Prognostic Impact of Age and Treatment Strategies in Elderly Patients With Oral Squamous Cell Carcinoma: An 11-Year Retrospective Analysis.

BACKGROUND: Oral squamous cell carcinoma (OSCC) is becoming increasingly common in elderly populations, particularly in aging societies such as Japan. METHODS: This study retrospectively evaluated 312 OSCC patients treated at Akita University Hospital between 2011 and 2021, focusing on prognostic differences between elderly (≥ 75 years) and non-elderly (< 75 years) patients. RESULTS: Among the elderly patients, curative intent was associated with significantly better disease-specific survival (DSS) and overall survival (OS) in comparison to palliative intent. The DSS for the elderly group wa

Extensive surgical therapy for oral cancer and the impact on a patient's social life and ability to cope with everyday stress: three-year results.

Extensive tumour resection remains the gold standard in the treatment of oral cancer. However, this can lead to severe functional and psychosocial complications. The study aimed to evaluate the social life and everyday stress in a long-term cohort of patients with oral cancer and who required surgical intervention over a period of three years. Patients undergoing surgery for malignant oral tumours in the Department of Maxillofacial Surgery at the University Hospital of Schleswig-Holstein in Kiel, Germany, participated in the study from August 2016 to July 2021. The questionnaires "Perceived St

Evaluation of the Antitumor and Antiproliferative Potential of Synthetic Peptides Derived from IsCT1, Associated with Cisplatin, in Squamous Cell Carcinoma of the Oral Cavity.

Head and neck squamous cell carcinoma (SCC), particularly in the oral cavity, is among the most prevalent and lethal forms of cancer globally. Current therapeutic strategies, predominantly involving cisplatin, face challenges like chemoresistance and toxicity to normal cells, justifying the exploration of new approaches. This study evaluates the antitumor, antiproliferative, and immunomodulatory potential of a synthetic peptide derived from IsCT1 (Isalo scorpion cytotoxic peptide), named AC-AFPK-IsCT1, in combination with cisplatin in oral squamous cell carcinoma cellular models. Tumor and nor

Oral Lachnoanaerobaculum Levels and Survival in Patients With Head and Neck Cancer.

IMPORTANCE: The oral microbiome plays a critical role in cancer treatment responses, yet its influence on outcomes in patients with head and neck squamous cell carcinoma (HNSCC) undergoing (chemo)radiotherapy remains poorly understood. Identifying specific microbiome signatures associated with treatment effectiveness could provide novel prognostic biomarkers and therapeutic targets. OBJECTIVE: To investigate the association between salivary Lachnoanaerobaculum spp abundance and treatment outcomes in patients with HNSCC undergoing (chemo)radiotherapy and to explore potential mechanisms. DESIG

Associated variables of peri-implant bone resorption for dental implants placed in the fibular reconstructed maxilla and mandible of patients with oral cancer: A retrospective clinical study.

STATEMENT OF PROBLEM: Peri-implant bone resorption is a critical factor in evaluating the long-term success of dental implants. However, information regarding the difference in peri-implant bone resorption when implants are placed in microvascular fibula free (MFF) flaps is limited, and how jaw location may influence long-term peri-implant bone stability under functional occlusal loading over extended follow-up periods is unclear. PURPOSE: The purpose of this retrospective clinical study was to investigate the variables related to peri-implant fibular bone resorption under functional occlusal

Tumor Bed Margins Versus Specimen Margins in Oral Cavity Cancer: Too Close to Call?

INTRODUCTION: The routine assessment of intraoperative margins has long been the standard of care for oral cavity cancers. However, there is a controversy surrounding the best method for sampling surgical margins. The aim of our study is to determine the precision of a new technique for sampling tumor bed margins (TBMs), to evaluate the impact on survival and the rate of free flap reconstructions. METHODS: This retrospective cohort study involved 156 patients with primary cancer of the tongue or floor of the mouth who underwent surgery as initial curative treatment. Patients were separated in

SATB2 expression in oral sarcomatoid (spindle cell) squamous cell carcinoma: clinicopathologic and immunophenotypic characterization of 10 cases.

OBJECTIVE: Sarcomatoid squamous cell carcinoma (sSCC) represents an uncommon histopathologic variant of squamous cell carcinoma (SCC). We examined the clinicopathologic and immunophenotypic characteristics, including SATB2 expression, of 10 cases of oral sSCCs. STUDY DESIGN: Archived sSCC cases diagnosed during the period 2000 to 2023 were retrieved. Lesions lacking proper histomorphological features or adequate immunohistochemical confirmation were excluded. Patient age, sex, and lesion location were recorded. All cases were immunostained against SATB2 (Clone EP281; Cell Marque). RESULTS: T

SPP1+ TAM Regulates the Metastatic Colonization of CXCR4+ Metastasis-Associated Tumor Cells by Remodeling the Lymph Node Microenvironment.

Lymph node metastasis, the initial step in distant metastasis, represents a primary contributor to mortality in patients diagnosed with oral squamous cell carcinoma (OSCC). However, the underlying mechanisms of lymph node metastasis in OSCC remain incompletely understood. Here, the transcriptomes of 56 383 single cells derived from paired tissues of six OSCC patients are analyzed. This study founds that CXCR4+ epithelial cells, identified as highly malignant disseminated tumor cells (DTCs), exhibited a propensity for lymph node metastasis. Importantly, a distinct subset of tumor-associated mac

The spatial distribution of intermediate fibroblasts and myeloid-derived cells dictate lymph node metastasis dynamics in oral cancer.

BACKGROUND: Oral cancer poses a significant health challenge due to limited treatment protocols and therapeutic targets. We aimed to investigate the invasive margins of gingivo-buccal oral squamous cell carcinoma (GB-OSCC) tumors in terms of the localization of genes and cell types within the margins at various distances that could lead to nodal metastasis. METHODS: We collected tumor tissues from 23 resected GB-OSCC samples for gene expression profiling using digital spatial transcriptomics. We monitored differential gene expression at varying distances between the tumor and its microenviron

Malignant transformation rate of erosive oral lichen planus: a retrospective study.

Oral lichen planus (OLP) is a chronic inflammatory oral condition previously known to have a rate of malignant transformation of around 1%. Additionally, erosive lichen planus, a subtype of lichen planus, which is a known risk factor for malignant transformation, has previously been unquantified in a large cohort of patients. In a single unit observation between 2005 and 2021 we retrospectively assessed 1,920 patients who underwent histological analysis for suspected oral lichen planus, and followed the progression of their disease to assess the rate of malignant transformation. In total, 1.39

Correlation between serum heavy metals and the risk of oral squamous cell carcinoma and oral potentially malignant disorders.

Oral squamous cell carcinoma (OSCC) is a serious public health problem in various Asian countries, including Sri Lanka, and a combination of cultural practices, lifestyle factors, and genetic predispositions influences the incidence of these cancers. The examination of the connection between exposure to heavy metals and the probability of developing oral potentially malignant disorders (OPMD) and OSCC has been limited in its scope, and the overall consequences of such exposure remain largely unknown. This study aims to clarify the link between serum levels of heavy metals and the risk of OSCC

Promoter Hypermethylation of LATS1 Gene in Oral Squamous Cell Carcinoma (OSCC) among North Indian Population.

BACKGROUND: LATS1 (Large Tumor Suppressor, isoform 1) is a gene that forms a complex with the cyclin-dependent kinase, CDK1, and regulates cell cycle progression. Genetic modifications lead to a loss in the activity of LATS1 gene. OSCC is the most commonly emerging cancer caused by genetic as well as epigenetic changes. Epigenetics changes vary from one population to another because these are influenced by dietary factors and environmental factors.  Tobacco chewing and smoking has been reported as major risk factors in OSCC. No report was found in the previous literature showing promoter hyper

Dental Calculus as a Potential Biosource for Human Papillomavirus Detection in Oral Squamous Cell Carcinoma.

OBJECTIVE: The infection of human papillomaviruses (HPVs) plays a role in the development of oral squamous cell carcinoma (OSCC). A poor oral hygiene and dental calculus may cause the infection to persist. Therefore, this study aimed to assess whether this dental calculus could serve as a potential biosource in early detection of HPVs in patients with OSCC. METHODS: DNA was isolated from the dental calculus of people diagnosed with OSCC, and MY09/11 primer set was used to detect the presence of HPV. The positive samples were further sequenced and aligned using megablast NCBI BLAST tool to ide
