarkers to therapeutic targets.

Aortic dissection (AD) is the rupture of the aortic intima, causing the blood in the cavity to enter the middle of the arterial wall. Without urgent and proper treatment, the mortality rate increases to 50% within 48 hours. Most patients present with acute onset of symptoms, including sudden severe pain and complex and variable clinical manifestations, which can be easily misdiagnosed. Despite this, the molecular mechanisms underlying AD are still unknown. Recently, non-coding RNAs have emerged as novel regulators of gene expression. Previous studies have proven that ncRNAs can regulate severa

Impact of Nutrition on Pulmonary Arterial Hypertension.

Pulmonary arterial hypertension (PAH) is characterized by sustained vasoconstriction, vascular remodeling, inflammation, and in situ thrombosis. Although there have been important advances in the knowledge of the pathophysiology of PAH, it remains a debilitating, limiting, and rapidly progressive disease. Vitamin D and iron deficiency are worldwide health problems of pandemic proportions. Notably, these nutritional alterations are largely more prevalent in PAH patients than in the general population and there are several pieces of evidence suggesting that they may trigger or aggravate disease

The Novel Development of an Experimental Model of Dihydropyridine Calcium Channel Blocker Poisoning using Intravenous Amlodipine.

BACKGROUND: Cardiovascular drug poisoning remains a leading cause of fatality. Within this class, calcium channel blockers (CCBs) account for the majority of deaths. CCBs are typically categorized as dihydropyridines (i.e. amlodipine or nifedipine) versus the non-dihydropyridine (i.e. verapamil and diltiazem) which are the most potent and once considered the CCB type responsible for all CCB-related deaths. Most recently, dihydropyridine deaths have increased. While there are established models of nondihydropyridine poisoning there currently are no established experimental models of dihydropyri

Spontaneous ocular and neurologic deficits in transgenic mouse models of multiple sclerosis and noninvasive investigative modalities: a review.

Multiple sclerosis (MS) is an autoimmune, inflammatory, neurodegenerative, demyelinating disease of the central nervous system, predominantly involving myelinated neurons of the brain, spinal cord, and optic nerve. Optic neuritis is frequently associated with MS and often precedes other neurologic deficits associated with MS. A large number of patients experience visual defects and have abnormalities concomitant with neurologic abnormalities. Transgenic mice manifesting spontaneous neurologic and ocular disease are unique models that have revolutionized the study of MS. Spontaneous experimenta

Cyclophilin-CD147 interactions: a new target for anti-inflammatory therapeutics.

CD147 is a widely expressed plasma membrane protein that has been implicated in a variety of physiological and pathological activities. It is best known for its ability to function as extracellular matrix metalloproteinase inducer (hence the other name for this protein, EMMPRIN), but has also been shown to regulate lymphocyte responsiveness, monocarboxylate transporter expression and spermatogenesis. These functions reflect multiple interacting partners of CD147. Among these CD147-interacting proteins cyclophilins represent a particularly interesting class, both in terms of structural consider

Soluble epoxide hydrolase: a new target for cardioprotection.

Arachidonic acid is metabolized to a number of bioactive eicosanoid molecules by several enzymes, including enzymes of the COX, lipoxygenase and cytochrome P450 (CYP) monooxygenase pathways. Inhibition of the CYP omega-hydroxylase pathway, stimulation of the CYP-epoxygenase pathway and administration of exogenous epoxyeicosatrienoic acids resulted in cardioprotection in animal models of ischemia; contractile function was improved in mouse hearts subjected to global ischemia/reperfusion, and infarct size was reduced in canine and rat hearts. Cardioprotective effects were also achieved when meta

Effects of HMG-CoA reductase inhibitors (statins) on progression of kidney disease.

Chronic kidney disease, especially in the setting of proteinuria, is characterized by hyperlipidemia. In animal models, hyperlipidemia causes glomerular foam cells and glomerulosclerosis. Treatment with 3-hydroxy-3-methylglutaryl coenzyme A reductase inhibitors (statins) ameliorates kidney disease in these models. The data of the role of hyperlipidemia in progression of human kidney disease are less clear. Data from small studies in glomerular disease suggest that statins decrease proteinuria. Data mainly from cardiovascular studies suggest that statins decrease the loss of glomerular filtrati

Tea and health.

The possible beneficial effects of tea consumption in the prevention of cancer and cardiovascular diseases have been demonstrated in animal models and suggested by studies in vitro. Similar beneficial effects, however, have not been convincingly demonstrated in humans: beneficial effects have been demonstrated in some studies but not in others. If such beneficial effects do exist in humans, they are likely to be mild, depending on many other lifestyle-related factors, and could be masked by confounding factors in certain populations. Another concern is that the amounts of tea consumed by human

Divalent cations in hypertension with implications to heart disease: calcium, cadmium interactions.

Heart disease still pervades all socioeconomic classes within the United States. Understanding the effects of environmentally-related pathogenesis (e.g., heavy metal accumulation) may aid in developing novel treatments for the prevention of heart dysfunction. The aim of this report was to use experimental investigation in an attempt to expand upon the multivariate importance of divalent cation interactions in the development of heart disease. Calcium and cadmium levels were measured by flame atomic absorption spectrophotometry in various tissues derived from two types of hypertensive rabbit mo

Nonlinear association between the erythrocyte distribution width-to-albumin ratio and mortality in older adults: A retrospective cohort study using NHANES 1999 to 2018 data.

Older adults face high risks of all-cause and cardiovascular disease (CVD) mortality; however, simple, inexpensive biomarkers for early risk identification are limited. The red cell distribution width-to-albumin ratio (RAR) is a low-cost, routinely measured marker, but its prognostic value in older adults is unclear. We examined whether RAR is independently associated with all-cause and CVD mortality in older adults. We conducted a retrospective cohort study using data from the United States National Health and Nutrition Examination Survey (1999-2018). The study included 16,558 adults aged ≥60

Life-course socioeconomic position and cardiovascular diseases among older adults in low- and middle-income countries: A systematic review.

BACKGROUND: In low- and middle-income countries (LMICs), economic instabilities may alter people's socioeconomic position (SEP) throughout their life course affecting their health. We conducted a systematic review to identify which life-course models link SEP over the lifespan and cardiovascular diseases (CVD) among older adults in LMICs. METHODS: Four databases were searched (PUBMED, EMBASE, SocINDEX, PsycInfo), yielding 3097 English articles after removing duplicates. Studies were graded using the Newcastle-Ottawa Quality Assessment Scale and results were reported through narrative synthesi

Machine learning prediction of cardiovascular disease risk progression from sulfur dioxide exposure in longitudinal population studies in China.

Cardiovascular disease (CVD) is a prevalent global health issue and one of the leading causes of death. Aging and air pollution are well-established risk factors for CVD. This study aims to investigate the association between air pollutants (sulfur dioxide, carbon monoxide, PM1, PM2.5, nitrogen dioxide, ozone) and the risk of heart disease. Utilizing data from the China Health and Retirement Longitudinal Study (CHARLS) and the China High Air Pollutants (CHAP) database, we employed multivariable-adjusted logistic regression to analyze the relationship between pollutants and heart disease. Increased mortality rate in patients with Takayasu arteritis is largely driven by early disease activity and damage: analysis of two cohorts.

OBJECTIVE: To evaluate mortality risk and pre-treatment predictors of mortality in Takayasu arteritis (TAK). METHODS: From two cohorts, outcomes at last visit (survival/mortality) were recorded. Standardized mortality ratios (SMR) were computed (indirect standardization to population death rates from India and Turkiye). Cox proportional hazards regression was used to estimate predictors of mortality [hazard ratios (HR) with 95%CI]. The final multivariable-adjusted models included significant predictors of mortality from initial analyses based on demographic features, disease activity, damage

Vascular Regenerative Cell Flux in Diabetes: Mechanistic and Clinical Implications.

UNLABELLED: Type 2 diabetes mellitus (T2DM) disrupts the quantity, phenotype, and function of circulating vascular regenerative (VR) progenitor cells that are critical to vessel repair. These impairments include reduced progenitor clonogenicity, mobilization and homing to areas of ischemia, and imbalances in secretory function, collectively contributing to poor vascular healing and complications such as peripheral artery disease, critical limb ischemia, and cardiovascular disease. Despite growing recognition of VR cell exhaustion, regenerative strategies remain hindered by imprecise cell pheno
