Noncanonical Taste Transduction of Sugars Elicits Responses in a Dedicated Subset of Gustatory Afferent Neurons.

In taste buds, the GPCR heterodimer TAS1R2 + TAS1R3 is considered the canonical taste receptor for sugars and noncaloric sweeteners. Nevertheless, evidence has accumulated for the presence of an alternative noncanonical transduction pathway that detects sugars, particularly at high concentration. Sodium-glucose transporter 1 (SGLT1) has been proposed as this transducer, selectively transporting glucose into a subset of taste bud cells which then transmit the signal to taste afferent neurons. To test for TAS1R-independent sweet taste detection, we conducted in vivo Ca2+ imaging on geniculate ga

The multilevel exploration test, a novel paradigm to measure exploratory behavior in depression animal models and the involvement of the PL-ZI circuit.

Diminished drive is one of the core symptoms of major depressive disorder (MDD) diagnosis, yet its underlying neural mechanisms remain elusive, primarily due to a lack of appropriate animal models. We developed a novel Multilevel Exploration Test (MET) apparatus to evaluate exploratory behavior, which is captured as a dynamic, stage-dependent process involving "search", "attend/investigate", and "approach" phases. We employed fiber photometry to measure real-time dopamine dynamics in the nucleus accumbens. We further combined cFos staining and neural circuit tracing to identify relevant brain

Anterior midcingulate cortex activity during pain catastrophizing predicts and sequentially tracks reduction in pain interference after cognitive behavioral therapy in fibromyalgia.

Fibromyalgia (FM) is a chronic pain disorder characterized by widespread pain, fatigue, and functional disability. Cognitive behavioral therapy (CBT) has been shown to alleviate FM symptoms, but the neural mechanisms underlying CBT's effectiveness in reducing functional impact remain unclear. In this study, we investigated the temporal dynamics of clinical and neural responses to CBT. We conducted a longitudinal randomized controlled neuroimaging trial with 98 patients with FM assigned to an 8-week CBT or education intervention. Functional MRI (fMRI) data were collected at baseline (PRE), midt

High-frequency visual stimulation can increase medial temporal lobe ripple oscillation density.

BACKGROUND: Flickering visual stimulation can evoke neural oscillations, which can influence ongoing brain activity. Electrophysiological recordings of neural oscillations in the ripple band (80-180 Hz) show that these high-frequency oscillations occur in the neocortex and the hippocampus, that they phase-synchronize across long distances, and that ripple oscillations in the neocortex often precede those in the hippocampus during wakefulness. It is therefore possible that the neocortical ripple oscillations propagate beyond sensory areas to the hippocampus, inducing ripple oscillations. METHO

Machine learning classification and regional differentiation of neuropathologically-confirmed Alzheimer's disease and comorbid Lewy body disease.

BACKGROUND: Alzheimer's disease (AD) and dementia with Lewy bodies (DLB) co-occur frequently, and growing evidence, including neuropathology, supports synergistic interplay between the diseases. We tested whether a single T1-weighted MRI scan may differentiate neuropathologically confirmed comorbid AD/DLB and AD controls using heterogeneously acquired neuroimaging. METHODS: We obtained structural neuroimaging, on two groups, AD with and without DLB pathology. Convolutional neural networks are trained across dimensions. We introduce a triple-ensemble strategy consisting of majority voting sche

Gut microbiota and pediatric eye diseases: current insights, mechanistic underpinnings, and future outlook.

BACKGROUND: As a core component of the human microecosystem, the gut microbiota modulates systemic immune-inflammatory responses, metabolic homeostasis, and neural signal transmission via the gut-eye axis. Dysbiosis of the gut microbiota has been closely linked to the onset and progression of multiple ophthalmic disorders in adults. Childhood represents a critical window for both gut microbial colonization and maturation, as well as a formative period for ocular development and visual function establishment. OBJECTIVE: This review systematically examines the evidence connecting gut microbiota

Cross-species neural co-culture uncovers metabolic signatures of cellular crosstalk.

Primates are distinguished by large brains relative to body size, with humans showing the greatest expansion. This increase in brain size evolved alongside advanced cognitive abilities as well as an elevated energetic demand. Importantly, allometric scaling alone does not explain this increased metabolic requirement, suggesting that other cellular mechanisms may be driving the unique energetic capacity of the human brain. Brain metabolism is critical for neurological function by providing the energy necessary for neuron firing. Much of metabolism in the brain is carried out by astrocytes: a ty

Multi-day recordings and adaptive stimulation protocols for in-home collection of deep brain stimulation intracranial recordings.

BACKGROUND: While open-loop deep brain stimulation (DBS) is an effective therapy for the motor symptoms of Parkinson's Disease (PD), recent work has explored whether closed-loop adaptive DBS (aDBS) may better address fluctuating symptoms through patient-specific and symptom-relevant neurophysiological biomarkers. To aid these investigations, we designed an interface for the research-enabled Summit Medtronic RC+S (RC+S) implanted neurostimulator (INS) to collect multi-day recordings along with the implementation of aDBS therapy. NEW METHOD: We developed applications in MATLAB for investigating

Understanding the development of neural abnormalities in adolescents with mental health problems: A longitudinal study.

BACKGROUND: Many mental health problems are neurodevelopmental in nature and have an onset during childhood. These disorders are associated with neural abnormalities, but it is unclear when these emerge and how this relates to the development of different mental health problems. METHODS: We identified children who developed mental health problems over two years and controls who remained healthy from the Adolescent Brain Cognitive Development (ABCD) study. Six mental health conditions (N = 58 to N = 173) were compared to controls (N = 2500) using separate linear models to assess group differen

Top-down modulation of visual action perception: distinct task effects in the action observation network.

Perceiving others' actions is essential for survival and social interaction. Cognitive neuroscience research has identified a network of brain regions crucial to visual action perception, known as the Action Observation Network (AON), comprising the posterior superior temporal cortex (pSTS), posterior parietal cortex, and premotor cortex. Recent research highlights the importance of integrating top-down processes, such as attention, to gain a deeper understanding of action perception. This study investigates how attention modulates the AON during human action perception. We conducted a two-ses
