Dense cataract and microphthalmia (dcm) in BALB/c mice is caused by mutations in the GJA8 locus.

A spontaneous mutation in BALB/c mice that causes congenital dense cataract and microphthalmia (dcm) was reported previously. This abnormality was found to be inheritable and the mode of inheritance indicated that this phenotype is due to mutation of an autosomal recessive gene. We performed genetic screen to identify the underlying mutations through linkage analysis with the dcm progenies of F(1) intercross. We identified the region of mutation on chromosome 3 and further mapping and sequence analysis identified the mutation in the GJA8 gene that encodes for connexin 50. The mutation represen

[Molecular mechanism and mental function of postnatal neurogenesis].

Postnatal neurogenesis has been observed in two brain regions: the subventricular zone (SVZ) of the lateral ventricle and the subgranular zone (SGZ) of the dentate gyrus in the hippocampus, among vertebrates including human. Accumulating evidence has indicated the molecular mechanisms commonly underlying embryonic and adult neurogenesis. Genetic factors essential for neural development, i.e., Pax6, Fabp7, Sox2, Wnt3, Notch1, etc., are also expressed in adult neurogenic regions. Adult neurogenesis, however, is distinct from embryonic neurogenesis in that the former is activity dependent; enviro

Pharmacogenetics of Crohn's disease.

The considerable interindividual differences in efficacy and side effects of commonly used medications in Crohn's disease are partly owing to genetic polymorphisms. Many genetic variants have been studied in genes possibly involved in the metabolism or mechanism of action of therapeutic agents such as glucocorticosteroids, azathioprine/6-mercaptopurine, methotrexate, calcineurin inhibitors or anti-TNF agents. However, the only test translated into clinical practice is thiopurine S-methyltransferase (TPMT) genotyping for hematological toxicity of thiopurine treatment. To date, there are no othe

Autoimmunity to type VII collagen: epidermolysis bullosa acquisita.

Epidermolysis bullosa acquisita (EBA) is an acquired, mechanobullous disease characterized by autoimmunity to type VII collagen. Type VII collagen makes anchoring fibrils, structures that connect the epidermis and its underlying basement membrane zone to the papillary dermis. EBA patients exhibit skin fragility, blisters, scars and milia formation reminiscent of genetic dystrophic epidermolysis bullosa (DEB). DEB patients have diminutive or absent anchoring fibrils due to a genetic defect in the gene encoding type VII collagen. EBA patients have a decrease in normally functioning anchoring fib

Erythrocyte variants and the nature of their malaria protective effect.

The malaria threat to global health is exacerbated by widespread drug resistance in the Plasmodium parasite and its insect vector, and the lack of an efficacious vaccine. Infection with Plasmodium parasites can cause a wide spectrum of pathologies, from a transient mild form of anaemia to a severe and rapidly fatal cerebral disease. Epidemiological studies in humans and experiments in animal models have shown that genetic factors play a key role in the onset, progression, type of disease developed and ultimate outcome of malaria. The protective effect of polymorphic variants in erythrocyte-spe

Pathological gambling: a comprehensive review of biobehavioral findings.

In this review, findings of biobehavioral research into pathological gambling (PG) are discussed, focusing on neuropsychological, psychophysiological, neuroimaging, neurochemical and genetic studies. Neuropsychological studies indicate deficiencies in certain executive functions. Psychophysiological studies indicate that arousal in PG is of importance when reward is present. Neuroimaging studies point to abnormalities in brain functioning. Recent research into the neurochemistry of PG indicates that abnormalities exist in different neurotransmitter systems. Finally, genetic studies indicate th

Toward an integrative understanding of social phobia.

Our objective was to examine the neurobiology of social phobia from the perspectives of basic sciences, genetics, immunology, neuroendocrinology, neurotransmission and neuroimaging and to provide an integrated understanding of social phobia in the framework of a hypothetical neural circuit. Family and twin studies provide evidence that social phobia is heritable with significant genetic influence, and molecular genetics offers possibilities in understanding the nature of the trait that is transmitted. The biologic distinctiveness of social phobia from anxiety disorders and physiological valida

[Search for new tuberculosis vaccines].

Studies in animal models and analysis of human genetic defects leading to severe mycobacteriosis, establish the major role of cellular immune responses in the control of tuberculosis. The BCG vaccine protects infants against severe forms of tuberculosis (meningitis and miliary tuberculosis). However its protective efficacy against the adult forms of tuberculosis is highly variable, ranging from 0% to 90% in different vaccine trials. Different attempts are being made to isolate new vaccines. Protein antigens exported by Mycobacterium tuberculosis provide some level of protection when delivered

Genetic defects in the thalassemias.

In summary, the beta-thalassemias are models for the study of human genetic disease. Defining the genetic defects in and surrounding the beta-globin gene in the beta(+)- and the beta(0)-thalassemias has resulted in new insights into the relationships between changes in gene structure and abnormalities in gene function. The region 5' to the beta-gene, the coding regions within the gene, and the IVS have all been found to contain single nucleotide defects which diminish or abolish beta-globin mRNA production and the production of beta-globin. The ability to isolate beta-globin genes by cloning,

MetaPredictomics: A Comprehensive Approach to Predict Postsurgical Non-Small Cell Lung Cancer Recurrence Using Clinicopathologic, Radiomics, and Organomics Data.

BACKGROUND: Non-small cell lung cancer (NSCLC) is a complex disease characterized by diverse clinical, genetic, and histopathologic traits, necessitating personalized treatment approaches. While numerous biomarkers have been introduced for NSCLC prognostication, no single source of information can provide a comprehensive understanding of the disease. However, integrating biomarkers from multiple sources may offer a holistic view of the disease, enabling more accurate predictions. In this study, we present MetaPredictomics, a framework that integrates clinicopathologic data with PET/CT radiomic

Locus-aware characterization of wheat LOG-related genes and their transcriptional responses to abiotic stress.

LONELY GUY (LOG) phosphoribohydrolases catalyze the final direct step in cytokinin activation, but their composition and stress-responsive regulation remain difficult to resolve in allohexaploid wheat because homoeology, alternative transcript models and annotation errors inflate apparent gene-family size. We conducted a locus-aware genome-wide analysis integrating comparative phylogenetics, chromosomal distribution, exon-intron and conserved-motif architecture, predicted subcellular localization, promoter cis-elements, Gene Ontology enrichment, protein-association and miRNA-target networks, p

Artificial intelligence techniques for classification of Alzheimer's disease using neuroimaging data: a review.

Alzheimer's disease (AD) is a gradually advancing brain disorder marked by memory impairment. The incurable, progressive nature of the disease leads to the dementia stage. Treatment is effective in the early stage, and it can be controlled but not cured. Artificial Intelligence (AI) learning models are used in medical science to detect and classify diseases into specific categories. Features are extracted from medical images and trained using AI learning models to perform an accurate diagnosis of AD. Recent advancements in machine learning (ML) and deep learning (DL) models have demonstrated s

Machine Learning in Amyotrophic Lateral Sclerosis: Achievements, Pitfalls, and Future Directions.

Background: Amyotrophic Lateral Sclerosis (ALS) is a relentlessly progressive neurodegenerative condition with limited therapeutic options at present. Survival from symptom onset ranges from 3 to 5 years depending on genetic, demographic, and phenotypic factors. Despite tireless research efforts, the core etiology of the disease remains elusive and drug development efforts are confounded by the lack of accurate monitoring markers. Disease heterogeneity, late-stage recruitment into pharmaceutical trials, and inclusion of phenotypically admixed patient cohorts are some of the key barriers to suc

Ischemic stroke: experimental models and reality.

The vast majority of cerebral stroke cases are caused by transient or permanent occlusion of a cerebral blood vessel ("ischemic stroke") eventually leading to brain infarction. The final infarct size and the neurological outcome depend on a multitude of factors such as the duration and severity of ischemia, the existence of collateral systems and an adequate systemic blood pressure, etiology and localization of the infarct, but also on age, sex, comorbidities with the respective multimedication and genetic background. Thus, ischemic stroke is a highly complex and heterogeneous disorder. It is

Contribution of in vivo models of thrombosis to the discovery and development of novel antithrombotic agents.

Cardiovascular and cerebrovascular diseases continue to be the leading cause of death throughout the world. Over the past two decades, great advances have been made in the pharmacological treatment and prevention of thrombotic disorders (e.g., tissue plasminogen activators, platelet GPIIb/IIIa antagonists, ADP receptor antagonists such as clopidogrel, low-molecular weight heparins, and direct thrombin inhibitors). New research is leading to the next generation of antithrombotic compounds such as direct coagulation FVIIa inhibitors, tissue factor pathway inhibitors, gene therapy, and orally act
